The reaction of 7-chlorotetrazolo[1,5-α]quinoxaline 5-oxide 6a with acetic anhydride gave 7-chloro-5-(7-chlorotetrazolo[1,5-α]quinoxalin-4-yl)-4,5-dihydro-4-oxotetrazolo[1,5-α]quinoxaline 7a , while the reaction of 7-chloro-1,2,4-triazolo[4,3-α]quinoxaline 5-oxide 6b with acetic anhydride afforded 7-chloro-5-(7-chloro-1,2,4-triazolo[4,3-α]quinoxalin-4-yl)-4,5-dihydro-4-oxo-1,2,4-triazolo[4,3-α]quinoxaline 7b and 7-chloro-4,5-dihydro-4-oxo-1,2,4-triazolo[4,3-α]quinoxaline 8b . Abstract The synthesis of acetic 2-{[1-methyl-2-(4-oxo-5,6,7,8-tetrahydro-4H-benzo [4,5]-thieno [2,3-d] [1,3-oxazin-2-yl)ethylidene]amino}-4,5,6,7-tetrahydrohenzo[b]thiophene ?3-carboxylic acid anhydride 5 and 2-(oxopropyl)-5,6,7,8-tetrahydro-4H-benzo-[4,5] thieno[2,3-d][1,3]oxazin-4-one 7, has been achieved in three steps from ethyl 2-amino-4,5,6,7-tetrahydrobenzo(b]thiophene-3-carboxlate 1 via the reaction with ethyl acetoacetate followed by hydrolysis and acetic anhydride-induced cyclization. The 2-substituent in compound 5 has two functional groups i.e. active methylene and acid anhydride which are suitably located for intramolecular transformation. Thermal and/or base catalyzed intramolecular cyclization of 5 afforded 2-(4-acetoxy-(hydroxyl)-2-methyl-5,6,7,8-tetrahydrobenzo[4,5] thieno[2,3-b]pyridin-3-yl)-5,6,7,8- tetrahydro-4H-benzo[4,5]thieno[2,3-d] [1,3]oxazin-4-one 10 and 9 respectively. Treatment of 5 with hydrazine hydrate, aromatic and/or heterocyclic amines induced the same intramolecular cyclization with a concomitant oxazine-pyrimidine interconversion to give 3-amino(aryl or heteryl)-2-(4-hydroxy-5,6,7,8-tetrdhydrobenzo-[4,5]thieno[2,3-b]pyridin-3-yl)-3,4,5,6,7,8-hexahydrobenzo[4,5] thieno[2,3-d] pyrimid in-4-one 11–14 respectively. 相似文献

In reaction with acylating agents, 2-aryl-2-methyl-7,8-dihydro-6H-pyrimido[4,5-b]pyrindin-4-ones are acylated at the methyl group and also enter into reaction with diethyl oxalate. 1-Aryl-2-phenacyl-7,8-dihydro-6H-pyrimido[4,5-b]pyrindin-4-ones undergo dehydration under the influence of concentrated sulfuric acid. On the basis of the PMR and UV spectra, it was concluded that 1-aryl-2-acetonyl(phenacyl)-7,8-dihydro-6H-pyrimido[4,5-b]pyrindin-4-ones exist in two tautomeric forms with strong intramolecular hydrogen bonds of the chelate type — enaminocarbonyl and enol. 相似文献

4-Chloroimidazo[4,5-d]-1,2,3-triazine was obtained by oxidative chlorinatlon from imidazo[4,5-d]-1,2,3-triazine-4-thione, and its reaction with nucleophilic agents and cleavage of the triazine ring to give 5-diazoimidazole-4-carbonitrile were studied. The newly synthesized diazoimidazole was subjected to C- and N-diazo coupling. A new, general method for the synthesis of spiro[4,5]trienones is described by the intramolecular ipso-halocyclization of 4-(p-unsubstituted-aryl)-1-alkynes. In the presence of halide electrophiles, a variety of 4-(p-unsubstituted-aryl)-1-alkynes underwent the intramolecular ipso-halocyclization with water smoothly, affording the corresponding halo-substituted spiro[4,5]trienones in moderate to good yields. The obtained spiro[4,5]trienones can be applied in constructing the azaquaternary tricyclic skeleton via Pd-catalyzed Heck reaction. Notably, the prepared spiro[4,5]trienones and azaquaternary tricycles are of importance in the areas of pharmaceuticals and agrochemicals. The mechanism of the intramolecular ipso-halocyclization reaction is also discussed according to the (18)O-labeling experiments and DFT calculations. 相似文献

