An electrochemical DNA biosensor was fabricated by immobilizing DNA probe on aluminum ion films that were electrodeposited on the surface of the stearic acid‐modified carbon paste electrode (CPE). DNA immobilization and hybridization were characterized with cyclic voltammetry (CV) by using methylene blue (MB) as indicator. MB has a couple of well‐defined voltammetric redox peaks at the CPE. The currents of redox peaks of MB decreased after depositing aluminum ion films on the CPE (Al(III)/CPE) and increased dramatically after immobilizing DNA probe (ssDNA/Al(III)/CPE). Hybridization of DNA probe led to a marked decrease of the peak currents of MB, which can be used to detect the target single‐stranded DNA. The conditions for the preparation of Al(III)/CPE, and DNA immobilization and hybridization were optimized. The specific sequences related to bar transgene in the transgenic corn and the PCR amplification of CP4 epsps gene from the sample of transgenic roundup ready soybean were detected by differential pulse voltammetry (DPV) with this new electrochemical DNA biosensor. The difference between the peak currents of MB at ssDNA/Al(III)/CPE and that at hybridization DNA modified electrode (dsDNA/Al(III)/CPE) was applied to determine the specific sequence related to the target bar gene with the dynamic range comprised between 1.0×10?7 mol/L to 1.0×10?4 mol/L. A detection limit of 2.25×10?8 mol/L of oligonucleotides can be estimated.  相似文献

A graphite electrode modified with silver (Ag‐CPE) has been applied to detect mercury(II) using differential pulse voltammetry (DPV). Under optimized conditions, the calibration curve is linear in the range from 5.0×10?8 mol L?1 to 1.0×10?4 mol L?1 of mercury(II). The detection limit was found to be 3.38×10?8 mol L?1 with a relative standard deviation (RSD) of 2.25 % (n=8). The proposed method was successfully applied for the detection of mercury(II) in leachate samples. The Ag‐CP composites were characterized using X‐ray diffraction (XRD), BET adsorption analysis and scanning electron microscopy (SEM).  相似文献

A room temperature ionic liquid N‐butylpyridinium hexafluorophosphate (BPPF6) was used as a binder to make an ionic liquid modified carbon paste electrode (IL‐CPE), which showed good characteristics such as simple preparation procedure, fast electrochemical response and good conductivity. The electrochemical oxidation of ascorbic acid (AA) on the new IL‐CPE was carefully studied. The oxidation peak potential of AA on the IL‐CPE appeared at 109 mV (vs. SCE), which was about 338 mV decrease of the overpotential compared to that obtained on the traditional carbon paste electrode (CPE) and the oxidation peak current was increased for about four times. The electrochemical parameters of AA on the IL‐CPE were calculated with the charge transfer coefficient (α) and the electrode reaction rate constant (ks) as 0.87 and 0.800 s?1, respectively. Based on the relationship of the oxidation peak current and the concentration of AA a sensitive analytical method was established with cyclic voltammetry. The linear range for AA determination was in the range from 1.0×10?5 to 3.0×10?3 mol/L with the linear regression equation as Ip (μA)=?2.52–0.064C (μmol/L) (n=13, γ=0.9942) and the detection limit was calculated as 8.0×10?6 mol/L (3σ). The proposed method was free of the interferences of coexisting substances such as dopamine (DA) and amino acids etc., and successfully applied to the vitamin C tablets determination.  相似文献

Simultaneous determination of dihydroxybenzene isomers was investigated at a multi‐wall carbon nanotubes (MWCNTs)/β‐cyclodextrin composite modified carbon ionic liquid electrode in phosphate buffer solution (pH 7.0, 1/15 mol/L) in the presence of cationic surfactant cetylpyridinium bromide (CPB). With the great enhancement of surfactant CPB, the voltammetric responses of dihydroxybenzene isomers were more sensitive and selective. The oxidation peak potential of hydroquinone was about 0.024 V, catechol was about 0.140 V and resorcinol 0.520 V in differential pulse voltammetric (DPV) measurements, which indicated that the dihydroxybenzene isomers could be separated entirely. The electrode showed wide linear behaviors in the range of 1.2×10?7–2.2×10?3, 7.0×10?7–1.0×10?3, 2.6×10?6–9.0×10?4 mol/L for hydroquinone, catechol and resorcinol, respectively. And the detection limits of the three dihydroxybenzene isomers were 4.0×10?8, 8.0×10?8, 9.0×10?7 mol/L, respectively. The proposed method could be applied to the determination of dihydroxybenzene isomers in artificial wastewater, and the recovery was from 97.4% to 104.2%.  相似文献

