A HPLC method with automated column switching and UV-diode array detection is described for the simultaneous determination of Vitamin D3 and 25-hydroxyvitamin D3 (25-OH-D3) in a sample of human plasma. The system uses a BioTrap precolumn for the on-line sample cleanup. A sample of 1 ml of human plasma was treated with 2 ml of a mixture of ethanol–acetonitrile (2:1 (v/v)). Following centrifugation, the supernatant was evaporated to dryness under a stream of dry and pure nitrogen. The residue was reconstituted in 250 μL of a solution of methanol 5 mmol l−1 phosphate buffer, pH 6.5 (4:1 (v/v)), and a 200 μl aliquot of this solution was injected onto the BioTrap precolumn. After washing during 5 min with a mobile phase constituted by a solution of 6% acetonitrile in 5 mmol l−1 phosphate buffer, pH 6.5 (extraction mobile phase), the retained analytes were then transferred to the analytical column in the backflush mode. The analytical separation was then performed by reverse-phase chromatography in the gradient elution mode with the solvents A and B (Solvent A: acetonitrile–phosphate buffer 5 mmol l−1, pH 6.5; 20:80 (v/v); solvent B: methanol–acetonitrile–tetrahydrofuran, 65:20:15 (v/v)). The compounds of interest were detected at 265 nm. The method was linear in the range 3.0–32.0 ng ml−1 with a limit of quantification of 3.0 ng ml−1. Quantitative recoveries from spiked plasma samples were between 91.0 and 98.0%. In all cases, the coefficient of variation (CV) of the intra-day and inter-day-assay precision was ≤2.80%. The proposed method permitted the simultaneous determination of Vitamin D3 and 25-OH-D3 in 16 min, with an adequate precision and sensitivity. However, the overlap of the sample cleanup step with the analysis increases the sampling frequency to five samples h−1. The method was successfully applied for the determination of Vitamin D3 and 25-OH-D3 in plasma from 46 female volunteers, ranging from 50 to 94 years old. Vitamin D3 and 25-OH-D3 concentrations in plasma were found from 4.30–40.70 ng ml−1 (19.74 ± 9.48 ng ml−1) and 3.1–36.52 ng ml−1 (7.13 ± 7.80 ng ml−1), respectively. To investigate the disposition of unbound cocaine in the rat blood, brain and bile, we demonstrate an in vivo multiple sampling microdialysis system coupled with liquid chromatography for cocaine assay and verified by tandem mass spectrometry. Three microdialysis probes were concurrently inserted into the jugular vein, bile duct and brain striatum of each anesthetized rat. After a period of 2 h post-surgical stabilization, cocaine (10 mg kg(-1)) was administered through the femoral vein. Separation of unbound cocaine from various biological fluids was applied to a reversed-phase C(18) column (250 x 4.6 mm I.D., 5 microm). The mobile phase consisted of acetonitrile--10 mm potassium dihydrogen phosphate buffer (25:75, v/v, pH 4.0) and 0.8% diethylamine at a flow rate of 1 mL min(-1). The UV detector wavelength was set at 235 nm. The results indicate that cocaine penetrates the blood--brain barrier with a rapid distribution. However, unbound cocaine in the bile dialysate was not detectable in the UV detection. We therefore use LC--tandem mass spectrometry to detect the bile fluid after cocaine administration (3 mg kg(-1), i.v.). The results indicate that cocaine goes through hepatobiliary excretion.  相似文献

A simple procedure was developed for the direct determination of As(III) and As(V) in water samples by flow injection hydride generation atomic absorption spectrometry (FI–HG–AAS), without pre-reduction of As(V). The flow injection system was operated in the merging zones configuration, where sample and NaBH4 are simultaneously injected into two carrier streams, HCl and H2O, respectively. Sample and reagent injected volumes were of 250 μl and flow rate of 3.6 ml min−1 for hydrochloric acid and de-ionised water. The NaBH4 concentration was maintained at 0.1% (w/v), it would be possible to perform arsine selective generation from As(III) and on-line arsine generation with 3.0% (w/v) NaBH4 to obtain total arsenic concentration. As(V) was calculated as the difference between total As and As(III). Both procedures were tolerant to potential interference. So, interference such as Fe(III), Cu(II), Ni(II), Sb(III), Sn(II) and Se(IV) could, at an As(III) level of 0.1 mg l−1, be tolerated at a weight excess of 5000, 5000, 500, 100, 10 and 5 times, respectively. With the proposed procedure, detection limits of 0.3 ng ml−1 for As(III) and 0.5 ng ml−1 for As(V) were achieved. The relative standard deviations were of 2.3% for 0.1 mg l−1 As(III) and 2.0% for 0.1 mg l−1 As(V). A sampling rate of about 120 determinations per hour was achieved, requiring 30 ml of NaBH4 and waste generation in order of 450 ml. The method was shown to be satisfactory for determination of traces arsenic in water samples. The assay of a certified drinking water sample was 81.7±1.7 μg l−1 (certified value 80.0±0.5 μg l−1).  相似文献

