The traditional overlapping resolution mapping (ORM) scheme was modified for application in HPLC with the flow rate of the mobile phase as a new factor. The mobile phase composition and a flow rate were taken as parameters to influence effective resolution and elution time. Chromatograms were obtained for the separation of the 16 compounds for ten trial conditions. Computer-assisted mathematical simulation was used to obtain optimum conditions for resolution and separation time of the compounds. The validity of the model developed was confirmed by experiments.  相似文献

A liquid chromatographic chiral stationary phase based on (+)‐(18‐crown‐6)‐2,3,11,12‐tetracarboxylic acid was applied to the resolution of 15 analytes, including racemic rasagiline, a chiral drug for the treatment of Parkinson's disease, and its analogues. The composition of mobile phase was optimized to be ethanol/acetonitrile/acetic acid/triethylamine (80:20:0.2:0.3, v/v/v/v) by evaluating the chromatographic results for the resolution of five selected analytes under various mobile phase conditions. Under the optimized mobile phase conditions, racemic rasagiline was resolved quite well with a separation factor of 1.48 and resolution of 2.71 and its 14 analogues were also resolved reasonably well with separation factors of 1.06–1.54 and resolutions of 0.54–2.11. Among 15 analytes, racemic rasagiline was resolved best except for just one analyte. The analyte structure–enantioselectivity relationship indicated that racemic rasagiline has the most appropriate structural characteristics for resolution on the chiral stationary phase.  相似文献

Some of the optimization methods in reversed phase high-performance liquid chromatography (RP-HPLC) are based on resolution of the critical band pair. Mobile phase composition is changed systematically to establish those conditions giving an acceptable resolution for such a critical band pair, but sometimes the critical pair may change with the separation conditions, which obliges to identify it for each of those conditions. In the case of ionizable compounds, more than two bands may be involved in resolution, showing—in some cases—changes in the elution order when the mobile phase composition was modified. In this paper, an alternative way that does not identify the critical pair after changing experimental conditions is proposed. The relative separation of the three bands involved in two alternating critical band pairs is evaluated as a sort of conjugate or combined resolution, represented as contour maps vs. two variables (content of organic modifier and pH). These maps are obtained from data of chromatograms made under different separation conditions; these conditions were generated by experimental design and data was mathematically processed with a computer program. Analytes of three families that have acid–base properties, triazines, phenoxyacids, and phenols, were used for this purpose. The chromatographic behavior when elution order reversion of ionizable compounds exists is studied.  相似文献

A novel methylated beta-cyclodextrin chiral stationary phase (CSP-ME), which was chemically immobilised onto porous silica via multiple urea-linkages was synthesised. The CSP-ME chiral stationary phase depicted good enantiomer separation abilities for some well-known flavour as well as fragrance compounds using high-performance liquid chromatography under reverse phase conditions. The optimum resolution for alpha-ionone, 3-methyl-alpha-ionone, flavanone, 5-methoxyflavanone, 6-methoxyflavanone, 7-methoxyflavanone, hesperetin, naringenin and taxifolin was achieved using a mobile phase composition consisting of 1 wt.% triethylammonium acetate buffer (pH 4.68)-methanol. The effects of pH of triethylammonium acetate buffer and the methanol-acetonitrile content of the mobile phase composition on their retention time and resolution were examined to optimise the separation conditions.  相似文献

以纤维素-三(3,5-二甲基苯基氨基甲酸酯)(CDMPC)高效液相色谱手性固定相(CSP)成功地拆分了戊唑醇和三唑酮对映异构体,通过考察流动相中异丙醇的含量、柱长及温度对手性拆分的影响,优化色谱分离条件。结果显示:该固定相对两种手性农药有较好的拆分效果,使用250mm色谱柱,流动相中含5%异丙醇时戊唑醇得到最佳分离,分离度为1.22;三唑酮在异丙醇含量为2%时有最大分离度1.47。减少异丙醇含量对两种农药均有利于增大分离度,温度升高则降低分离效果。  相似文献

以S,S-酒石酸正十二酯和H3BO生成的配合物为手性载体, 通过量热分析结果初步证明该配合物的存在, 采用厚体液膜拆分了普萘洛尔外消旋体. 考察了S,S-酒石酸十二酯浓度和水相pH值对拆分性能的影响, 优化了其拆分操作条件, 同时进行了动力学分析. 得出厚体液膜拆分的一般规律, 即其拆分属于动力学拆分过程, 适当提高手性载体的浓度有利于提高对映单体流量和拆分的立体选择性, 水相pH值对于拆分性能有着重要的影响. 其拆分过程中萃取反应过程为控速步骤, 对映单体的跨膜传递可以用两个串联的准一级不可逆过程进行描述.  相似文献

Summary The chiral separation of eight racemic compounds has been investigated on a cellulose tris(3,5-dimethylphenylcarbamate) chiral column under reversed-phase conditions. The discrimination mechanism under reversed-phase conditions is discussed. Addition of acid to the mobile phase is necessary for resolution of acidic racemic compounds. The presence of ion-pair reagent in the mobile phase is a key factor in the resolution of basic racemic compounds. Retention of the racemates is also affected by addition of acid or salt. The anion in buffer not only interacts with the racemete, but also with the polysaccharide derivative on the silica gel surface.  相似文献

