High-grade epithelial ovarian cancer is a fatal disease in women frequently associated with drug resistance and poor outcomes. We previously demonstrated that a marine-derived compound MalforminA1 (MA1) was cytotoxic for the breast cancer cell line MCF-7. In this study, we aimed to examine the effect of MA1 on human ovarian cancer cells. The potential cytotoxicity of MA1was tested on cisplatin-sensitive (A2780S) and cisplatin-resistant (A2780CP) ovarian cancer cell lines using AlamarBlue assay, Hoechst dye, flow cytometry, Western blot, and RT-qPCR. MA1 had higher cytotoxic activity on A2780S (IC50 = 0.23 µM) and A2780CP (IC50 = 0.34 µM) cell lines when compared to cisplatin (IC50 = 31.4 µM and 76.9 µM, respectively). Flow cytometry analysis confirmed the cytotoxic effect of MA1. The synergistic effect of the two drugs was obvious, since only 13% of A2780S and 7% of A2780CP cells remained alive after 24 h of treatment with both MA1 and cisplatin. Moreover, we examined the expression of bcl2, p53, caspase3/9 genes at RNA and protein levels using RT-qPCR and Western blot, respectively, to figure out the cell death mechanism induced by MA1. A significant down-regulation in bcl2 and p53 genes was observed in treated cells compared to non-treated cells (p < 0.05), suggesting that MA1 may not follow the canonical pathway to induce apoptosis in ovarian cancer cell lines. MalforminA1 showed promising anticancer activity by inducing cytotoxicity in cisplatin-sensitive and cisplatin-resistant cancer cell lines. Interestingly, a synergistic effect was observed when MA1 was combined with cisplatin, leading to it overcoming its resistance to cisplatin.  相似文献

A new series of 3,4-disubstituted quinazolin-2-ones, with potential T-type calcium channel antagonist activity, and new 4-methylene-quinazolin-2-ones, promising catalysts as N-heterocyclic olefins, have been prepared in good yield by a simple reaction between 2-aminobenzophenone, or 2-aminoacetophenone, and cyanomethyl anion electrogenerated by acetonitrile reduction at a graphite electrode, followed by the addition of different organic isocyanates and subsequent heterocyclization.  相似文献

This work aims to develop a new mineral porous tubular membrane based on mineral coal fly ash. Finely ground mineral coal powder was calcinated at 700 °C for about 3 h. The elaboration of the mesoporous layer was performed by the slip casting method using a suspension made of the mixture of fly ash powder, water and PVA. The obtained membrane was submitted to a thermal treatment which consists in drying at room temperature for 24 h then a sintering at 800 °C. SEM photographs indicated that the membrane surface was homogeneous and did not present any macro defects (cracks, etc.). The average pore diameter of the active layer was 0.25 μm and the thickness was around 20 μm. The membrane permeability was 475 l/h m2 bar.This membrane was applied to the treatment of the dying effluents generated by the washing baths in the textile industry. The performances in term of permeate flux and efficiency were determined and compared to those obtained using a commercial alumina microfiltration membrane. Almost the same stabilised permeate flux was obtained (about 100 l/h m2). The quality of permeate was almost the same with the two membranes: the COD and color removal was 75% and 90%, respectively.  相似文献

Surfactant-modified bentonite was synthesized by replacing adsorbed Na+ with long-chain alkyl quaternary ammonium cation, hexadecyltrimethylammonium bromide (HDTMAB). The sorption isotherms of phenol, p-chlorophenol, and 2,4-dichlorophenol were modeled according to the Langmuir and Freundlich equations. The Langmuir isotherm was found to describe the equilibrium adsorption data well. The mechanisms and characteristics of sorption of these ionizable organic contaminants onto surfactant-modified bentonite from water were investigated systematically and described quantitatively. The sorption properties are affected by the treatment conditions, such as amount of organobentonite, and the properties of organic compounds. Nickel(II)-based immobilized metal affinity chromatography (IMAC) has been used to capture from standard samples the hydroxamate-containing siderophores, acetohydroxamic acid (ahaH), suberodihydroxamic acid (shaH(2)) or desferrioxamine B (DFOB) in recoveries ranging between 35-90%. The capacity of a 1 mL Ni(II)-charged IMAC column towards DFOB capture at the pH optima of 8.9 is approximately 3000 nmol. This method has been used for the direct capture of DFOB (approximately 65% recovery) from the untreated supernatant of iron-deprived cultures of Streptomyces pilosus, the soil bacterium from which DFOB was first discovered. In addition to selecting for DFOB and a related siderophore, two other Fe(III)-responsive species have been identified from RP-HPLC analysis of the IMAC-processed eluant from the S. pilosus supernatant. Since the characterisation of siderophores from natural systems is hampered by the low yields obtained from traditional purification methods, this IMAC-based affinity method offers significant potential for improving yields of this key class of bioligands and other small molecule metabolites with affinities to IMAC-compatible metal ions.  相似文献

