Herbs and herbal based products have been widely used, but some products might contain no herbal ingredient as claimed in the product label. Therefore, it is crucial to develop a fast, sensitive and reliable method to analyze the herbs and their finished products. The roots of Eurycoma longifolia, and the health supplement of E. longifolia and Cordyceps were used as the sample matrices in this study. Sonication assisted extraction was applied to extract the marker compounds, namely eurycomanone and cordycepin from E. longifolia and Cordyceps, respectively. The presence of the marker compounds was established by high throughput ultra-performance liquid chromatography tandem mass spectrometry (UPLC?MS/MS) using a highly sensitive and selective technique of multiple reaction monitoring (MRM). The positive ion transitions in MRM for eurycomanone and cordycepin are m/z 409 → 391 and m/z 252 → 136, respectively. The test method was validated for its robustness, accuracy, precision, linearity, detection and quantitation limits, as well as estimated for its measurement uncertainty.  相似文献

Novel substances of expected doping activity are constantly introduced to the market. β-Methylphenethylamine (BMPEA) is classified as a doping agent by the World Anti-Doping Agency as it is a positional isomer of amphetamine. In this work, the development and application of a simple and rapid analytical procedure that enables discrimination between both isomers is described. The analytes of interest were extracted from urine by a two-step liquid–liquid extraction and then analyzed by UPLC/MS/MS under isocratic conditions. The entire analytical procedure was validated by evaluating its selectivity, discrimination capabilities, carry-over, sensitivity, and influence of matrix effects on its performance. Application of the method resulted in detection of BMPEA in eight anti-doping samples, including the first report of adverse analytical finding regarding its use. Further analysis showed that BMPEA may be eliminated unchanged along with its phase II conjugates, the hydrolysis of which may considerably improve detection capabilities of the method. Omission of the hydrolysis step may therefore, produce false-negative results. Testing laboratories should also carefully examine their LC/MS/MS-based amphetamine and BMPEA findings as both isomers fragment yielding comparable collision-induced dissociation spectra and their insufficient chromatographic separation may result in misidentification. This is of great importance in case of forensic analyses as BMPEA is not controlled by the public law, and its manufacturing, distribution, and use are legal.  相似文献

A modular approach to α,β-unsaturated N-aryl ketonitrones has been developed. Specifically, condensation of anilines and enals followed by alkylation of the resulting α,β-unsaturated imines provided N-allyl anilines, which were subjected to oxidation with Oxone® to form α,β-unsaturated N-aryl ketonitrones. This modular approach is general and provides rapid access to diversely substituted α,β-unsaturated N-aryl ketonitrones with a single purification step in good yields.  相似文献

Bisphenol A (BPA) is a synthetic industrial reactant used in the production of polycarbonate plastics, and genistein is a natural phytoestrogen abundant in the soybean. Current studies investigating the endocrine-disrupting effects of concomitant exposures to BPA and genistein have warranted the development of an analytical method for the simultaneous measurement of BPA and genistein, as well as their primary metabolites, bisphenol A ?-d-glucuronide (BPA gluc) and genistein 4′-?-d-glucuronide (genistein gluc), respectively. All four analytes were extracted from rat plasma via solid phase extraction (SPE). Three SPE cartridges and four elution schemes were tested. Plasma extraction using Bond Elut Plexa cartridges with sequential addition of ethyl acetate, methanol, and acetonitrile yielded optimal average recoveries of 98.1 ± 1.8% BPA, 94.9 ± 8.0% genistein, 91.4 ± 6.1% BPA gluc, and 103 ± 6.1% genistein gluc. Identification and quantification of the four analytes were performed by a validated HPLC-MS/MS method using electrospray ionization and selective reaction monitoring. This novel analytical method should be applicable to the measurement of BPA, genistein, BPA gluc, and genistein gluc in urine, cultures, and tissue following in vivo exposures. While reports of the determination of BPA and genistein independently exist, the simultaneous optimized extraction and detection of BPA, genistein, BPA gluc, and genistein gluc have not previously been reported.  相似文献

建立超高效液相色谱–串联质谱法检测动物源食品中克伦特罗、莱克多巴胺、沙丁胺醇、特布他林、氯丙那林、西马特罗、菲诺特罗、妥布特罗、喷布特罗等9种β-受体激动剂的方法。样品经β-葡萄糖醛苷酶酶解,用0.2mol/L乙酸铵溶液(pH 5.2)提取,阳离子固相萃取柱净化,以乙腈–0.2%甲酸水溶液作为流动相进行洗脱,用Eclipse Plus C_(18)(50 mm×2.1 mm,1.8μm)色谱柱分离,采用多反应监测(MRM)模式进行定性和定量分析。9种β-受体激动剂的质量浓度在0.1~10.0 ng/mL范围内与定量离子丰度呈良好的线性关系,线性相关系数均大于0.999,检出限为0.1μg/kg。平均回收率为85.0%~101.2%,测定结果的相对标准偏差为2.8%~9.5%(n=6)。该方法精密度好,灵敏度高,能简便、快速、准确地测定动物源食品中的9种β-受体激动剂。  相似文献

