A new series of peptidic nanostructures bearing two intercalating moieties was designed and synthesized to achieve selective recognition of DNA sequences. A cationic porphyrin was attached to a glutamic acid side chain and the latter introduced into a peptidic sequence by standard solid-phase peptide synthesis methodology. Conformation of the hydrosoluble peptidic structures bearing two cationic porphyrins was studied by circular dichroism. Using UV-visible spectroscopy and induced circular dichroism, we demonstrate that the compounds are fully intercalated upon binding to double-stranded DNA and that the compounds exhibit a tremendous preference for GC over AT sequences for intercalation.  相似文献

The ascorbic acid/organic base photoreduction of zinc(II) porphyrins was investigated. It was established that certain substituents can direct the photoreduction to the site of the macrocycle to which they are attached. For example, zinc(II) vinylporphyrins (8, 12, 16, 20) are photoreduced with cis stereochemistry on the ring bearing the vinyl group to give the corresponding chlorins. Zinc(II) acetylporphyrins (22, 24) were likewise reduced to chlorins such that cis-hydrogenation took place on the ring bearing the acetyl group. Zinc(II) formylporphyrins 33 also appear to reduce at the ring bearing the formyl group. When the zinc(II) acrylic porphyrin 28 was photoreduced, reduction did take place at the ring bearing the acrylic side chain, but migration of the acrylate double bond was very rapid, and the product isolated was the corresponding porphyrin propionate 30. Reduction of a zinc(II) porphyrin 35 bearing both a vinyl group and a nuclear carboxylic ester took place at the ring bearing the carboxylic ester. The reaction provides a general method for regioselective synthesis of chlorins from zinc(II) porphyrins without any evidence of formation of over-reduction products characteristic of many other procedures for formation of chlorins from porphyrin precursors.  相似文献

A series of new porphyrin derivatives possessing the side arms of alkyl-substituted thiophene oligomer were synthesized. The effects of solvent polarity on the formation of supramolecular assembly have been studied by UV-vis absorption, fluorescence emission, and TEM measurement. The linear-shape porphyrin derivatives bearing two thiophene pentamers at meso-position showed the significant spectral changes in both blue-shift and band broadening of Soret bands which indicate the formation of a relatively larger porphyrin H-aggregate that occurred in nonpolar solvent such as n-hexane.  相似文献

在纳秒(ns)领域中,利用Z-扫描技术测定了Zn(o-BocTyr)TAPP(主体1)和Zn(p-BocTyr)TAPP(主体2)两种手性锌卟啉的三阶非线性光学性质以及对咪唑类客体的分子识别行为的构象研究. 结果表明,(1) 两种手性锌卟啉都具有反饱和吸收效应和自散焦效应;(2) 由于两种主体中侧链位置的差异,造成两种主体分子极化程度的不同,主体1具有较大的三阶非线性折射率(n2)值;(3) 主体1中侧链苯环与卟啉环之间存在一定的相互作用;(4) 分子识别出现了配位方向的选择性,客体咪唑(Im)与主体1侧链中的苯环能够形成π-π相互作用,选择从有侧链一方配位于主体1,而2-甲基咪唑(2-MeIm)选择从没有侧链一方进攻主体1,但对于主体2,则没有出现配位方向的选择性.  相似文献

Threading of a polymer through a macrocyclic ring may occur directly, that is, by finding the end of the polymer chain, or by a process in which the polymer chain first folds and then threads through the macrocyclic ring in a hairpin‐like conformation. We present kinetic and thermodynamic studies on the threading of a macrocyclic porphyrin receptor ( H21 ) onto molecular threads that are blocked on one side and are open on the other side. The open side is modified by groups that vary in ease of folding and in bulkiness. Additionally, the threads contain a viologen binding site for the macrocyclic receptor, which is located close to the blocking group. The rates of threading of H21 were measured under various conditions, by recording as a function of time the quenching of the fluorescence of the porphyrin, which occurs when receptor H21 reaches the viologen binding site. The kinetic data suggest that threading is impossible if the receptor encounters an open side that is sterically encumbered in a similar way as a folded polymer chain. This indicates that threading of polymers through macrocyclic compounds through a folded chain mechanism is unlikely.  相似文献

