Six new nor-beta-lapachones have been synthesized from reaction of 3-bromo-nor-beta-lapachone with arylamines. These derivatives have potent anticancer properties against several cell lines. Here, we report complete unambiguous assignments of (1)H and (13)C chemical shifts of the new compounds. The assignments were made using a combination of one- and two-dimensional NMR techniques ((1)H, (13)C, (1)H-(1)H COSY, (1)H-(13)C HSQC, and (1)H-(13)C HMBC).  相似文献

A series of KF/Al2O3 catalyzed Michael-addition reactions between malononitrile and α,β-unsaturated cycloketones in DMF solution were studied. At room temperature, 2-cyano-3-aryl-3-(1,2,3,4-tetrahydronaphthalen-1-one-2-yl) propionitrile derivatives were synthesized by the reaction between 2-arylmethylidene-1,2,3,4-tetra-hydronaphthalen-1-one and malononitrile. However, if the temperature was increased to 80℃, 2-amino-3-cyano-4-aryl-4H-benzo[h]chromene derivatives were obtained in high yields. When the α,β-unsaturated ketones were replaced by 2,6-biarylmethylidenecyclohexanone or 2,5-biarylmethylidenecyclopentanone, another series of 2-amino-3-cyano-4H-pyran derivatives was isolated successfully. The structures of the products were confirmed by X-ray diffraction analysis.  相似文献

The probable structure of the inclusion complex of beta-cyclodextrin (beta-CD) and (-)-epigallocatechin gallate (EGCg) in D2O was investigated using several NMR techniques. EGCg formed a 1:1 complex with beta-CD, in which the A ring and a portion of the C ring of EGCg were included at the head of the phenolic hydroxyl group attached to C7 of EGCg in the beta-CD cavity from the wide secondary hydroxyl group side. In the 1:1 complex with beta-CD, EGCg maintained the conformation in which the B and B' rings of EGCg took pseudoequatorial and pseudoaxial positions with respect to the C ring, respectively. The structure of the inclusion complexes of beta-CD and EGCg obtained from NMR experiments supported those determined from AM1 semiempirical SCF MO calculations well.  相似文献

(1)H and (13)C NMR chemical shifts of alpha- and beta-anomers of adenosine, 2'-deoxyadenosine and their acetate derivatives were completely and definitely assigned using the concerted application of one- and two-dimensional experiments (gCOSY, gNOESY, gHSQC and gHMBC). A new 2(3 --> 20)abeotaxane with an unusual 13beta-substitution pattern and a new 6/8/6-ring taxane were isolated from the methanol extract of the needles of Taxus cuspidata. The structures were established as 2alpha,7beta-diacetoxy-5alpha,10beta,13beta-trihydroxy-2(3 --> 20)abeotaxa-4(20), 11-dien-9-one (1) and 2alpha,5alpha,7beta,9alpha,13alpha-pentahydroxy-10beta-acetoxytaxa-4(20),11-diene (2) on the basis of 1D and 2D NMR spectral data and high-resolution FAB-MS analyses.  相似文献

In this work, the detailed NMR studies and full (13)C NMR assignments for five diterpenoids isolated from Sideritis caesarea and Sideritis athoa are described. Three 3beta,8beta-epoxymexicanolides, including xyloccensin K, 6-acetoxycedrodorin and a new one named xyloccensin W, were isolated from the fruit of a Chinese mangrove Xylocarpus granatum. Their structures were determined by spectroscopic analyses. The first complete assignment of (1)H and (13)C NMR data for xyloccensin W was achieved by means of 2D NMR techniques, including (1)H-(1)H COSY, HSQC, HMBC and NOESY spectra. In addition, the confusion of (1)H and (13)C NMR data previously reported for xyloccensin K was clarified.  相似文献

A chemoselective route for the synthesis of chromeno[2,3‐c]pyrazole‐2(3H)‐carbothioamide derivatives by a five‐component reaction of salicylaldehyde, malononitrile, NH2NH2?H2O, aryl isothiocyanate, and H2O in EtOH/AcOH mixture is reported. This new protocol has the advantages of high yields, short reaction times, ease of operation, and simple purification. All structures were confirmed by IR, 1H‐ and 13C‐NMR, and MS analyses. A plausible mechanism for this type of reaction is proposed (Scheme 2).  相似文献