A new rapid flow injection procedure for the simultaneous determination of nitrate, nitrite and ammonium in single flow injection analysis system is proposed. The procedure combines on-line reduction of nitrate to nitrite and oxidation of ammonium to nitrite with spectrophotometric detection of nitrite by using the Griess-llosvay reaction. The formed azo dye was measured at 543 nm. The influence of reagent concentration and manifold parameters were studied. Nitrite, nitrate and ammonium can be determined within the range of 0.02–1.60 μg mL−1, 0.02–1.60 μg mL−1 and 0.05–1.40 μg mL−1, respectively. R.S.D. values (n = 10) were 2.66; 1.41 and 3.58 for nitrate, nitrite and ammonium, respectively. This procedure allows the determination and speciation of inorganic nitrogen species in soils with a single injection in a simple way, and high sampling rate (18 h−1). Detection limits of 0.013, 0.046 and 0.047 μg mL−1were achieved for nitrate, nitrite and ammonium, respectively. In comparison with others methods, the proposed one is more simple, it uses as single chromogenic reagent less injection volume (250 mL in stead of 350 mL) and it has a higher sampling rate.  相似文献

Aceto Balsamico Tradizionale of Modena (ABTM) is a typical product (PDO denomination) of the province of Modena produced by cooked grape must which undergoes a long ageing period (at least 12 years) in series of wooden casks (batterie). The study of the transformations of this product during ageing is extremely relevant in order to control the authenticity of ABTM towards succedaneous products and mislabelling of age.

This paper presents the results of the investigation of sugars and fixed organic acids in ABTM samples of different ages, coming from different batterie. The analytes were simultaneously determined by a gas chromatographic method optimised for this peculiar matrix.

The method shows good separation and resolution of the investigated chemical species and allows their determination in the concentration ranges reported in brackets: malic (7.6–15.5 g kg−1), tartaric (4.0–9.7 g kg−1), citric (0.6–1.5 g kg−1) and succinic (0.36–0.62 g kg−1) acid and glucose (153–294 g kg−1), fructose (131–279 g kg−1), xylose (011–0.39 g kg−1), ribose (0.078–0.429 g kg−1), rhamnose (0.061–0.195 g kg−1), galactose (0.136–0.388 g kg−1), mannose (0.41–1.46 g kg−1), arabinose (0.33–1.00 g kg−1) and sucrose (0.46–6.84 g kg−1), with mean associated errors ranging from 5 to 19% depending on the analytes.

Moreover, the recovery values are always satisfactory, being close to one for most of the analytes.

Furthermore, in order to assess the degree of variability of the different analytes content with vinegar ageing and the similarity/dissimilarity among series of casks a three-way data analysis method (Tucker3) is proposed. The chemometric technique applied on the data set shows differences between the samples on the bases of their different ageing period, and between the batterie, which traditionally have an own peculiar production procedure.  相似文献

A low solvent consumption method for Fourier transform infrared spectroscopy (FT-IR) determination of doxylamine succinate in pharmaceuticals has been developed. The analyte was continuous and selectively extracted with a 13% (v/v) ethanol:chloroform solvent mixture, recirculating the solvent through the sample and monitoring the process by FT-IR. Doxylamine succinate was determined by on-line standard addition measuring the peak area in the regions 1730–1710 and 1485–1462 cm−1 corrected with a two-point baseline established between 2000 and 1800 cm−1. This new method implies low volumes of chloroformic solvent mixture, only 2.6 mL per sample, in front of classical batch FT-IR methods, improving analytical efficiency and reducing waste generation. The on-line extraction and standard addition determination of doxylamine succinate allowed a throughput of 10 h−1.  相似文献

