Dithiothreitol (DTT)-based displacement is widely utilized for separating ligands from their gold nanoparticle (AuNP) conjugates, a critical step for differentiating and quantifying surface-bound functional ligands and therefore the effective surface density of these species on nanoparticle-based therapeutics and other functional constructs. The underlying assumption is that DTT is smaller and much more reactive toward gold compared with most ligands of interest, and as a result will reactively displace the ligands from surface sites thereby enabling their quantification. In this study, we use complementary dimensional and spectroscopic methods to characterize the efficiency of DTT displacement. Thiolated methoxypolyethylene glycol (SH-PEG) and bovine serum albumin (BSA) were chosen as representative ligands. Results clearly show that (1) DTT does not completely displace bound SH-PEG or BSA from AuNPs, and (2) the displacement efficiency is dependent on the binding affinity between the ligands and the AuNP surface. Additionally, the displacement efficiency for conjugated SH-PEG is moderately dependent on the molecular mass (yielding efficiencies ranging from 60 to 80?% measured by ATR-FTIR and ≈90?% by ES-DMA), indicating that the displacement efficiency for SH-PEG is predominantly determined by the S–Au bond. BSA is particularly difficult to displace with DTT (i.e., the displacement efficiency is nearly zero) when it is in the so-called normal form. The displacement efficiency for BSA improves to 80?% when it undergoes a conformational change to the expanded form through a process of pH change or treatment with a surfactant. An analysis of the three-component system (SH-PEG?+?BSA?+?AuNP) indicates that the presence of SH-PEG decreases the displacement efficiency for BSA, whereas the displacement efficiency for SH-PEG is less impacted by the presence of BSA.

A dimeric organophosphorus hydrolase (OPH; EC 3.1.8.1; 72 kDa) was isolated from wild-type bacteria, analyzed for its 16s rRNA sequence, purified, and immobilized on gold nanoparticles (AuNPs) to form the transducer part of a biosensor. The isolated strain was identified as Pseudomonas aeruginosa. The AuNPs were characterized by transmission electron microscopy and localized surface plasmon resonance. Covalent binding of OPH to the AuNPs was confirmed by spectrophotometry, enzymatic activity assays, and FTIR spectroscopy. Coumarin 1, a competitive inhibitor of OPH, was used as a fluorogenic probe. The bioconjugates quench the emission of coumarin 1 upon binding, but the addition of paraoxon results in an enhancement of fluorescence that is directly proportional to the concentration of paraoxon. The gold-OPH conjugates were then used to determine paraoxon in serum samples spiked with varying levels of paraoxon. The method works in the 50 to 1,050 nM concentration range, has a low standard deviation (with a CV of 5.7–11 %), and a detection limit as low as 5?×?10?11 M.

We report the development of a hyphenated instrument with the capacity to quantitatively characterize aqueous suspended gold nanoparticles (AuNPs) based on a combination of gas-phase size separation, particle counting, and elemental analysis. A customized electrospray-differential mobility analyzer (ES-DMA) was used to achieve real-time upstream size discrimination. A condensation particle counter and inductively coupled plasma mass spectrometer (ICP-MS) were employed as downstream detectors, providing information on number density and elemental composition, respectively, of aerosolized AuNPs versus the upstream size selected by ES-DMA. A gas-exchange device was designed and optimized to improve the conversion of air flow (from the electrospray) to argon flow required to sustain the ICP-MS plasma, the key compatibility issue for instrumental hyphenation. Our work provides the proof of concept and a working prototype for utilizing this construct to successfully measure (1) number- and mass-based distributions; (2) elemental compositions of nanoparticles classified by size, where the size classification and elemental analysis are performed within a single experiment; (3) particle concentrations in both solution (before size discrimination) and aerosol (after size discrimination) phases; and (4) the number of atoms per nanoparticle or the nanoparticle density.  相似文献

