苯基锂、2-甲氧基苯基锂和2,4-二甲氧基苯基锂分别与肉桂酸反应以相当好的收率生成相应的查尔酮;2,6-二甲氧基苯基锂与肉桂酸不发生反应,但能与肉桂酸甲酯反应生成2',6'-二甲氧基查尔酮;类似的还制得了2',4',6'-三甲氧基查尔酮和2',4',4,6'-四甲氧基查尔酮.2,4-二甲氧基苯基锂和2,4,6-三甲氧基苯基锂也能与γ-丁酸内酯反应分别生成γ-羟丙基-(2,4-二甲氧基苯基)酮和γ-羟丙基-(2,4,6-三甲氧基苯基)酮.没发现三级醇生成.  相似文献

以生物质来源的对甲氧基苯甲醛(大茴香醛)为原料,经还原反应制备得到对甲氧基苄醇,经氯代反应制备得到对甲氧基氯苄,再与乙酰乙酸乙酯经取代反应制备得到2-乙酰基-3-(4-甲氧基苯基)丙酸乙酯,再经串联的水解、脱羧反应制备得到4-(4-甲氧基苯基)-2-丁酮,最后经脱甲基反应制备得到4-对羟基苯基-2-丁酮,总收率为60.7%。具有路线简捷、易于操作、环境友好、收率高等优点。  相似文献

基于Perkin反应策略合成了具有强效抗肿瘤、抗血管活性的天然产物Combretastatin A-1(CA1)和Combretastatin B-1(CB1).以2,3,4-三羟基苯甲醛(1)为起始物, 经单甲基化反应得到2,3-二羟基-4-甲氧基苯甲醛(2), 再经酚羟基保护得到2,3-二异丙基-4-甲氧基苯甲醛(3), 该化合物与3,4,5-三甲氧基苯乙酸(4)发生Perkin反应分离得到E-2-(3,4,5-三甲氧基苯基)-3-(2',3'-二异丙氧基-4'-甲氧基)丙烯酸(E-5), 经脱羧反应得到Z-3,4,4',5-四甲氧基-2',3'-二异丙氧基二苯乙烯(6), 最后经脱保护反应得到CA1.另外, 将E-2-(3,4,5-三甲氧基苯基)-3-(2',3'-二异丙氧基-4'-甲氧基)丙烯酸(E-5)脱去保护基得到E-2-(3,4,5-三甲氧基苯基)-3-(2',3'-二羟基-4'-甲氧基)丙烯酸(7), 该化合物经脱羧-异构化反应得到E-3,4,4',5-四甲氧基-2',3'-二羟基二苯乙烯(E-CA1), 最后经催化氢化得到CB1.  相似文献

通过EPR技术观察到一系列典型的烷基或甲氧基取代的苯基烷基酮ArCOR(Ar=2,5-二甲基苯基(1);2,5-二甲氧基苯基(2);4-叔丁基-2,5-二甲氧基苯基(3))与t-BuMgCl反应中形成的自由基阴离子中间体[ArC(R)OMgCl]。产物分析表明,反应主要生成还原产物[ArCH(R)OH]和1,2-加成产物[ArC(R)(t-Bu)OH]。降低溶剂极性及反应温度都有利于还原产物的生成。对上述反应的可能历程作了简单讨论。  相似文献

以(S)-2-氨甲基-1-乙基吡咯烷为原料,合成了新型手性双功能有机催化剂(S)-1-[3,5-二(三氟甲基)苯基]-3-(1-乙基吡咯烷基-2-甲基)硫脲(2,产率82.2%).用2催化对甲氧基苯甲醛与硝基甲烷的不对称Henry反应,制得(R)-1-(4-甲氧基苯基)-2-硝基乙醇(3, 84%ee);3经Pd/C催化氢化得到(R)-2-氨基-1-(4-甲氧基苯基)乙醇(4);4与反式肉桂酰氯反应合成了(R)-aegeline,总产率37.6%(以对甲氧基苯甲醛计).化合物的结构经1H NMR, 13C NMR和MS表征.  相似文献

2-苯基吲哚 (1a) 在甲醇中的染料敏化光氧化反应给出2-苯基-2-(2'-苯基-3'-吲哚基)二氢吲哚-3-酮 (2a) 和2-甲氧基-2-苯基二氢吲哚-3-酮 (4a), 相应N-甲基取代产物由1-甲基-2-苯基吲哚 (1b) 的类似反应获得。发现反应产物分布随吲哚 (1) 的浓度和介质酸度的变化而变化。对反应机理进行了推测, 其中当1a的反应在乙腈中进行时, 分离到了相应的反应中间体: 2-苯基-3H-吲哚-3-酮 (3a)。  相似文献

为了研究吲哚衍生物的生理活性与结构之间的关系, 我们采用 Fischer反应取代苯乙酮的苯腙与多聚磷酸(PPA)一起加热, 合成了一系列2-芳基吲哚。我们发现: 当取代苯乙酮分子中的芳基分别为2,3,4-三甲氧基苯基, 3,4,5-三甲氧基苯基或4,6-二甲基-2-甲氧基苯基时, 可以顺利地得到相应的2-芳基吲哚, 当芳基为2,4,6-三-甲氧基苯基时, 主要产物为4,6-二甲氧基-3-甲基-1-苯基吲唑。本文讨论了反应机理。  相似文献

以2-萘酚为起始原料,经过氧化偶联,消旋体的拆分得到手性骨架2-氨基-2'-羟基-1,1'-联萘(NOBIN),并以S-NOBIN为原料,经过六步反应合成了两个新型配体S-(+)-2-(2-吡啶酰胺基)-2'-二苯基膦基-1,1'-联萘(1a)和S-(+)-2-(6-甲基-2-吡啶酰胺基)-2'-二苯基膦基-1,1'-联萘(1b)。并进行了铜配合物催化的二乙基锌对2-环己烯酮的1,4-共轭加成反应的研究。反应产物3-乙基环己酮(13)的e.e.值高达92%。  相似文献

