采用高效液相色谱-质谱联用技术及高效液相色谱法对生熟普洱茶中的主要成分进行定性和定量分析。鉴定出普洱茶水溶液中8种主要成分,分别为没食子酸(GA)、没食子酸儿茶素(GC)、表没食子酸儿茶素(EGC)、儿茶素(C)、咖啡因(CAF)、表儿茶素(EC)、表没食子酸儿茶素没食子酸酯(EGCG)和表儿茶素没食子酸酯(ECG)。以这8种成分的含量为指标,对普洱生茶和熟茶各20批进行主成分分析、聚类分析和判别分析,能准确地区分普洱生茶与熟茶。  相似文献

Hydrogen/deuterium exchange reactions in a quadrupole ion trap mass spectrometer are used to differentiate galloylated catechin stereoisomers (catechin gallate and epicatechin gallate; gallocatechin gallate and epigallocatechin gallate) and the nongalloylated analogs (catechin and epicatechin, gallocatechin and epigallocatechin). Significant differences in the hydrogen/deuterium exchange behavior of the four pairs of deprotonated catechin stereoisomers are observed upon reaction with D(2)O. Interestingly, the nongalloylated catechins undergo H/D exchange to a much greater extent than the galloylated species, incorporating deuterium at both aromatic/allylic and active phenolic sites. Nongalloylated catechin isomers are virtually indistinguishable by their H/D exchange kinetics over a wide range of reaction times (0.05 to 10 s). Our experimental results are explained using high-level ab initio calculations to elucidate the subtle structural variations in the catechin stereoisomers that lead to their differing H/D exchange kinetics.  相似文献

Attomole quantities of catechins were determined by a capillary liquid chromatography system with electrochemical detection (CLC-ECD) and the system is applied to the determination of catechins in human plasma. The eight catechins: catechin (C), epicatechin (EC), gallocatechin (GC), epigallocatechin (EGC), catechin gallate (Cg), epicatechin gallate (ECg), gallocatechin gallate (GCg), and epigallocatechin gallate (EGCg), were separated within 10 min using a capillary column (0.2 mm i.d.) and a mobile phase of phosphoric acid (85%)-methanol-water (0.5:27.5:72.5, v/v/v), and were detected at +0.85 V vs. Ag/AgCl. Peak heights were found to be linearly related to the amount of catechins injected, from 200 amol to 500 fmol (r > 0.998). The detection limits of the catechins were 61 amol for EGC, 75 amol for EC, 54 amol for GC, 61 amol for C, 67 amol for GCg, 75 amol for EGCg, 75 amol for ECg and 89 amol for Cg (S/N = 3). Because the present method is highly sensitive and allows facile pretreatment for plasma sample, the time courses of concentrations of catechins (GCg, EC, EGCg, ECg, and Cg) and their conjugates in human plasma obtained from a 10 microl plasma sample after ingestion of green tea could be determined.  相似文献

建立了高效液相色谱-串联质谱法快速测定葡萄酒中白藜芦醇、黄酮类、多酚类功效成分的分析方法。葡萄酒样品直接稀释后进样,用C18柱进行分离,以乙腈-0.1%(体积分数)甲酸水溶液为流动相进行梯度洗脱,通过多反应监测(MRM)模式进行检测。13种功效成分在各自线性范围内呈良好的线性关系,相关系数均大于0.99。除表没食子儿茶素、没食子儿茶素、儿茶素没食子酸酯、花旗松素的检出限为1.0、1.0、3.0、3.0μg/L外,其他9种化合物的检出限均小于1.0μg/L。回收率为80.9%~112.3%,相对标准偏差小于10%。该方法快速、准确、灵敏度高,适用于葡萄酒中功效成分的快速分析。对实际样品的检测表明,所测葡萄酒样品中均含有儿茶素、表儿茶素、表没食子儿茶素、没食子儿茶素、表儿茶素没食子酸酯/儿茶素没食子酸酯、白藜芦醇、大豆黄素等功效成分,不同品种葡萄酒中这些功效成分含量差异显著。  相似文献

