A selective, highly sensitive, precise, and novel bioanalytical method has been developed and validated to quantify sinococuline, an active constituent present in the phytopharmaceutical drug product containing Cocculus hirsutus plant extract, in vivo. Chromatographic separation was achieved on a Luna Omega Polar-C18 bonded analytical column maintained at 45°C. The isocratic mobile phase consisted of methanol and ammonium formate buffer (60:40, v/v) at acidic pH with a low flow rate of 0.250 mL/min. Detection was performed on an API 4000 mass spectrometer using electrospray ionization in positive polarity and multiple reaction monitoring mode to achieve a lower limit of quantification of 1.50 ng/mL. Excellent accuracy and precision were obtained after extracting the analyte from plasma samples using a chemical analogue as an internal standard in the absence of an isotope-labeled compound. The extraction efficacy was evidenced from recovery study, and the analyte was found to be stable in plasma. Validation study demonstrated linearity with coefficient of correlation, r ≥ 0.99, and minimal matrix effect. This bioanalytical method was successfully applied to evaluate pharmacokinetic parameters of sinococuline from a phase I clinical trial of an aqueous extract of C. hirsutus in healthy human volunteers.  相似文献

Given a planar continuum Gaussian free field h𝒰 in a domain 𝒰 with Dirichlet boundary condition and any δ > 0, we let be a real-valued smooth Gaussian process where is the average of h𝒰 along a circle of radius δ with center v. For γ > 0, we study the Liouville first-passage percolation (in scale δ), i.e., the shortest path distance in 𝒰 where the weight of each path P is given by . We show that the distance between two typical points is for all sufficiently small but fixed γ > 0 and some constant c* > 0. In addition, we obtain similar upper bounds on the Liouville first-passage percolation for discrete Gaussian free fields, as well as the Liouville graph distance, which roughly speaking is the minimal number of euclidean balls with comparable Liouville quantum gravity measure whose union contains a continuous path between two endpoints. Our results contradict some reasonable interpretations of Watabiki's prediction (1993) on the random distance of Liouville quantum gravity at high temperatures.© 2019 Wiley Periodicals, Inc.  相似文献

Self-immolative aryl azides can react with trans-cyclooctenes (TCO), triphenylphosphines or hydrogen sulfide (H2S) to activate prodrugs, imaging probes and drug delivery systems. To date, the synthesis of polymers containing these aryl azide self-immolative linkers and their reactivity with a strained alkene (i.e., in a bioorthogonal reaction) has not been explored. Also, due to the instability of aryl azides towards light and high temperatures, the polymerization methods compatible with aryl azides are limited. Through systematic investigation of the reversible addition-fragmentation chain transfer (RAFT) and atom transfer radical polymerization (ATRP) methods, a self-immolative PEG-aryl azide block copolymer (PEG45-b-ABOC28 2 ) and a non-responsive 4-fluoroaryl block copolymer (PEG45-b-FBOC24 3 ) was prepared. ATRP provided the desired polymers in a highly controlled manner, whereas the RAFT conditions led to higher levels of aryl azide polymer degradation. The ATRP derived polymers 2 and 3 were formulated into nanoparticles of approximately 200 nm diameter, and particle triggering was demonstrated by the [3+2]-cycloaddition reaction of TCO with PEG45-b-ABOC28 2 in solution (pure polymer) and as a formulated nanoparticle. Preliminary in vitro cell viability studies suggested that the stimuli-responsive aryl azide polymers/nanoparticles are not cytotoxic up to 200 μg/ml concentrations.  相似文献