The reaction of 7-chloro-4-(2-cyano-2-hydroxyvinyl)tetrazolo[1,5-α]quinoxaline 2a with 4-aminopyridine, p-toluidine or p-aminophenol gave 7-chloro-4-(4-pyridylcarbamoylmethylene)-4,5-dihydrotetrazolo[1,5-α]-quinoxaline 7a , 7-chloro-4-(p-tolylcarbamoylmethylene)4, 5-dihydrotetrazolo[1,5-α]quinoxaline 8a or 7-chloro-4-(p-hydroxyphenylcarbamoylmethylene)-4,5-dihydrotetrazolo[1,5-α]quinoxaline 9a , respectively. The reaction of 7-chloro-4-(2-cyano-2-hydroxyvinyl)-1,2,4-triazolo[4,3-α]quinoxaline 2b with 4-aminopyridine, p-toluidine or p-aminophenol afforded 7-chloro4-(4-pyridylcarbamoylmethylene)-4,5-dihydro-1,2,4-triazolo-[4,3-α]quinoxaline 7b , 7-chloro-4-(p-tolylcarbamoylmethylene)-4,5-dihydro-1,2,4-triazolo[4,3-α]quinoxaline 8b or 7-chloro-4-(p-hydroxyphenylcarbamoylmethylene)-4,5-dihydro-1,2,4-triazolo[4,3-α]quinoxaline 9b , respectively. The reaction of compound 2a with 2-aminopyridine or 3-aminopyridine provided 7-chloro-4-(2-pyridyl-carbamoylmethylene)-4,5-dihydrotetrazolo[1,5-α]quinoxaline 10 or 7-chloro-4-(3-pyridyl-carbamoylmethylene)-4,5-dihydrotetrazolo[1,5-α]quinoxaline 11 , respectively. Compounds 7a,b (4-pyridylcarbamoyl) predominated as the enamine tautomer A in a trifluoroacetic acid solution, while compounds 8a,b (p-tolylcarbamoyl) and compounds 9a,b (p-hydroxyphenylcarbamoyl) coexisted as the enamine A and methylene imine B tautomers in a trifluoroacetic acid solution. Moreover, the ratio of the enamine tautomer A elevated in an order of compound 11 (3-pyridylcarbamoyl), compound 10 (2-pyridylcarbamoyl) and compound 7a (4-pyridylcarbamoyl), reflecting an order of the increase in the pKa values of the aminopyridine side chain moieties. In general, the ratio of the enamine tautomer A was higher in the basic carbamoyl derivatives 7–11 than in the neutral ester derivatives 3a,b . From these results, the basic side chain moiety of the tetrazolo[1,5-α]quinoxalines 7a-11 or 1,2,4-triazolo[4,3-α]quinoxalines 7b-9b was found to increase the ratio of the enamine tautomer A in trifluoroacetic acid media. 相似文献

Quinoline-2,4-diones reacted with 2-[bis(methylthio)methylene]malononitrile in DMF/Et3N to produce 3-(methylthio)-4-oxo-4,5-dihydrofuro[3,2-c]quinolone-2-carbonitriles and 3-(methylthio)-4-oxo-4,5-dihydrofuro[3,2-c]quinolone-2-carboxamides in state of 2-imino-substituted 4-(methylthio)-5,6-dihydro-2H-pyrano[3,2-c]quinolone-3-carbonitriles.
