The electrogenerated chemiluminescent (ECL) behavior of hemin at a platinum electrode in the alkaline solution has been investigated in detail. Under the optimum conditions the linear response range of hemin is 1.0×10−5–1.0×10−8 g ml−1, the detection limit was 1.0×10−8 g ml−1, and the relative standard derivation for 1×10−7 g ml−1 hemin was 2.8%. It has been also found that hemin would catalyze the ECL of lucigenin at a platinum electrode in a neutral solution in the presence of hydrogen peroxide, the catalytic ECL intensity was linear with the concentration of hemin in the range of 1.0×10−14–1.0×10−10 g ml−1. IgG labeled with hemin was used to examine the ECL catalytic activity of hemin after conjugating to protein, and the results showed that hemin retained ECL catalytic activity when conjugated to protein.  相似文献

Pullulanase from Bacillus acidopullulyticus was purified on a packed bed and a fluidized bed of calcium alginate beads. The binding of enzyme activity to the medium was found to follow Langmuir isotherm pattern. The maximum binding capacity was 1476 U ml−1 matrix and the dissociation constant was 142 U ml−1. The dynamic binding capacities at 5% breakthrough in the packed and fluidized beds were 472 U ml−1 and 644 U ml−1, respectively. In the packed bed as well as the fluidized bed, an activity recovery of more than 95% with fold purification in the range of 46–59 was observed. The elution with a competitive inhibitor, viz. maltose, and high-fold purification indicate an affinity-based process. The purification process worked equally well with columns of bed volumes of 3.8 and 10 ml.  相似文献

Composite diazotization-coupling reagents containing sulfanilamide (SAM), sulfapyridine (SP) or sulfathiazole (ST) as the diazotizable aromatic amines and sodium 1-naphthol-4-sulfonate (NS) as the coupling agent using column preconcentration on naphthalene-tetradecyldimethylbenzylammonium(TDBA)-iodide adsorbent have been used for the spectrometric determination of trace nitrate and nitrite in soil and water samples. Nitrite ion reacts with SAM in the pH range 2.0–5.0, SP in the pH range 2.0–2.5 and ST in the pH range 2.0–3.3 in HCl medium to form water-soluble colourless diazonium cations. These cations were coupled with NS in the pH range 9.0–12.0 for the SAM system, 9.6–12.0 for the SP system and 8.5–12.0 for the ST system to be retained on naphthalene-TDBA-I material packed in a column. The solid mass is dissolved from the column with 5 ml of dimethylformamide and the absorbance is measured spectrometerically at 543 nm for SAM-NS, 533 nm for SP-NS and 535 nm for ST-NS. Nitrate is reduced to nitrite by a copper-coated cadmium reductor column and the nitrite is then treated with the diazotization-coupling reagent by column preconcentration. The absorbance due to the sum of nitrate and nitrite is measured and nitrate is determined by difference. The calibration graph was linear over the range 2–40 ng NO2-N ml−1 and 1.5–30 ng NO3-N ml−1 in aqueous samples for the SAM and ST systems and 2–48 ng NO2-N ml−1 and 1.5–36 ng NO3-N ml−1 in aqueous samples for the SP system, respectively. The sensitivity, accuracy and precision of the systems decreased in the order STSAMSP. The detection limits were 1.4 ng NO2-N ml−1 and 1.1 ng NO3-N ml−1 for SAM, 1.6 ng NO2-N ml−1 and 1.2 ng NO3-N ml−1 for SP, and 1.0 ng NO2-N ml−1 and 0.75 ng NO3-N ml−1 for ST, respectively. The preconcentration factors are 8, 5 and 6 for SAM-NS, SP-NS and ST-NS, respectively. Interferences from various foreign ions have been studied and the methods have been applied to the determination of ng ml−1 levels of nitrite and nitrate in soil and water samples. The mean recovery was 95–102% for all three systems.  相似文献

A sensitive method for the post-column reaction detection of organophosphorus pesticides is described. The method relies on photolysis of the organocompounds by irradiation with a low-pressure mercury lamp (main spectral line, 254 nm) in the presence of peroxydisulfate. The resultant orthophosphate was reacted with molybdate to form molybdophosphoric acid, which subsequently reacted with thiamine to generate thiochrome. Finally, the fluorescence intensity of thiochrome was measured at 440 nm with excitation at 375 nm. Factors affecting the rate of these reactions were optimized so that its contribution to the total band-broadening was negligible.