应用高效液相色谱法测定绿茶中没食子酸(GA)、咖啡因(CAF)及5种儿茶素类化合物,包括儿茶素(C)、表儿茶素(EC)、表没食子儿茶素没食子酸酯(EGCG)、表儿茶素没食子酸酯(ECG)、没食子儿茶素没食子酸酯(GCG)。对国家标准方法GB/T 8313-2008中流动相组成进行了调整,用0.5%乙酸代替2%的乙酸,在AT.Lichrom ODS色谱柱(4.6mm×250mm,5μm)上进行分离,流量为1.0mL.min-1,在波长278nm处进行紫外检测。7种组分在40min内达到完全分离。7种组分的质量浓度分别在一定的范围内与其峰面积呈线性关系。方法用于绿茶的分析,7种组分的加标回收率在96.0%~102.8%之间。  相似文献

Column liquid chromatography on a C18-bonded silica column with water-methanol-acetic acid as eluent was used to determine polyphenols and caffeine in tea. Without any pretreatment, catechin, epicatechin gallate, epigallocatechin gallate, epigallocatechin, epicatechin and caffeine were separated successfully within 15 min. The detection limits (S/N = 3) of polyphenols studied were 1.8-24 mg/l at a detection wavelength 270 nm. The linear range of the peak area calibration curves for the analytes were over two orders of magnitude with a correlation coefficient of 0.996-0.999. Using this method, some Chinese tea samples were analyzed with a good reproducibility (RSD are below 5%).  相似文献

A new ultra high performance liquid chromatography coupled with tandem mass spectrometry method for a fast and sensitive determination of eight polyphenols (hydroxytyrosol, catechin, epicatechin, epigallocatechin gallate, oleuropein, quercetin, rutin, tyrosol) and panthotenic acid in extra‐virgin olive oil was developed. The method does not require long sample pre‐treatment and presents the lowest limit of detection and limit of quantitation values present in literature. Inter‐ and intra‐day variability, linear dynamic range of the calibration curve, recovery and matrix effect were also determined and investigated. The method was applied to several oil samples of different type and origin. Given its accuracy, precision and rapidity, the method is characterized by an interestingly high throughput, reliability, and sensitivity.  相似文献

Flavan‐3‐ol monoglycosides, having four aglycons (+)‐catechin, (?)‐epicatechin, (?)‐epigallocatechin and epicatechin gallate monomeric units, are detected for the first time in Vitis vinifera L. cv. Merlot grape seeds and wine. These compounds were analyzed in red wine, seed and skin extracts by electrospray ionization quadrupole time of flight mass spectrometry (MS) in negative mode. Fragment ions derived from retro‐Diels Alder, heterocyclic ring fragmentation, benzofuran forming fragmentation and glycoside fragmentations were detected in targeted MS/MS mode. These compounds were not detected in skins; the comparative study showed evidence that these glycosylated compounds originate only from grape seeds. Our method allows for the identification of these glycosylated compounds based on their exact mass and their specific fragmentation pattern. However, exact glucose position on the monomeric units can not be determined. This work allowed us to partially identify 14 new flavan‐3‐ol monoglycosides, based on the exact mass of the molecular ions and their specific retro‐Diels Alder, heterocyclic ring fragmentation, benzofuran forming fragmentation and glycoside fragmentations. Potential of an “electronic tongue” multisensor system in identification of various tea samples and in quantitative analysis of separate tea components was studied. As a reference method for quantitative analysis of antioxidants of the polyphenolic type served micellar electrokinetic chromatography with a UV detector. The contents of epicatechin gallate, epigallocatechin gallate, epicatechin, epigallocatechin, gallic acid, gallocatechin gallate, and caffeine alkaloid in various tea samples were quantitatively estimated.  相似文献