Electrochemical detection of nitrite was achieved via electrodeposition of gold nanoparticles (AuNPs) onto glassy carbon electrodes, followed by 3‐mercaptopropionic acid (MPA) self‐assembly, enabling attachment of an iron(III) monoamino‐phthalocyanine (FeMAPc) catalyst via amide bond formation. The use of scanning electron microscopy, energy dispersive X‐ray spectroscopy and ultraviolet‐visible spectroscopy realised surface characterisation while cyclic voltammetry and electrochemical impedance spectroscopy techniques were applied for electrochemical interrogation. The electrochemical behaviour of nitrite at the bare (GCE), AuNPs/GCE, FeMAPc/GCE and FeMAPc‐MPA/AuNPs/GCE was further scrutinised using differential pulse voltammetry in phosphate buffer solution (0.1 M PBS, pH 5.8). Overall the FeMAPc‐MPA/AuNPs/GCE resulted in sensitivity 14.5 nA/µM, which was double that of AuNPs/GCE, 2.4 times FeMAPc/GCE and 3.5 times the response at a bare GCE, with linear range 1.9 µM–2.04 mM (PBS, pH 5.8) and LOD 0.21 µM. An interference study revealed that the proposed sensor (FeMAPc‐MPA/AuNPs/GCE) exhibited a selective response in the presence of interfering anions and the analytical capability of the sensor was demonstrated via nitrite ion determination in real water samples.  相似文献

Expanded bed adsorption of bovine serum albumin (BSA) directly from a feedstock containing whole yeast cells has been investigated with an anion-exchanger DEAE Spherodex M. In the presence of 6% (w/w) yeast cells, the axial liquid-phase dispersion coefficient was found in the order of 10(-6) m2/s, which felled into the common range of 1.0 x 10(-6)-1.0 x 10(-5) m2/s observed previously without the use of cell suspensions as mobile phase. We found that the static and dynamic binding capacity of BSA decreased with increasing the yeast cell concentration due to the competitive adsorption of cells onto the outer surface of the anion-exchanger. However, because of the small size of the adsorbent, the large pore diffusivity of protein and the favorable column efficiency (low axial dispersion coefficient), the dynamic binding capacity of BSA in the presence of 6% (w/w) cells in the expanded bed reached 86% that of the equilibrium adsorption density. Then, the whole expanded bed adsorption process of BSA in the presence of cells, including feedstock loading, washing and elution steps, was predicted using a mathematical model with parameters all determined independently. In the elution stage, the steric mass-action adsorption isotherm with salt concentration as one of the model parameters was used to predict the step-gradient elution process with salt concentration increases. Computer simulations showed that the model was in good agreement with the experimental results for the whole operation process.  相似文献

A new colorimetric sensor for sensing Hg2+ in a high-salinity solution has been developed using gold nanoparticles (AuNPs) decorated with 3-mercaptopropionate acid (MPA) and adenosine monophosphate (AMP). Because of the high negative charge density of AMP on each AuNP surface, MPA/AMP-capped AuNPs are well dispersed in a high-salt solution. In contrast, the aggregation of MPA-capped AuNPs was induced by sodium ions, which shield the negative charges of the carboxylic groups of MPA. Through the coordination between the carboxylic group of MPA and Hg2+, the selectivity of MPA/AMP-capped AuNPs for Hg2+ in a high-salt solution is remarkably high over that of the other metals without the addition of a masking agent or a change in the temperature. We have carefully investigated the effect of the AMP concentration on the stability and sensitivity of MPA/AMP-capped AuNPs. Under optimum conditions, the lowest detectable concentration of Hg2+ using this probe was 500 nM on the basis of the measurement of the ratio of absorption at 620 nm to that at 520 nm. The sensitivity to Hg2+ can be further improved by modifying the MPA/AMP-capped AuNPs with highly fluorescent rhodamine 6G (R6G). By monitoring the fluorescence enhancement, the lowest detectable concentration of Hg2+ using R6G/MPA/AMP-capped AuNPs was 50 nM.  相似文献

An immunosensor for determination of salbutamol was developed. It based on glass carbon electrode (GCE) modified with a conductive multilayer film comprised of multi-wall carbon nanotubes, polythionine and gold nanoparticles. Salbutamol antibody was immobilized on the surface of the modified GCE which then was blocked with bovine serum albumin (BSA). The stepwise self-assembly process of the immunosensor was studied by cyclic voltammetry. The detection scheme is based on competitive binding of salbutamol to the sensor surface whose differential pulse voltammetric signal decreases after competitive binding of the salbutamol-BSA conjugate and free salbutamol to the salbutamol antibody. The sensor responds to salbutamol in 5 to 150 nM concentration range, with a detection limit of 1 nM. This method was applied to the precise and sensitive determination of salbutamol in spiked feed samples.