应用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)研究了一系列阳离子性卟啉化合物的质谱行为.结果表明,阳离子性卟啉与非离子性卟啉化合物的激光解吸电离方式有明显不同.对于四-氮R基(R=甲基,乙基,丙基,苄基)吡啶基卟啉,吡啶基氮上的侧链基团(R)可明显影响该类化合物在MALDI-TOF-MS测试过程中离子形成方式;R基团的增大以及平衡阴离子半径的增大可增加平衡阴离子与卟啉环阳离子之间相互作用力的共价成分,因此在MALDI-TOF-MS测定过程中能够得到卟啉环阳离子与多个平衡阴离子结合在一起的较高质量数的离子峰.另外,还初步探讨了阳离子性卟啉化合物的激光解吸电离机理.  相似文献

本文合成了一种新型Boc(叔丁氧羰基)保护手性苏氨酸修饰的自由卟啉及其锌配合物。通过元素分析、紫外-可见光谱、荧光光谱、核磁共振氢谱、CD光谱等多种谱图对结构进行了表征,并结合理论计算采用模拟退火的方法搜索了手性锌卟啉配合物的最低能量构象。此外,利用Z-扫描技术测定了手性卟啉化合物的三阶非线性折射率。  相似文献

A series of water-soluble porphyrin electrolytes with quaternary ammonium ions side chains (TEMZnPN and THMZnPN) and sulfonated fullerene derivatives were synthesised and characterised. The self-assembled system was fabricated through the electrostatic interaction between the quaternary ammonium ions on the porphyrin derivatives and the sulfonic ions on the fullerene derivatives. The optical properties of the self-assembled system in aqueous solution were recorded by ultraviolet–visible and fluorescence spectroscopy. Scanning electron microscopy indicated that the morphology of the self-assembled films was highly relative to the distance between the quaternary ammonium ions and the porphyrin core, which could be easily adjusted by changing the length of the alkyl chain in the porphyrin derivatives.  相似文献

A modular building-block approach has been developed for the construction of linear amphipathic porphyrin arrays. The reaction of meso-(trifluoromethyl)dipyrromethane and an aldehyde under the conditions of the two-step room temperature porphyrin synthesis affords the trans-substituted porphyrin (13-56% yields). A similar reaction with two different aldehydes provides access to porphyrins bearing two different functional groups. An ethyne porphyrin and an iodo porphyrin (either free base or zinc) are selectively joined via Pd(0)-catalyzed coupling reactions, affording a linear array with porphyrins in defined metalation states. Coupling of a zinc-porphyrin bearing iodo and ester groups with a free base porphyrin bearing ethyne and ester groups yielded the zinc-free base porphyrin dimer. Coupling of a bis-ethyne porphyrin with a porphyrin bearing iodo and ester groups afforded the porphyrin trimer. Cleavage of the esters yielded the amphipathic porphyrin dimer and trimer arrays. The arrays with adjacent zinc and free base porphyrins undergo efficient electronic energy transfer. Both amphipathic porphyrin arrays have been incorporated into L-alpha-phosphatidylcholine vesicles. This versatile synthetic strategy provides access to a family of porphyrin arrays for studies of photophysical processes in supramolecular assemblies.  相似文献

