Naturally contaminated corn samples of different origin were extracted using two conventional techniques (blending and shaking) and three alternative approaches (ultrasonic extraction, accelerated solvent extraction, and microwave-assisted extraction). Use of the same extraction mixture for all trials enabled the efficiency of the various extraction techniques to be compared. Extracts were filtered and directly analyzed by LC–ESI–MS, without further clean-up. The yield from the alternative extraction techniques showed efficiency to be higher than for conventional techniques. In particular, microwave-assisted extraction was slightly superior to other techniques.  相似文献

A simple, fast and sensitive RP-HPTLC method is developed for simultaneous quantitative determination of vanillin and related phenolic compounds in ethanolic extracts of Vanilla planifolia pods. In addition to this, the applicability of accelerated solvent extraction (ASE) as an alternative to microwave-assisted extraction (MAE), ultrasound-assisted extraction (UAE) and Soxhlet extraction was also explored for the rapid extraction of phenolic compounds in vanilla pods. Good separation was achieved on aluminium plates precoated with silica gel RP-18 F(254S) in the mobile phase of methanol/water/isopropanol/acetic acid (30:65:2:3, by volume). The method showed good linearity, high precision and good recovery of compounds of interest. ASE showed good extraction efficiency in less time as compared to other techniques for all the phenolic compounds. The present method would be useful for analytical research and for routine analysis of vanilla extracts for their quality control.  相似文献

Three commonly applied extraction techniques for persistent organic chemicals, Soxhlet extraction (SE), accelerated solvent extraction (ASE) and microwave-assisted extraction (MAE), were applied on soil and fish samples in order to evaluate their performances. For both PCBs and PBDEs, the two more recent developed techniques (ASE and MAE) were in general capable of producing comparable extraction results as the classical SE, and even higher extraction recoveries were obtained for some PCB congeners with large octanol-water partitioning coefficients (Kow). This relatively uniform extraction results from ASE and MAE indicated that elevated temperature and pressure are favorable to the efficient extraction of PCBs from the solid matrices. For PBDEs, difference between the results from MAE and ASE (or SE) suggests that the MAE extraction condition needs to be carefully optimized according to the characteristics of the matrix and analyte to avoid degradation of higher brominated BDE congeners and improve the extraction yields.  相似文献

The concentrations of dioxins in fish oil and fish meal were determined with accelerated solvent extraction, using a novel integrated carbon fractionation extraction cell followed by a miniturized multilayer silica column and bioanalysis on a recently-developed chemically-activated fluorescent gene expression cell bioassay. The developed method allows for simultaneous gravimetric lipid weight determination, which was shown for both matrices under study (about 100% lipid recovery of each sample). Initial results practically meet the quality criteria on screening methods for control of dioxins in food and feedstuffs laid down in the EU Commission Directives 2002/69/EC (food) and 2002/70/EC (feed). This demonstrates that the developed method can be used as a screening tool for monitoring dioxins in food and feed after some additional improvements and testing on a greater number of matrices.  相似文献

以快速溶剂萃取–凝胶渗透色谱净化–气相色谱质谱法测定土壤中邻甲苯胺的含量。土壤样品经ASE–350快速溶剂萃取仪萃取,萃取液用凝胶渗透色谱净化浓缩后,用全扫描模式(SCAN)采集气相色谱–质谱法测定土壤中的邻甲苯胺含量。该方法检出限为0.01 mg/kg,加标回收率为78.3%~95.5%,测定结果的相对标准偏差为2.35%~14.9%(n=6)。该方法具有分离效果好、灵敏度高、重现性好、前处理操作简便及纯化效果好等优点,可用于测定土壤中邻甲苯胺的含量。  相似文献