In this work, we constructed a salbutamol immunosensor which was based on salbutamol-Ab adsorbed on the AuNPs/PTH/MWCNTs/GCE. Just as the procedures shown in Graph 1, competitive immunoreaction was the experimental principle. We quantify the adsorption and desorption of a monoclonal immunoglobulin-G antibody, rituxamab (RmAb), on silica capillary surfaces using electrospray-differential mobility analysis (ES-DMA). We first develop a theory to calculate coverages and desorption rate constants from the ES-DMA data for proteins adsorbing on glass capillaries used to electrospray protein solutions. This model is then used to study the adsorption of RmAb on a bare silica capillary surface. A concentration-independent coverage of ≈4.0 mg/m(2) is found for RmAb concentrations ranging from 0.01 to 0.1 mg/mL. A study of RmAb adsorption to bare silica as a function of pH shows maximum adsorption at its isoelectric point (pI of pH 8.5) consistent with literature. The desorption rate constants are determined to be ≈10(-5) s(-1), consistent with previously reported values, thus suggesting that shear forces in the capillary may not have a considerable effect on desorption. We anticipate that this study will allow ES-DMA to be used as a "label-free" tool to study adsorption of oligomeric and multicomponent protein systems onto fused silica as well as other surface modifications.  相似文献

BSA adsorption onto negatively and positively charged polystyrene nanoparticles was investigated. The nanoparticles were characterized in terms of particle size, zeta potential, surface group density, and morphology. The adsorption behavior of BSA on the particle surface, as a function of pH and overall charge of the particle, was studied using ITC. Different thermodynamic data such as enthalpy changes upon binding and stoichiometry of the systems were determined and discussed. The degree of surface coverage with BSA was calculated using the thermodynamic data. The cellular uptake of particles before and after BSA adsorption was studied using HeLa cells in the presence and absence of supplemented FCS in the cell culture medium.  相似文献

An electrochemical sensor was developed based on gold nanoparticles incorporated in mesoporous MFI zeolite for the determination of purine bases. Au nanoparticles (AuNPs) were incorporated into the mesoporous MFI zeolite (AuNPs/m‐MFI) by post‐grafting reaction. The composite materials were characterized by transmission electron microscopy (TEM), X‐ray photoelectron spectroscopy (XPS) and electrochemical methods. Au nanoparticles with a size of 5‐20 nm are uniformly dispersed in the pores of mesoporous MFI zeolite. And the morphology of MFI zeolite can be perfectly kept after pore expansion and Au nanoparticles incorporation. The electrocatalytic oxidation of purine bases (guanine and adenine in DNA) is investigated by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). The surface‐confined Au nanoparticles provide the good catalytic activity for oxidation of purine bases. The simultaneous detection of guanine and adenine can be achieved at AuNPs/m‐MFI composites modified glassy carbon electrode (GCE). The electrochemical sensor based on AuNPs/m‐MFI exhibits wide linear range of 0.5–500 μM and 0.8–500 μM with detection limit of 0.25 and 0.29 μM for guanine and adenine, respectively. Moreover, the electrochemical sensor is applied to evaluation of guanine and adenine in herring sperm DNA samples with satisfactory results.  相似文献

This research deals with the potential of metallic nanoparticles in DNA/RNA stabilization. In this regard, the interaction of adenosine monophosphate (AMP) as a representative of the nucleotide family was theoretically investigated through density functional theory. AMP is a short form of DNA or RNA macromolecules that can interact with gold (Au) and silver (Ag) nanoparticles. The literature represents the thiolation of oligonucleotides as a suitable way for DNA/RNA adsorption on the stated nanoparticles. In this study, the role of different solvents on the thiolation of AMP is scrutinized. The results showed that acetonitrile is the best solvent for the thiolation of AMP with reaction enthalpy (ΔH) of −66.3 kJ/mol. The adsorption of thiolated adenosine monophosphate (TAMP) on the surface of Ag/Au nanoparticles (Ag/AuNP) was studied in different solvents, and in the gas phase. Our results revealed the interaction of TAMP with adsorption energies of −65.3 and −74.1 kJ/mol (gas phase), −54.8 and −62.5 kJ/mol (acetonitrile), and −59.2 and −70.6 kJ/mol (water) on AgNPs and AuNPs, respectively. AuNPs have a shorter bond distance in adsorption of TAMP compared to AgNPs (2.61 vs. 2.78 Å). For all systems, the frontier molecular orbital, the electronic charge passed through the nanoparticles, the density of states, and the molecular electrostatic potential were investigated. The results confirm the suitability of these nanoparticles as carriers for nucleotides that may be useful in gen delivery applications.  相似文献

