采用反相悬浮交联法制备壳聚糖微球,对微球进行羟丙基氯化及氨基化,并偶联色素配体Cibacron Blue F3GA,得到一种新型染料亲和吸附剂.以牛血清白蛋白(BSA)为目标蛋白,考察了该染料亲和吸附剂的吸附性能,发现其对BSA有较高的吸附量(95.2mg/g),吸附行为满足Langmuir吸附等温式.负载牛血清白蛋白的微球容易洗脱,洗脱率高达99%.  相似文献

通过分子设计和过程策划,制备了高性能的牛血清白蛋白(BSA)分子表面印迹材料.首先以甲基丙烯酰氯为试剂,使交联聚乙烯醇(CPVA)微球表面的羟基发生酯化反应,将大量可聚合双键引入到CPVA微球表面.然后以含有可聚合双键的CPVA微球为载体,阳离子单体丙烯酰氧乙基三甲基氯化铵(DAC)为功能单体,N,N’-亚甲基双丙烯酰胺(MBA)为交联剂,牛血清白蛋白(BSA)为模板分子,在水溶液体系中,基于主-客体之间的强静电相互作用,采用接枝聚合与印迹过程同步进行的方式,制备了高性能BSA分子表面印迹微球MIP-PDAC/CPVA.采用红外光谱(IR)和扫描电子显微镜(SEM)对产物微球进行了表征.研究了印迹聚合物微球MIP-PDAC/CPVA对BSA的大分子识别特性.研究结果表明,微球MIP-PDAC/CPVA对BSA具有优良的结合亲和性和特异的识别选择性,结合容量高达108 mg/g,对牛血红蛋白(BHb)却基本不结合;相对于BHb,MIP-PDAC/CPVA对BSA的选择性系数高达60.2.  相似文献

由大分子单体法制得了聚醋酸乙烯酯接枝聚苯乙烯(PVAc-g-PSt)微球,使该微球在碱性条件下醇解形成了聚乙烯醇接枝聚苯乙烯(PVA-g-PSt)微球.经X射线光电子能谱对PVA-g-PSt微球表层组成的表征,发现微球具有以PVA为壳、PSt为核的核壳结构.进而利用汽巴蓝F3GA(CB)与PVA-g-PSt微球进行亲核反应,制得了CB功能化的PVA-g-PSt微球,由元素分析定量出了固定在微球表面的CB含量.用透射电子显微镜对微球的粒径与形态进行了表征,发现微球表面在CB功能化后变得相对粗糙,粒径大小基本保持不变,但形态更加规整.比较了3种不同微球对蛋白质的吸附,考察了吸附动力学和影响吸附的因素,发现起始牛血清蛋白(BSA)浓度、pH值、离子强度对微球吸附BSA有明显的影响,并利用Zeta电位探讨了微球与蛋白质间的相互作用机理.最后用硫氰酸钠(NaSCN)进行解吸,计算解吸率最高可达到95.7%,该CB功能化微球可以重复利用,吸附率仅减少5.3%.  相似文献

合成了具有较高荧光量子产率(0.69)和良好光稳定性的可聚合荧光染料单体,该荧光染料的光稳定性高于商品化的染料罗丹明B。 通过氧化还原引发剂引发乳液聚合制备了超细荧光聚合物纳米微球,将染料分子共价连接在聚合物链上。 使用非离子表面活性剂Triton X-100作为乳化剂、甲基丙烯酸甲酯(MMA)作为单体和助乳化剂,制备的超细纳米微球平均粒径为22 nm,而不加MMA时制备出的纳米微球平均粒径在150 nm左右。 由于微球表面带有苄氯基团,为进一步的微球功能化提供了途径。  相似文献

本文制备了聚合物基质弱酸型阳离子交换固相萃取填料,以甲基丙烯酸和苯乙烯为原料,二乙烯基苯为交联剂,聚乙烯醇为分散剂,过氧化苯甲酰为引发剂,采用悬浮聚合法制备了具有亲脂和弱阳离子交换性能的球形固相萃取填料,并对其结构和形貌进行了表征。以该聚合物微球作为填料制备固相萃取小柱,萃取水中联硝氯酚、硫双二氯酚、吡喹酮和丙硫苯咪唑4种杀虫剂,乙腈为洗脱剂,洗脱液采用液相色谱分析。分别考察了样品的p H值和流速、洗脱剂的体积和流速对萃取回收率的影响,确定了最佳固相萃取条件。选择了最佳色谱分析条件。结果表明,该聚合物微球单分散性好,自制固相萃取小柱对水中4种杀虫剂的吸附性能好,与HPLC联用测定结果重现性好,联硝氯酚、硫双二氯酚、吡喹酮和丙硫苯咪唑的最低检测限分别为0.26μg·L-1、0.31μg·L-1、0.42和0.63μg·L-1。  相似文献

