Immobilized polysaccharide-based chiral stationary phases (CSPs) are gaining importance in the resolution of racemic compounds due to their stable nature on working with normal solvents and those prohibited for use with coated phases (tetrahydrofuran, chloroform, dichloromethane, acetone, 1,4-dioxane, ethyl acetate, and certain other ethers). This review discusses the use of immobilized polysaccharide CSPs in the chiral resolution of various racemates by liquid chromatography. The discussion includes immobilization methodologies, enantioselectivities, efficiencies, and a comparison of chiral recognition capabilities of coated vs. immobilized CSPs. Some applications of immobilized CSPs to the chiral resolution of racemic compounds are also presented.  相似文献

手性分离在生物医药等领域具有重要意义。高效液相色谱(HPLC)因其经济、快速、高效等特点被广泛应用于手性化合物的分离分析中。手性固定相(CSP)是HPLC实现手性分离的核心,而制备有效CSP的关键在于手性选择剂的筛选。近年来,大量文献报道了新型CSPs的制备,其中键合型CSPs因具有溶剂耐受性和较高稳定性等优点受到了广泛关注。该文对近年来以手性单分子、多糖、环糊精、大环抗生素、冠醚、杯芳烃及生物碱等为手性选择剂制备的新型键合型CSPs进行了归纳整理,并对其发展前景进行了展望。  相似文献

The HPLC separation of the enantiomers of six phthalans (1,3-dihydrobenzo[c]furans) synthesized as racemic mixtures from ortho-lithiated aryloxiranes was accomplished in the normal-phase mode using seven polysaccharide-derived and Pirkle-type chiral stationary phases (CSPs) and n-hexane/2-propanol mixtures as mobile phases. Separation and resolution factors up to 1.6 and 4.2, respectively, were obtained. The performances of various CSPs with regard to the same compound were, however, quite different not only between the two types of CSPs but also within the same type (polysaccharide-derived or Pirkle-type). Also diastereomeric pairs of phthalans show different enantioseparation using the same CSP.  相似文献

利用手性HPLC法对天然产物(+)-巴拉苏酰胺(balasubramide)及其对映体进行分离和光学纯度测定。在手性分离过程中,考察了两种不同的手性固定相和不同比例的流动相(正己烷和异丙醇),以进行手性分离方法的优化。结果表明:正己烷和异丙醇(70/30,V/V)在手性柱Chiralpak AD-H上获得最佳分离。光学活性的巴拉苏酰胺的对映体过量值高于98%,其分离因子(α)和分离度为2.15和21.80。本研究为光学活性的巴拉苏酰胺及其后续衍生物光学纯度控制提供了方法学基础。  相似文献

In rationalizing the odd chromatographic behavior for the separation of the enantiomers of N-(3,5-dinitrobenzoyl)-alpha-arylalkylamines on HPLC chiral stationary phases (CSPs) derived from alpha-(6,7-dimethyl-1-naphthyl)alkylamines, we initially suggested the occurrence of two competing, opposite sense chiral recognition processes termed the "dipole-stacking process" and the "hydrogen-bonding process". A simplified "single mechanism" model was later suggested with the importance of face to edge pi-pi interaction between aromatic rings come to recognized. The initial and subsequent chiral recognition models can be differentiated by noting the chromatographic trends for the enantioseparation of a homologous series of N-(3,5-dinitrobenzoyl)-alpha-(p-alkylphenyl)ethylamines on the aforementioned CSPs. Data so obtained were consistent with the second "single mechanism" model but not with the first "two competing mechanism" model. From these results, it has been concluded that the "single mechanism" model is more plausible than the "two competing mechanism" model.  相似文献

A pair of cis-trans isomeric chiral stationary phases (CSPs) derived from (S)-1-(1-naphtyl)ethylamine was prepared. The chromatographic behaviours on both CSPs with regard to the resolution of enantiomeric amino acids, amino alcohols, amines, and carboxylic acid were studied. According to separation factors, the trans-CSP showed better chiral recognition ability for the separation of most analytes chosen in this study. Three homologous series of the alkyl esters of racemic amino acids were resolved on both CSPs using n-hexane-2-propanol and n-hexane-dichloromethane as mobile phases. The trans-CSP also showed better enantioselectivity for the resolution of homologues. A reverse of elution order was observed for the resolution of the homologous series of phenylglycine alkyl esters on both CSPs. It was found that the relationship between the separation factor and the alkyl chain length of the ester homologous series depended upon the components of mobile phase. A higher magnitude of difference between the two CSPs in enantioselectivity for the resolution of a given homologue was obtained when n-hexane-dichloromethane was used as a mobile phase. A chiral recognition process, in which steric repulsion, face-to-face π-π interaction, face-to-edge π-π interaction and hydrogen bonding interaction were involved, was also suggested to describe the separation of enantiomeric homologues on both CSPs. This study clearly indicates that the chiral resolution is influenced by the geometry of the double bond in a CSP.  相似文献