利用LB (Langmuir-Blodgett)技术将含不同链长的卟啉化合物(C4Py, C6Py和C8Py)单层膜转移到ITO (indium-tin oxide)导电玻璃上, 发现其具有良好的光电转换性质. 卟啉化合物修饰后的紫外吸收光谱与光电流工作谱重叠, 表明卟啉化合物起到了敏化光电流产生的效果; 而且电子给体、电子受体和偏压对其敏化效果的实验结果表明: 光诱导电子转移是产生光电响应的主要原因. 而且, 这三个卟啉化合物的光电响应性质与碳链长度相关, 其中含有六个碳链的C6Py表现出最佳的光电转化效果.  相似文献

We report synthesis and coordination properties of β-alkyl porphyrin derivatives bearing terpyridylphenyl substituents at the meso-position and propionate side chains at the β-positions. Rhenium(I) carbonyl ion was encapsulated not in the core but in the external terpyridyl ligand. Such porphyrin-terpyridine hybrid porphyrins can be potentially available for less-invasive imaging like SPECT.  相似文献

Prior syntheses of porphyrins bearing meso-formyl groups have generally employed the Vilsmeier formylation of an acid-resistant copper or nickel porphyrin. A new approach for the synthesis of free base porphyrins bearing one or two (cis or trans) meso-formyl substituents entails the use of a dipyrromethane bearing an acetal group at the 5-position, a dipyrromethane-1-carbinol bearing an acetal group at the 5-position or carbinol position, or a dipyrromethane-1,9-dicarbinol bearing an acetal group at a carbinol position. Treatment of the resulting meso-acetal-substituted free base porphyrin to gentle acidic hydrolysis yields the corresponding meso-formyl porphyrin.  相似文献

Three new Mn(III) porphyrin catalysts of O2.-dismutation (superoxide dismutase mimics), bearing ether oxygen atoms within their side chains, were synthesized and characterized: Mn(III) 5,10,15,20-tetrakis[N-(2-methoxyethyl)pyridinium-2-yl]porphyrin (MnTMOE-2-PyP(5+)), Mn(III)5,10,15,20-tetrakis[N-methyl-N'-(2-methoxyethyl)imidazolium-2-yl]porphyrin (MnTM,MOE-2-ImP(5+)) and Mn(III) 5,10,15,20-tetrakis[N,N'-di(2-methoxyethyl)imidazolium-2-yl]porphyrin (MnTDMOE-2-ImP(5+)). Their catalytic rate constants for O2.-dismutation (disproportionation) and the related metal-centered redox potentials vs. NHE are: log k(cat)= 8.04 (E(1/2)=+251 mV) for MnTMOE-2-PyP(5+), log k(cat)= 7.98 (E(1/2)=+356 mV) for MnTM,MOE-2-ImP(5+) and log k(cat)= 7.59 (E(1/2)=+365 mV) for MnTDMOE-2-ImP(5+). The new porphyrins were compared to the previously described SOD mimics Mn(III) 5,10,15,20-tetrakis(N-ethylpyridinium-2-yl)porphyrin (MnTE-2-PyP(5+)), Mn(III) 5,10,15,20-tetrakis(N-n-butylpyridinium-2-yl)porphyrin (MnTnBu-2-PyP(5+)) and Mn(III) 5,10,15,20-tetrakis(N,N'-diethylimidazolium-2-yl)porphyrin (MnTDE-2-ImP(5+)). MnTMOE-2-PyP(5+) has side chains of the same length and the same E(1/2), as MnTnBu-2-PyP(5+)(k(cat)= 7.25, E(1/2)=+ 254 mV), yet it is 6-fold more potent a catalyst of O2.-dismutation , presumably due to the presence of the ether oxygen. The log k(cat)vs. E(1/2) relationship for all Mn porphyrin-based SOD mimics thus far studied is discussed. None of the new compounds were toxic to Escherichia coli in the concentration range studied (up to 30 microM), and protected SOD-deficient E. coli in a concentration-dependent manner. At 3 microM levels, the MnTDMOE-2-ImP(5+), bearing an oxygen atom within each of the eight side chains, was the most effective and offered much higher protection than MnTE-2-PyP(5+), while MnTDE-2-ImP(5+) was of very low efficacy.  相似文献