A rapid and sensitive system of liquid chromatography coupled with microdialysis was developed for the simultaneous determination of unbound thalidomide in rat blood, brain and bile for pharmacokinetic study. Microdialysis probes were concurrently inserted into the jugular vein toward the right atrium, the brain striatum and the bile duct of the anesthetized Sprague-Dawley rats for biological fluid sampling after the administration of thalidomide (5 mg kg(-1)) through the femoral vein. Thalidomide and dialysates were separated using a Zorbax ODS C(18) column and a mobile phase comprising acetonitrile-methanol-0.1 mm 1-octanesulufonic acid (32:3:65, v/v/v, pH 5.3) at flow rate of 1 mL min(-1). The UV wavelength was set at 220 nm. The concentration-response relationship was linear (r(2)>0.995) over a concentration range of 0.025--25 microg mL(-1). The intra-assay and inter-assay precision and accuracy of thalidomide fell within 7%. The average in vivo recoveries were 0.31+/- 0.02,0.046+/- 0.004 and 0.57+/- 0.02 (n=6), respective to the dialysates of blood, brain and bile, with thalidomide at concentrations 2, 5 and 10 microg mL(-1). The disposition of thalidomide in the blood, brain and bile fluid suggests that there is a rapid thalidomide exchange and equilibration between the blood and brain systems. In addition, thalidomide undergoes hepatobiliary excretion.  相似文献

The paper presents a rapid method for the determination of dioxopromethazine hydrochloride (DPZ), an antihistamine drug, by the capillary electrophoresis with electrochemiluminescene detection (CE–ECL) using tris(2,2′-bipyridyl)ruthenium(II) (Ru(bpy)32+) reagent. This CE–ECL detection method has high sensitivity, good selectivity and reproducibility for DPZ analysis. Under the optimized conditions: separation capillary, 38 cm length (25 μm i.d.); sample injection, 10 s at 8 kV; separation voltage, 12.5 kV; running buffer, 20 mmol L−1 sodium phosphate of pH 6.0; detection potential, 1.15 V; 50 mmol L−1 of phosphate buffer (pH 7.14) containing 5 mmol L−1 of Ru(bpy)32+ in ECL detection cell, the detection limit of DPZ was 0.05 μmol L−1 (S/N = 3). The linear range extended from 5 to 100 μmol L−1. The linear curve obtained was Y = 181.62 + 9.28X with a correlation coefficient of 0.9970. The relative standard deviations of the ECL intensity and the migration time for six continuous injections of 5 μmol L−1 DPZ were 3.7% and 0.92%, respectively. The CE–ECL method was applied to analyze DPZ in real samples including tablets, rat serum and human urine, and satisfactory results were obtained without interference from samples matrix. The CE–ECL technique was proved to be a potential method for the detection of DPZ in clinic analysis.  相似文献

A novel, rapid and specific ultra performance liquid chromatography electrospray ionization tandem mass spectrometry (UPLC-ESI-MS/MS) method has been developed for simultaneous determination and pharmacokinetic studies of three bufadienolides (bufalin, cinobufagin and resibufogenin) in rat plasma. The analytes, bufalin, cinobufagin, resibufogenin and the internal standard, diphenhydramine were extracted from rat plasma samples by a one-step liquid–liquid extraction and separated on an ACQUITY UPLC™ BEH C18 column with gradient elution using a mobile phase composed of acetonitrile and water (containing 0.1% formic acid) at a flow rate of 0.20 mL min−1. Detection was carried out on a triple-quadrupole tandem mass spectrometer in the multiple reaction monitoring (MRM) mode via an electrospray ionization (ESI) interface. The three bufadienolides could be simultaneously determined within 3.0 min. Linear calibration curves were obtained over the concentration ranges of 1.0–200 ng mL−1 for all the analytes. The intra- and inter-day precisions (relative standard deviation (R.S.D.)) were less than 11.35 and 10.87%, respectively. The developed method was applied for the first time to the pharmacokinetic studies of bufadienolides in rats following a single intravenous administration of 2.10 mg kg−1 bufadienolides.  相似文献