Presented here is the quantitative analysis of green tea NIST standard reference materials (SRMs) via liquid chromatography-particle beam/electron ionization mass spectrometry (LC-PB/EIMS). Three different NIST green tea standard reference materials (SRM 3254 Camellia sinesis Leaves, SRM 3255 C. sinesis Extract and SRM 3256 Green Tea-containing Oral Dosage Form) are characterized for the content of caffeine and a series of catechin species (gallic acid, catechin, epicatechin, epigallocatechin, epicatechin gallate and epigallocatechin gallate (EGCG)). The absolute limits of detection for caffeine and the catechin species were determined to be on the nanogram level. A reversed-phase chromatographic separation of the green tea reference materials was carried out on a commercial C18 column using a gradient of water (containing 0.1% TFA) and 2:1 methanol:acetonitrile (containing 0.1%TFA) at 0.9 mL min−1 and an analysis time of 50 min. Quantification of caffeine and the catechin species was carried out using the standard addition and internal standard methods, with the latter providing appreciable improvements in precision and recovery.  相似文献

 筛选出HypersilBDSC18和ZorbaxSBC18两种适合同时分离茶叶和茶多酚中 7种儿茶素和咖啡因的反相柱。采用甲醇 水 醋酸 (或三氟醋酸 )作流动相 ,分别以等强度洗脱和梯度洗脱 (均在 30min内 )分离测定了我国 6种不同产地茶叶样品和 3种茶多酚样品中 7种儿茶素的含量。考察了 7种儿茶素和咖啡因的保留值与流动相组成及柱温的关系 ,优化了色谱条件及样品前处理方法。用电喷雾电离质谱 (ESI MS)定性确认没食子儿茶素没食子酸酯(GCG)和儿茶素没食子酸酯 (CG)两组分 ,并用高效液相色谱制备两对照品用于定量分析。  相似文献

