用3%三氯醋酸从榆耳子实体中提取出水溶性粗多糖,经乙醇分级、SepharoseCL6B纯化得到单一组分GIA.SepharoseCL6B柱层析测得GIA的相对分子质量为1 26×105.GC分析表明GIA由Man和Glc两种单糖构成,其物质的量比为1 42∶1.多糖GIA经部分酸水解、高碘酸氧化、Smith降解和甲基化分析其结构主要为β(1→3)Man和β(1→3)Glc,且在6~0处构成分枝结构,平均每11个己糖残基有2个分枝,多糖GIA的支链部分由(1→4)Glc构成,末端残基为Man,Glc.  相似文献

白术(Atractylodes macrocephala Koidz)水溶性粗多糖,经乙醇分级及Sepha-dex G-75层析柱制备得到组分AMP.经Sephadex G-75、DEAE-阴离子交换纤维素层析及HPLC验证,AMP相对分子质量及极性为均一组分,HPLC测得相对分子质量为8 374.气相色谱(GC)分析表明其单糖组成为Glc,Gal,Rha和Man,物质的量比为7.36∶1.00∶3.05∶1.52.多糖AMP经部分酸水解、高碘酸氧化、Smith降解、甲基化分析,其结构主体部分由Glc,Rha和Man组成,平均每6个残基上有1个分支,分支点为Glc的4-O处和Man的6-O处,末端残基为Glc.  相似文献

以波谱分析为主要手段研究沙棘果多糖Hn的结构,GC分析表明Hn由Ara,Gal,Man和Glc组成,单糖物质的量比与利用GC分析Hn甲基化产物的结果一致.^13C—NMR分析表明Hn中存在α,β型糖苷键.利用GC—MS分析进一步确定Hn主链结构由β(1→4)Glc,β(1→6)Man和β(1→4)Gal构成,在3-O处有分支,支链由α(1→4)Glc,(1→6)Gal或末端Ara,Glc构成.  相似文献

玉米芯用PH=3的HCl煮提得到酸提水溶性粗多糖.该粗多糖组成为Glc,Xyl,Gal,经乙醇分级和Sepharose CL-6B柱层析纯化,得到多糖CCCP.经Sephadex A-25柱层析、比旋光度测定、醋酸纤维薄膜电泳等方法鉴定CCCP为均一多糖.经唾液淀粉酶解、纤维素酶解、部分酸水解、高碘酸氧化、Smith降解、甲基化分析及IR,GC和GC/MS等方法分析表明:CCCP为少分枝结构;主链由吡喃型(1→3)Xyl构成,在O(4)处有分枝;支链主要由(1→4)Glc构成,在O(6)处有分枝,支链还存在1→3,1→6键型连接的Glc,Gal;末端基为Xyl,Glc,Gal.  相似文献

独脚金粗多糖(Strigaasiatica(L.)O.Ktze. polysaccharide,SKPc)经DEAE-Sephadex A-25以及Sephadex G-100柱层析分离纯化后得到独脚金多糖组分(SKP~*_1)。采用高效液相色谱(HPLC)法检测SKP~*_1,表明多糖SKP~*_1极性、相对分子质量是均一的,且其相对分子质量为14852u。采用气相色谱(GC)分析法研究SKP~*_1的单糖组成为葡萄糖(Glu)、阿拉伯糖(Ara)、半乳糖(Gal)、木糖(Xyl)、甘露糖(Man)、鼠李糖(Rha),相对摩尔比是2.5∶2.0∶0.85∶0.4∶0.5∶1.0;可能主要以β型糖苷键连接。采用化学分析和仪器分析结合进行SKP~*_1结构分析,表明SKP~*_1结构是Ara以β-(1→3)糖苷键连接构成主链之一,2,4或2,5位有支链;Gal以β-(1→6)糖苷键连接构成SKP~*_1的主链之一,且在3位上有分枝。支链由β-(1→4或1→5)Xyl、β-(1→4,1→6,1→2)Glc构成。Rha、Ara、Man、Gal、Glc都有一部分构成糖分子的末端残基。体外清除羟基实验结果表明,SKP~*_1有较好地清除羟基自由基的活性。  相似文献

沙棘果皮水溶性多糖经酸性乙醇分级和DEAE -SephadexA - 2 5柱层析纯化 ,得到多糖Hn.经SepharoseCL - 4B柱层析、玻璃纤维纸电泳、比旋光度测定等方法鉴定Hn 为均一多糖 .经唾液淀粉酶解、部分酸水解、高碘酸氧化、Smith降解、甲基化分析及IR ,1 3C -NMR ,GC和GC -MS等方法 ,确定其基本结构中主链由 1→ 4Glc基、1→ 6Man基和 1→ 6Gal基构成 .三者摩尔比为nGlc∶nMan∶nGal=3∶1 .5∶1 ,分支点在 3—O处 ,分支率为 43% ,侧链由 1→ 6Gal基、1→ 4Glc基、末端Glc或末端Ara基构成 .  相似文献

从玉米芯中提取到水提水溶性粗多糖,通过乙醇分级、冻融、凝胶过滤层析,生物测定导向等手段,分离到一种能延长体外凝血时间,而且具有外源抗凝血功能的多糖CCⅢ.CCⅢ对活化部分凝血酶原时间(APTT)无显著影响,但可显著延长体外抗凝血时间(PT).将CCⅢ纯化,经糖组成分析、甲基化、高碘酸氧化、Smith降解和GC-MS分析,确定该多糖结构为:β-(1→4)Glc,(1→3)Xyl构成主链,Glc在6-O处有分枝.平均每10个糖残基有1个分枝,支链由β-(1→3)Xyl,(1→3)Glc构成.  相似文献