Concentrations of pharmaceuticals and personal care products (PPCPs) in natural solids remain largely unknown. Contributing to this, is a lack of methods permitting the simultaneous detection of the diverse, low-level contaminants present in these complex matrices. We have developed a microwave-assisted solvent extraction (MASE)-based method targeting seven diverse PPCPs (caffeine, 17β-estradiol, ibuprofen, ketoprofen, musk ketone, naproxen, and triclosan) and a molecular marker for fecal waste (epicoprostanol). The method consisted of optimizing the following variables: derivatization of the polar target analytes, silica gel open column clean-up, and gas chromatographic-mass spectrometric (GC-MS) analysis of sample extracts for analysis and detection of the compounds noted above. Testing of the method on spiked soil allowed for 89.6 ± 2.89% recovery of three target compounds and 25.0 ± 1.93% recovery of five of the compounds. Although the latter recoveries were low, the precision across all recoveries was high, suggesting good reproducibility in application of the method. Furthermore, we suspect that matrix effects are likely responsible for the lower recoveries. Techniques with the exclusive incorporation of organic solvents were found inapplicable in the study of a pharmaceutical salt, diphenhydramine HCl. Application of the developed method to sediment collected directly downstream of the effluent pipe of a wastewater treatment plant allowed detection of ibuprofen, naproxen, ketoprofen, and epicoprostanol at ng-μg per gram dry weight concentrations. The observation of acidic pharmaceuticals, previously believed to exhibit insignificant sorption to solid matrices, in the tested sediment samples, coupled with application of biosolids for agricultural purposes, demonstrates the need for expanded investigation of PPCP contamination of natural solid matrices.  相似文献

建立了加速溶剂萃取-凝胶色谱净化-气质联用同时测定土壤中15种有机氯农药的分析方法。优化了凝胶色谱(GPC)净化的条件,比较了凝胶色谱净化及浓硫酸净化法对15种有机氯回收率的影响。结果表明,采用GPC净化能够有效避免浓硫酸净化对氯丹、异狄氏剂、硫丹、甲氧滴滴涕等农药回收率的影响,GPC净化的最佳收集时间为10~15 min,15种有机氯农药的回收率为56%~122%。15种有机氯农药在0.03~6.0 mg/L范围内具有较好的线性,相关系数达0.999以上,方法的检出限为0.1~5.0μg/kg,定量下限为0.4~16.0μg/kg。采用该方法对实际样品进行加标回收率实验,土壤样品的加标回收率为68%~122%,相对标准偏差为1.2%~5.9%。该方法简单、快捷、灵敏度高,已用于实际土壤样品的检测。  相似文献

建立了超声提取-固相萃取纯化/正相高效液相色谱测定大豆及大豆油中磷脂酰胆碱(PC)、磷脂酰乙醇胺(PE)、磷脂酰肌醇(PI)的方法。考察了提取溶剂、超声功率、超声时间、提取温度及净化方式的影响,并研究了不同色谱固定相对磷酸甘油酯分离效果的影响。优化的实验条件为:以氯仿-甲醇(2∶1,体积比)为提取溶剂,1 500 W功率超声提取30 min;氨基固相萃取柱为纯化小柱;正己烷-异丙醇-1%HAc(8∶8∶1,体积比)为流动相。在该条件下,PC、PE、PI的线性范围分别为0.08~8.00、0.15~15.00、0.30~20.00 g.L-1,定量下限分别为0.021、0.050、0.060 g.L-1,检出限在8~23 mg.L-1之间,其在大豆和大豆油中的回收率为85%~108%。日内与日间精密度分别不大于4.7%和8.6%。  相似文献

A fast, simple, non-destructive method for the direct screening of polycyclic aromatic hydrocarbons (PAHs) in vegetable oil samples is proposed. The method uses a supercritical fluid extraction (SFE) system coupled on-line with a fluorimetric detector to determine PAHs. This special assembly avoids the main problems encountered in the determination of PAHs in complex matrices such as vegetable oils. PAHs are selectively extracted by using silica gel in the thimble and cleaned up by passage through a C18 column. Interferences are preferentially retained by the silica gel during the SFE process while PAHs are adsorbed in the C18 column and the remainder of the matrix is sent to waste. Finally, the C18 column is purged to remove residual CO2 gas and adsorbed PAHs are recovered by desorption with a solvent. The extracts from positive samples are subsequently analyzed by liquid chromatography (LC) with fluorescence detection. The proposed method allows the confirmation of vegetable oil safety and hence provides a new tool for consumer protection.  相似文献