Cyclodextrins (CDs) can be bound on silica to prepare chiral stationary phases (CSPs) for liquid chromatography. The cyclodextrin ring is connected to a spacer previously bonded on the silica surface. Three different CD-CSPs were prepared with three different spacers. (i) A dimethylethoxysilane with a linear 6 carbon chain produced a monomeric layer. The bonded CD units can move and rotate freely. (ii) The corresponding trimethoxysilane produced a polymeric layer. The bonded CD units were not located at the same distance of the silica surface, but they could still move and rotate freely. (iii) The third spacer contained a cyclohexyl ring that may introduce some conformational rigidity in the CD connection to silica. The three CSPs prepared contained a CD surface coverage of about 0.3 micromol/m(2) which is approximately half of the maximum theoretical CD coverage. The first spacer was the most efficient to bond CDs with an average value of 1.7 spacers per CD ring, whereas 5.5 spacers per CD ring were needed with the two other spacers. The chiral recognition capabilities of the three phases were compared using 14 racemic compounds. No pronounced differences were noted, but the CSP prepared with the dimethylethoxysilane monomeric spacer seems to be the most efficient for chiral recognition.  相似文献

通过区域选择性方法制备了两种新型淀粉衍生物,分别为淀粉2-苯甲酸酯-3-(4-甲基苯基氨基甲酸酯)-6-(3,5-二氯苯基氨基甲酸酯)和淀粉2-苯甲酸酯-3-(3,5-二氯苯基氨基甲酸酯)-6-(4-甲基苯基氨基甲酸酯),将二者分别涂覆于氨丙基硅胶后用作液相色谱手性固定相。研究表明:所制备的手性固定相显示出特异的手性识别能力,其手性识别能力明显高于均匀取代淀粉衍生物——淀粉三(3,5-二氯苯基氨基甲酸酯),取代基的性质及在葡萄糖单元上的位置对手性固定相的手性识别能力有较大的影响。一些未在商品化的手性柱Chiralpak AD上得到有效分离的手性化合物在所制备的固定相上得到了更好的分离。所测试的8对对映体在淀粉2-苯甲酸酯-3-(4-甲基苯基氨基甲酸酯)-6-(3,5-二氯苯基氨基甲酸酯)固定相上均得到了分离,因而此固定相的手性识别能力较强,具有潜在的应用价值。  相似文献

Liquid chromatographic comparisons for enantiomer resolution of α-amino acids and chiral primary amino compounds were made using chiral stationary phases (CSPs) prepared by covalently bonding (+)-(18-crown-6)-2,3,11,12-tetracarboxylic acid (18-C-6-TA) of the same chiral selector. The resolution of all α-amino acids on CSP 1 developed in our group was found to be better than that on CSP 2 reported by Machida et al. All α-amino acids examined in this study were well enantioseparated on CSP 1 (α=1.22–2.47), while four analytes were not resolved or all the other analytes were poorly resolved on CSP 2 than on CSP 1. However, in resolving the primary amino compounds without a carbonyl group, CSP 1 was comparable with CSP 2. Although (+)-18-C-6-TA of the same chiral selector was used to prepare CSP 1 and CSP 2, this study showed that different connecting methods for the CSPs might influence their ability to resolve the analytes depending on their structures related to the chiral recognition mechanism.  相似文献

Several structurally well-defined perfunctionalised cyclodextrin chiral stationary phases (CD CSPs) for high performance liquid chromatography have been successfully prepared by immobilisation of perfunctionalised cyclodextrins on silica through urea linkage(s) using the Staudinger reaction. These CSPs show high chiral recognition efficiency and are utilised in the resolution of various types of racemic compounds. This paper reviews the development of sixteen perfunctionalised cyclodextrin-based CSPs, their preparation, and their application to enantioseparation of seventy-seven racemic compounds under a range of separation conditions.  相似文献