A study on transport, kinetic selectivity and stability in SLM operations using a new carrier, the molecule 2-hydroxy-5-dodecylbenzaldehyde (2H5DBA) in kerosene, is described. A simple transport model is derived to evaluate the mass transfer coefficient in the membrane. Finally a comparison with the di-(2-ethylhexyl) phosphoric acid (D2EHPA) carrier in kerosene is made. The SLM system was employed and tested in the removal of Cu2+ from wastewater by using the operating conditions obtained from L–L extraction tests. Studies on the kinetics of copper extraction by using the 2H5DBA showed that complexation reaction is very fast. Transport tests were performed at different carrier concentrations (10%, 30%, 50% (v/v)) showing the improvement of SLM performance with increasing its concentration. Operating the SLM at optimum conditions (50% (v/v) 2H5DBA concentration in kerosene, feed pH 5, strip pH 2.2) final copper concentrations in the feed and strip phases were, respectively, 2.0 and 47.0 mg L−1, starting from 50 mg L−1 in the feed, meaning a significant up-hill transport. The fluxes (J) were calculated by fitting the experimental data of copper concentration in the feed by an exponential equation. They were used to calculate the transport (kinetic) selectivities of Cu2+ SLM separation over Ni2+, Zn2+ and Mn2+, given by the ratio J0(Cu)/J0(M), where M = Ni, Zn and Mn. The values were 37.4, 48.2 and 42.1, respectively. Transport and stability tests at the optimal carrier concentration by using the 2H5DBA and the D2EHPA in kerosene were carried out to compare them in terms of flux, lifetime and mass transfer coefficients. Experimental data evidenced for 2H5DBA a lower copper flux (8.67 mmol h−1 m−2 versus 36.71 mmol h−1 m−2), a lower lifetime (20 h versus 57 h) and lower mass transfer coefficient in the membrane (3.00 × 10−7 m s−1 versus 2.00 × 10−6 m s−1) but the selectivity of the separation process can overcome the disadvantages.  相似文献

An improved flow-based procedure is proposed for turbidimetric sulphate determination in waters. The flow system was designed with solenoid micro-pumps in order to improve mixing conditions and minimize reagent consumption as well as waste generation. Stable baselines were observed in view of the pulsed flow characteristic of the systems designed with solenoid micro-pumps, thus making the use of washing solutions unnecessary. The nucleation process was improved by stopping the flow prior to the measurement, thus avoiding the need of sulphate addition. When a 1-cm optical path flow cell was employed, linear response was achieved within 20–200 mg L−1, described by the equation S = −0.0767 + 0.00438C (mg L−1), r = 0.999. The detection limit was estimated as 3 mg L−1 at the 99.7% confidence level and the coefficient of variation was 2.4% (n = 20). The sampling rate was estimated as 33 determinations per hour. A long pathlength (100-cm) flow cell based on a liquid core waveguide was exploited to increase sensitivity in turbidimetry. Baseline drifts were avoided by a periodical washing step with EDTA in alkaline medium. Linear response was observed within 7–16 mg L−1, described by the equation S = −0.865 + 0.132C (mg L−1), r = 0.999. The detection limit was estimated as 150 μg L−1 at the 99.7% confidence level and the coefficient of variation was 3.0% (n = 20). The sampling rate was estimated as 25 determinations per hour. The results obtained for freshwater and rain water samples were in agreement with those achieved by batch turbidimetry at the 95% confidence level.  相似文献

The far infrared spectrum from 370 to 50 cm−1 of gaseous 2-bromoethanol, BrCH2CH2OH, was recorded at a resolution of 0.10 cm−1. The fundamental O–H torsion of the more stable gauche (Gg′) conformer, where the capital G refers to internal rotation around the C–C bond and the lower case g to the internal rotation around the C–O bond, was observed as a series of Q-branch transitions beginning at 340 cm−1. The corresponding O–H torsional modes were observed for two of the other high energy conformers, Tg (285 cm−1) and Tt (234 cm−1). The heavy atom asymmetric torsion (rotation around C–C bond) for the Gg′ conformer has been observed at 140 cm−1. Variable temperature (−63 to −100°C) studies of the infrared spectra (4000–400 cm−1) of the sample dissolved in liquid xenon have been recorded. From these data the enthalpy differences have been determined to be 411±40 cm−1 (4.92±0.48 kJ/mol) for the Gg′/Tt and 315±40 cm−1 (3.76±0.48 kJ/mol) for the Gg′/Tg, with the Gg′ conformer the most stable form. Additionally, the infrared spectrum of the gas, and Raman spectrum of the liquid phase are reported. The structural parameters, conformational stabilities, barriers to internal rotation and fundamental frequencies have been obtained from ab initio calculations utilizing different basis sets at the restricted Hartree–Fock or with full electron correlation by the perturbation method to second order. The theoretical results are compared to the experimental results when appropriate. Combining the ab initio calculations with the microwave rotational constants, r0 adjusted parameters have been obtained for the three 2-haloethanols (F, Cl and Br) for the Gg′ conformers.  相似文献

