Economic evaluations of the capital costs for anaerobic digestion systems for gas production show that the reactor is a significant
cost component. The successful application of high solids digestion of processed MSW (e.g., greater than 10% solids within
the digester) would allow a decrease in reactor volume with maintenance of relatively high gas production rates. However,
high solids slurries do not mix well in conventional stirred tank reactors. A horizontal shaft, hydraulically driven reactor
was designed and fabricated to test the anaerobic digestion of high solids concentrations. Digester performance was evaluated
as a function of experimental parameters such as nutrient requirements, feeding rates, pH control, and agitator design/ rotation
speed; horsepower of mixing was also evaluated for the reactor. Several startup protocols were examined to obtain a biologically

The process of converting lignocellulosic biomass to ethanol via fermentation depends on developing economic sources of cellulases.Trichoderma reesei cellobiohydrolase (CBH) I is a key enzyme in the fungal cellulase system; however, specific process application requirements make modification of the enzyme by site-directed mutagenesis (SDM) an attractive goal. To undertake SDM investigations, an efficient, cellulase-free host is required. To test the potential ofEscherichia coli as a host, T.reesei CBH I cDNA was expressed inE. coli strain GI 724 as a C-terminal fusion to thermostable thioredoxin protein. Full-length expression of CBH I was subsequently verified by molecular weight, Western blot analysis, and activity on soluble substrates.
A series of correlations was made between the performance of 15 wood species in simultaneous saccharification and fermentation (SSF) and their respective chemical compositions. A compelling inverse trend (p < 0.001) was demonstrated between the percent conversion of glucan to ethanol during SSF and the Klason lignin content of the wood samples before dilute acid pretreatment. No significant relationships were found between the glucan, xylan, and ash compositions of the native wood samples and ethanol yield. This observation is unique and provides a convenient predictor of biomass conversion efficiency. 相似文献
Tone languages such as Thai and Mandarin Chinese use differences in fundamental frequency (F0, pitch) to distinguish lexical meaning. Previous behavioral studies have shown that native speakers of a non-tone language have difficulty discriminating among tone contrasts and are sensitive to different F0 dimensions than speakers of a tone language. The aim of the present ERP study was to investigate the effect of language background and training on the non-attentive processing of lexical tones. EEG was recorded from 12 adult native speakers of Mandarin Chinese, 12 native speakers of American English, and 11 Thai speakers while they were watching a movie and were presented with multiple tokens of low-falling, mid-level and high-rising Thai lexical tones. High-rising or low-falling tokens were presented as deviants among mid-level standard tokens, and vice versa. EEG data and data from a behavioral discrimination task were collected before and after a two-day perceptual categorization training task.
Results
Behavioral discrimination improved after training in both the Chinese and the English groups. Low-falling tone deviants versus standards elicited a mismatch negativity (MMN) in all language groups. Before, but not after training, the English speakers showed a larger MMN compared to the Chinese, even though English speakers performed worst in the behavioral tasks. The MMN was followed by a late negativity, which became smaller with improved discrimination. The High-rising deviants versus standards elicited a late negativity, which was left-lateralized only in the English and Chinese groups.