A sensitive, specific, accurate HPLC‐MS/MS method was developed and validated for the simultaneous quantification of catechin, epicatechin, liquiritin, isoliquiritin, liquiritigenin, isoliquiritigenin, piperine and glycyrrhetinic acid from Longhu Rendan pills in rat plasma. Chromatographic separation was performed with a Hypersil Gold C18 column using a gradient of methanol and 0.01% acetic acid containing 0.2 mm ammonium acetate as mobile phase. The analytes were quantified on a triple quadrupole mass spectrometer, operating in selected reaction monitoring mode and switching the electrospray ion source polarity between positive and negative modes in a single run. The calibration curves of catechin, epicatechin, liquiritin, isoliquiritin, liquiritigenin, isoliquiritigenin, piperine and glycyrrhetinic acid were linear over the concentration ranges of 5–2000, 5–2000, 0.5–200, 0.5–200, 0.25–100, 0.25–100, 0.025–10 and 0.50–200 ng mL?1, respectively. The intra‐ and inter‐assay precisions and accuracies were <11.6 and 91.9–108.2%, respectively, for all analytes. Matrix effects for all analytes were between 88.2 and 114.2%. Stability testing showed that all analytes were stable in plasma at 24 °C for 3 h, at 4 °C for 24 h, after three freeze–thaw cycles, and at ?80 °C for 15 days. The method was successfully applied to an in vivo study evaluating the pharmacokinetics of multiple nonvolatile compounds following intragastric administration of Longhu Rendan pills to rats. A rapid and quantitative analytical method for the simultaneous determination of green tea catechins using ultra‐performance liquid chromatography/electrospray ionization–mass spectrometry was developed. Total analytical run time was 3.5 min for the detection of (?)‐epicatechin (EC), (?)‐epicatechin‐3‐O‐gallate (ECG), (?)‐epigallocatechin (EGC), (?)‐epigallocatechin‐3‐O‐gallate (EGCG) and myricetin as the internal standard (IS) in rat plasma. The calibration curves were linear over the range of 10–5000 ng/mL for all the catechins. The inter‐ and intra‐day precision (relative standard deviation) and accuracy (percentage deviation) of the method were both lower than 10%. The average extraction recoveries in plasma ranged from 68.5 to 86.5%, and the lower limits of quantification of EC, EGC, ECG and EGCG were 10 ng/mL with a signal‐to‐noise ratio of >10. The assay developed was successfully applied to a pharmacokinetic study of catechins following intravenous and intragastric administrations of green tea extract in rats. Plasma concentrations of four catechins were detected up to 5–24 h after administration, and the pharmacokinetic parameters of catechins were in agreement with previous studies. From these findings, taken together with the high productivity and precision, the developed method could be a reliable and reproducible tool for the evaluation of pharmacokinetic properties of catechins. A micellar electrokinetic capillary chromatography (MEKC) method, using UV detection, was developed for the determination of polyphenols in Toona sinensis (Meliaceae); the procedure involved precipitation of polyphenols from the leaves of T. sinensis using methanol. The structures can be established with fifteen compounds including methyl gallate, gallic acid, kaempferol, quercitin, quercitrin, rutin, kaempferol‐glucoside, catechin, epicatechin, stearic acid, palmitic acid, β‐sitosterol, stigmasterol, β‐sitosteryl‐glucoside, and stigmasteryl‐glucoside by spectroscopic analysis. However, there has been no investigation to quantitate the polyphenols that form T. sinensis. Thus, seven polyphenols of T. sinensis with UV absorbance, catechin (C), epicatechin (EC), methyl gallate (MG), rutin (R), gallic acid (G), quercitrin (Q), and kaempferol (K) were separated within 10 min with a 40 cm uncoated fused‐silica column, with the RSD < 3% (migration times), voltage at 15 kV using this method. On‐column detection was carried out at 254 nm. The detection limit of this method for all analytes ranged from 19.5 to 0.02 μM (RSD < 3.1%). The method provided a rapid and sensitive identification of polyphenols of interest in T. sinensis and is suitable for biological activity studies.  相似文献

Polyphenols occur naturally in a vast variety of plants. One of their predominant properties is their antioxidant activity. To provide a deeper understanding of the antioxidant mechanism, 1H CIDNP spectroscopy (CIDNP=chemically induced dynamic nuclear polarization) is used to study model hydrogen abstraction reactions with four catechin‐based polyphenols: catechin (CA), gallocatechin (GC), epigallocatechin (EGC), and epigallocatechin gallate (EGCG). The experiments involve photoinduced hydrogen‐atom transfer to a hydrogen abstractor (e.g., excited isopropylthioxanthone) followed under steady‐state conditions and in a time‐resolved fashion (resolution 500 ns–1 ms). It is found that hydrogen abstraction is an essentially stochastic process with a slight preference for the B rings in the catechin‐based polyphenols. Remarkably, analogous reactivity patterns could be followed in the “real systems”, green tea and red wine. We also show that CIDNP can be used as a semiquantitative tool to assess chemical reactivity.  相似文献

An emerging green solvent called a deep eutectic solvent(DES) was applied to the extraction and determination of catechin(C),(+)epicatechin gailate(ECG) and (-)epigallocatechin gallate(EGCG) from Chinese green tea.After evaluating different combinations of them by extraction methods and DESs,a DES-based extraction method was established and optimized by a systematic investigation of the influencing factors.As a result,a total of 3.629,35.25 or 114.2 mg/g catechin,(+)epicatechin gallate or (-)epigaliocatechin gallate were extracted respectively under optimal conditions with extraction efficiencies of 82.7%,92.3% and 97.0%,respectively.By comparing with other common used solvents for extracting catechin compounds,DESs were proved to be potential extraction solvents for bioactive ingredients.  相似文献