A novel voltammetric method for the determination of β-d-glucose (GO) is proposed based on the reduction of Cu(II) ion in Cu(II)(NH3)42+–GO complex at lanthanum(III) hydroxide nanowires (LNWs) modified carbon paste electrode (LNWs/CPE). In 0.1 mol L−1 NH3·H2O–NH4Cl (pH 9.8) buffer containing 5.0 × 10−5 mol L−1 Cu(II) ion, the sensitive reduction peak of Cu(II)(NH3)42+–GO complex was observed at −0.17 V (versus, SCE), which was mainly ascribed to both the increase of efficient electrode surface and the selective coordination of La(III) in LNW to GO. The increment of peak current obtained by deducting the reduction peak current of the Cu(II) ion from that of the Cu(II)(NH3)42+–GO complex was rectilinear with GO concentration in the range of 8.0 × 10−7 to 2.0 × 10−5 mol L−1, with a detection limit of 3.5 × 10−7 mol L−1. A 500-fold of sucrose and amylam, 100-fold of ascorbic acid, 120-fold of uric acid as well as gluconic acid did not interfere with 1.0 × 10−5 mol L−1 GO determination.  相似文献

An inductively coupled plasma mass spectrometer (ICP-MS) was used as an ion chromatographic (IC) detector for the speciation analysis of arsenic and selenium. The arsenic and selenium species studied included arsenite [As(III)], arsenate [As(V)], monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), arsenobetaine (AsB), selenite [Se(IV)] and selenate [Se(VI)]. Gradient elution using (NH4)2CO3 and methanol at pH 9 allowed the chromatographic separation of all species in less than 12 min. Effluents from the IC column were delivered to the nebulization system of ICP-DRC-MS for the determination of arsenic and selenium. The potentially interfering 38Ar40Ar+ and 40Ar40Ar+ at the selenium masses m/z 78 and 80 were reduced in intensity by approximately 3 orders of magnitude by using 0.6 mL min−1 CH4 as reactive cell gas in the DRC while an Rpq value of 0.3 was used. Meanwhile, arsenic was determined as the adduct ion 75As12CHH+ at m/z 89, which is more sensitive than 75As. The limits of detection for arsenic and selenium were in the range of 0.002–0.01 ng mL−1 and 0.01–0.02 ng mL−1, respectively, based on peak height. The relative standard deviation of the peak areas for five injections of 5 ng mL−1 As and Se mixture was in the range of 2–4%. The concentrations of arsenic and selenium species have been determined in urine samples collected locally. The major As and Se species in urines were AsB, DMA and probably selenosugar at concentration of 20–40, 15–19 and 17–31 ng mL−1, respectively. The recoveries were in the range of 94–105% for all the determinations. This method has also been applied to determine various arsenic compounds in two fish samples. In this study, a simple and rapid microwave-assisted extraction method was used for the extraction of arsenic compounds from fish. The arsenic species were quantitatively leached with an 80% v/v methanol solution in a focused microwave field during a period of 5 min.  相似文献