在离子液体均相体系中合成了一种新型两亲性窄分子量分布的低聚壳聚糖衍生物月桂基-琥珀酰化壳聚糖(LSCOS). 以LSCOS为载体材料, 以牛血清蛋白(BSA)为模板蛋白, 以戊二醛为交联剂, 用油包水(W/O)乳化交联法制备了包载BSA的BSA/LSCOS缓释载药微球. 通过扫描电子显微镜(SEM)、 透射电子显微镜(TEM)及紫外-可见光谱(UV-Vis)研究了BSA/LSCOS比率和戊二醛/LSCOS比率对微球的形貌结构、 包埋率、 载药率和体外药物释放特性的影响. 结果表明, 在离子液体中合成的LSCOS包覆了BSA, 形成的微球粒径约为1 μm, 微球表面随BSA用量的增加变得光滑, 随戊二醛用量的增加变得粗糙. BSA的累积释放率与BSA包载量成正比, 与交联剂添加量成反比, 因此, 可通过控制蛋白质药物的添加比率和交联剂用量来控制蛋白质药物体外释放率.  相似文献

用改良的乳化冷凝法制备载牛血清蛋白(BSA)的大粒径明胶微球. 结果表明, 明胶水溶液的质量分数为25%、水相与油相体积比3∶20、搅拌速度300 r/min、交联剂用0.1 mL质量分数为25%的戊二醛、 表面活性剂用0.1 g span-80为制备平均直径约250 μm明胶微球的理想条件. 所制备微球的后处理方法不同, 则明胶微球的表面形貌也不同, 细胞粘附率不同. 空白明胶微球在体外可以完全降解, 载BSA的明胶微球对BSA具有良好的缓释性, 释放时间可长达30 d. 显微镜观察成纤维细胞在明胶微载体上生长良好.  相似文献

将水凝胶膜样品置于牛血清蛋白(BSA)的水溶液中,沉积一定时间后,采用染料染色结合分光光度法分析沉积前后BSA浓度的变化,计算沉积量,并采用洗脱法测试BSA与水凝胶材料的结合程度.结果表明,BSA的沉积过程为不可逆过程;BSA的沉积作用不受共聚物水凝胶材料中NVP(N-乙烯基吡咯烷酮)结构单元吸附作用的控制;BSA与水凝胶材料松散结合,很容易洗脱;沉积在水凝胶膜表面上的BSA对后续沉积过程无影响.  相似文献

本研究发展了一种采用磁性介孔微球Fe3O4@nSiO2@mSiO2测定溶液中多环芳烃(PAHs)和牛血清蛋白(BSA)结合常数(KBSA)的方法.在合成Fe3O4@nSiO2@mSiO2微球时,将少量的十六烷基三甲基铵阳离子(CTAM+)模板保留在介孔内作为萃取水体中自由溶解态PAHs的吸附剂,而与溶液中的BSA及其结合的PAHs则由于体积排阻作用不被萃取.微球外层很薄的介孔层(≈50 nm)显著缩短了水体中PAHs向介孔内CTAM+迁移的路径,而微球较大的表面积(186m2/g)和孔体积(0.16 cm3/g)增大了微球在水体中与PAHs的接触面积,使其能够快速富集水体中的自由溶解态PAHs,萃取平衡时间(t90%)仅为66 min.实验测定了不同BSA浓度条件下PAHs自由溶解态分数,并据此计算得到PAHs与BSA的结合常数lgKBSA=3.94~5.00,与微耗损固相微萃取方法测得的结果基本一致,而平衡时间仅为后者的1/200,显著地提高了工作效率.  相似文献

