Alkyne hydroamination is an effective approach for the production of enamines and enamine-containing N-heterocycles. However, stereoselectivity control is a considerable challenge in this reaction because of the electronic repulsion between an incoming nitrogen lone pair and the alkyne π-system. Herein, we propose a methodology involving β-regio- and Z-selective alkyne hydroamination by using tetrafluoro-λ6-sulfanyl (SF4) alkynes under superbasic, naked anion conditions. The reaction is compatible with a wide variety of N-heterocycles, including indoles, carbazoles, pyrazoles, and imidazoles, and selectively furnishes SF4-linked Z-vinyl enamines with β-regioselectively. Moreover, the method can be extended to the β- and Z-controlled, base-mediated alkyne hydrophenoxylation with phenols to provide SF4-linked Z-vinyl ethers in high yields. As the SF4 unit has attracted attention as a bioisostere for alkynes, p-benzenes, bicyclo[1.1.1]pentyl (BCP) groups, and cubanes in medicinal chemistry, this chemistry represents an effective approach to creating novel drug candidates incorporating SF4-containing molecules.  相似文献

A tandem approach for the regio- and stereoselective synthesis of oxazolo-fused pyrroloquinolines 3al via the reaction of o-alkynylaldehydes 1ai with chiral amino alcohols 2ac under mild reaction conditions is described. The possible participation of the pyridine ring in the regioselective formation of 5-exo-dig cyclized products was supported by the controlled experiments. The structures and stereochemistry of the products were confirmed by NOESY and X-ray crystallographic studies.  相似文献

Current medical diagnostic echo systems are mostly using harmonic imaging. This means that a fundamental frequency (e.g., 2 MHz) is transmitted and the reflected and scattered higher harmonics (e.g., 4 and 6 MHz), produced by nonlinear propagation, are recorded. The signal level of these harmonics is usually low and a well-defined transfer function of the receiving transducer is required. Studying the acoustic response of a single contrast bubble, which has an amplitude in the order of a few Pascal, is another area where an optimal receive transfer function is important.

We have developed three methods to determine the absolute transfer function of a transducer. The first is based on a well-defined wave generated by a calibrated source in the far field. The receiving transducer receives the calibrated wave and from this the transfer functions can be calculated. The second and third methods are based on the reciprocity of the transducer. The second utilizes a calibrated hydrophone to measure the transmitted field. In the third method, a pulse is transmitted by the transducer, which impinges on a reflector and is received again by the same transducer. In both methods, the response combined with the transducer impedance and beam profiles enables the calculation of the transfer function.

The proposed methods are useful to select the optimal piezoelectric material (PZT, single crystal) for transducers used in reception only, such as in certain 3D scanning designs and superharmonic imaging, and for selected experiments like single bubble behavior.

We tested and compared these methods on two unfocused single element transducers, one commercially available (radius 6.35 mm, centre frequency 2.25 MHz) the other custom built (radius 0.75 mm, centre frequency 4.3 MHz). The methods were accurate to within 15%.  相似文献

In this paper, we define the Baskakov-Durrmeyer type operators based on generalized Appell polynomials. Here, we establish moment estimates, an estimate via weighted modulus of continuity and a Voronovoskaya type asymptotic result. Further, we study a quantitative-Voronovoskaya-type theorem and Grüss Voronovskaya-type theorem. Lastly, we give the approximation result for functions having derivatives of bounded variation.  相似文献

The bioassay guided fractionation of methanolic extract of Murraya koenigii (L.) Spreng. leaves resulted in the isolation of seven pyranocarbazoles. These were evaluated against four bacterial strains and ten Candida sp. including two matched pair of fluconazole sensitive/resistant clinical isolates. Out of seven, three i.e. Koenine (mk279), Koenigine (mk309) and Mahanine (mk347) exhibited significant antibacterial activity MIC90 3.12–12.5 μg/mL against bacterial strains Streptococcus aureus and Klebsiella pneumonia compared with standard drug Kanamycin MIC90 12.5 μg/mL. However, only mk309 was found active against variety of Candida species MIC90 12.5–100 μg/mL. It was observed that hydroxylation at C-6 and C-7 positions in the studied pyranocarbazoles activate the bioactivity. Simultaneously, decrease in Log P value compares with ?H and ?O?CH3 substituted derivatives. The study is focused on selective antifungal and antibacterial activity of pyranocarbazoles on bacterial strains S. aureus, K. pneumonia and variety of Candida species with structure activity relationship observations.  相似文献