A simple and rapid flow injection (FI) method is reported for the determination of phosphate (as molybdate reactive P) in freshwaters based on luminol chemiluminescence (CL) detection. The molybdophosphoric heteropoly acid formed by phosphate and ammonium molybdate in acidic conditions generated chemiluminescence emission via the oxidation of luminol. The detection limit (3× standard deviation of blank) was 0.03 μg P l−1 (1.0 nM), with a sample throughput of 180 h−1. The calibration graph was linear over the range 0.032–3.26 μg P l−1 (r2=0.9880) with relative standard deviations (n=4) in the range 1.2–4.7%. Interfering cations (Ca(II), Mg(II), Ni(II), Zn(II), Cu(II), Co(II), Fe(II) and Fe(III)) were removed by passing the sample through an in-line iminodiacetate chelating column. Silicate interference (at 5 mg Si l−1) was effectively masked by the addition of tartaric acid and other common anions (Cl, SO42−, HCO3, NO3 and NO2) did not interfere at their maximum admissible concentrations in freshwaters. The method was applied to freshwater samples and the results (26.1±1.1–62.0±0.4 μg P l−1) were not significantly different (P=0.05) from results obtained using a segmented flow analyser method with spectrophotometric detection (24.4±4.45–84.0±16.0 μg P l−1).  相似文献

Amberlite XAD-16 resin has been functionalized using nitrosonaphthol as a ligand and characterized employing elemental, thermogravimetric analysis and FT-IR spectroscopy. The sorption of Ni(II) and Cu(II) ions onto this functionalized resin is investigated and optimized with respect to the sorptive medium (pH), shaking speed and equilibration time between liquid and solid phases. The monitoring of the influence of diverse ions on the sorption of metal ions has revealed that phosphate, bicarbonate and citrate reduce the sorption up to 10–14%. The sorption data followed Langmuir, Freundlich, and Dubinin–Radushkevich (D–R) isotherms. The Freundlich parameters computed are 1/n = 0.56 ± 0.03 and 0.49 ± 0.05, A = 9.54 ± 1.5 and 6.0 ± 0.5 mmol g−1 for Ni(II) and Cu(II) ions, respectively. D–R isotherm yields the values of Xm = 0.87 ± 0.07 and 0.35 ± 0.05 mmol g−1 and of E = 9.5 ± 0.23 and 12.3 ± 0.6 kJ mol−1 for Ni(II) and Cu(II) ions, respectively. Langmuir characteristic constants estimated are Q = 0.082 ± 0.005 and 0.063 ± 0.003 mmol g−1, b = (4.7 ± 0.2) × 104 and (7.31 ± 0.11) × 104 l mol−1 for Ni(II) and Cu(II) ions, respectively. The variation of sorption with temperature gives thermodynamic quantities of ΔH = −58.9 ± 0.12 and −40.38 ± 0.11 kJ mol−1, ΔS = −183 ± 10 and −130 ± 8 J mol−1 K−1 and ΔG = −4.4 ± 0.09 and −2.06 ± 0.08 kJ mol−1 at 298 K for Ni(II) and Cu(II) ions, respectively. Using kinetic equations, values of intraparticle transport and of first order rate constant have been computed for both the metal ions. The sorption procedure is utilized to preconcentrate these ions prior to their determination in tea, vegetable oil, hydrogenated oil (ghee) and palm oil by atomic absorption spectrometry using direct and standard addition methods.  相似文献

A procedure for chromium preconcentration and speciation with a dual mini-column sequential injection system coupled with electrothermal atomic absorption spectrometry (ETAAS) was developed. At pH 6, the sample solution was firstly aspirated to flow through a Chlorella vulgaris cell mini-column on which the Cr(III) was retained. The effluent was afterwards directed to flow through a 717 anion exchange resin mini-column accompanied by the retention of Cr(VI). Thereafter, Cr(III) and Cr(VI) were eluted by 0.04 mol L− 1 and 1.0 mol L− 1 nitric acid, respectively, and the eluates were quantified with ETAAS. Chemical and flow variables governing the performance of the system were investigated. By using a sampling volume of 600 µL, sorption efficiencies of 99.7% for Cr(III) and 99% for Cr(VI) were achieved along with enrichment factors of 10.5 for Cr(III) and 11.6 for Cr(VI), within linear ranges of 0.1–2.5 µg L− 1 for Cr(III) and 0.12–2.0 µg L− 1 for Cr(VI). Detection limits of 0.02 µg L− 1 for Cr(III) and 0.03 µg L− 1 for Cr(VI) along with RSD values of 1.9% for Cr(III) and 2.5% for Cr(VI) (1.0 µg L− 1, n = 11) were obtained. The procedure was validated by analyzing a certified reference material of GBW08608 and further demonstrated by chromium speciation in river and tap water samples.  相似文献