The direct enantioseparation of a novel aminothiazolecarboxamide fungicide, ethaboxam, on polysaccharide-derived chiral stationary phases (CSPs) is described. Good resolution is achieved with several polysaccharide-derived CSPs. Chiralcel OD (OD-H) and Chiralpak AS are excellent for direct enantiomer separation of ethaboxam. The elution behavior and the effects of eluent composition on the resolution of ethaboxam are also investigated. Furthermore, the mechanism for chiral recognition using molecular mechanics is discussed.  相似文献

Nine new quinine (QN) carbamate C9-dimers (QN-X-QN), with different aliphatic and cyclic spacers (X), have been synthesized and immobilized onto porous silica gel for HPLC. The chiral discriminating behavior of these "dimeric" anion-exchange type chiral stationary phases (CSPs) has been investigated in detail, to elucidate the role of the presence of a second QN subunit on the chiral selector (SO), as well as the influence of the structure and length of the spacer, on the overall chiral recognition of a set of N-derivatized amino acids and other acidic drugs. The bulkiness of the intermediate spacer tuned the chiral recognition abilities of these SOs, with the 1,3-adamantylen-derived CSP being the one that led to the best separations. Shorter spacers reduced the chiral discrimination abilities of the "dimeric" selectors, with the n-hexylen bridge being the most favorable distance to allow a nearly independent interaction of the two QN subunits with the racemic analytes. The comparison to five "monomeric" CSPs showed that the "dimeric" ones usually retain the chiral analytes more strongly, though the enantioseparation is not improved. Nevertheless, the exceptional resolution abilities of dimeric SOs with a trans- 1,2-diaminocyclohexylen-bridge for the separation of DNP-derivatives of amino acids and certain acidic drugs of therapeutical interest (e.g., profens) seemed to be superior to most of the other CSPs.  相似文献

Two bonded chiral stationary phases (CSPs), 8-aminoquinoline-2-ylmethyl- and 8-aminoquinoline-7-ylmethyl-diaza-18-crown-6-capped [3-(2-O-beta-cyclodextrin)-2-hydroxypropoxy]propylsilyl silica particles (non-porous, 1.5 microm), have been prepared and evaluated using capillary liquid chromatography at high pressures (> or = 8000 p.s.i.). High column efficiency (up to 400 000 plates m(-1)) was achieved for chiral separations. These CSPs with two recognition sites, i.e. substituted-diaza-18-crown-6 and beta-cyclodextrin combined with high chromatographic efficiency provide good resolution of a variety of enantiomers and positional isomers in relatively short times under reversed-phase conditions. After inclusion of a Ni (II) ion from the mobile phase, the positively charged crown ether-capped beta-cyclodextrin facilitates specific static, dipolar, and host-guest complexation interactions with solutes.  相似文献

A series of fourteen anilide derivatives of ibuprofen were resolved on six chiral stationary phases (CSPs) derived from N-arylcarbamoyl derivatives of (S)-phenylglycine. Excellent chiral resolutions were achieved on these CSPs. The ionic-type CSPs showed better chiral recognition abilities than the corresponding covalent-type CSPs, and the CSP bearing two chiral centers has better performance than the CSPs bearing only one chiral center. The highest separation factor was achieved using the ionic-type CSP bearing two chiral centers for the resolution of the 3,5-dinitroanilide derivative of ibuprofen. This result is better than those reported in literature for the resolution of ibuprofen on the CSPs derived from amino acids, According to the chromatographic behaviors, the hydrogen bonding interaction, the π-π interactions provided by the phenyl groups in CSPs bearing one chiral center, and the phenylethylcarbamoyl moiety in CSPs bearing two chiral centers dominate the chiral recognition.  相似文献

采用"一锅法",以1,6-二异氰酸正己酯作间隔臂,制备了万古霉素及苯异氰酸酯衍生化的万古霉素手性固定相。对拉米夫定、拉米夫定的L-薄荷醇酯、酞胺哌啶酮和盐酸氟西汀进行了手性分离研究,在极性有机相模式下,研究了流动相甲醇中冰醋酸-三乙胺浓度和比例对手性分离的影响,观察到两种手性固定相具有不同的手性识别能力。在万古霉素手性固定相上4种溶质都获得了基线分离;在苯异氰酸酯衍生化的手性固定相上除盐酸氟西汀外也均获得基线分离。  相似文献