The size distribution of adenylate cyclase from the rat renal medulla solubilized with the nonionic detergents Triton X-100 and Lubrol PX was determined by gel filtration and by centrifugation in sucrose density gradients made up in H2O or D2O. The physical parameters of the predominant form in Triton X-100 are s20,w, 5.9S; Strokes radius, 62 A; partial specific volume (v), 0.74 ml/g; mass, 159,000 daltons; f/f0, 1.6; axial ratio (prolate ellipsoid), 11. For the minor form the values are: s20w, 3.0; Stokes radius, 28 A; mass, 38,000 daltons; f/f0, 1.2. The corresponding values determined in Lubrol PX are similar. The value for V for the enzyme indicates that it binds less than 0.2 mg detergent/mg protein. Since interactions with detergents probably substitute for interactions with lipids and hydrophobic amino acid side chains, these findings suggest that no more than 5% of the surface of adenylate cyclase is involved in hydrophobic interactions with other membrane components. Thus, most of the mass of the enzyme is not deeply embedded in the lipid bilayer of the plasma membrane. Similar studies have been performed on the soluble guanylate cyclase of the rat renal medulla. In the absence of detergent, the molecular properties of this enzyme are: s20w, 6.3S; Stokes radius, 54 A, V, 0.75 ml/g; mass, 154,000 daltons f/f0, 1.4; Axial ratio, 7. The addition of 0.1% Lubrol PX to this soluble enzyme increases it activity two- to fourfold and changes the physical properties to: s20,w, 5.5S; Stokes radius, 62 A; V, 0.74 ml/g; mass, 148,000 daltons, f/f0, 1.6; axial ratio, 11. These results show that Lubrol PX activates the enzyme by causing a conformational change with unfolding on the polypeptide chain. Guanylate cyclase from the particulate cell fraction can be solubilized with Lubrol PX but has properties quite different from those of the enzyme in the soluble cell fraction. It is a heterogeneous aggregate with s20,w, 10S; Stokes radius, 65 A; mass about 300,000 daltons. The conditions which solubilize guanylate cyclase also solubilize adenylate cyclase and the two activities can be separated on the same sucrose gradient.  相似文献

Platinum dendrimer-encapsulated nanoparticles (DENs) containing an average 147 atoms were prepared within sixth-generation, hydroxyl-terminated poly(amidoamine) dendrimers (G6-OH). The hydrodynamic radii (R(h)) of the dendrimer/nanoparticle composites (DNCs) were determined by quasi-elastic light scattering (QLS) at high (pH ~10) and neutral pH for various salt concentrations and identities. At high pH, the size of the DNC (R(h) ~4 nm) is close to that of the empty dendrimer. At neutral pH, the size of the DNC approximately doubles (R(h) ~8 nm) whereas that of the empty dendrimer remains unchanged. Changes in ionic strength also alter the size of the DNCs. The increase in size of the DNC is likely due to electrostatic interactions involving the metal nanoparticle.  相似文献

Burned greasy deposits were found inside shells of the large Nile bivalve Chambardia rubens, excavated in an eight- to tenth- century AD church of the Coptic monastery of Bawit, Egypt, and supposedly used as oil lamps. The residues were subjected to a combination of chromatographic residue analysis techniques. The rather high concentrations of unsaturated fatty acids, as analysed by gas chromatography (GC) in the methylated extract, suggest the presence of a vegetal oil. Analysis of the stable carbon isotopes (δ 13C values) of the methyl esters also favoured plants over animals as the lipid source. In the search for biomarkers by GC coupled to mass spectrometry on a silylated extract, a range of diacids together with high concentrations of 13,14-dihydroxydocosanoate and 11,12-dihydroxyeicosanoate were found. These compounds are oxidation products of erucic acid and gondoic acid, which are abundantly present in seeds of Brassicaceae plants. Liquid chromatography coupled to mass spectrometry analysis showed low concentrations of unaltered triglycerides, but revealed sizeable amounts of triglycerides with at least one dihydroxylated acyl chain. The unusual preservation of dihydroxylated triglycerides and α,ω-dicarboxylic acids can be related to the dry preservation conditions. Analysis of the stereoisomers of the dihydroxylated fatty acids allows one to determine whether oxidation took place during burning of the fuel or afterwards. The results prove that the oil of rapeseed (Brassica napus L.) or radish (Raphanus sativus L.) was used as illuminant in early Islamic Egypt, and that not only ceramic lamps but also mollusk shells were used as fuel containers.  相似文献
