Potential genotoxic impurities in pharmaceuticals at trace levels are of increasing concern to both pharmaceutical industries and regulatory agencies due to their possibility for human carcinogenesis. Molecular functional groups that render starting materials and synthetic intermediates as reactive building blocks for small molecules may also be responsible for their genotoxicity. Determination of these genotoxic impurities at trace levels requires highly sensitive and selective analytical methodologies, which poses tremendous challenges on analytical communities in pharmaceutical research and development. Experimental guidance for the analytical determination of some important classes of genotoxic impurities is still unavailable in the literature. Therefore, the present review explores the structural alerts of commonly encountered potential genotoxic impurities, draft guidance of various regulatory authorities in order to control the level of impurities in drug substances and to assess their toxicity. This review also describes the analytical considerations for the determination of potential genotoxic impurities at trace levels and finally few case studies are also discussed for the determination of some important classes of potential genotoxic impurities. It is the authors’ intention to provide a complete strategy that helps analytical scientists for the analysis of such potential genotoxic impurities in pharmaceuticals.  相似文献

土壤硝态氮反映土壤短期氮素供应水平,实时了解土壤硝态氮的含量为精准农业和农业面源污染防控提供支撑,因此,在线实时检测土壤硝态氮方法突破就显得十分迫切。土壤硝态氮中的硝酸根离子在土壤中的高水溶性和流动性为全固态硝酸根离子选择电极高敏感检测土壤中硝态氮提供了条件,固态硝态氮离子选择电极的离子选择膜反应硝酸根离子在被测溶液中的浓度。采用全固态硝酸根离子选择电极,且与温度电极和pH电极融合组成电极阵列对土壤饱和溶液中的硝态根离子进行检测。设计了高输入阻抗运算放大电路对电极信号进行采集,并通过微处理控制蠕动泵完成土壤硝态氮待测溶液连续流动测量及实时传输结果。实验结果表明,电极响应时间≤15 s,斜率-51.63 mV/decade,线性范围10-5-10-2.2 mol/L,最低检测限10-5.23 mol/L。相对标准差在0.78%-4.47%范围内,加标回收率均在90%-110%以内。与国家标准紫外可见分光光度法测试结果相比,相关系数(R2)为0.9952,为土壤硝态氮在现场检测奠定技术基础。  相似文献

Polyhydroxyalkanoates (PHAs) are a family of biodegradable and biocompatible polymers produced by several species microorganisms that possess favorable mechanical properties (e.g. strength and elongation properties). Different types of PHA polymers have been used in medical applications. However, in order to better understand the use of this polymer in the different applications, a thorough understanding of the kinetics of in vivo degradation is one of the major requirements. In this study, poly(3‐hydroxybutyrate) (PHB) was subcutaneously implanted in mice and incubated for 2, 4, 8, or 16 weeks. After removal from the animal, the strength, elongation, mass loss, and enthalpy of the PHB were tested for each time point. From these data, a mathematical model was generated by Rayleigh's method of dimensional analysis, where polymer strength over tissue contact time could be predicted. To prove the model, previous data obtained by our group were used: poly(3‐hydroxybutyrate‐co‐3‐hydroxyhexanoate) [P(HB‐co‐HHx)] incubation in the presence of human embryonic kidney cells (HEK). It was found that the developed model was aligned with experimental results, could predict the strength of the polymer when in contact with cells, and the predicted strength follows the trend of the experimental data. Also, the dimensionless constant (K) value associated with the model is different for both experiments, where this constant, produced via experimental data, is used for construction of a homogeneous equation. Field measurements were made of soil density and moisture patterns under different vehicle tire paths with varying external pressures and number of passes. In addition, laboratory index tests were performed to determine the compaction behaviour of the same soil. Using these results, a prediction equation of dry density in terms of applied pressure and moisture content was obtained for the clay soil. A previously developed equation for sandy soil was modified for the complete range of moisture contents encountered. Estimation of shear strength for the clay soil was made using plastic and liquid limits.  相似文献

A novel one-pot synthesis of four new classes of amidothiophosphoric esters and trisamidothiophosphoric esters is developed. (1-Alkyl-2-pyrimidinylidenamido)bis(diethylamido)thiophosphates, (1-alkyl-2-pyrimidinylidenamido)bis (O-2/4-methylphenyl)thiophosphates, (3-alkyl-2-thiazolinylidenamido)bis (diethylamido)thiophosphate and (3-alkyl-2-thiazolinylidenamido)bis(O-2/4-methylphenyl)thiophosphates are obtained from the nucleophilic substitution and oxidation of N-alkyl-2-cycloiminylidenaminodichlorophosphines generated in situ from the reaction of the corresponding N-alkyl-2-aminocycloiminium halide with phosphorus trichloride and triethylamine. The synthesized thiophosphoric esters have been investigated for fungicidal properties.  相似文献
