Synthesis and crystal structures of novel glycoluril carboxylic acids conglomerates

The two novel conglomerates were obtained by crystallization of racemic (2'S,3aS,6aR)/(2'R,3aR,6aS) (glycoluril-1-yl)-3-methylbutanoic acid and (2'R,3aR,6aR)/(2'S,3aS,6aS) (4,6-dimethylglycoluril-1-yl)pentanoic acid synthesized by highly diastereoselective condensation of 4,5-dihydroxy- imidazolidin-2-ones with racemic ureido acids. The differences in the molecular geometry of synthesized racemates were studied by X-ray diffraction that showed them to crystallize as conglomerates in non-centrosymmetric space groups Pna2₁ and P2₁2₁2₁, respectively     

Chiral resolution of multichiral center racemates by different modalities of chromatography

It is a continuous chase of researchers to separate chiral racemates due to their dominance in the pharmaceutical industry, agrochemicals, and food additives. There are many analytical techniques which have been used to resolve one chiral center racemates, but, little the literature report is scanty for the resolution of multiple stereogenic center racemates. Therefore, in this article the efforts were made to describe the enantiomeric resolution of multiple stereogenic center racemates by different modalities of chromatography. The various modalities discussed are high-performance liquid chromatography, capillary electrophoretic chromatography, miceller electrokinetic chromatography, supercritical fluid chromatography, nanoliquid chromatography, and gas chromatography. Besides, the attempts have also been made to discuss the future challenges and prospectives of the enantiomeric resolution of multichiral racemates.     

Thermoanalytical methods in pharmaceutical technology

A short survey is given of some aspects of the application of thermoanalytical methods, especially differential thermal analysis (DTA), differential scanning calorimetry (DSC) and thermogravimetry (TG), in solid-dosage technology. The usefulness of these methods in the prediction of drug-excipient compatibility, studies of solid-dispersion systems, the analysis of enantiomers and racemates, measurement of the time of tablet disintegration, the analysis of drug formulations and studies of the processes of grinding and drying of drugs is discussed.

Zusammenfassung Es wird ein kurzer Überblick über einige Aspekte der Anwendung von thermoanalytischen Methoden, insbesondere von DTA, DSC und TG in der Feststoffdosierung gegeben. Weiterhin wird die Nützlichkeit dieser Methoden bei der Voraussage der Bindemittelverträglichkeit, bei der Untersuchung disperser Feststoffsysteme, bei der Analyse von Enantiomeren und Racematen, bei der Messung der Tablettenzerfallszeit, bei der Analyse der Arzneimittelformierung und bei der Untersuchung von Mahl- und Trocknungsprozessen bei Arzneimitteln diskutiert.

     

Binary solid-liquid phase diagram of the two diastereomer racemates of triglycidyl isocyanurate (TGIC)

Triglycidyl isocyanurate [2451-62-9] as a trifunctional epoxy monomer is mostly used as curing agent. It exists in the form of two diastereomer racemates according to the configuration of the three chiral carbon atoms in the molecule, i.e. beta- or RRR/SSS and alpha- or RRS/SSR. The binary solid-liquid phase diagram of the two diastereomer racemates has been determined by differential scanning calorimetry (DSC). The two diastereomer racemates have different crystalline structures and are physical mixtures with an eutectic phase diagram. The eutectic mixture melts at 92.3 ± 1.0 °C and has alpha-TGIC content of 93.44% by mass or mole. With the help of the phase diagram the ratio of the two diastereomer racemates in TGIC may be determined. The binary phase diagram shows that beta-TGIC can be separated with efficiency from the mixture and may serve for enantiomer synthesis of high optical activity giving rise to the development of novel polymers.     

Kinetic resolution of heterocyclic amines by reaction with optically active acid chlorides. The effect of reaction conditions on the diastereoselectivity of acylation of (±)-3-methyl-2,3-dihydro-4H-1,4-benzoxazine

The influence of the reaction conditions (solvent, base) on the diastereoselectivity of acylation of (±)-3-methyl-2,3-dihydro-4H-1,4-benzoxazine with (S)-naproxen and N-tosyl-(S)-proline chlorides was studied. The highest diastereoselectivity was observed for the reaction carried out in benzene in the presence of aliphatic tertiary amines.

A New Easy Access to Enantiomerically Pure 2,2'-Dihydroxy-1,1'-Binaphthyl

Optically pure binaphthyl structures make up the most important family of auxiliaries, ligands and catalysts used in enantioselective reactions. Syntheses of 2,2'-disubstituted-1,1'-binaphtyl derivatives were carried out one pot in water by oxidative coupling with FeCl₃ in the presence of - and -cyclodextrin (CD) derivatives. A new efficient and inexpensive preparation of trihydroxyethyl- and tri-2-hydroxypropyl- - and -CD afforded these powerful solubilizing agents. The resolution of racemic 2,2'-dihydroxy-1,1'-binaphthyl was easily achieved by semipreparative HPLC separating diastereomers obtained from the reaction with (-)menthyl chloroformate. Final basic hydrolysis afforded the enantiomerically pure product.