A sample solution was passed at 20 ml min−1 through a column (150×4 mm2) of Amberlite IRA-410Stron anion-exchange resin for 60 s. After washing, a solution of 0.1% sodium borohydride was passed through the column for 60 s at 5.1 ml min−1. Following a second wash, a solution of 8 mol l−1 hydrochloric acid was passed at 5.1 ml min−1 for 45 s. The hydrogen selenide was stripped from the eluent solution by the addition of an argon flow at 150 ml min−1 and the bulk phases were separated by a glass gas–liquid separator containing glass beads. The gas stream was dried by passing through a Nafion® dryer and fed, via a quartz capillary tube, into the dosing hole of a transversely heated graphite cuvette containing an integrated L’vov platform which had been pretreated with 120 μg of iridium as trapping agent. The furnace was held at a temperature of 250°C during this trapping stage and then stepped to 2000°C for atomization. The calibration was performed with aqueous standards solution of selenium (selenite, SeO32−) with quantification by peak area. A number of experimental parameters, including reagent flow rates and composition., nature of the gas–liquid separator, nature of the anion-exchange resin, column dimensions, argon flow rate and sample pH, were optimized. The effects of a number of possible interferents, both anionic and cationic were studies for a solution of 500 ng 1−1 of selenium. The most severe depressions were caused by iron (III) and mercury (II) for which concentrations of 20 and 10 mg  1−1 caused a 5% depression on the selenium signal. For the other cations (cadmium, cobalt, copper, lead,. magnesium, and nickel) concentrations of 50–70 mg 1−1 could be tolerated. Arsenate interfered at a concentration of 3 mg−1, whereas concentrations of chloride, bromide, iodide, perchlorate, and sulfate of 500–900 mg l−1 could be tolerated. A linear response was obtained between the detection limit of 4 ng 1−1, with a characteristic mass of 130 pg. The RSDs for solutions containing 100 and 200 ng 1−1 selenium were 2.3% and 1.5%, respectively.  相似文献

An amide-linked 2,6-bis{[(2-hydroxy-5-tert-butylbenzyl)(pyridyl-2-methyl)-amino]-methyl}-4-methylphenol-ruthenium(II) tris(bipyridine) 2PF6 complex, 1, was first used to recognize Co(II) in EtOH/H2O (1:1, v/v) solution, with the ruthenium(II) tris(bipyridine) moiety selected as a fluorophore and the multi-substituted phenol unit chosen as a receptor. The fluorescence quenching of 1 was attributed to the formation of an inclusion complex between multi-substituted phenol unit and Co(II) by 1:1 complex ratio (K = 2.5 × 105), which has been utilized as the basis of the fabrication of the Co(II)-sensitive fluorescent chemosensor. The analytical performance characteristics of the proposed Co(II)-sensitive chemosensor were investigated. The sensor can be applied to the quantification of Co(II) with a linear range covering from 1.0 × 10−7 to 5.0 × 10−5 M and a detection limit of 5 × 10−8 M. The experiment results show that the response behavior of 1 to Co(II) is pH-independent in medium condition (pH 4.5–9.5) and show excellent selectivity for Co(II) over transition metal cations except Cu(II). The chemosensor has been used for determination of Co(II) in water samples.  相似文献

Electrochemical studies of famotidine were carried out using voltammetric techniques: cyclic voltammetry, linear sweep and square wave adsorptive stripping voltammetry. The dependence of the current on pH, buffer concentration, nature of the buffer, and scan rate was investigated. The best results for the determination of famotidine were obtained in MOPS buffer solution at pH 6.7. This electroanalytical procedure enabled to determine famotidine in the concentration range 1 × 10−9–4 × 10−8 mol L−1 by linear sweep adsorptive stripping voltammetry (LS AdSV) and 5 × 10−10–6 × 10−8 mol L−1 by square wave adsorptive stripping voltammetry (SW AdSV). Repeatability, precision and accuracy of the developed methods were checked. The detection and quantification limits were found to be 1.8 × 10−10 and 6.2 × 10−10 mol L−1 for LS AdSV and 4.9 × 10−11 and 1.6 × 10−10 mol L−1 for SW AdSV, respectively. The method was applied for the determination of famotidine in urine.  相似文献
