The development of nanotechnology has led to the design of cutting‐edge nanomaterials with increasing levels of complexity. Although “traditional” solid, uniform nanoparticles are still the most frequently reported structures, new generations of nanoparticles have been constantly emerging over the last several decades. The outcome of this nano‐art extends beyond nanomaterials with alternative compositions and/or morphologies. The current state‐of‐the‐art allows for the design of nanostructures composed of different building blocks that exhibit diverse properties. Furthermore, those properties can be a reflection of either individual features, which are characteristic of a particular building block alone, and/or synergistic effects resulting from interactions between building blocks. Therefore, the unique structures as well as the outstanding properties of nanorattles have attracted increasing attention for possible biomedical and industrial applications. Although these nanoparticles resemble core–shell particles, they have a distinctive feature, which is a presence of a void that provides a homogenous environment for the encapsulated core. In this Review, we give a comprehensive insight into the fabrication of nanorattles. A special emphasis is put on the choice of building blocks as well as the choice of preparation method, because those two aspects further influence properties and thus possible future applications, which will also be discussed.  相似文献

The purpose of this study was to develop and validate a high-sensitivity methodology for identifying one of the most used drugs—ketamine. Ketamine is used medicinally to treat depression, alcoholism, and heroin addiction. Moreover, ketamine is the main ingredient used in so-called “date-rape” pills (DRP). This study presents a novel methodology for the simultaneous determination of ketamine based on the Dried Blood Spot (DBS) method, in combination with capillary electrophoresis coupled with a mass spectrometer (CE-TOF-MS). Then, 6-mm circles were punched out from DBS collected on Whatman DMPK-C paper and extracted using microwave-assisted extraction (MAE). The assay was linear in the range of 25–300 ng/mL. Values of limits of detection (LOD = 6.0 ng/mL) and quantification (LOQ = 19.8 ng/mL) were determined based on the signal to noise ratio. Intra-day precision at each determined concentration level was in the range of 6.1–11.1%, and inter-day between 7.9–13.1%. The obtained precision was under 15.0% (for medium and high concentrations) and lower than 20.0% (for low concentrations), which are in accordance with acceptance criteria. Therefore, the DBS/MAE/CE-TOF-MS method was successfully checked for analysis of ketamine in matrices other than blood, i.e., rose wine and orange juice. Moreover, it is possible to identify ketamine in the presence of flunitrazepam, which is the other most popular ingredient used in DRP. Based on this information, the selectivity of the proposed methodology for identifying ketamine in the presence of other components of rape pills was checked.  相似文献

Background: Carnosine is a dipeptide molecule (β-alanyl-l-histidine) with anti-inflammatory, antioxidant, anti-glycation, and chelating properties. It is used in exercise physiology as a food supplement to increase performance; however, in vitro evidence suggests that carnosine may exhibit anti-cancer properties. Methods: In this study, we investigated the effect of carnosine on breast, ovarian, colon, and leukemic cancer cell proliferation. We further examined U937 promonocytic, human myeloid leukemia cell phenotype, gene expression, and cytokine secretion to determine if these are linked to carnosine’s anti-proliferative properties. Results: Carnosine (1) inhibits breast, ovarian, colon, and leukemic cancer cell proliferation; (2) upregulates expression of pro-inflammatory molecules; (3) modulates cytokine secretion; and (4) alters U937 differentiation and phenotype. Conclusion: These effects may have implications for a role for carnosine in anti-cancer therapy.  相似文献

In implantable materials, surface topography and chemistry are the most important in the effective osseointegration and interaction with drug molecules. Therefore, structural and surface modifications of nanostructured titanium dioxide (TiO2) layers are reported in the present work. In particular, the modification of annealed TiO2 samples with —OH groups and silane derivatives, confirmed by X-ray photoelectron spectroscopy, is shown. Moreover, the ibuprofen release process was studied regarding the desorption-desorption-diffusion (DDD) kinetic model. The results proved that the most significant impact on the release profile is annealing, and further surface modifications did not change its kinetics. Additionally, the cell adhesion and proliferation were examined based on the MTS test and immunofluorescent staining. The obtained data showed that the proposed changes in the surface chemistry enhance the samples’ hydrophilicity. Moreover, improvements in the adhesion and proliferation of the MG-63 cells were observed.  相似文献

Cisplatin is a drug frequently used in chemotherapy of various types of tumors due to its strong cytostatic activity against cancer cells. However, this therapy is not free from severe side effects related to the nonselective action of the drug. The solution to this problem could be the application of drug-targeted delivery systems (DTDSs). Gold nanoparticles can be used in such systems as selective drug carriers, ensuring its transportation through the bloodstream to destination tissue. The method of DTDSs analysis providing qualitative and quantitative information about the formation of this conjugation is crucial to establish the kinetics of reaction and stoichiometry of reagents, which ensures the best drug binding rate. Moreover, the status of so far proposed techniques/methods dedicated to elaborating the course of DTDSs formation is preliminary and in majority guarantee only the confirmation of drug‒carrier conjugate formation. In this paper, we demonstrate the procedures of reagents’ preparation and cisplatin‒gold nanoparticles DTDS formation, which have a significant influence on the rate and stoichiometry of the reaction. We also present the novel application of CE-ICP-MS hyphenation for effective separation and online monitoring of all components of the reaction mixture.  相似文献

Trichothecene mycotoxins are sesquiterpenoid compounds primarily produced by fungi in taxonomical genera such as Fusarium, Myrothecium, Stachybotrys, Trichothecium, and others, under specific climatic conditions on a worldwide basis. Fusarium mold is a major plant pathogen and produces a number of trichothecene mycotoxins including deoxynivalenol (or vomitoxin), nivalenol, diacetoxyscirpenol, and T-2 toxin, HT-2 toxin. Monogastrics are sensitive to vomitoxin, while poultry and ruminants appear to be less sensitive to some trichothecenes through microbial metabolism of trichothecenes in the gastrointestinal tract. Trichothecene mycotoxins occur worldwide however both total concentrations and the particular mix of toxins present vary with environmental conditions. Proper agricultural practices such as avoiding late harvests, removing overwintered stubble from fields, and avoiding a corn/wheat rotation that favors Fusarium growth in residue can reduce trichothecene contamination of grains. Due to the vague nature of toxic effects attributed to low concentrations of trichothecenes, a solid link between low level exposure and a specific trichothecene is difficult to establish. Multiple factors, such as nutrition, management, and environmental conditions impact animal health and need to be evaluated with the knowledge of the mycotoxin and concentrations known to cause adverse health effects. Future research evaluating the impact of low-level exposure on livestock may clarify the potential impact on immunity. Trichothecenes are rapidly excreted from animals, and residues in edible tissues, milk, or eggs are likely negligible. In chronic exposures to trichothecenes, once the contaminated feed is removed and exposure stopped, animals generally have an excellent prognosis for recovery. This review shows the occurrence of trichothecenes in food and feed in 2011–2020 and their toxic effects and provides a summary of the discussions on the potential public health concerns specifically related to trichothecenes residues in foods associated with the exposure of farm animals to mycotoxin-contaminated feeds and impact to human health.