用自制的新型L-异亮氨酸配体交换固定相在配体交换模式下对11种DL-氨基酸进行了拆分研究,详细考察了流动相中铜离子浓度、甲醇含量、流速及温度对拆分效果的影响,并探讨了可能的拆分机理。研究结果表明:流动相中高浓度的铜离子不利于DL-氨基酸的拆分;增加流动相中甲醇的含量,降低流动相流速以及提高色谱柱温度均可改善拆分效果。  相似文献

High-performance liquid chromatography (HPLC) has been commonly used as method of separating and identifying photosynthetic pigments such as chlorophylls and carotenoids because of such advantages as speed, high resolution and sensitivity. In this technique, high separation relies largely on the type of column material. This study compared the efficiency of five reverse-phase columns, C8, C18, C18 monolithic, π-NAP, and cholester, for separation of photosynthetic pigments at several fixed conditions of mobile phase and temperature. This investigation also analysed the parameters of ΔtR and tR ratio for selected pigments and resolution for structural isomers, such as α- and β-carotene. Among above columns tested, cholester column is suitable for separation of pigments not only for a broad range of polarity, but also for hydrophobic pigments in a simple mobile phase. This finding can help in the selection of column and HPLC parameters in separating photosynthetic pigments.  相似文献

The theory of resolution optimization by isochronal analysis is further extended to the cases involving simultaneous changes in mobile phase velocity and column temperature, and in mobile phase composition and temperature. In each case, the resolution equation, when written in terms of isochronal conditions, shows that improvements can be obtained without sacrificing analysis time. Several subcases, which relate to the dependence of the selectivity factors on the changed parameters, are described. For each system, the contributions of the various parameters to the resolution are examined. The results of these discussions are plotted as resolution surfaces, on which lines of constant analysis time are indicated. The experimental routes that chromatographers must take in order to apply isochronal analysis are described.  相似文献

A new chiral chemically bonded stationary phase for direct resolution of optical isomers in HPLC was prepared. The experiemental data showed that the chiral stationary phase gave good resolution for the racemic α-amino acid derivatives and that there was no significant influence on the resolution as the eluting rate varied from 0.6 to 2.0 mL/min. The content of isopropanol in the eluent significantly affected the resolution of the enantiomers. The eluent consisting of 1% isopropanol in hexane gave most favorable result, but the retention times were a little longer. It is evident that the resolution was also influenced by the structure of the solute. In all cases, the resolution of N-Ac-DL-Phe-OMe is lower than that of N-Ac-DL-Leu-OMe and N-Ac-DL-Val-OMe. Although the capacity factor of N-Ac-DL-Tyr-Ome varied greatly in different eluting systems, the separation factor and resolution were very close. It was observed that D-amino acid derivatives eluted faster than the corresponding L-isomers. In the above chromatographic conditions N-acetyl-DL-α-phenylethylamine could not be resolved.  相似文献

The phenyl isothiocyanate, an electrophilic reagent for peptide chain sequencing, is used to pre‐column derivatize a variety of α‐amino acids in alkaline medium before their enantioresolution on a vancomycin bonded chiral phase using the acetonitrile‐based mobile phase. The observed resolution is believed to be due to the re‐location of the hydrogen receptor site from sulfur to nitrogen on the isothiocyanyl fragment of derivatizing reagent, which in turn changes the enantioselectivity. Under the same chromatographic conditions, the resolution for N‐benzoylated, 3,5‐dinitrobenzoylated and N‐carbobenzyloxylated amino acids is either not found or unsatisfactory. Also, no resolution is obtained in the reversed‐ or normal phase mode for all phenyl isothiocyanated amino acids examined in this study.  相似文献

Summary Polynuclear aromatics (PNA) were collected and enriched from diesel engine exhausts by means of a probe and an absorber system. Group separation of PNA from other constituents of exhaust was accomplished through elution with n-heptane on a silica column. A variety of phase systems ranging from unpolar to strong polar packings and eluents, respectively, were tested to resolve PNA into single species. Of these a n-octadecyl bonded silica packing and methanol/water was found to be the most optimal with regard to resolution for real PNA mixtures. Abstract

Efficiency and resolution have been studied with a CCC apparatus classified by Y. Ito1 as a HDES type J, and known as the HSCCC or the Multilayer Coil Separator Extractor. It has been shown that the efficiency decreases when increasing the linear velocity of the mobile phase in the column, which can be done by increasing the flow rate, or by decreasing the volume of the mobile phase in the column. The two ways of changing the linear velocity are not equivalent, since, at constant linear velocity, the efficiency increases when the volume of the mobile phase is well balanced with that of stationary phase. For each flow rate there is a minimum value for the volume of the mobile phase, and the relationship is linear. Working under these conditions, changing both the flow rate and the volume of the mobile phase leads to an efficiency which goes through a minimum at medium flow rates.

The resolution does not show this minimum: it is best at low flow rates and low volumes of mobile phase, and its variations are more important at low flow rates. This means that, if the resolution is good at moderate flow rates, it will remain good at higher flow rates. Combining three equations, it has been possible to predict the resolution when working with the minimum volume of mobile phase for a given flow rate.  相似文献