This paper reported the enzymatic deposition of Au nanoparticles (AuNPs) on the designed 3-mercapto-propionic acid/glucose oxidase/chitosan (MPA/GOD/Chit) modified glassy carbon electrode and its application in glucose detection. Chit served as GOD immobilization matrix and interacted with MPA through electrostatic attraction. AuNPs, without nano-seeds presented on the electrode surface, was produced through the glucose oxidase catalyzed oxidation of glucose. The mechanism of production of AuNPs was confirmed to be that enzymatic reaction products H(2)O(2) in the solution reduce gold complex to AuNPs. The characterizations of the electrode modified after each assembly step was investigated by cyclic voltammetry and electrochemical impedance spectroscopy. Scanning electron microscopy showed the average particle size of the AuNPs is 40nm with a narrow particle size distribution. The content of AuNPs on the electrode surfaces was measured by differential pulse stripping voltammetry. The electrochemical signals on voltammogram showed a linear increase with the glucose concentration in the range of 0.010-0.12mM with a detection limit of 4μM. Nanoscience research aims to produce nanoparticles without adverse effects for medical applications. The pulsed laser ablation (PLA) technique was utilized in this study to synthesize gold nanoparticles (AuNPs) using bovine serum albumin (BSA) in simulated body fluid (SBF) at the fundamental wavelength of the Nd: YAG laser (1064 nm). BSA acted as a stabilizer, reducing and capping agent to produce spherically shaped AuNPs (diameter 3–10 nm). The successful synthesis of AuNPs was confirmed through color changes and UV–vis spectroscopy. The agglomeration and precipitation of AuNPs are attributed to the presence of BSA in the solution, and electrostatic repulsion interactions between BSA and Au nanoclusters. The effect of salt concentration of SBF on BSA stability as well as the interaction of BSA conjugated AuNPs to form complexes was studied using molecular dynamic simulations. Our results show that the stability of AuNPs-BSA conjugates increase with the salt concentration of BSA. Moreover, the synthesized AuNPs exhibit low toxicity and high biocompatibility, supporting their application in drug delivery. Investigation of the cytotoxic effect of the synthesized AuNPs show that normal fibroblast cells (L929) remain intact after treatment whereas a dose-dependent inhibition effect on the growth of cervix cancer cells (HeLa) is observed. In general, this study presents an effective, environmentally-friendly, and facile approach to the synthesis of multifunctional AuNPs using the PLA technique, as a promising efficacious therapeutic treatment of cervical cancer.  相似文献

Polyethylene glycol (PEG) surface conjugations are widely employed to render passivating properties to nanoparticles in biological applications. The benefits of surface passivation by PEG are reduced protein adsorption, diminished non-specific interactions, and improvement in pharmacokinetics. However, the limitations of PEG passivation remain an active area of research, and recent examples from the literature demonstrate how PEG passivation can fail. Here, we study the adsorption amount of biomolecules to PEGylated gold nanoparticles (AuNPs), focusing on how different protein properties influence binding. The AuNPs are PEGylated with three different sizes of conjugated PEG chains, and we examine interactions with proteins of different sizes, charges, and surface cysteine content. The experiments are carried out in vitro at physiologically relevant timescales to obtain the adsorption amounts and rates of each biomolecule on AuNP-PEGs of varying compositions. Our findings are relevant in understanding how protein size and the surface cysteine content affect binding, and our work reveals that cysteine residues can dramatically increase adsorption rates on PEGylated AuNPs. Moreover, shorter chain PEG molecules passivate the AuNP surface more effectively against all protein types.  相似文献