Extraction is the most important step in the purification of bioactive compounds from natural products. This study introduces a simple online extraction strategy coupled with high‐speed counter‐current chromatography for efficient extraction and purification of bioactive components from solid natural products. For online extraction strategy, 1.0 g of ground Mangnolia officinalis or Piper nigrum was loaded into a guard column, which was then positioned on the manual injection valve instead of the sample loop. Bioactive components were directly extracted by the mobile phase of high‐speed counter‐current chromatography, and then transferred into high‐speed counter‐current chromatography for purification. In addition, the compatibility of the developed methodology for direct purification of bioactive components from fresh M. officinalis was successfully demonstrated. Obviously, in comparison with traditional offline heat‐reflux extraction, online extraction avoided the instrument, time, solvent, and energy consumption, and purified two phenolic compounds (honokiol and magnolol) from M. officinalis and three alkaloids (piperyline, piperine, and piperanine) from P. nigrum with high extraction efficiency. The superiority of the developed methodology is to establish an easy, rapid, and efficient technique for the purification of a wide variety of bioactive components from solid natural products.  相似文献

The comparison of four extraction techniques for isolation of five native and one labelled steroid oestrogens from sediment was described. The three conventional extraction techniques Soxhlet warm extraction (SWE), accelerated solvent extraction (ASE), microwave-assisted extraction (MAE) and a promising technique QuEChERS were tested for isolation of low environmentally relevant oestrogen concentrations using different extraction conditions. The least expensive and time-consuming method QuEChERS provided the best extraction recoveries (53–84%) from all techniques. MAE achieved the highest recovery from conventional techniques for less polar oestrogens using dichloromethane: acetone 3:1 mixture as an extraction solvent (50–71%), but for extraction of the whole group of oestrogens including more polar estriol acetone or methanol must be used. ASE provided higher extraction recoveries using dichloromethane at 60°C (53–74%) for less polar oestrogens. However, the repeatability of results was unsatisfactory and recoveries using other extraction conditions were lower than for MAE. The most time-consuming SWE achieved the worst extraction recoveries and for isolation of low oestrogen concentrations from sediments, it is completely unsuitable.  相似文献

以烟草中有机氯类农药为研究对象,从检出限、回收率、重复性和色谱图4个方面系统比较了机械振荡萃取法、超声波溶剂萃取法、微波辅助萃取法和加速溶剂萃取法4种常用前处理方法。由于加速溶剂萃取法是在高温高压下进行萃取,在提取出残留农药的同时,也提取出更多的干扰物。机械振荡法、超声波溶剂萃取法和微波辅助萃取法各项结果差别较小,其中机械振荡萃取法回收率和重复性最好,但提取时间较长。考虑溶剂消耗和提取时间等方面的因素,微波辅助萃取是较理想的样品前处理方法。  相似文献

An on-line Soxtec extraction and cleanup method is optimized for PCBs determination in mussel samples. Soxtec extraction conditions have been optimized using two factorial designs. Optimized extraction parameters were sample weight, anhydrous sodium sulfate weight, solvent volume, alumina weight, and boiling and rinsing times. The results suggest that all PCBs congeners extracted by Soxtec appeared statistically affected by solvent volume. Soxtec quantitative recoveries (8– ?115%) were achieved for all PCBs and method precision (RSD <12%) was satisfactory. The optimized method is compared with three different applied extraction methods, Soxhlet, accelerated solvent extraction, and microwave-assisted extraction. This latter presents the cleanup step on column or assisted by polypropylene membranes. The extraction effectiveness of the four different applied extraction methods for the determination of PCBs in mussel was similar.  相似文献

In this study, low temperature vacuum microwave-assisted extraction, which simultaneous performed microwave-assisted extraction (MAE) in low temperature and in vacuo environment, was proposed. The influencing parameters including solid/liquid ratio, extraction temperature, extraction time, degree of vacuum and microwave power were discussed. The predominance of low temperature vacuum microwave-assisted extraction was investigated by comparing the extraction yields of vitamin C, β-carotene, aloin A and astaxanthin in different foods with that in MAE and solvent extraction, and 5.2–243% increments were obtained. On the other hand, the chemical kinetics of vitamin C and aloin A, which composed two different steps including the extraction step of analyte transferred from matrix into solvent and the decomposition step of analyte degraded in the extraction solvent, were proposed. All of the decomposition rates (K2) for the selected analyte in low temperature, in vacuo and in nitrogen atmosphere decreased significantly comparing with that in conventional MAE, which are in agreement with that obtained from experiments. Consequently, the present method was successfully applied to extract labile compound from different food samples. These results showed that low temperature and/or in vacuo environment in microwave-assisted extraction system was especially important to prevent the degradation of labile components and have good potential on the extraction of labile compound in foods, pharmaceutical and natural products.  相似文献

