Despite its high morbidity and mortality, contrast‐induced acute kidney injury (CIAKI) remains a diagnostic dilemma because it relies on in vitro detection of insensitive late‐stage blood and urinary biomarkers. We report the synthesis of an activatable duplex reporter (ADR) for real‐time in vivo imaging of CIAKI. ADR is equipped with chemiluminescence and near‐infrared fluorescence (NIRF) signaling channels that can be activated by oxidative stress (superoxide anion, O2.?) and lysosomal damage (N‐acetyl‐β‐d ‐glucosaminidase, NAG), respectively. By virtue of its high renal clearance efficiency (80 % injected doses after 24 h injection), ADR detects sequential upregulation of O2.? and NAG in the kidneys of living mice prior to a significant decrease in glomerular filtration rate (GFR) and tissue damage in the course of CIAKI. ADR outperforms the typical clinical assays and detects CIAKI at least 8 h (NIRF) and up to 16 h (chemiluminescence) earlier. 相似文献

Photoelectrochemical(PEC) water splitting is a promising approach for renewable hydrogen production.However,the practical PEC solar-to-fuel conversion efficiency is still low owing to poor light absorption and rapid recombination of charge carriers in photoelectrode.In this work,we report a ternary photoanode with simultaneously enhancement of light absorption and water oxidation efficiency by introducing copper phthalocyanine(CuPc) and nickel iron-laye red double hydroxide(NiFe-LDH) on TiO_2(denoted as TiO_2/CuPc/NiFe-LDH).An experimental study reveals that CuPc loading on TiO_2 bring strong visible light absorption;NiFe-LDH as an oxygen evolution reaction catalyst efficiently accelerates the surface water oxidation reaction.This synergistic effect of CuPc and NiFe-LDH gives enhanced photocurrent density(2.10 mA/cm2 at 0.6 V vs.SCE) and excellent stability in the ternary TiO_2/CuPc/NiFeLDH photoanode. 相似文献

Exosomes hold great potential in therapeutic development. However, native exosomes usually induce insufficient effects in vivo and simply act as drug delivery vehicles. Herein, we synthesize responsive exosome nano‐bioconjugates for cancer therapy. Azide‐modified exosomes derived from M1 macrophages are conjugated with dibenzocyclooctyne‐modified antibodies of CD47 and SIRPα (aCD47 and aSIRPα) through pH‐sensitive linkers. After systemic administration, the nano‐bioconjugates can actively target tumors through the specific recognition between aCD47 and CD47 on the tumor cell surface. In the acidic tumor microenvironment, the benzoic‐imine bonds of the nano‐bioconjugates are cleaved to release aSIRPα and aCD47 that can, respectively, block SIRPα on macrophages and CD47, leading to abolished “don't eat me” signaling and improved phagocytosis of macrophages. Meanwhile, the native M1 exosomes effectively reprogram the macrophages from pro‐tumoral M2 to anti‐tumoral M1. 相似文献

Some efficient polymer‐supported palladium catalysts have been synthesized. Their catalytic effects were evaluated in the Heck reaction of iodobenzene with acrylates or styrene. High catalytic activities were achieved with turn over frequencies (TOF) up to 28000 and 6250, respectively. For the reaction of iodobenzene with styrene, high stereoselectivity was obtained. Additionally, the catalysts could be recovered by a simple filtration progress and can be reused for at least 5 times with a slow progressive decrease in activity. 相似文献

利用有限元分析方法探究镁合金空心螺钉内固定治疗股骨颈骨折的生物力学特性, 旨在为临床治疗股骨颈骨折提供一种新的思路. 选取一例青年健康志愿者的下肢CT数据建立股骨三维模型, 在Mimics插件3-matic中建立股骨网格并赋予网格材料属性, 利用Solidworks 17.0软件建立空心螺钉模型, 将上述2个三维模型导入Workbench 2020软件中, 分别赋予空心螺钉镁合金、钛合金材料属性, 进行对比分析, 记录股骨及空心螺钉的峰值、应力分布值和股骨位移. 结果表明, 钛合金螺钉内固定模型最大应力为357.27MPa, 最大位移为0.034mm; 镁合金螺钉内固定模型最大应力为311.27MPa, 最大位移为0.034mm; 2组不同材料的空心钉结果数值差异不大. 研究表明镁合金空心螺钉和传统钛合金空心螺钉力学性能相似, 且镁合金更加亲和人体, 在临床方面可以镁合金空心螺钉代替钛合金空心螺钉治疗股骨颈骨折. 相似文献

研究了基于NZ30K合金开发的新型Mg-3Nd-0.2Zn-0.4Zr-0.2Mn镁合金耐腐蚀性、体外降解行为特性及浸提液细胞生物毒性. 采用金相显微镜得到新型镁合金金相显微图, 采用扫描电镜(Scanning Electron Microscope, SEM)获取SEM图; 采用武汉科思特电化学工作站进行电化学测试, 并绘制动电位极化曲线, 以磷酸盐缓冲液(Phosphate Buffer Solution, PBS)模拟体液环境, 记录氢气析出体积并计算腐蚀速率; 利用细胞完全培养基测定pH值、重量变化曲线; 获取大鼠骨髓间充质干细胞(Bone Mesenchymal Stem Cells, BMSCs), 并利用完全细胞培养基制作新型镁合金浸提液, 检测细胞生物活性, 以ZA75镁合金为基础添加0.3%Mn元素制成合金作为对照组, 比较腐蚀电位、体外降解情况以及细胞活性. 结果表明: 新型Mg-3Nd-0.2Zn-0.4Zr-0.2Mn镁合金横截面等轴晶体组织细小均匀性较好, 纵截面呈长条状组织均匀性稍差; 自腐蚀电位较高, 为-1.3912V; 自腐蚀电流密度较低, 为7.37×10-7 A?cm-2; 体外析氢量低, 失重量、pH值变化幅度相对较小; 降解速率下降后呈现小范围上升后趋于平缓; 具有良好的细胞相容性, 可以促进BMSCs细胞增殖分化. 相似文献

This paper studies the distribution of finite-time ruin quantities. It gives the probability mass function of finite time number of claims, and find the distribution function of aggregate claims by using discretise method and compared with exact distribution function, which shows that the approximation works very well. In addition, by applying decomposition for density function, it gives the explicit expression for joint density of ruin time and deficit at ruin. 相似文献

An optimized method based on GC-MS with ethyl chloroformate derivatization has been developed for the comprehensive analysis
of endogenous metabolites in serum. Twenty-two reference standards and serum samples were used to validate the proposed method.
The correlation coefficient was higher than 0.9900 for each of the standards, and the LOD varied from 125 to 300 pg on-column.
The analytical equipment exhibited good repeatability (RSD<10%) for all of the standards. Both the repeatability and the within-48-h
stability of the analytical method were satisfactory (RSD<10%) for the 18 metabolites identified in the serum samples. Mean
recovery was acceptable for the 18 metabolites, ranging from 70% to 120% with RSDs of less than 10%. Using the optimized protocol
and a subsequent multivariate statistical technique, complete differentiation was achieved between the metabolic profile of
uremic patients and that of age- and sex-matched normal subjects. Significantly decreased levels of valine, leucine, and isoleucine
and increased levels of myristic acid and linoleic acid were observed in the patient group. This work demonstrated that this
method is suitable for serum-based metabolic profiling studies. 相似文献