用溶胶-凝胶法以磷钼酸(MPA)的镍盐溶液水解钛酸四丁酯制备了NiPMo/TiO2催化剂.使用ICP、 XRD、 TG-DTA、 IR、 TPD-MS和微反应技术研究了催化剂的化学组成、热稳定性、化学吸附性质和催化反应性能.杂多钼酸盐与TiO2通过O2-在TiO2表面发生了键合.在623 K下,杂多阴离子仍保持原有的Keggin结构.CO2在Lewis酸位Ni(Ⅱ)和Lewis碱位Ni-O-Mo的桥氧协同作用下生成CO2卧式吸附态Ni(Ⅱ)←O-(CO)←(O--Ni).丙烯有多种吸附态在催化剂上吸附.在563 K、 1 MPa和空速1500 h-1的反应条件下,丙烯的摩尔转化率为3.2%,产物MAA选择性为95%.  相似文献

A new and simple synthesis of novel N-protected methyl 5-substituted-4-hydroxypyrrole-3-carboxylates, which exist in equilibrium with their 4-oxo tautomers, has been developed in two steps starting from N-protected α-amino acids. The key intermediates are enaminones, which can also be isolated, characterized, and used for the construction of other functionalized heterocycles, before they spontaneously decompose to pyrrole products. 4-Hydroxypyrroles are prone to partial aerial oxidation but can be efficiently alkylated or reduced to stable polysubstituted pyrrolidine derivatives.  相似文献

The chemoselectivity in the intramolecular CH insertion of various diazosulfonamides has been experimentally studied. The results reveal that the aliphatic 1,4-, 1,5-, or 1,6-C(sp3)?H insertions of diazosulfonamides are not accessible, while the aromatic 1,5-C(sp2)?H insertion can be realized specifically by adjusting the diazo-adjacent group. In addition, the general chemoselectivities in the intramolecular CH insertions of diazosulfonyl compounds are summarized. Generally, diazosulfones undergo both aromatic 1,5-C(sp2)?H and aliphatic 1,5- and 1,6-C(sp3)?H insertions, while diazosulfonates undergo aliphatic 1,5- and 1,6-C(sp3)?H insertions. However, diazosulfonamides only undergo aromatic 1,5-C(sp2)?H insertion.  相似文献

An efficient iodine-mediated oxidative Pictet-Spengler reaction in dimethyl sulphoxide (DMSO) using terminal alkynes as the 2-oxoaldehyde surrogate for the synthesis of aryl (9H-pyrido[3,4-b]indol-1-yl)methanones is described. The scope of the protocol includes the total synthesis of Fascaplysin, Eudistomins Y1 and Y2. The methodology is extended for preparing pyrrolo[1,2-a]-quinoxaline and indolo[1,5-a]quinoxaline derivatives. The utility of 1-aroyl-β-carbolines was demonstrated by performing palladium-catalyzed β-carboline directed ortho-C(sp2)-H functionalization of the phenyl ring with thiomethyl (SMe) group using DMSO as source and for accessing 4-aryl-canthin-6-ones.  相似文献

In this Letter, we described a facile method for constructing fused bicyclic 1-arylpyrazol-5-one ring system. We employed various methylene-containing carboxylic acids as the substrates and proved that the pyrazolone ring closure requires activated methylene group in intermediate II. Accordingly, a series of structurally diversified, fused bicyclic 1-arylpyrazol-5-ones was prepared in moderate to high yields using the requisite substrates.  相似文献

The Langevin paramagnetic theory can’t describe the relation between magnetization of ferrofluids and applied magnetic field. The structuralization of ferrofluids, which is considered the main influence factor of the magnetization, is regarded. The part of magnetization works is deposited when the structure is forming. This action influences the magnetization of ferrofluids directly or indirectly. On the base of the “compressing” model, the Langevin function that usually describes the magnetization of ferrofluid is modified, and a well-fitted curve is obtained. An equation of the relation between the equivalent volume fraction after being “compressed” and the intensity of magnetic field is discovered, which approximately describes the process of magnetization. The relation between the approximate initial susceptibility and the volume fraction can be obtained from modified formula.  相似文献

KMnO4-mediated oxidative CN bond cleavage of tertiary amines producing secondary amine was introduced, which was trapped by electrophiles (acyl chloride and sulfonyl chloride) to form amides and sulfonamides. The highly regioselective Buchwald–Hartwig amination at C-2 of the cheap and readily accessible reagent, 2,4-dichloropyridine with a range of anilines and heterocyclic amines is described. This new methodology is robust and provides a facile access to 4-chloro-N-phenylpyridin-2-amines on 0.25 mol scale. These intermediates undergo a further Buchwald–Hartwig amination at higher temperature to enable rapid exploration of the chemical space at C-4 and to provide a library of 2,4-bisaminopyridines.  相似文献