We developed an ultra-high-performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) method to determine four antibacterial drugs in human plasma for clinical usage. Samples were prepared using protein precipitation with methanol. Chromatographic separation was accomplished in 4.5 min on a BEH C18 column (2.1 × 50 mm, 1.7 μm) using a gradient elution of methanol and water (containing 7.71 g/L concentrated ammonium acetate, adjusted to pH 6.5 with acetic acid) at a flow rate of 0.4 mL/min. Positive electrospray was used for ionization. The method was linear in the concentration range 1–100 μg/mL for vancomycin, norvoncomycin, and meropenem; and 0.5–50 μg/mL for R-isomer of moxalactam and S-isomer of moxalactam. For all analytes, the intra- and inter-day accuracies and precisions were −8.47%–10.13% and less than 12%, respectively. The internal standard normalized recoveries and matrix effect were 62.72%–105.78% and 96.67%–114.20%, respectively. All analytes were stable at six storage conditions, with variations of less than 15.0%. The method was applied in three patients with central nervous system infection. The validated method might be useful for routine therapeutic drug monitoring and pharmacokinetic study.  相似文献

In the present work, a new method based on a sample treatment by dispersive liquid–liquid microextraction (DLLME) for the extraction of six bisphenols (bisphenol A, bisphenol S, and monochloro-, dichloro-, trichloro-, and tetrachlorobisphenol A), four parabens (methyl-, ethyl-, propyl-, and butylparaben), and six benzophenones (benzophenone-1, benzophenone-2, benzophenone-3, benzophenone-6, benzophenone-8, and 4-hydroxybenzophenone) in human urine samples, followed by ultrahigh-performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) analysis, is validated. An enzymatic treatment allows determining the total content of the target EDCs. The extraction parameters were accurately optimized using multivariate optimization strategies. Ethylparaben ring-13C6, benzophenone-d10, and bisphenol A-d16 were used as surrogates. Limits of quantification ranging from 0.1 to 0.6 ng mL?1 and interday variabilities (evaluated as relative standard deviations) from 2.0 to 13.8 % were obtained. The method was validated using matrix-matched standard calibration followed by a recovery assay with spiked samples. Recovery rates ranged from 94 to 106 %. A good linearity, for concentrations up to 300 ng mL?1 for parabens and 40 ng mL?1 for benzophenones and bisphenols, was also obtained. The method was satisfactorily applied for the determination of target compounds in human urine samples from 20 randomly selected individuals.  相似文献

We show here that baseline separation of dansylated estrone, 17β-estradiol, and 17α-estradiol can be done, contrary to previous reports, within a short run time on a single RP-LC analytical column packed with particles bonded with phenyl-hexyl stationary phase. The chromatographic method coupled with isotope dilution tandem MS offers a simple assay enabling the simultaneous analysis of these analytes. The method employs 13C-labeled estrogens as internal standards to eliminate potential matrix effects arising from the use of deuterated estrogens. The assay also offers adequate accuracy and sensitivity to be useful for biological samples. The practical applicability of the validated method is demonstrated by the quantitative analyses of in vivo samples obtained from rats treated with Premarin®.

Application of gas chromatography-mass spectrometry (GC-MS) can significantly improve trace analyses of compounds in complex matrices from natural environments compared to gas chromatography only. A GC-MS/MS technique for determination of poly-β-hydroxybutyrate (PHB), a bacterial storage compound, has been developed and used for analysis of two soils stored for up to 319 d, fresh samples of sewage sludge, as well as a pure culture of Bacillus megaterium. Specific derivatization ¶of β-hydroxybutyrate (3-OH C4?:?0) PHB monomer units by N-tert-butyl-dimethylsilyl-N-methyltrifluoracetamide (MTBSTFA) improved chromatographic and mass spectrometric properties of the analyte. The diagnostic fragmentation scheme of the derivates tert-butyldimethylsilyl ester and ether of β-hydroxybutyric acid (MTBSTFA-HB) essential for the PHB identification was shown. The ion trap MS was used, therefore the scan gave the best sensitivity and with MS/MS the noise decreased, so the S/N was better and also with second fragmentation the amount of ions increased compared to SIM. The detection limit for MTBSTFA-HB by GC-MS/MS was about 10–13 g μL–1 of injected volume, while by GC (FID) and GC-MS (scan) it was around 10–10 g μL–1 of injected volume. Sensitivity of GC-MS/MS measurements of PHB in arable soil and activated sludge samples was down to 10 pg of PHB g–1 dry matter. Comparison of MTBSTFA-HB detection in natural soil sample by GC (FID), GC-MS (scan) and by GC-MS/MS demonstrated potentials and limitations of the individual measurement techniques.  相似文献

A novel sensitive screening method based on liquid chromatography–tandem mass spectrometry (LC–MS/MS) has shown the feasibility of separation and detection of thiodiglycolic acid in aqueous samples. The analysis of this compound is of interest since it is specific microbiological metabolite of thiodiglycol, which is precursor and degradation product of chemical warfare agent sulfur mustard. The LC–electrospray ionisation (ESI)–MS method provides a sensitive and direct approach for thiodiglycolic acid identification and quantification using non-extracted non-derivitised samples from aqueous solutions. Chromatographic separation of the thiodiglycolic acid was produced using a reverse phase LC column with gradient mobile phases consisting of 0.1% formic acid in water and acetonitrile. Identification and quantification of species were achieved using ESI–tandem MS monitoring two precursor-to-product ion transitions for thiodiglycolic acid. The method demonstrates linearity over at least two orders of magnitude and detection limit of 10 ng...mL1 in environmental water samples.  相似文献