The electrochemical properties of L ‐cysteic acid studied at the surface of p‐bromanil (tetrabromo‐p‐benzoquinone) modified carbon paste electrode (BMCPE) in aqueous media by cyclic voltammetry (CV) and double step potential chronoamperometry. It has been found that under optimum condition (pH 7.00) in cyclic voltammetry, the oxidation of L ‐cysteic acid at the surface of BMCPE occurs at a half‐wave potential of p‐bromanil redox system (e.g., 100 mV vs. Ag|AgCl|KClsat), whereas, L ‐cysteic acid was electroinactive in the testing potential ranges at the surface of bare carbon paste electrode. The apparent diffusion coefficient of spiked p‐bromanil in paraffin oil was also determined by using the Cottrell equation. The electrocatalytic oxidation peak current of L ‐cysteic acid exhibits a linear dependency to its concentration in the ranges of 8.00×10?6 M–6.00×10?3 M and 5.2×10?7 M–1.0×10?5 M using CV and differential pulse voltammetry (DPV) methods, respectively. The detection limits (2σ) were determined as 5.00×10?6 M and 4.00×10?7 M by CV and DPV methods. This method was used as a new, selective, rapid, simple, precise and suitable voltammetric method for determination of L ‐cysteic acid in serum of patient's blood with migraine disease.  相似文献

Electrochemical behavior of dopamine (DA) was investigated at the gold nanoparticles self‐assembled glassy carbon electrode (GNP/LC/GCE), which was fabricated by self‐assembling gold nanoparticles on the surface of L ‐cysteine (LC) modified glassy carbon electrode (GCE) via successive cyclic voltammetry (CV). A pair of well‐defined redox peaks of DA on the GNP/LC/GCE was obtained at Epa=0.197 V and Epc=0.146 V, respectively. And the peak separation between DA and AA is about 0.2 V, which is enough for simultaneous determination of DA and AA. The peak currents of DA and AA were proportional with their concentrations in the range of 6.0×10?8–8.5×10?5 mol L?1 and 1.0×10?6–2.5×10?3 mol L?1, with the detection limit of 2.0×10?8 mol L?1 and 3.0×10?7 mol L?1 (S/N=3), respectively. The modified electrode exhibits an excellent reproducibility, sensibility and stability for simultaneous determination of DA and AA in human serum with satisfactory result.  相似文献

We report on a novel electrochemical method to detect trace pentachlorophenol (PCP) by using a chitosan (CS) modified carbon paste electrode (CS/CPE). Compared with that at a bare carbon paste electrode (CPE), the current response was greatly improved at the CS/CPE due to the enhancement effect of CS. Under optimal working conditions, the oxidation peak current of PCP was proportional to its concentration in the range of 1.0 × 10?7 to 5.0 × 10?6 and 5.0 × 10?6 to 1.0 × 10?4 mol/L, with an extremely low detection limit of 4.0 × 10?8 mol/L. Our method was successfully used to detect the PCP concentration in vegetable samples.  相似文献

Differential pulse voltammetry (DPV) at a carbon fibre rod electrode (CFRE) and a capillary carbon paste electrode (CPE) have been used for the determination of pesticides cymoxanil and famoxadone, respectively. In the cathodic potential range, optimum conditions were found for the determination of cymoxanil by DPV at CFRE at pH 4 with limit of quantification (LQ) of 5.9×10?7 mol L?1. In the anodic area, determination of famoxadone by DPV at CPE was performed at optimum pH 2 with LQ=1.4×10?7 mol L?1. Practical applicability of the newly developed methods was verified on spiked samples of river water and soil.  相似文献

