将自制的在线固相萃取柱与液相色谱串联四极杆静电轨道离子阱质谱技术相结合,建立了快速测定牛奶中15种糖皮质激素类药物的新方法.样品采用乙腈均质提取2 min,经自制在线固相萃取小柱富集净化,在Waters CORTECS T3(150 mm ×2.1 mm,5μm)分析柱上实现良好分离,外标法定量.15种目标物质量浓度在0.10~100 ng/mL范围内线性关系良好,相关系数均大于0.99;在0.20,0.40,2.0μg/kg3个添加水平下,15种糖皮质激素的回收率在87.1% ~ 116.0%之间,相对标准偏差(RSD)在3.2%~8.7%(n=6)之间;方法的检出限(LOD,S/N≥3)和定量限(LOQ,S/N≥1O)分别为0.10和0.20μg/kg.该法适用于大批量牛奶中糖皮质激素类药物的快速定性筛查和定量分析.  相似文献

High‐field asymmetric waveform ion mobility spectrometry (FAIMS) separates ions by utilizing the characteristics of nonlinear ion mobility at high and low electric fields. Accurate ion discrimination depends on the precise solution of nonlinear relationships and is essential for accurate identification of ion species for applications. So far, all the nonlinear relationships of ion mobility obtained are based at low electric fields (E/N <65 Td). Microchip FAIMS (μ‐FAIMS) with small dimensions has high electric field up to E/N = 250 Td, making the approximation methods and conclusions for nonlinear relationships inappropriate for these systems. In this paper, we deduced nonlinear functions based on the first principle and a general model. Furthermore we considered the hydrodynamics of gas flow through microchannels. We then calculated the specific alpha coefficients for cocaine, morphine, HMX, TNT and RDX, respectively, based on their FAIMS spectra measured by μ‐FAIMS system at ultra‐high fields up to 250 Td. The results show that there is no difference in nonlinear alpha functions obtained by the approximation and new method at low field (<120 Td), but the error induced by using approximation method increases monotonically with the increase in field, and could be as much as 30% at a field of 250 Td. 57例胃癌癌灶内锌值为(13.740±4.150)×10~(-6),低于癌边缘组织(15.677±4.567)×10~(-6)(P<0.01),亦低于正常组织(15.897±3.580)×10~(-6)(P<0.01)。癌灶铜值为(7.91±6.19)×10~(-6),高于正常组织(5.62±12.01)×10~(-6)(P<0.05),癌灶铜/锌比值亦高于正常组织(P<0.01),但与边缘组织无显著性差异。分化较好的胃腺癌与未分化癌间七种微量元素含量均无显著性差异。这些结果提示,胃癌组织中锌值的降低与铜/锌比值的升高可作为胃癌的辅助诊断指标,而且锌值的检测更具意义。但与分化程度无关。  相似文献

The effect of the fluoroethylene carbonate (FEC) addition in electrolyte on LiFePO4 cathode performance was investigated in low-temperature electrolyte LiPF6/EC/PC/EMC (0.14/0.18/0.68). Cyclic voltammetry, electrochemical impedance spectroscopy, and charge/discharge tests were conducted in this work. In the presence of FEC, the polarization of LiFePO4 electrode decreased both at room and low temperatures. Meanwhile, the exchange current density increased. The rate capability of LiFePO4 electrode was greatly enhanced as well. The morphology of the solid electrolyte interphase (SEI) on LiFePO4 surface was modified with the addition of FEC as confirmed by scanning electron microscopy measurement. A compact film with small impedance was formed on LiFePO4 surface compared to the case of FEC-free. The compositions of the film were analyzed by X-ray photoelectron spectroscopic measurement. The contents of Li x PO y F z , LiF, and the carbonate species generated from solvents decomposition were reduced. The modified SEI promoted the migration of lithium ion through the electrode/electrolyte interphase and enhanced the electrochemical performance of the cathode.  相似文献

The demand for reduced chemical preservative usage is currently growing, and natural preservatives are being developed to protect seafood. With its excellent antibacterial properties, linalool has been utilized widely in industries. However, its antibacterial mechanisms remain poorly studied. Here, untargeted metabolomics was applied to explore the mechanism of Shewanella putrefaciens cells treated with linalool. Results showed that linalool exhibited remarkable antibacterial activity against S. putrefaciens, with 1.5 µL/mL minimum inhibitory concentration (MIC). The growth of S. putrefaciens was suppressed completely at 1/2 MIC and 1 MIC levels. Linalool treatment reduced the membrane potential (MP); caused the leakage of alkaline phosphatase (AKP); and released the DNA, RNA, and proteins of S. putrefaciens, thus destroying the cell structure and expelling the cytoplasmic content. A total of 170 differential metabolites (DMs) were screened using metabolomics analysis, among which 81 species were upregulated and 89 species were downregulated after linalool treatment. These DMs are closely related to the tricarboxylic acid (TCA) cycle, glycolysis, amino acid metabolism, pantothenate and CoA biosynthesis, aminoacyl-tRNA biosynthesis, and glycerophospholipid metabolism. In addition, linalool substantially affected the activity of key enzymes, such as succinate dehydrogenase (SDH), pyruvate kinase (PK), ATPase, and respiratory chain dehydrogenase. The results provided some insights into the antibacterial mechanism of linalool against S. putrefaciens and are important for the development and application of linalool in seafood preservation.  相似文献

This review focuses on recent and potential advances in chemometric methods in relation to data processing in metabolomics, especially for data generated from mass spectrometric techniques. Metabolomics is gradually being regarded a valuable and promising biotechnology rather than an ambitious advancement. Herein, we outline significant developments in metabolomics, especially in the combination with modern chemical analysis techniques, and dedicated statistical, and chemometric data analytical strategies. Advanced skills in the preprocessing of raw data, identification of metabolites, variable selection, and modeling are illustrated. We believe that insights from these developments will help narrow the gap between the original dataset and current biological knowledge. We also discuss the limitations and perspectives of extracting information from high-throughput datasets.  相似文献

基于现有低碳钢焊条性能的特点,针对典型低碳钢焊条(E4303型焊条)抗裂性较差的问题,在新型低碳钢焊条药皮配方开发研制中采取药皮过渡微量的镧钇元素.通过冲击韧性、拉伸、硬度、金相等试验研究镧钇元素对低碳钢焊条熔敷金属组织和力学性能的影响,利用扫描电子显微镜对显微组织进行观察分析,用X射线荧光光谱仪进行光谱分析.结果表明:加入适量的镧钇元素能够细化熔敷金属显微组织,提高熔敷金属的冲击韧性.当镧钇元素加入量最佳(3.5%镧和0.6%钇)时,熔敷金属的冲击韧性提高了38.8%,延伸率提高了25%,抗拉强度提高了11.9%,屈服强度提高了10.9%,硬度却不下降.  相似文献