Co atoms were reacted with ethene at 77 K and the paramagnetic products studied by electron spin resonance (ESR) at X- and K-bands. The ESR spectra of the major product at both frequencies showed eight cobalt multiplets (ICo=7/2) indicating a mono-cobalt complex. The spectra have orthorhombic g and cobalt hyperfine tensors and were simulated by the parameters; g1=2.284, g2=2.0027, g3=2.1527; A1<-25 MHz, A2=-109 MHz, A3=-198 MHz. Proton and 13C (1% natural abundance) hyperfine couplings were lower than the line widths (<2 MHz) indicating less than 0.5 spin transfer to the ethene ligands. We assigned the spectrum to a Jahn-Teller-distorted planar trigonal mono-cobalt tris-ethene [Co(eta-C2H4)3] complex in C2v symmetry. The SOMO is either a 3dx2-y2 (2a1) orbital in a T-geometry or a 3dxy (b1) orbital in a Y-geometry but there is only a spin density, a2, of 0.30 in these d orbitals. The spin deficiency of 0.70 is attributed to two factors; spin transfer from the Co to ethene pi/pi* orbitals and a 4p orbital contribution, b2, to the SOMO. Calculations of a2 and b2 have been made at three levels of spin transfer, theta. At theta=0.00a2 is 0.23 and b2 is 0.78, at theta=0.25a2 is 0.25 and b2 is 0.52 and at theta=0.50a2 is 0.28 and b2 is 0.23. The other possible assignment to a mono-cobalt bis-ethene complex [Co(eta-C2H4)2] cannot be discounted from the ESR data alone but is considered unlikely on other grounds. The complex is stable up to approximately 220 K indicating a barrier to decomposition of approximately 50 kJ Mol-1  相似文献

用氢谱、红外光谱、X-射线粉末衍射、热分析、元素分析等测试方法研究了Veronicafolin (3,5,4′-三羟基-6,7,3′-三甲氧基黄酮) 和β-环糊精 (β-CD) 的固体包合物的谱学特征。元素分析结果显示形成Veronicafolin-β-CD·20H2O包合物,其中C:39.58%, H: 5.75%,表明包合物中主客体比为1∶1。该包合类型属于AL-型。通过紫外-可见分光光度法研究了在羟丙基-β-环糊精(HP-β-CD)的存在下Veronicafolin的相溶解度曲线,测得校正曲线为y = 24148x + 0.0075 (r=0.9999),相溶解曲线为y=0.4738x-2.0×10-7 (r=0.9490),包结平衡常数Ks为4.5×106mol-1。HP-β-CD提高了黄酮醇Veronicafolin的溶解度。  相似文献

Although the medicinal plant Artemisia rupestris L. has been widely researched for several decades, its alkaloids have never been isolated before. To our surprise, the alkaloids in the plant were not detected in the stems but detected in the flowers. Herein, a novel and strange guaipyridine sesquiterpene alkaloid with a carboxyl group named rupestine was purified successfully from the total alkaloids extracted from the flowers by high-speed counter-current chromatography (HSCCC). The two-phase solvent system used was composed of ethyl acetate-methanol-water (8:1:7, v/v/v). Fifty six milligrams of rupestine was obtained at over 97% purity and 95% recovery from 200 mg of the total alkaloids in one-step separation. Its structure was elucidated by spectroscopic methods including high resolution ESI-MS, (1)H NMR, (13)C NMR, Heteronuclear Multiple Bond Correlation (HMBC), Heteronuclear Single Quantum Coherence (HSQC), and Nuclear Overhauser Enhancement Spectroscopy (NOESY).  相似文献

A series of novel ketimines with intrinsic protecting group anchoring was synthesized and allowed to react with various silylketene acetals in the presence of 5-10 mol % of a chiral Zn(OTf)(2)-(R,R)-Ph-pybox-aqua complex. The corresponding optically active quaternary alpha-amino acid derivatives were obtained in high yields and with enantioselectivities ranging from 34 % up to 95 % ee. The catalyst was studied by (1)H NMR spectroscopy and X-ray crystallography, and a dynamic equilibrium of two species was identified in solution. These are a homo-chiral 1:2 metal-ligand complex and a 1:1 metal-ligand complex, of which the latter is expected to be the actual catalyst of the diastereo- and enantioselective reaction. A strong positive nonlinear effect was observed due to the formation of a catalytically inactive 1:2 metal-ligand hetero-chiral complex. On the basis of DFT calculations and the absolute stereochemistry of the products, simultaneous coordination of the imino electrophile and a single molecule of H(2)O to the chiral Lewis acid complex is proposed. Coordination of the imine-nitrogen atom in the axial position of an octahedral complex can account for the facial selectivity as well as the diastereoselectivity observed.  相似文献