Abstract

The interaction between clozapine (CLZ) as an orally administrated antipsychotic drug with double stranded calf thymus DNA (dsDNA) was investigated at electrode surface using differential pulse voltammetry (DPV). Activated carbon paste electrode (CPE) was modified with dsDNA and used for monitoring the changes of the characteristics peak of CLZ in 0.05 M acetate buffer (pH 4.3). The adsorptive stripping voltammetry on dsDNA‐modified carbon paste electrode (dsDNA‐CPE) was used for determination of very low concentration of CLZ. Under optimal conditions, the oxidation peak current is proportional to CLZ concentration in the range of 7×10?9?1.2×10?6 mol l?1 with a detection limit of 1.5×10?9 mol l?1 for 180 s accumulation time by DPV. The proposed dsDNA‐CPE was successfully used for determination of CLZ in human serum samples with recovery of 97.0±2.5%.  相似文献

The electrochemical behavior of D ‐penicillamine (D ‐PA) studied at the surface of ferrocene carboxylic acid modified carbon paste electrode (FCAMCPE) in aqueous media using cyclic voltammetry and double step potential chronoamperometry. It has been found that under optimum condition (pH 7.00), the oxidation of D ‐PA at surface of such an electrode is occurred about 420 mV less positive than that an unmodified carbon paste electrode (CPE). The catalytic oxidation peak current was linearly dependent on the D ‐PA concentration and a linear calibration curve was obtained in the ranges 7.5×10?5 M – 1.0×10?3 M and 6.5×10?6 M?1.0×10?4 M of D ‐PA with cyclic voltammetry (CV) and differential pulse voltammetry (DPV) methods respectively. The detection limits (3σ) were determined as 6.04×10?5 M and 6.15×10?6 M. This method was also used for the determination of D ‐PA in pharmaceutical preparation (capsules) by standard addition method.  相似文献

A sensor for cytochrome c (cyt c) was developed using a carbon paste electrode (CPE) modified with cellulose‐DNA. Cyt c was adsorbed on the cellulose‐DNA modified CPE through the electrostatic interaction between them. Owing to this process, a pair of well‐defined peaks appeared at +48 mV/85 mV (Epc/Epa vs. Ag/AgCl). This property of the cellulose‐DNA modified CPE was utilized for the analysis of cyt c in a biological sample. The optimum experimental conditions for analysis were investigated and a calibration plot was obtained between 1.0×10?4 M and 1.0×10?6 M (±5% at n=5) at the optimized condition. The detection limit for cyt c at the optimized experimental condition was 5.0×10?7 M (S/N=3) for 30 min of deposition time with differential pulse voltammetry (DPV). The real sample analysis was carried out with the standard addition method to evaluate the developed method. The content of cyt c in total proteins of 80.0 mg/mL in rat mitocondria fractions was determined to be 0.12 (±0.02) mg/mL.  相似文献

This work reports the novel application of carbon‐coated magnetite nanoparticles (mNPs@C) as catalytic nanomaterial included in a composite electrode material (mNPs@C/CPE) taking advantages of their intrinsic peroxidase‐like activity. The nanostructured electrochemical transducer reveals an enhancement of the charge transfer for redox processes involving hydrogen peroxide. Likewise, mNPs@C/CPE demonstrated to be highly selective even at elevated concentrations of ascorbic acid and uric acid, the usual interferents of blood glucose analysis. Upon these remarkable results, the composite matrix was further modified by the addition of glucose oxidase as biocatalyst, in order to obtain a biosensing strategy (GOx/mNPs@C/CPE) with enhanced properties for the electrochemical detection of glucose. GOx/mNPs@C/CPE exhibit a linear range up to 7.5×10?3 mol L?1 glucose, comprising the entirely physiological range and incipient pathological values. The average sensitivity obtained at ?0.100 V was (1.62±0.05)×105 nA L mol?1 (R2=0.9992), the detection limit was 2.0×10?6 M while the quantification limit was 6.1×10?6 mol L?1. The nanostructured biosensor demonstrated to have an excellent performance for glucose detection in human blood serum even for pathological values.  相似文献