A novel and simple method was established for the extraction and determination of seven compounds in Anemarrhena asphodeloides Bge. using silica gel‐based vortex‐homogenized matrix solid‐phase dispersion and ultra‐high performance liquid chromatography quadrupole‐time of‐flight mass spectrometer. The conditions for the extraction were optimized. Silica gel was used as the dispersant, 50% methanol–water was selected as an elution solvent and the grinding time was 3 min. Compared with the traditional ultrasonic‐assisted extraction, the developed method was rapid and efficient. In order to screen potential antioxidants, extract dealing with the optimized method was applied to a polyamide chromatography column and a D‐101 macroporous resin column. Fr.2.2 showed the highest antioxidant activities with the most content of flavonoid. A total of 25 peaks were identified from the active fraction. A 2,2′‐diphenyl‐1‐picrylhydrazyl ultra‐high performance liquid chromatography coupled with mass spectrometry approach was adopted for the rapid and exact screening and identification of antioxidant compounds. It indicated that flavonoids exhibited potential antioxidant activities. The antioxidant activities of nine monomeric compounds in vivo were tested. Structure–activity relationships were discussed. Five flavonoids with the concentration of 500 µg/mL would reduce the oxidative stress of PC12 cells that were induced with 2,2′‐azobis[2‐methylpropionamidine] dihydrochloride.  相似文献

A method of microwave-assisted extraction coupled with high-speed counter-current chromatography was established for separation and purification of isopimpinellin, pimpinellin and phellopterin from Toddalia asiatica (L.) Lam. The conditions of MAE including the extraction solvent, size of sample, solid/liquid ratio, extraction temperature and extraction time were optimized by a mono-factor test. That is, 2.0 g dried powder of T. asiatica (L.) Lam of 0.30-0.15 mm size was extracted with 20 mL (solid/liquid ratio of 1:10, g/mL) methanol under 50 °C for 1 min. The crude extract was separated and purified by high-speed counter-current chromatography with hexane-ethyl acetate-methanol-water (5:5:5.5:4.5, v/v/v/v) solvent system. 0.85 mg/g of isopimpinellin, 2.55 mg/g of pimpinellin and 0.95 mg/g of phellopterin were obtained from original sample in one-step within 240 min, the purity determined by high performance liquid chromatography was 95.0%, 99.1% and 96.4%, respectively. Their chemical structures were further identified by mass spectroscopy and nuclear magnetic resonance spectroscopy. The results demonstrated that microwave-assisted extraction coupled with high-speed counter-current chromatography was a feasible, economical and efficient technique for rapid extraction, separation and purification of effective compounds from natural products.  相似文献

The extraction and determination of chlorinated biphenyls (CBs) in soils and solid wastes is an ongoing subject of study. This is an overview article that compares the microwave-assisted extraction (MAE) and accelerated solvent extraction (ASE) techniques. The extraction of CBs by ASE has been optimised taking into account the variation of pressure, temperature and extraction time by means of experimental design and the Simplex approach. The recoveries obtained under the optimum conditions are compared and discussed with those obtained from MAE and Soxhlet extractions.  相似文献

An efficient microwave-assisted extraction (MAE) method has been developed and evaluated for the quantification of eight major polybrominated diphenylethers (PBDEs) in sewage sludge. The PBDEs were extracted from wet and dry sludge in a microwave extraction unit using a hexane/acetone mixture for 35 min at a controlled temperature of 130 °C. The extract was concentrated, cleaned up on a silica gel column, and analyzed by gas chromatography/mass spectrometry (GC/MS) in the negative chemical ionization (NCI) mode. The MAE procedure exhibited higher extraction efficiency, specifically for BDE (brominated diphenylether) 209, than the conventional Soxhlet extraction. The test congeners were clearly separated under specific instrumental operating conditions, at a source temperature of 230 °C and a column length of 20 m. The present analytical method showed recovery efficiencies ranging from 80 to 110% when applied to the PBDE-free sludge spiked with eight PBDE congeners. The efficiency of the MAE method was confirmed using sludge obtained from four sewage treatment plants (STPs). The results indicate that BDE 47, 99, and 209 are the most abundant congeners present in these sewage sludges, which is consistent with previous reports.  相似文献