Budu is a famous Malaysian fish sauce, usually used as seasoning and condiment in cooking. Budu is produced by mixing fish and salt at certain ratio followed by fermentation for six months in closed tanks. In this study, four commercial brands of Budu were analyzed for their chemical properties (pH, salt content and volatile compounds). The pH of Budu samples ranged from 4.50-4.92, while the salt (NaCl) content ranged between 11.80% and 22.50% (w/v). For tentative identification of volatile flavor compounds in Budu, two GC columns have been used, DB-WAX and HP-5MS. A total of 44 volatile compounds have been detected and 16 were common for both columns. 3-Methyl-1-butanol, 2-methylbutanal, 3-methylbutanal, dimethyl disulfide, 3-(methylthio)-propanal, 3-methylbutanoic acid and benzaldehye have been identified as the aroma-active compounds in Budu due to their lower threshold values.  相似文献

The goal of this investigation was to evaluate the in vitro and in vivo efficiency of Raphanus raphanistrum extracts against Fusarium and Pythiaceae species associated with apple and peach seedling decline in Tunisian nurseries. A chemical composition of organic extracts was accomplished using liquid chromatography, thin layer chromatography, and gas chromatography analysis. The in vitro test of three aqueous extract doses of R. raphanistrum against some apple and peach decline agents showed its efficacy in reducing mycelia growth. The in vivo assay of fine powder of this plant on peach seedlings revealed that treatment 8-weeks before the inoculation and planting was more efficient than the treatment before one week. This experiment revealed that the root weight of peach seedlings inoculated by F. oxysporum was improved to 207.29%. For apple seedlings, the treatment 8 weeks before the inoculation and plantation was more efficient than the treatment one week before; it reduced the root browning index. The study of R. raphanistrum chemical composition and its efficiency showed that the glucosinolates products: nitrile (4-Hydroxy-3-(4-methylphenylthio) butane nitrile, benzene acetonitrile, 4-fluoro,butane nitrile, 4-hydroxy-3-[(4-methylphenyl) thio] nitrile), and thiocyanate molecules (thiocyanic acid, ethyle) are responsible for the anti-fungal activities.  相似文献

The reactions of Cu (II), Zn (II) and Cd (II) chloride or bromide with (E)-1-(3,4-dimethoxybenzylidene)-4-methylthiosemicarbazone (MTSVT) lead to the formation of new complexes. They were characterized by spectroscopic studies: IR, 1H and 13C NMR. The crystal structures of the compounds [MTSVT] ( L ), [ZnBr2(MTSVT)2] ( 2 ), [CdCl2(MTSVT)2] ( 3 ) and [CdBr2(MTSVT)2.H2O] ( 4 ) were determined by X-ray diffraction. For complexes 2 – 4 , the ion is coordinated through the sulfur atom. All compounds were tested for their antifungal activity against human pathogenic fungi Candida albicans and Aspergillus fumigatus, and for their antibacterial activity against Gram (+) Bacillus subtilis and Enterococcus faecalis as well as against Gram (−) bacteria such as Paracoccus yeei and Acinetobacter baumanii. The results indicated that the metal complexes exhibited a marked enhancement in antibacterial activity compared with the parent Schiff base.  相似文献

Desferrioxamine B (DFOB) was biotinylated at the pendant amine using solid-phase organic synthesis (SPOS) on a matrix used conventionally for metal affinity chromatography. The strength of the DFOB-matrix coordinate bonds was functionally equivalent to a covalent bond which underpinned the veracity of the SPOS format. After washing excess reagents, biotin-DFOB was eluted from the matrix with water at pH 6.  相似文献