An efficient and novel synthesis of chromeno[2,3‐d]pyrimidine‐2,4‐dione derivatives with a nitroketene‐aminal moiety at C(5) via four‐component reaction of salicylaldehydes, barbituric acid, diamines, and 1,1‐bis(methylsulfanyl)‐2‐nitroethene in EtOH and in the presence of AcOH is reported. Easy performance, good yields, and easy purification are the main advantages of this method. All structures were confirmed by IR, MS, and 1H‐ and 13C‐NMR, and by X‐ray crystal‐structure analyses. Lophenol, cholest-4α-methyl-7-en-3β-ol (1), obtained from Dracaena cochinchinensis (Lour.) S. C. Chen, was structurally modified. It was acetylated to protect 3β-hydroxyl group, and then oxidised by selenium dioxide in acetic acid to give cholest-4a-methyl-8-en-3β, Ta-diol diacetate (3). This compound 3 is unstable in chloroform solution or when heated and easily converted to a diene compound, cholest-4a-methyl-7,14-dien-3β-ol acetate (4). The structures of 3 and 4 were elucidated by means of IR, ^1H NMR, ^13C NMR and MS, and the absolute configuration of 3 was established by X-ray crystallography. The property of 3 was also discussed in this paper. Both 3 and 4 are new compounds and were reported for the first time.  相似文献

The stereochemistry of gamma-butyrolactons tetrahydro-6a-phenylfuro[3,4-b]furan-2(3H)-one (1), 1,4,5,9b-tetrahydro-3a-methylnaphtho[2,1-b]furan-2(3aH)-one (2), 1,4,5,9 b-tetrahydro-3a-methylfuro[2,3-c]quinolin-2(3aH)-one (3) and hexahydro-furo[3,2-c]benzofuran-2-one (4) was studied using DPFGSE-NOE experiments. Compounds 1-3 contain two stereocenters, while 4 contains three. Both (1)H and (13)C spectra showed a single diastereomer of all the compounds. Routine 2D experiments (DQF)-COSY, HMQC/HSQC, and HMBC were used to assign (1)H and (13)C spectra completely. Diastereotopic methylene protons with resolved (1)H NMR signals as well as protons of cyclohexane served as references for the construction of the spatial arrangement in the molecules. NOE contacts between protons attached to the stereocenter and the diastereotopic protons were thus used to determine the configuration of the molecules. Vicinal coupling constants (3)J assisted the assignment of the conformational arrangement of the cyclohexane ring of 4.  相似文献

3‐Butylaminoquinolinediones ( 1 ) react with NaNO2 in AcOH to give the corresponding N‐nitrosoderivatives ( 2 ). The analogous reactions of 4‐hydroxy‐3‐butylaminoquinolinediones ( 5 ), prepared by the reduction of 1 with NaBH4, produce the corresponding nitrosamines ( 4 ). The reduction of both 2 and 4 with Zn under different conditions was non‐productive, but the reduction of both compounds with LiAlH4 at the oxo and lactame groups yielded impure products, generating new tricyclic benzoxadiazocines ( 9 ) by a reaction with HNCO. All compounds were characterized by IR, 1H‐, and 13C‐NMR (in some cases, 15N‐NMR also) spectroscopy and EI and/or ESI mass spectrometry. The X‐ray structure of compound 9g was determined.  相似文献

Three new abietane diterpene lactones, 3α-hydroxyjolkinolide A (1), ent-8α, 14β-dihydroxy-13(15)-ene-16(12α)-abietanolide (2) and ent-8α,14α-dihydroxy-13(15)-ene-16(12α)-abietanolide (3) as well as a known abietane diterpene jolkinolide A (4) were isolated from the roots of Euphorbia wallichii. (1)H and (13)C NMR spectroscopic data for 5alpha-androstanes and halo-5alpha-androstanes with different substituents at positions C-3, C-9, C-11 and C-17 were examined and assigned by a combination of 1D and 2D NMR experiments. The substituent effects on the (13)C chemical shifts were compared with those of epi-androsterone, used as a reference compound.
