Human poisoning due to consumption of seafood contaminated with phycotoxins is a worldwide problem, and routine monitoring programs have been implemented in various countries to protect human consumers. Following successive episodes of unexplained shellfish toxicity since 2005 in the Arcachon Bay on the French Atlantic coast, a national research program was set up to investigate these atypical toxic events. Part of this program was devoted to fit-for-purpose cell-based assays (CBA) as complementary tools to collect toxicity data on atypical positive-mouse bioassay shellfish extracts. A collaborative study involving five laboratories was conducted. The responses of human hepatic (HepG2), human intestinal (Caco2), and mouse neuronal (Neuro2a) cell lines exposed to three known lipophilic phycotoxins-okadaic acid (OA), azaspiracid-1 (AZA1), and pectenotoxin-2 (PTX2)-were investigated. A screening strategy composed of standard operating procedures and a decision tree for dose-response modeling and assay validation were designed after a round of "trial-and-error" process. For each toxin, the shape of the concentration-response curves and the IC(50) values were determined on the three cell lines. Whereas OA induced a similar response irrespective of the cell line (complete sigmoid), PTX2 was shown to be less toxic. AZA1 induced cytotoxicity only on HepG2 and Neuro2a, but not on Caco2. Intra- and inter-laboratory coefficients of variation of cell responses were large, with mean values ranging from 35 to 54 % and from 37 to 48 %, respectively. Investigating the responses of the selected cell lines to well-known toxins is the first step supporting the use of CBA among the panel of methods for characterizing atypical shellfish toxicity. Considering these successful results, the CBA strategy will be further applied to extracts of negative, spiked, and naturally contaminated shellfish tissues.  相似文献

Copolymers of acrylonitrile with cinnamonitrile (I), ethyl benzylidenecyanoacetate (II), and benzylidenemalononitrile (III) were prepared in suspension up to high conversions. Films and molded specimens were made from the copolymers and their basic physical and mechanical properties, such as solubility, viscosity, glass transition temperature, and tensile and compressive strength were determined. Further, treatment by heat and ultraviolet light, the permeability of water vapors, and the behavior of films in a weathering tester were studied.  相似文献

The French Food Safety Agency is a public body incorporating 12 laboratories that perform research to support expertise and public decisions taken in the fields of sanitary safety of food, animal health, and veterinary drugs. On the request of the General Management of the Agency a quality-management system in research (QMSR) is being implemented in the Food and Food Process Quality Research Laboratory. The experimental QMSR is based on existing standards and documents, describing the provisions required for scientific and technical competence, quality management, and project management. Furthermore, this QMSR also incorporates specific notions of great importance for research activities such as the positive and negative non-conformities, the non-confirmation of hypotheses, and the principle of evaluation of a research activity by peers for both quality and scientific aspects.  相似文献

Split Hopkinson Pressure Bar tests are commonly used to determine material stress-strain relationship at high deformation rates. Obtaining this relationship is dependant both on certain assumptions and substantial post-processing of the data recorded during the test. Measurement uncertainty rarely appears on the resulting curves. This article introduces a simple method of estimating the measurement uncertainty associated with SHPB tests.  相似文献

Reaction products of vacuum and oxidative degradation of poly-p-xylylene have been quantitatively determined by chromatographic analysis as function of time, temperature and oxygen pressure. Respective Arrhenius parameters were also ascertained for some of the reaction products and for the sums of all products. The energies of activation for the sums agree quite satisfactorily with the energies of activation obtained previously by uninterrupted experiments in quartz-spoon reaction vessels. The results found here can be described in terms of mechanisms previously postulated on the basis of the total loss in weight (or volatile production) data. Scission of “weak” links (due to abnormal structures) takes place followed by formation of various products. The whole process is governed by the initial chain scission reaction; however, the energies of activation for each of the products do not need to be identical with that of the chain scission reaction. Each product is formed by a reaction which has its own characteristic number average kinetic chain lengths; the latter have their specific energy of activation values. Oxidative degradation produces the same organic compounds as vacuum degradation and in addition CO, CO2, and H2O. Oxidized intermediate compounds are apparently fairly rapidly decarboxylated and decarbonylated. Oxidative chain scission is appreciably faster than that in vacuum. Almost simultaneous “weak” link and “normal” chain scission are taking place initiating the formation of a number of products.  相似文献

This paper was devoted to the three-dimensional nonlinear finite element analysis of inflatable beams. The beams under consideration are made of modern textile materials and can be used as a load-bearing beams or arches when inflated. A 3D Timoshenko beam with a homogeneous orthotropic woven fabric (OWF) was proposed. The model took into account the geometric nonlinearities and the follower force resulting from the inflation pressure. The use was made of the usual total Lagrangian form of the virtual work principle to perform the nonlinear equilibrium equations which were discretized by the finite element method. Two kinds of solutions were then investigated: finite elements solutions for linearized problems which were obtained by the means of the linearization around the prestressed reference configuration of the nonlinear equations and nonlinear finite element solutions which were performed by the use of an optimization algorithm based on the Quasi-Newton method. As an example, the bending problem of a cantilever inflated beam under concentrated load was considered and the deflection results improve the existing theoretical models. As these beams are made from fabric, the beam models were validated through their comparison with a 3D thin-shell finite element model. The influence of the material effective properties and the inflation pressure on the beam response was also investigated through a parametric study. The finite elements solutions for linearized problems were found to be close to the theoretical results existing in the literature. On the other hand, the results for the nonlinear finite element model were shown to be close to the results for the linearized finite elements model in the case of high mechanical properties and the nonlinear finite element model was used to improve the linearized model when the mechanical properties of the fabric are low.  相似文献

The potential of solid phase extraction (SPE) clean-up has been assessed to reduce matrix effects (signal suppression or enhancement) in the liquid chromatography-tandem mass spectrometry (LC–MS/MS) analysis of lipophilic marine toxins. A large array of ion-exchange, silica-based, and mixed-function SPE sorbents was tested. Polymeric sorbents were found to retain most of the toxins. Optimization experiments were carried out to maximize recoveries and the effectiveness of the clean-up. In LC–MS/MS analysis, the observed matrix effects can depend on the chromatographic conditions used, therefore, two different HPLC methods were tested, using either an acidic or an alkaline mobile phase. The recovery of the optimized SPE protocol was around 90% for all toxins studied and no break-through was observed. The matrix effects were determined by comparing signal response from toxins spiked in crude and SPE-cleaned extracts with those derived from toxins prepared in methanol. In crude extracts, all toxins suffered from matrix effects, although in varying amounts. The most serious effects were observed for okadaic acid (OA) and pectenotoxin-2 (PTX2) in the positive electrospray ionization mode (ESI+). SPE clean-up on polymeric sorbents in combination with the alkaline LC method resulted in a substantial reduction of matrix effects to less than 15% (apparent recovery between 85 and 115%) for OA, yessotoxin (YTX) in ESI and azaspiracid-1 (AZA1), PTX2, 13-desmethyl spirolides C (SPX1), and gymnodimine (GYM) in ESI+. In combination with the acidic LC method, the matrix effects after SPE were also reduced but nevertheless approximately 30% of the matrix effects remained for PTX2, SPX1, and GYM in ESI+. It was concluded that SPE of methanolic shellfish extracts can be very useful for reduction of matrix effects. However, the type of LC and MS methods used is also of great importance. SPE on polymeric sorbents in combination with LC under alkaline conditions was found the most effective method.  相似文献