[reaction: see text] Lipase-catalyzed resolution of (2R*,3S*)-3-methyl-3-phenyl-2-aziridinemethanol, (+/-)-2, at low temperatures gave synthetically useful (2R,3S)-2 and its acetate (2S,3R)-2a with (2S)-selectivity (E = 55 at -40 degrees C), while a similar reaction of (2R*,3R*)-3-methyl-3-phenyl-2-aziridinemethanol, (+/-)-3, gave (2S,3S)-3 and its acetate (2R,3R)-3a with (2R)-selectivity (E = 73 at -20 degrees C). Compound (+/-)-2 was prepared conveniently via diastereoselective addition of MeMgBr to tert-butyl 3-phenyl-2H-azirine-2-carboxylate, (+/-)-1a, which was successfully prepared by the Neber reaction of oxime tosylate of tert-butyl benzoyl acetate 7a. The tert-butyl ester was requisite to promote this reaction. For determination of the absolute configuration of (2S,3R)-2a, enantiopure (2S,3R)-2 was independently prepared in three steps involving diastereoselective methylation of 3-phenyl-2H-azirine-2-methanol, (S)-10, with MeMgBr. The absolute configuration of (2S,3S)-3 was determined by X-ray analysis of the corresponding N-(S)-2-(6-methoxy-2-naphthyl)propanoyl derivative (S,S,S)-13.  相似文献

A novel chemiluminescence (CL) system was evaluated for the determination of hydrogen peroxide, glucose and ascorbic acid based on hydrogen peroxide, which has a catalytic-cooxidative effect on the oxidation of luminol by KIO(4). Hydrogen peroxide can be directly determined by luminol-KIO(4)-H(2)O(2) CL system. The detection limit was 3.0x10(-8) mol l(-1) and the calibration graph was linear over the range of 2.0x10(-7)-6.0x10(-4) mol l(-1). The relative standard deviation of H(2)O(2) was 1.1% for 2.0x10(-6) mol l(-1) (N=11). Glucose was indirectly determined through measuring the H(2)O(2) generated by the oxidation of glucose in the presence of glucose oxidase at pH 7.6. The present method provides a source for H(2)O(2), which, in turn, coupled with the luminol-KIO(4)-H(2)O(2) CL reaction system. The CL was linearly correlated with glucose concentration of 0.6-110 mug ml(-1). The relative standard deviation was 2.1% for 10 mug ml(-1) (N=11). Detection limit of glucose was 0.08 mug ml(-1). Ascorbic acid was also indirectly determined by the suppression of luminol-KIO(4)-H(2)O(2) CL system. The calibration curve was linear over the range of 1.0x10(-7)-1.0x10(-5) mol l(-1) of ascorbic acid. The relative standard deviation was 1.0% for 8.0x10(-7) mol l(-1) (N=11). Detection limit of ascorbic acid was 6.0x10(-8) mol l(-1). These proposed methods have been applied to determine glucose, ascorbic acid in tablets and injection.  相似文献

4-(2-Chlorophenyl)-3-cyano-6-hydroxy-5-(2-thienoyl)-6-trifluoromethylpiperidin-2-thione, which was used for the synthesis of 2-allylthio-4-(2-chlorophenyl)-3-cyano-6-hydroxy-5-(2-thienoyl)-6-trifluoromethyl-1,4,5,6-tetrahydropyridine, was obtained by the reaction of 2-thienoyltrifluoroacetone with 2-chlorophenylphenylmethylenecyanothioacetamide or with a mixture of 2-chlorobenzaldehyde and cyanothioacetamide in the presence of N-methylmorpholine. The molecular and crystal structure of the piperidinthione have been established by X-ray crystallography.  相似文献

The enantiomers of 5-hydroxy-3-(4-methoxyphenyl)-2-(3,4,5-trimethoxyphenyl)-cyclopent-2-en-1-one, a novel anticancer agent, were separated by derivatisation with caronaldehyde, separation of the resulting diastereoisomers of the corresponding esters by silica gel column chromatography and regeneration of alcohols (S)-5-hydroxy-3-(4-methoxyphenyl)-2-(3,4,5-trimethoxyphenyl)-cyclopent-2-en-1-one and (R)-5-hydroxy-3-(4-methoxyphenyl)-2-(3,4,5-trimethoxyphenyl)-cyclopent-2-en-1-one under aqueous conditions. The absolute configuration of the enantiomers was determined by 1H NMR studies of the corresponding Mosher esters. Alternatively, the enantiomers were separated by preparative HPLC to collect the (S)- and (R)-5-hydroxy-3-(4-methoxyphenyl)-2-(3,4,5-trimethoxyphenyl)-cyclopent-2-en-1-ones with high purity which was comparable with that obtained by the chemical method. We describe the practical synthetic route for (2S)-7-methoxy-1,2,3,4-tetrahydro-2-naphthylamine 1(2S)-2-amino-7-methoxytetraline; (S)-AMT]. (2R)-2-(3-Methoxybenzyl)succinic acid [(R)-1] was obtained by the optical resolution of 2-(3-methoxybenzyl)succinic acid (1) as the salt of (1R,2S)-2-(benzylamino)cyclohexylmethanol (7), and (R)-1 was converted to the optically active (2S)-7-methoxy-1,2,3,4-tetrahydro-2-naphthoic acid [(S)-2] by the intramolecular Friedel-Crafts reaction followed by catalytic hydrogenation. (S)-AMT was obtained from the acid (S)-2 by Hofmann rearrangement without racemization.  相似文献