A facile method of obtaining chainlike assemblies of gold nanoparticles (AuNPs) on a chemically modified glass surface based on NaBH(4) treatment is developed. Citrate-stabilized AuNPs (17 nm) are immobilized on a glutaraldehyde-functionalized glass surface and assembled into chainlike structures after treatment with aqueous sodium borohydride (NaBH(4)) solution. The production and morphology of the AuNP chainlike assemblies are controlled by the density of the immobilized NPs, the concentration of NaBH(4) solution, and the treatment time. The AuNP assemblies are stable in water and can undergo drying. X-ray photoelectron spectroscopic data show that the number of citrate ions on the AuNPs decreased by 43% after treatment with 5 mg/mL NaBH(4) solution. The NaBH(4)-induced partial removal of the citrate ions and the roughness of the glass surface greatly affect the binding force of AuNPs on the substrate. The immobilized AuNPs begin to move at the solid-liquid interface without desorbing when the strength of the binding force was decreased. These mobile NPs form chainlike assemblies under the driving force of van der Waals interaction and diffusion. This interface-based formation of chainlike assemblies of AuNPs may provide a simple protocol for the 1D assembly of other Au-coated colloidal nanoparticles.  相似文献

Although nanomaterials facilitate significant technological advancement in our society, their potential impacts on the environment are yet to be fully understood. In this study, two environmentally relevant bacteria, Shewanella oneidensis and Bacillus subtilis, have been used as model organisms to elucidate the molecular interactions between these bacterial classes and Au nanoparticles (AuNPs) with well-controlled and well-characterized surface chemistries: anionic 3-mercaptopropionic acid (MPA), cationic 3-mercaptopropylamine (MPNH2), and the cationic polyelectrolyte poly(allylamine hydrochloride) (PAH). The data demonstrate that cationic, especially polyelectrolyte-wrapped AuNPs, were more toxic to both the Gram-negative and Gram-positive bacteria. The levels of toxicity observed were closely related to the percentage of cells with AuNPs associated with the cell surface as measured in situ using flow cytometry. The NP concentration-dependent binding profiles were drastically different for the two bacteria strains, suggesting the critical role of bacterial cell surface chemistry in determining nanoparticle association, and thereby, biological impact.  相似文献

The adsorption characteristics of three proteins [bovine serum albumin (BSA), myoglobin (Mb), and cytochrome c (CytC)] onto self-assembled monolayers of mercaptoundecanoic acid (MUA) on both gold nanoparticles (AuNP) and gold surfaces (Au) are described. The combination of quartz crystal microbalance measurements with dissipation (QCM-D) and pH titrations of the zeta-potential provide information on layer structure, surface coverage, and potential. All three proteins formed adsorption layers consisting of an irreversibly adsorbed fraction and a reversibly adsorbed fraction. BSA showed the highest affinity for the MUA/Au, forming an irreversibly adsorbed rigid monolayer with a side-down orientation and packing close to that expected in the jamming limit. In addition, BSA showed a large change in the adsorbed mass due to reversibly bound protein. The data indicate that the irreversibly adsorbed fraction of CytC is a monolayer structure, whereas the irreversibly adsorbed Mb is present in form of a bilayer. The observation of stable BSA complexes on MUA/AuNPs at the isoelectric point by zeta-potential measurements demonstrates that BSA can sterically stabilize MUA/AuNP. On the other hand, MUA/AuNP coated with either Mb or CytC formed a reversible flocculated state at the isoelectric point. The colloidal stability differences may be correlated with weaker binding in the reversibly bound overlayer in the case of Mb and CytC as compared to BSA.  相似文献

Determination of the true surface areas, concentrations, and particle sizes of gold nanoparticles (AuNPs) is a challenging issue due to the nanoparticle morphological irregularity, surface roughness, and size distributions. A ligand adsorption-based technique for determining AuNP surface areas in solution is reported. Using a water-soluble, stable, and highly UV–vis active organothiol, 2-mercaptobenzimidazole (MBI), as the probe ligand, we demonstrated that the amount of ligand adsorbed is proportional to the AuNP surface area. The equivalent spherical AuNP sizes and concentrations were determined by combining the MBI adsorption measurement with Au3+ quantification of aqua regia-digested AuNPs. The experimental results from the MBI adsorption method for a series of commercial colloidal AuNPs with nominal diameters of 10, 30, 50, and 90 nm were compared with those determined using dynamic light scattering, transmission electron microscopy, and localized surface plasmonic resonance methods. The ligand adsorption-based technique is highly reproducible and simple to implement. It only requires a UV–vis spectrophotometer for characterization of in-house-prepared AuNPs.  相似文献