The SAM nanoSe0/Vc/SeCys‐film modified Au electrode has been prepared to determine selenocystine and selenomethionine. The AFM and SEM showed the special three‐dimensional (3D) network structure of the sol‐gel films. The affinity between nanoparticles and biomolecules created special chemical characters analyzed by the XRD and fluorescence. The modified electrode was characterized by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). The modified films partly had resistance in the charge transduction of Fe(CN) , but the less electron‐transfer resistance. Differential pulse voltammetric (DPV) determination of selenoamino acids using SAM nanoSe0/Vc/SeCys‐film modified Au electrode was presented. In PBS (pH 7.0)+0.1 mol L?1 NaClO4 solution, selenoamino acids yielded a sensitive reduction peak at about +400±50 mV. The peak current had a linear relationship with the concentration of selenoamino acids in the range of 5.0×10?8–1.0×10?5 mol L?1, and a 3σ detection limit of selenoamino acids was 1.2×10?8 mol L?1. The relative standard deviation of DPV signals of 0.50×10?6 mol L?1 selenoamino acids was 3.8% (n=8) using the same electrode and was 4.4% (n=5) when using three modified electrodes prepared at different times. The content of selenoamino acids in the organo‐selenium powder were determined by DPV. The results showed 71.5 μg g?1 of SeCys and 65.1 μg g?1 of SeMet in the organo‐selenium powder.  相似文献

Voltammetric properties and possibility of the determination of carcinogenic aminoderivatives of polycyclic aromatic hydrocarbons, namely 1‐ and 2‐aminonaphthalene and 2‐aminobiphenyl, have been investigated. Carbon paste electrodes (CPE) modified with monomeric α‐, β‐ or γ‐cyclodextrin and carbon‐based screen‐printed electrodes (SPE) surface‐modified with a thin film of β‐cyclodextrin (β‐CDP) or carboxymethylated β‐cyclodextrin (β‐CDPA) condensation polymer were used for that purpose as simple electrochemical biosensors. Analytical procedure for the DPV determination of tested amines using these biosensors was proposed. Linear calibration plots within the concentration range of 2×10?8 – 2×10?7 mol dm?3 and 2×10?7–1×10?6 mol dm?3 with limits of quantification of the order of 10?9 mol dm?3 were obtained for the modified CPE and modified SPE.  相似文献

A novel electrochemical sensor for sensitive detection of methyldopa at physiological pH was developed by the bulk modification of carbon paste electrode (CPE) with graphene oxide nanosheets and 3‐(4′‐amino‐3′‐hydroxy‐biphenyl‐4‐yl)‐acrylic acid (3,′AA). Applying square wave voltammetry (SWV), in phosphate buffer solution (PBS) of pH 7.0, the oxidation current increased linearly with two concentration intervals of methyldopa, one is 1.0×10?8–1.0×10?6 M and the other is 1.0×10?6–4.5×10?5 M. The detection limit (3σ) obtained by SWV was 9.0 nM. The modified electrode was successfully applied for simultaneous determination of methyldopa and hydrochlorothiazide. Finally, the proposed method was applied to the determination of methyldopa and hydrochlorothiazide in some real samples.  相似文献

The electrochemical impedance spectroscopy (EIS) has been used to study the interaction between paraquat and carbon modified by silver (Ag? CPE) and silver particles‐impregnated natural phosphate (Ag/NPh? CPE). This study was developed using spectrophotometry (UV? Vis) and infrared spectroscopy. The resulting interaction was controlled by adsorption at lower concentration (≤1.0×10?5 mol L?1) and by diffusion in the opposite case. Both electrodes are used to determining paraquat with a low detection limit (<1.0×10?12 mol L?1). The precision expressed as relative standard deviation RSD for the concentration level 1.0×10?5 mol L?1 of paraquat, (n=8) were 0.93 % and 1.1 % for Ag/NPh? CPE and Ag? CPE respectively.  相似文献

