Luciferin‐regenerating enzyme (LRE) contributes to in vitro recycling of D‐luciferin. In this study, reinvestigation of the luciferase‐based LRE assay is reported. Here, using quick change site‐directed mutagenesis seven T‐LRE (Lampyris turkestanicusLRE) mutants were constructed and the most functional mutant of T‐LRE (T69R) was selected for this research and the effects of D‐ and L‐cysteine on T69R T‐LRE‐luciferase‐coupled assay are examined. Our results demonstrate that bioluminescent signal of T69R T‐LRE‐luciferase‐coupled assay increases and then reach equilibrium state in the presence of 5 mm D‐cysteine. In addition, results reveal that 5 mm D‐ and L‐cysteine in the absence of T69R T‐LRE cause a significant increase in bioluminescence intensity of luciferase over a long time as well as decrease in decay rate. Based on activity measurements, far‐UV CD analysis, ANS fluorescence and DLS (Dynamic light scattering) results, D‐cysteine increases the activity of luciferase due to weak redox potential, antiaggregatory effects, induction of changes in conformational structure and kinetics properties. In conclusion, in spite of previous reports on the effect of LRE on luciferase bioluminescent intensity, the majority of increase in luciferase light output and time‐course originate from the direct effects of D‐cysteine on structure and activity of firefly luciferase.  相似文献

This research paper comprises of the synthesis of polypyrrole (PPy)-Fe2O3 nanocomposites by employing the in situ chemical oxidative polymerization method. The concentration of the filler material was adjusted between 10–50 wt % of PPy. The synthesized nanocomposites were characterized by using X-ray diffraction (XRD). Magnetic analysis and DC electrical conductivity of the samples were carried out using vibrating sample magnetometer (VSM) and two probe DC conductivity method, point towards magnetically active and electrically conductive samples. The magnetic parameters under applied magnetic field demonstrated that the values of coercivity (H c ), saturation magnetization (M s ) and remanence (M r ) can be tailored by carefully controlling the amount of dopant material into the nanocomposites indicating their suitability for controllable switching devices and microwave absorption applications. The DC electrical conductivity showed an increase up to 20 wt % of filler material and thereafter a decrease in the conductivity of nanocomposites with increase in filler content is observed. Thermogravimetric analysis (TGA) showed an increase in thermal stability with an increase in ferrite content in nanocomposites.  相似文献

We experimentally investigate the gain saturation effect of a piece of 8 m long Erbium doped fiber (EDF), we introduce a theoretical model for the EDF ring loop, and our simulation results show very good cross-gain modulation (XGM) and wavelength conversion. We also experimentally investigate the XGM in an EDF ring loop system. Based on the study of the XGM in the EDF ring loop system, a wavelength conversion is designed with the EDF ring loop system. The EDF ring loop systems as a wavelength conversion is experimentally demonstrated by converting a sinusoidal modulated optical signal at wavelength of 1551 nm to an optical signal at wavelength of 1553.3 nm.  相似文献

Fluorine doped SnO2 nanostructures were grown using ultrasonic assisted sol–gel method. The gel was obtained by dissolving stannous chloride in methanol with ammonium fluoride as dopant followed by irradiation with ultrasonic vibrations. Obtained samples were characterized by structural, morphological and optical studies. All the peaks in the X-ray diffractograms are identified and indexed as tetragonal cassiterite structure. Negative slope of Williamson–Hall plots indicates compressive strain. Particle size of SnO2 nanostructures is decreases with increases in concentration of fluorine doping. Atomic force microscopy, scanning electron microscopy and transmission electron microscopy studies confirm the formation of ring like porous structures and then hollow tube like growth with increase in the fluorine concentration. Peaks in Raman spectra also indicate strong confinement in SnO2 particles. Distinct peaks in the PL spectra make the structure suitable for photovoltaic applications.  相似文献