This detection system was used for the determination of phosphate, acephate and methamidophos, which were separated on an ODS column by isocratic reversed phase chromatography with acetonitrile–water as the mobile phase. A linear relationship between analyte concentration and peak area was obtained within the range 0.016–7.0 μg ml−1 with correlation coefficients greater than 0.9995 and detection limits between 4 and 12 ng ml−1. Intra- and inter-day precision values of about 1.2% R.S.D. (n = 10) and 2.1% R.S.D. (n = 30), respectively, were obtained.

Pesticide residues below ng ml−1 levels could be determined in environmental waters when a preconcentration device was coupled on-line with the HPLC system. Detection limits as low as 0.01 ng ml−1 were achieved for only 250 ml of sample. In the analyses of vegetables and grains, the detection limit was about 1 μg kg−1.  相似文献

Abstract— Ultraviolet B irradiation has been believed to decrease or impair the activity of reactive oxygen species (ROS) scavenging enzymes such as superoxide dismutase (SOD) in the skin. It has been recently reported that two isozymes of SOD, namely copper-zinc SOD (Cu-Zn SOD) and manganese SOD (Mn SOD), exist in mammalian cells and that the two enzymes play different roles in living systems. The aim of this study was to investigate changes in SOD activities and protein levels in cultured human keratinocytes after acute UVB irradiation. In addition, the protein levels of Cu-Zn SOD and Mn SOD were quantified separately. A single exposure to UVB irradiation produced an increase in SOD activity and protein level that peaked immediately after UVB irradiation, after which a decline was observed, with subsequent recovery to baseline levels 24 h after irradiation. In individual assays of Mn SOD and Cu-Zn SOD, the amount of Mn SOD protein decreased and then gradually recovered 24 h after irradiation. In contrast, the amount of Cu-Zn SOD protein increased immediately after UVB irradiation, and then gradually declined. To evaluate the mechanisms of these changes, we examined the effects of the cytokines, interleukin-1α (IL-1α) and tumor necrosis factor-α (TNF-α), which can be secreted from keratinocytes after UVB irradiation, on the SOD activity and protein levels in keratinocytes. Interleukin-la and TNF-α enhanced both the SOD activity and protein level of Mn SOD, while these cytokines had no effect on Cu-Zn SOD protein levels in cultured human keratinocytes after incubation for 24 h. Furthermore, when neutralizing antibodies against IL-1α and TNF-α were added separately or together to the culture medium before UVB irradiation, the recovery of total SOD activity and Mn SOD protein level were markedly inhibited 24 h after irradiation. Our results suggest that significant increases in SOD activity and protein level occur as a cutaneous antioxidant defense mechanism that protects against the cytotoxicity as a result of UVB irradiation, and that this increase in SOD is attributed to Cu-Zn SOD. The Cu-Zn SOD and Mn SOD protein levels changed in a different manner after UVB irradiation. The former may participate in an early phase and the latter in a late phase defense mechanism directed against oxidant cytotoxicity through UVB irradiation. In addition, the recovery of Mn SOD to baseline levels 24 h after UVB irradiation seems to be mediated through cytokines such as IL-1α and TNF-α, which are secreted from keratinocytes.  相似文献

By designing a novel flow-through electrolytic cell (FEC), bromine was produced near to the surface of the platinum electrode by electrochemical oxidation of acidic KBr. The fast and weak chemiluminescence signal produced by the chemical reaction of the electrogenerated bromine with H2O2 was greatly enhanced by tetracyclines Based on these observations, a new, sensitive and simple electrogenerated chemiluminescence (ECL) method for the determination of tetracyclines was developed. Under the optimum experimental conditions, the calibration graphs are linear over the range 3.0×10−8 to 5.0×10−5 g ml−1 for tetracycline, 2.0×10−7 to 2.4×10−5 g ml−1 for oxytetracycline and 1.0×10−7 to 5.0×10−5 g ml−1 for chlortetracycline. The limits of detection (S/N=3) are 1.0×10−8 g ml−1 for tetracycline, 7.0×10−8 g ml−1 for oxytetracycline and 1.5×10−7 g ml−1 for chlortetracycline. For the determination 5.0×10−7 g ml−1 tetracycline, the relative standard deviation was <5%. The proposed method was used to determine tetracyclines in pharmaceutical formulations.  相似文献