Summary A simple, rapid and reproducible method is presented for the analysis of green-tea extracts in different cosmetic formulations and in in-vitro skin extracts. Cosmetically active principal components were used for determination of complex assembled green-tea extracts. The catechins selected were catechin, epicatechin, epigallocatechin, epigallocatechin gallate, and epicatechin gallate, because of their efficacy and their concentrations in green-tea extracts. The determination was performed by high-performance liquid chromatography (HPLC), on a reversed-phase (RP) column, coupled to a single-quadrupole mass spectrometer (MS) via an electrospray ionization (ESI) interface. Detection was performed in the negative selected-ion-monitoring (SIM) mode. A detection limit between 5 and 15 ng g−1 was achieved in methanol-water-ascorbic acid extracts from different emulsions. A routine analytical procedure could be established with good quantitative reliability. During validation, the repeatabilities (relative standard deviation) for catechin standard solutions were found to be 1.1–2.7% (within one day) and 2.2–4.3% (day-to-day). Recoveries from spiked placebos were 98–105%. The method was successfully used to determine the storage stability of green-tea extract in cosmetic formulations and the in-vitro penetration of green-tea extract into the skin.  相似文献

Novel magnetic molecularly imprinted polymers (MMIPs) with multiple‐template based on silica were modified by four types of deep eutectic solvents (DESs) for the rapid simultaneous magnetic solid‐phase extraction (MSPE) of tanshinone Ⅰ, tanshinone ⅡA, and cryptotanshinone from Salvia miltiorrhiza bunge; glycitein, genistein, and daidzein from Glycine max (Linn.) Merr; and epicatechin, epigallocatechin gallate, and epicatechin gallate from green tea, respectively. The synthesized materials were characterized by Fourier transform infrared spectroscopy and field emission scanning electron microscopy. Single factor experiments were to explore the relationship between the extraction efficiency and four factors (the sample solution pH, amount of DESs for modification, amount of adsorbent, and extraction time). It was showed that the DES4‐MMIPs have better extraction ability than the MMIPs without DESs and the other three DESs‐modified MMIPs. The best extraction recoveries with DES4‐MMIP were tanshinone Ⅰ (85.57%), tanshinone ⅡA (80.58%), cryptotanshinone (92.12%), glycitein (81.65%), genistein (87.72%), daidzein (92.24%), epicatechin (86.43%), epigallocatechin gallate (80.92%), and epicatechin gallate (93.64%), respectively. The novel multiple‐template MMIPs materials modified by DES for the rapid simultaneous MSPE of active compounds were proved to reduce the experimental steps than single‐template technique, and increase the extraction efficiency.  相似文献

A high-performance liquid chromatographic method with electrochemical detection was developed for the determination of twelve tea catechins including four major catechins: epicatechin (EC), epigallocatechin (EGC), epicatechin gallate (ECG) and epigallocatechin gallate (EGCG); four of their epimers at the C-2 position, C, GC, CG and GCG; and four methylated catechin derivatives, epigallocatechin-3-O-(3-O-methyl)gallate, gallocatechin-3-O-(3-O-methyl)gallate, epigallocatechin-3-O-(4-O-methyl)gallate and epicatechin-3-O-(3-O-methyl)gallate. These catechins were separated on an ODS C18 reversed-phase column by isocratic elution with 0.1 M NaH2PO4 buffer (pH 2.5)-acetonitrile (87:13) containing 0.1 mM EDTA.2Na. The detection limits (S/N = 3) of these catechins were approximately 10-40 pmol ml-1 at an applied voltage of 600 mV. Extracting these catechins from tea leaf powder with H2O-acetonitrile (1:1) at 30 degrees C for 40 min inhibited the epimerization at C-2 significantly from these epicatechins compared to extraction with hot water at 90 degrees C. This analytical method is sensitive to and appropriate for the simultaneous determination of various biologically active catechins in green tea.  相似文献