CHIRALPAK IA, CHIRALPAK IB and CHIRALPAK IC are a new generation of chiral packing materials for resolution of enantiomers by chromatography. They are all prepared by chemical immobilisation of the respective polysaccharide derivatives onto a supporting silica matrix. The present article aims to establish an understanding of the global enantioselective profiles of this series of chiral stationary phases (CSPs). When used in combination, high success rates for enantiomer resolution could be achieved. These three CSPs exhibit an exceptional level of complementary chiral recognition characteristics by applying common screening and method optimisation strategies.  相似文献

While HPLC chiral stationary phases (CSPs) based on chiral crown ethers have been known useful for the resolution of only racemic primary amino compounds or some secondary amino compounds, in this study, we first demonstrated that the CSP based on (+)-(18-crown-6)-2,3,11,12-tetracarboxylic acid is also useful for the resolution of N-benzoyl-alpha-amino acids, which do not contain a primary or secondary amino group. Especially, N-(3,5-dinitrobenzoyl)-alpha-amino acids were resolved better than corresponding N-(3-nitrobenzoyl)- or N-benzoyl-alpha-amino acids, the separation (alpha) and the resolution factors (R(S)) for the resolution of eight N-(3,5-dinitrobenzoyl)-alpha-amino acids being in the range of 1.06-1.81 and 0.54-2.81, respectively. The optimum mobile phase condition was the mixture of acetic acid-triethylamine-acetonitrile with the ratio of 0.05/0.25/100 (v/v/v).  相似文献

Two novel polymers containing glucose units as the main-chain that only differ in terms of their regioregularity were synthesized to evaluate their chiral recognition abilities as chiral stationary phases(CSPs)for high performance liquid chromatography(HPLC).The regioregular polymer(poly-5)shows clear resolution ability for the racemate of cobalt(Ⅲ)acetylacetonate(Co(acac)3),whereas the corresponding regioirregular polymer(poly-3)does not show any chiral recognition for Co(acac)3.The regioregular polymer main-chain seems to play an important role not only in providing an efficient interaction with the racemate but also in expressing the chiral recognition ability as a CSP for HPLC.  相似文献

A set of 42 chiral compounds containing stereogenic sulfur was prepared. There were 31 chiral sulfoxide compounds, three tosylated sulfilimines and eight sulfinate esters. The separations were done using five different macrocyclic glycopeptide chiral stationary phases (CSPs), namely ristocetin A, teicoplanin, teicoplanin aglycone (TAG), vancomycin and vancomycin aglycone (VAG) and seven eluents, three normal-phase mobile phases, two reversed phases and two polar organic mobile phases. Altogether the macrocyclic glycopeptide CSPs were able to separate the whole set of the 34 sulfoxide enantiomers and tosylated derivatives. Five of the eight sulfinate esters were also separated. The teicoplanin and TAG CSPs were the most effective CSPs able to resolve 35 and 33 of the 42 compounds. The three other CSPs each were able to resolve more than 27 compounds. The normal-phase mode was the most effective followed by the reversed-phase mode with methanol-water mobile phases. Few of these compounds could be separated in the polar organic mode with 100% methanol mobile phases. Acetonitrile was also not a good solvent for the resolution of enantiomers of sulfur-containing compounds, neither in the reversed-phase nor in the polar organic mode. The structure of the chiral molecules was compared to the enantioselectivity factors obtained with the teicoplanin and TAG CSP. It is shown that the polarity, volume and shape of the sulfoxide substituents influence the solute enantioselectivity factor. Changing the oxidation state of the sulfur atom from sulfoxides to sulfinate esters is detrimental to the compound's enantioselectivity. The enantiomeric retention order on the teicoplanin and TAG CSPs was very consistent: the (S)-(+)-sulfoxide enantiomer was always the less retained enantiomer. In contrast, the (R)-(-)-enantiomer was less retained by the ristocetin A, vancomycin and vancomycin aglycone columns, showing the complementarity of these CSPs. The macrocyclic glycopeptide CSPs provided broad selectivity and effective separations of chiral sulfoxides.  相似文献
