A novel UPLC–MS/MS method for simultaneous quantification of trigonelline, 4-hydroxyisoleucine,and diosgenin from Trigonella foenum-graecum extract: Application to pharmacokinetic study in healthy and type 2 diabetic rats

Trigonelline (TR), 4-hydroxyisoleucine (4-HI), and diosgenin (DG) are the main bioactives of the purified standardized extract of the popular plant Trigonella foenum-graecum L. (TFG), and it has been proven effective for the treatment of various diseases. However, to the best of our knowledge, no study has investigated the pharmacokinetic parameters of purified standardized T. foenum-graecum extract in normal and diabetic Wistar rats. The present study has developed and validated a rapid, reliable, and sensitive simultaneous ultra-performance liquid chromatography MS method to estimate these bioactives. The chromatographic separation was achieved using methanol, acetonitrile, and 0.1% formic acid with the ideal gradient flow system on a BEH Shield RP 18 column. A positive electrospray ionization mode was selected to estimate m/z values of TR (138.14 > 94.63), 4-HI (148.19 > 74.08), and DG (415.54 > 271.33). The method was robust and reproducible over the linearity range of 60–5000, 6–5000, and 15–5000 ng/mL for TR, 4-HI, and DG, respectively. Using this novel validated method, we investigated the pharmacokinetic parameters of bioactives using Phoenix WinNonlin version 8.0 (Certera) in normal and diabetic rats. The assay was successfully applied for the estimation of pharmacokinetic parameters using noncompartmental analysis. This investigation shows that the absorption rate increased, whereas distribution and elimination processes slowed down in diabetic rats compared with normal rats.     

Redox-Switchable Complexes Based on Nanographene-NHCs

A series of rhodium and iridium complexes with a N-heterocyclic carbene (NHC) ligand decorated with a perylene-diimide-pyrene moiety are described. Electrochemical studies reveal that the complexes can undergo two successive one-electron reduction events, associated to the reduction of the PDI moiety attached to the NHC ligand. The reduction of the ligand produces a significant increase on its electron-donating character, as observed from the infrared spectroelectrochemical studies. The rhodium complex was tested in the [3+2] cycloaddition of diphenylcyclopropenone and methylphenylacetylene, where it displayed a redox-switchable behavior. The neutral complex showed moderate activity, which was suppressed when the catalyst was reduced.     

Synthesis of Polylactide: Effect of Dispersion of the Initiator

The ring opening polymerization of L-lactide was studied in bulk using stannous octoate as initiator. In some experiments, triphenylphosphine, a Lewis base was also used as co-initiator. The polymerization was carried out at 130°C up to 29 h. The monomer was used after recrystallizing three times with dry toluene. Experiments were carried out using a wide range of monomer to initiator ratio. The averages and distributions of molar masses of resulting PLA have been determined by means of size exclusion chromatography, SEC. It is shown that the (mode, process) procedure of dispersion of the catalyst in polymerization system affects the molar mass distribution of the product as is evidenced by the bimodality or even trimodality observed in the SEC chromatograms.     

Conformational Dynamics Underlies Different Functions of Human KDM7 Histone Demethylases

The human KDM7 subfamily histone H3 Nϵ-methyl lysine demethylases PHF8 (KDM7B) and KIAA1718 (KDM7A) have different substrate selectivities and are linked to genetic diseases and cancer. We describe experimentally based computational studies revealing that flexibility of the region linking the PHD finger and JmjC domains in PHF8 and KIAA1718 regulates interdomain interactions, the nature of correlated motions, and ultimately H3 binding and demethylation site selectivity. F279S an X-linked mental retardation mutation in PHF8 is involved in correlated motions with the iron ligands and second sphere residues. The calculations reveal key roles of a flexible protein environment in productive formation of enzyme-substrate complexes and suggest targeting the flexible KDM7 linker region is of interest from a medicinal chemistry perspective.     

Self-Assembly of Benzimidazole-Derived Tris-NHC Ligands and AgI-Ions to Hexanuclear Organometallic Cages and Their Unusual Transmetalation Chemistry

Multi-ligand self-assembly to attain the Ag^I-N-heterocyclic carbene (NHC)-built hexanuclear organometallic cages of composition [Ag₆( 3 a , b )₄](PF₆)₆ from the reaction of benzimidazole-derived tris(azolium) salts [H₃- 3 a , b ](PF₆)₃ with Ag₂O was achieved. The molecular structures of the cages were established by X-ray diffraction studies along with NMR and MS analyses. The existence of a single assembly in solution was supported by diffusion-ordered spectroscopy (DOSY) ¹H NMR spectra. Further, transmetalation reactions of these self-assembled complexes, [Ag₆( 3 a , b )₄](PF₆)₆, with Cu^I/Au^I-ions provided various coinage metal-NHC complexes having diverse molecular compositions, which included the first example of a hexanuclear Cu^I-dodecacarbene complex, [Cu₆( 3 b )₄](PF₆)₆.