In this paper a molecular wire modified carbon paste electrode (MW‐CPE) was firstly prepared by mixing graphite powder with diphenylacetylene (DPA). Then a graphene (GR) and chitosan (CTS) composite film was further modified on the surface of MW‐CPE to receive the graphene functionalized electrode (CTS‐GR/MW‐CPE), which was used for the sensitive electrochemical detection of adenosine‐5′‐triphosphate (ATP). The CTS‐GR/MW‐CPE exhibited excellent electrochemical performance and the electrochemical behavior of ATP on the CTS‐GR/MW‐CPE was carefully studied by cyclic voltammetry with an irreversible oxidation peak appearing at 1.369 V (vs. SCE). The electrochemical parameters such as charge transfer coefficient (α) and electrode reaction standard rate constant (ks) were calculated with the results of 0.53 and 5.28×10?6 s?1, respectively. By using differential pulse voltammetry (DPV) as detection technique, the oxidation peak current showed good linear relationship with ATP concentration in the range from 1.0 nM to 700.0 µM with a detection limit of 0.342 nM (3σ). The common coexisting substances, such as uric acid, ascorbic acid and guanosine‐5′‐triphosphate (GTP), showed no interferences and the modified electrode was successfully applied to injection sample detection.  相似文献

An electroanalytical study of the oxidation processes of umbelliferone and hymecromone at a glassy carbon electrode in micellar solution and emulsified medium by different voltammetric techniques is described. The non-ionic surfactant Triton X-405 in acetate-buffered medium at pH 4.8 was found to be the most suitable. Different ranges of linearity were obtained in the micellar solutions, depending on the technique used; the limits of determination for differential pulse voltammetry (DPV) at a stationary electrode were 2.9×10?6 mol l?1 and 3.3×10?6 mol l?1 for umbelliferone and hymecromone, respectively. In the emulsified medium formed with a mixture of toluene and ethyl acetate (3:2), the oxidation processes yielded similar results. With DPV, linear calibration plots were obtained in the ranges 1.0×10?5–9.0×10?7 mol l?1 umbelliferone and 1.0×10?5–2.0×10?6 mol l?1 hymecromone. The media used are predominantly aqueous so that special reference electrodes and solvent purification are not needed.  相似文献

The in‐site functionalization of 4‐aminothiophenol (4‐ATP) self‐assembled monolayer on gold electrode at physiological pH yields a redox active monolayer of 4′‐mercapto‐N‐phenylquinone diimine (MNPD). The functionalized electrode exhibits excellent electrocatalytic responses towards dopamine (DA) and ascorbic acid (AA), reducing the overpotentials by about 0.22 V and 0.34 V, respectively, with greatly enhanced current responses. Due to its different catalytic activities toward DA and AA, the modified electrode resolves the overlapping voltammetric responses of DA and AA into two well‐defined voltammetric peaks by differential pulse voltammetry (DPV), which can be used for the simultaneous determination of these species in a mixture. The catalytic peak current obtained from DPV was linearly related to DA and AA concentration in the ranges of 5.0×10?6?1.25×10?4 M and 8.0×10?6?1.3×10?4 M with correlation coefficient of 0.999 and 0.998, respectively. The detective limits (3σ) for DA and AA were found to be 1.2×10?6 M and 2.4×10?6 M, respectively. The modified electrode shows good sensitivity, selectivity and stability, and has been applied to the determination of DA and AA simultaneously in samples with satisfactory results.  相似文献

In this work, a sensitive electrochemical DNA biosensor for the detection of sequence‐specific target DNA was reported. Firstly, CuO nanospindles (CuO NS) were immobilized on the surface of a glassy carbon electrode (GCE). Subsequently, gold nanoparticles (Au NPs) were introduced to the surface of CuO NS by the electrochemical deposition mode. Probe DNA with SH (HS‐DNA) at the 5′‐phosphate end was covalently immobilized on the surface of the Au NPs through Au? S bond. Scanning electron microscopy (SEM) was used to elucidate the morphology of the assembled film, and electrochemical impedance spectroscopy technique (EIS) was used to investigate the DNA sensor assembly process. Hybridization detection of DNA was performed with differential pulse voltammetry (DPV) and the methylene blue (MB) was hybridization indicator. Under the optimal conditions, the decline of reduction peak current of MB (ΔI) was linear with the logarithm of the concentration of complementary DNA from 1.0×10?13 to 1.0×10?6 mol·L?1 with a detection limit of 3.5×10?14 mol·L?1 (S/N=3). In addition, this DNA biosensor has good selectivity, and even can distinguish single‐mismatched target DNA.  相似文献