The purpose of this study was to determine the effect of He---Ne laser irradiation (632.8 nm, 10 mW) on the induction of acrosome reaction and mortality in bull sperm cells in comparison with two important capacitation agents; calcium and heparin. Frozen-thawed bull sperm cells were washed in percoll gradient and suspended at a concentration of 1 × 106 ml−1 in sp-TALP medium, capacitated in the presence of 2 mM CaCl2, 10 μg ml−1 heparin, or irradiated at fluences from 2 to 16 J cm−2, and incubated for 0, 30, 60 and 90 minutes. At the end of the incubation period, the percentage of sperm that were acrosome-reacted and dead was determined. The results obtained indicated that laser irradiation at all fluences produced a significant increase (p < 0.001) in the percentage of sperm cells that were acrosome reacted, and a significant decrease (p < 0.001) in the percentage of dead sperm at 90 minutes of incubation in comparison to other capacitation agents and the control group. The percentage of sperm cells with acrosome reaction was increased with increasing fluences of laser irradiation and time of incubation. It is conclude that the application of He---Ne laser irradiation at fluences from 2 to 16 J cm−2 induced the acrosome reaction and decreased the sperm mortality percentage in vitro of bull sperm cells.  相似文献

Within the past several years significant advances have been made towards the development and incorporation of chemically modified electrodes as selective detectors for high performance liquid chromatography and flow injection analysis. In many cases the chemically modified electrode systems closely approach the “ideal” detector specifications of chemical and mechanical stability along with a significant linear response region. This paper will discuss the characterization and incorporation of ionomeric poly(ester-sulfonic acid) coated electrodes as nonaqueous electrochemical detectors. The orientation of the electrodes in the detector system as well as the increased sensitivity levels to 10−10 g ml−1 for cationic species and 10−9 g ml−1 for neutral species will be presented. Also the applicability of the ionomer coated electrodes as nonelectrolyte detectors achieved a reproducible response with detection limits to 10−6 g ml−1. Overall this system performed as well as, or better than, more specialized and expensive thin layer electrochemical detectors.  相似文献

The behavior of the ciprofloxacin (CPFX) complex with copper, Cu(II)L2, at a mercury electrode has been investigated in borax–boric acid buffer. The adsorption phenomena were observed by linear sweep voltammetry. The mechanism of the electrode reaction was found to be reduction of Cu(II)L2 adsorbed on the surface of the electrode by an irreversible charge transfer to metal amalgam, Cu(0)(Hg). In the presence of DNA, the formation of the electrochemically non-active complexes Cu(II)L2–DNA results in the decrease of the equilibrium concentration of Cu(II)L2 and its peak current. Under the optimum conditions, the decrease of the peak current is proportional to DNA concentration. The linear ranges are 6.67×10−8 to 1.20×10−5 g ml−1 for calf thymus DNA (ctDNA), 3.30×10−8 to 2.33×10−6 g ml−1 for fish sperm DNA (fsDNA) and 1.0×10−8 to 1.2×10−6 g ml−1 for yeast RNA. The detection limits are 5.00×10−9, 3.00×10−9 and 2.50×10−9 g ml−1, respectively. This method exhibits good recovery and high sensitivity.  相似文献

It was found that the weak chemiluminescence produced from the reaction of polyhydroxy phenols with luminol in alkaline solution could be strongly enhanced by ferricyanide and ferrocyanide. Based on this found, a new flow injection chemiluminescence method is proposed for the determination of four polyhydroxy phenols: pyrogallol, phlorglucinol, quinol and resorcinol. The detection limits of the method are 0.03 μg ml−1 pyrogallol, 0.03 μg ml−1 phlorglucinol, 0.04 μg ml−1 quinol, and 0.02 μg ml−1 resorcinol. The possible mechanism of CL reactions is also discussed briefly.  相似文献