Conclusion
Results showed that native speakers of English, Chinese and Thai recruited largely similar mechanisms when non-attentively processing Thai lexical tones. However, native Thai speakers differed from the Chinese and English speakers with respect to the processing of late F0 contour differences (high-rising versus mid-level tones). In addition, native speakers of a non-tone language (English) were initially more sensitive to F0 onset differences (low-falling versus mid-level contrast), which was suppressed as a result of training. This result converges with results from previous behavioral studies and supports the view that attentive as well as non-attentive processing of F0 contrasts is affected by language background, but is malleable even in adult learners. 相似文献
A suite of keV polyatomic or 'cluster' projectiles was used to bombard unoxidized and oxidized self-assembled monolayer surfaces. Negative secondary ion yields, collected at the limit of single ion impacts, were measured and compared for both molecular and fragment ions. In contrast to targets that are orders of magnitude thicker than the penetration range of the primary ions, secondary ion yields from polyatomic projectile impacts on self-assembled monolayers show little to no enhancement when compared with monatomic projectiles at the same velocity. Rising costs for landfill disposal of municipal sewage residues have prompted evaluation of alternative methods for reducing
the bulk of the final waste. Representative samples of municipal sewage sludge residues were obtained from three major treatment
plants in the United States, including Los Angeles (Hyperion), Denver (North Metro), and Chicago (Stickney). The majority
of the treated, dewatered sewage sludge solids was found to be volatile (50–60%) and, presumably, biodegradable. Additionally,
much of the volatile content was solubilized by both acid detergent fiber and neutral detergent fiber treatments, and was
presumed to be proteineous microbial biomass in nature. Both low- and high-solids anaerobic digester systems, as well as the
standard biochemical methane potential (BMP) assay, were utilized to evaluate the anaerobic digestibility of these sewage
sludge residues. The low methane yields and, thus, the poor organic waste conversion indicated the need for treatment prior
to bioconversion. The effectivenesss of various pretreatments based on assessment of increased soluble protein or organics
and anaerobic digestibility as determined by the BMP assay was evaluated. 相似文献
Anaerobic digestion represents one of several commercially viable processes to convert woody biomass, agricultural wastes, and municipal solid wastes to methane gas, a useful energy source. This process occurs in the absence of oxygen, and is substantially less energy intensive than aerobic biological processes designed for disposal purposes. The anaerobic conversion process is a result of the synergistic effects of various microorganisms, which serve as a consortium. The rate-limiting step of this conversion process has been identified as the hydrolysis of cellulose, the major polymeric component of most biomass and waste feedstocks. Improvements in process economics therefore rely on improving the kinetic and physicochemical characteristics of cellulose degrading enzymes. The most thoroughly studied cellulase enzymes are produced by aerobic fungi, namely Trichoderma reesei. However, the pH and temperature optima of fungal cellulases make them incompatible for use in anaerobic digestion systems, and the major populations of microorganisms involved in cellulase enzyme production under anaerobic digestion conditions are various bacterial producers. The current state of understanding of the major groups of bacterial cellulase producers is reviewed in this paper. Also addressed in this review are recently developed methods for the assessment of actual cellulase activity levels, reflective of the digester "hydrolytic potential," using a series of detergent extractive procedures. 相似文献
A new thermostable endoglucanase,Acidothermus cellulolyticus E1, and another bacterial endoglucanase, E5 fromThermomonospora fusca, each exhibit striking synergism with a fungal cellobiohydrolase (Trichoderma reesei CBH I) in the saccharification of microcrystalline cellulose. In neither case did the ratio of endoglucanase to exoglucanase
that demonstrated maximum synergism coincide exactly with the ratio that actually released the maximum quantity of soluble
sugar for a given total cellulase loading. The difference between the two ratios, after significant hydrolysis of the substrate,
was considerably larger in the case ofA. cellulolyticus E1. For both endoglucanase pairings with CBH I, the offset between the ratio for maximum synergism and the ratio for maximal

Two recombinant ferulic acid esterases from Penicillium funiculosum produced in Aspergillus awamori were evaluated for their ability to improve the digestibility of pretreated corn stover. The genes, faeA and faeB, were cloned from P. funiculosum and expressed in A. awamori using their native signal sequences. Both enzymes contain a catalytic domain connected to a family 1 carbohydrate-binding
module by a threonine-rich linker peptide. Interestingly, the carbohydrate binding-module is N-terminal in FaeA and C-terminal
in FaeB. The enzymes were purified to homogeneity using column chromatography, and their thermal stability was characterized
by differential scanning microcalorimetry. We evaluated both enzymes for their potential to enhance the cellulolytic activity