A simple, sensitive and high throughput liquid chromatography-tandem mass spectrometry method has been developed for the determination of lafutidine in human plasma. Sample clean-up involved solid phase extraction of lafutidine along with ranitidine as the internal standard from 100 μL of human plasma. The chromatographic separation is achieved within 2.5 min on a Grace Denali C18 (50 × 4.6 mm, 5 μ) column using 2 mM ammonium acetate, pH 3.0 adjusted with acetic acid and acetonitrile (20: 80, v/v) as the mobile phase. The precursor → product ion transitions for lafutidine (m/z 432.2 → 351.4) and IS (m/z 315.3 → 176.3) were monitored on a triple quadrupole mass spectrometer, operating in the multiple reaction monitoring and positive ion mode. The method is validated over a wide dynamic concentration range of 0.25–1000 ng/mL. The mean relative recovery for lafutidine across quality controls is 97.9%. The relative matrix effect between eight different plasma lots, expressed as coefficient of variation of the slopes of the calibration lines is 1.94. The method is applied to a bioequivalence study of 10 mg lafutidine tablet formulation in 26 healthy Indian male subjects under fasting condition. The reproducibility of study data is demonstrated by analysis of 93 incurred samples.  相似文献

Extant enzymes with precisely arranged multiple residues in their three-dimensional binding pockets are capable of exhibiting remarkable stereoselectivity towards a racemic mixture of substrates. However, how early protein folds that possibly featured short peptide fragments facilitated enantioselective catalytic transformations important for the emergence of homochirality still remains an intriguing open question. Herein, enantioselective hydrolysis was shown by short peptide-based nanotubes that could exploit multiple solvent-exposed residues to create chiral binding grooves to covalently interact and subsequently hydrolyse one enantiomer preferentially from a racemic pool. Single or double-site chiral mutations led to opposite but diminished and even complete loss of enantioselectivities, suggesting the critical roles of the binding enthalpies from the precise localization of the active site residues, despite the short sequence lengths. This work underpins the enantioselective catalytic prowess of short peptide-based folds and argues their possible role in the emergence of homochiral chemical inventory.  相似文献

The synthesis of calcium complexes ligated by three different chiral iminophosphonamide ligands, L- H ( L =[Ph2P{N(R)CH(CH3)Ph}2]), L′ -H ( L′ =[Ph2P{NDipp}{N(R)CH(CH3)Ph}]), (Dipp=2,6-iPr2C6H3), and L′′ -H ( L′′ =[Ph2P{N(R)CH(CH3)naph}2]), (naph=naphthyl) is presented. The resulting structures [ L 2Ca], [ L′ 2Ca], and [ L′′ 2Ca] represent the first examples of enantiopure homoleptic calcium complexes based on this type of ligands. The calcium complexes show blue–green photoluminescence (PL) in the solid state, which is especially bright at low temperatures. Whereas the emission of [ L′′ 2Ca] is assigned to the fluorescence of naphthyl groups, the PL of [ L 2Ca] and [ L′ 2Ca] is contributed by long-lived phosphorescence and thermally activated delayed fluorescence (TADF), with a strong variation of the PL lifetimes over the temperature range of 5–295 K. Furthermore, an excellent catalytic activity was found for these complexes in hydroboration of ketones at room temperature, although no enantioselectivity was achieved.  相似文献

The problem of the diffraction of surface waves, obliquely incident on a partially immersed fixed vertical barrier in deep water, is solved approximately by reducing it to the solution of an integral equation, for small angle of incidence of the incident wave. The corrections to the reflection and transmission coefficients over their normal incidence values for small angle of incidence are obtained and presented graphically for some intermediate values of wave numbers.  相似文献

We formulate a definition of isometric action of a compact quantum group (CQG) on a compact metric space, generalizing Banica's definition for finite metric spaces. For metric spaces (X,d)(X,d) which can be isometrically embedded in some Euclidean space, we prove the existence of a universal object in the category of the compact quantum groups acting isometrically on (X,d)(X,d). In fact, our existence theorem applies to a larger class, namely for any compact metric space (X,d)(X,d) which admits a one-to-one continuous map f:X→Rnf:XRn for some n   such that d0(f(x),f(y))=?(d(x,y))d0(f(x),f(y))=?(d(x,y)) (where d0d0 is the Euclidean metric) for some homeomorphism ?   of R+R+.  相似文献