Incorporation of 3-fluorotyrosine and site-specific mutagenesis have been used with stopped-flow spectrophotometry and pulse radiolysis to investigate the catalytic properties of human manganese superoxide dismutase (MnSOD). All of the nine tyrosine residues in each of the four subunits of the homotetramer of human MnSOD were replaced with 3-fluorotyrosine. Previous studies showed that the crystal structures of the unfluorinated and fluorinated human MnSOD are nearly superimposable with the root-mean-square deviation for 198 -carbon atoms at 0.3 Å. However, the catalytic activity kcat/Km of the fluorinated MnSOD at 30 μM−1 s−1 was less than unfluorinated wild type at 800 μM−1 s−1. Comparison of the values of kcat/Km for fluorinated and unfluorinated wild-type andY34F MnSOD showed that this decrease for the fluorinated enzyme was in significant part due to 3-fluorotyrosine residues distant (>7 Å) from the active-site metal, not to 3-fluorotyrosine at position 34 close (5 Å) to the metal. Although many rate constants for the catalysis are decreased by this fluorination, the rate of dissociation of the product-inhibited complex appears unchanged by the presence of fluorinated tyrosines. These results suggest that Tyr34 is not a proton donor in the release of the product-inhibited complex, which involves protonation of a peroxo complex of the metal with release of hydrogen peroxide.  相似文献

A highly sensitive and selective method is described for the determination of trace amounts of nitrite based on its effect on the oxidation of carminic acid with bromate. The reaction was monitored spectrophotometrically by measuring the decrease in absorbance of carminic acid at 490 nm after 3 min of mixing the reagents. The optimum reaction conditions were 1.8×10−1 mol l−1 H2SO4, 3.8×10−3 mol l−1 KBrO3, and 1.2×10−4 mol l−1 carminic acid at 30°C. By using the recommended procedure, the calibration graph was linear from 0.2 to 14 ng ml−1 of nitrite; the detection limit was 0.04 ng ml−1; the R.S.D. for six replicate determinations of 6 ng ml−1 was 1.7%. The method is mostly free from interference, especially from large amounts of nitrate and ammonium ions. The proposed method was applied to the determination of nitrite in rain and river water.  相似文献

A catalytic photometric method was developed for the determination of sub-nanogram levels of cobalt. The method is based on the catalytic effect of cobalt(II) on the oxidative coupling of 3-methyl-2-benzothiazolinone hydrazone with N-ethyl-N-(2-hydroxy-3-sulfopropyl)-3,5-dimethoxyaniline (DAOS) to form a colored dye (λmax=525 nm) in the presence of hydrogen peroxide. In this reaction system, 1,2-dihydroxybenzene-3,5-disulfonate (Tiron) acted as an effective activator for the catalysis of cobalt(II). Variation of reaction time between 5 and 10 min allows the determination range to be extended from 0.01 to 1.0 ng ml−1. The reaction system can also be successfully adapted to flow-injection analysis (FIA). The dynamic range of the proposed flow-injection method was 0.01–1.0 ng ml−1 and detection limit (signal/noise, S/N=3) was 5 pg ml−1 at a sampling rate of 30 h−1. Manual and flow-injection methods were applied to the direct determination of cobalt in pepperbush as a standard material.  相似文献

A spectrofluorimetric method to determine levofloxacin is proposed and applied to determine the substance in tablets and spiked human urine and serum. The fluorimetric method allow the determination of 20–3000 ng ml−1 of levofloxacin in aqueous solution containing acetic acid–sodium acetate buffer (pH 4) with λexc=292 and λem=494 nm, respectively. Micelle enhanced fluorescence improves the sensibility and allows levofloxacin direct measurement in spiked Human serum (5 μg ml−1) and urine (420 μg ml−1), in 8 mM sodium dodecyl sulphate solutions at pH 5.  相似文献

A catalytic flow-injection (FI) method was developed for the determination of 10−9 mol l−1 levels of vanadium(IV, V). The method is based on the catalytic effect of vanadium(V) on oxidation of N-(3-sulfopropyl)-3,3′,5,5′-tetramethylbenzidine (TMBZ·PS) using bromate as oxidant to form a yellow dye (λmax=460 nm). The use of 5-sulfosalicylic acid (SSA) as an activator enhanced the sensitivity of the method. The calibration graphs with a working range 0.05–8.0 ng ml−1 were obtained for vanadium(V). Vanadium(IV) was also determined, being oxidized by bromate. The detection limit (signal/noise, S/N=3) was 0.01 ng ml−1 (ca. 2×10−10 mol l−1) vanadium. The relative standard deviations (R.S.D.) for 15 determinations of 0.5 ng ml−1 vanadium, and for ten determinations of 0.1 and 1.0 ng ml−1 vanadium were 0.41, 2.6 and 0.25%, respectively, with a sampling rate of 15 samples h−1. The proposed method was successfully applied to the determination of vanadium in natural waters.  相似文献