The ability to attach redox-active molecules to oxide surfaces in controlled architectures (distance, orientation, packing density) is essential for the design of a variety of molecular-based information storage devices. We describe the synthesis of a series of redox-active molecules wherein each molecule bears a benzylphosphonic acid tether. The redox-active molecules include zinc porphyrins, a cobalt porphyrin, and a ferrocene-zinc porphyrin. An analogous tripodal tether has been prepared that is based on a tris[4-(dihydroxyphosphorylmethyl)phenyl]-derivatized methane. A zinc porphyrin is linked to the methane vertex by a 1,4-phenylene unit. The tripodal systems are designed to improve monolayer stability and ensure vertical orientation of the redox-active porphyrin on the electroactive surface. For comparison purposes, a zinc porphyrin bearing a hexylphosphonic acid tether also has been prepared. The synthetic approaches for introduction of the phosphonic acid group include derivatization of a bromoalkyl porphyrin or use of a dimethyl or diethyl phosphonate substituted precursor in a porphyrin-forming reaction. The latter approach makes use of dipyrromethane building blocks bearing mono or tripodal dialkyl phosphonate groups. The zinc porphyrin-tripodal compound bearing benzylphosphonic acid legs tethered to a SiO(2) surface (grown on doped Si) was electrically well-behaved and exhibited characteristic porphyrin oxidation/reduction waves. Collectively, a variety of porphyrinic molecules can now be prepared with tethers of different length, composition, and structure (mono or tripodal) for studies of molecular-based information storage on oxide surfaces.  相似文献

We have recently reported that aquo and thioether complexes of the ferric cytochrome c heme peptide N-acetylmicroperoxidase-8 (FeIII-1) exhibit greater low-spin character than do the corresponding complexes of a synthetic, water-soluble, monohistidine-ligated heme peptide (FeIII-2; Cowley, A. B.; Lukat-Rodgers, G. S.; Rodgers, K. R.; Benson, D. R. Biochemistry 2004, 43, 1656-1666). Herein we report results of studies showing that weak-field ligands bearing a full (fluoride, chloride, hydroxide) or partial (phenoxide, thiocyanate) negative charge on the coordinating atom trigger dissociation of the axial His ligand in FeIII-2 but not in FeIII-1. We attribute the greater sensitivity of His ligation in FeIII-1 to weak-field anionic ligands than to weak-field neutral ligands to the following phenomena: (1) anionic ligands pull FeIII further from the mean plane of a porphyrin than do neutral ligands, which will have the effect of straining the His-Fe bond in FeIII-2, and (2) heme in FeIII-2 is likely to undergo a modest doming distortion following anion binding that will render the His-ligated side of the porphyrin concave, thereby increasing porphyrin/ligand steric interactions. We propose that ruffling of the heme in FeIII-1 is an important factor contributing to its ability to resist His dissociation by weak-field anions. First, ruffling should allow His to more closely approach the porphyrin than is possible in FeIII-2, thereby reducing bond strain following anion binding. Second, the ruffling deformation in FeIII-1, which is enforced by the double covalent heme-peptide linkage, will almost certainly prevent significant porphyrin doming.  相似文献

An ortho-dimethyl substituted meso-tetrakisarylporphyrin prefunctionalized with triflate groups was prepared in good yield from an accessible 2,6-dimethyl-4-(triflyloxy)benzaldehyde. This porphyrin is an interesting building block, which could directly be engaged in Suzuki cross-coupling reactions, to be further tetra functionalized with 3-pyridyl ligands in yields equal or above 85%. The porphyrin core of the various compounds bearing four remote coordination sites was metalated with zinc(II). The molecular structures of the starting triflate porphyrin derivative and of the zinc(II) porphyrin substituted with four 3-pyridyl groups bearing a protected alcohol were determined using X-ray crystallography.  相似文献