从山茱萸中提取出水溶性粗多糖, 经柱色谱分离纯化得到一种酸性多糖组分FCP5-A. 采用高效凝胶渗透色谱法(HPGPC)测定其为均一性多糖, 平均分子量为8.7×104. 经IR、GC、部分酸水解、13C NMR及甲基化分析等方法对该多糖的化学结构进行了表征. 结果表明, 该多糖由鼠李糖、阿拉伯糖、半乳糖及半乳糖醛酸组成, 其摩尔比为1∶5.7∶0.6∶1.2. FCP5-A为多分支结构, 由-2)Rha(1-及-4)GalA(1-构成主链, 在鼠李糖的4位存在分支; 支链主要由高度分支的阿拉伯糖构成, 此外还存在-3)Gal(1-; 末端残基为Ara(1-及Gal(1-. 结果提示, FCP5-A为一种新的山茱萸酸性分支多糖.  相似文献

A water-soluble polysaccharide, HM_(41), was obtained from Halenia elliptica D. Don by acidic ethanol fractionation and gel filtration. Its homogeneity was confirmed by chromatography using multiple systems. HM_(41) was composed of rhamnose(Rha), arabinose(Ara), xylose(Xyl), mannose(Man),galactose(Gal), glucose(Glc) with a molar ratio of 1.0:5.5:1.8:3.0:9.4:21. The average molecular weight of HM_(41) was approximately 1.17 * 10~4. Periodate oxidation, Smith degradation, methylation and GC, IR,NMR, XRD, GC–MS analysis were used for the structural analysis of HM_(41). Its main chain was composed mainly of β-(1→4)Gal, β-(1→4)Glc and β-(1→6)Glc. β-(1 →4)Gal were substituted at 6-O and on average there were 14 branches among 23 main chain residues;(1→ 4)Glc had no branch;(1→6)Glc were substituted at 3-O and on average there were 9 branches among 14 main chain residues. The side chain was composed of(1→3,6)-Rha,(1→4)/(1→5)-Ara,(1 →4)/(1→5)-Xyl,(1→4,6)-Man and(1→2)-Glc. The terminal residue was composed of Ara, Xyl, Man, Gal, and Glc. Then, we demonstrated that HM and HM_(41) had strong scavenging activities in vitro hydroxyl. Overall, HM and HM_(41) may have potential applications in the antioxidants for medical and food industry.  相似文献

An aqueous acidic polysaccharide, named rhamnogalacturonan (designated as TIP-D2) was isolated from Tribulus terrestris L by means of DEAE-cellulose chromatography and gel filtration. The molecular mass of TTP-D2 was estimated to be 26 KDa by gel filtration.TTP-D2 is composed of galacturonic acid, rhamnose, arabinose, galactose,fucose,mannose,xylose and glucose in a ratio of 71.4:13.5:5.6:4.9:3.1:1.9:1.9:1.0. The main chain structure of TTP-D2 was elucidated as an acidic hetero-polysaccaride with the connection of α-(1-4) galacturonic acid with α-(1-3) rhamnose by GC analysis of partially hydrolyzed products and determination of ^1H,^13C-NMR spectra.  相似文献

In this paper, capillary zone electrophoresis with amperometric detection (CZE-AD) was applied to determine the compositions of hetero-polysaccharides from Chinese herbs, Angelica sinensis and flax by analyzing their hydrolyzed monosaccharides: fucose, galactose, glucose, arabinose, rhamnose and xylose. Under the selected optimum conditions, the six monosaccharides could be perfectly separated within 25 min and showed significant current responses at copper electrodes. The linear ranges of the six monosaccharides were all from 5.0 x 10(-6) to 2.0 x 10(-4) mol L(-1) and their detection limits were lower or near 1.0 x 10(-6) mol L(-1) (S/N = 3). Experiments showed that the Angelica sinensis polysaccharide was composed of fucose, galactose, glucose, arabinose, rhamnose and xylose (mole ratio 1.0:13.6:15.0:8.7:21.3:3.7), and the flax polysaccharide was composed of galactose, glucose and arabinose (mole ratio 1.0:4.98:1.1). The purity of these polysaccharides leached by the introduced leaching method was 98.3 and 97.6%, respectively. Analyzing polysaccharides by this method has some merits of speed, simple instrumentation and operation, high sensitivity and high reproducibility.  相似文献

研究了白芷多糖的结构和单糖组成。采用紫外光谱和红外光谱法对白芷多糖进行定性分析。结果表明,白芷多糖具有多糖特征性的紫外和红外吸收峰,其分子中存在酰胺结构;凝胶渗透色谱法测定结果表明,白芷多糖的峰值分子量为88538;采用气相色谱法测定白芷多糖中单糖的种类和构成比例,结果表明,白芷多糖由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖和半乳糖等7种单糖组成,并计算出其中6种单糖的摩尔组成比例。  相似文献

A crude polysaccharide was extracted from Physalis alkekengi L. fruit. HPLC was used for the component analysis of the polysaccharide. The results indicate that Physalis alkekengi L. polysaccharide(PAP) was composed of rhamnose, xylose, arabinose, galactose, and glucose. Free radicals scavenging activity of PAP was studied through 3 free radicals scavenging tests. PAP exhibited high scavenging effects on OH and DPPH radicals, and both the scavenging rates were about 80%. The scavenging rate of O2^- radical was about 22%.  相似文献