A new bulk acoustic wave (BAW) cellulase sensing technique, which is based on the enzymatic hydrolysis process of sodium carboxymethylcellulose (CMC) by cellulase, was established. The frequency shift curves of BAW sensor indicated that the viscosity of the tested solutions decreased during the hydrolysis process. The hydrolysis rate of CMC by cellulase was calculated from the frequency shift curves. The hydrolysis rate of CMC under different pH conditions at 30°C showed that cellulase had high hydrolysis ability approximately at pH 5.0. Kinetic parameters (the Michaelis constant Km and the maximum rate Vmax) of the process were estimated by using a linear method of Lineweaver–Burk plot. Km is 1.95±0.25 mg ml−1 and Vmax is −(4.25±0.58)×10−3 g1/2 cm−3/2 cP1/2 min−1. Also the activation energy (Ea) of the enzymatic hydrolysis, with a value of 51.99±1.26 kJ mol−1, was estimated in this work.  相似文献

A simple and rapid spectrophotometric method for the determination of tinidazole is presented. This method is based on the measurement of the absorbance of the signal at 368 nm yielded by bathochromic shift during alkaline hydrolysis of tinidazole in 0.1 N NaOH. The method is linear within the range of 1–30 μg ml−1, and the detection and quantification limits are 0.07 and 0.25 μg ml−1, respectively. The precision of the method, expressed as the relative standard deviation, is 0.19% for a tinidazole concentration of 15 (μg ml−1. The method was applied to the analysis of tinidazole in pharmaceutical formulations and serum.  相似文献

The very sensitive, fairly selective direct spectrophotometric method for the determination of trace amount of vanadium (V) with 1,5-diphenylcarbohydrazide (1,5-diphenylcarbazide) has been developed. 1,5-diphenylcarbohydrazide (DPCH) reacts in slightly acidic (0.0001–0.001 M H2SO4 or pH 4.0–5.5) 50% acetonic media with vanadium (V) to give a red–violet chelate which has an absorption maximum at 531 nm. The average molar absorption coefficient and Sandell’s sensitivity were found to be 4.23×104 l mol−1 cm−1 and 10 ng cm−2 of Vv, respectively. Linear calibration graph were obtained for 0.1–30 μg ml−1 of Vv: the stoichiometric composition of the chelate is 1:3 (V: DPCH). The reaction is instantaneous and absorbance remain stable for 48 h. The interference from over 50 cations, anions and complexing agents has been studied at 1 μg ml−1 of Vv. The method was successfully used in the determination of vanadium in several standard reference materials (alloys and steels), environmental waters (potable and polluted), biological samples (human blood and urine), soil samples, solution containing both vanadium (V) and vanadium (IV) and complex synthetic mixtures. The method has high precision and accuracy (s=±0.01 for 0.5 μg ml−1).  相似文献

A novel chemiluminescence (CL) flow system for the determination of chlortetracycline is described. It is based on the direct CL reaction of chlortetracycline and [Cu(HIO6)2]5− in KOH medium. The unstable [Cu(HIO6)2]5− was on-line electrogenerated by constant-current electrolysis. The CL intensity was linear with chlortetracycline concentration in the range of 0.1–100 μg ml−1. The determination limit was 5.3×10−8 g ml−1. The whole process could be completed in 1 min. The proposed method is suitable for automatic and continuous analysis, and has been applied satisfactorily to analysis of chlortetracycline in biological fluid.  相似文献

Investigation of the use of a single analytical procedure using the non-suppressed ion chromatographic method with direct spectrophotometric detection capable of determining eight oxoanions simultaneously is presented in this paper. Potassium phosphate was found to be the most suitable eluent for UV absorbance detection at 205 nm. Oxoanions AsO3−3, SeO2−3, AsO3−4, VO3, SeO2−4, WO2−4, MoO2−4 and CrO2−4 were detected at ng ml−1 levels with well separated peaks at retention time < 25 min. The working range is in the range ng ml−1 to 50 μg ml−1. The method is sufficiently sensitive to determine As (V), V(V), Mo(VI) and Cr(VI) anions (and NO3) directly in a river water sample. The accuracy of these results was established by comparison with conventional atomic absorption methods.  相似文献
