THE SYNTHESIS OF PROTECTIVE FRAGMENTS OF α-HUMAN ATRIAL NATRIURETIC POLYPEPTIDE (α-hANP)

In this paper the synthesis of α-human atrial natriuretic polypeptide fragments (positions, 1-4, 5-10, 11-16, 17-22, 23-28) by solution method is reported. In the synthesis of some peptides, the re-sults from mixed anhydride procedure and active ester procedure are compared. In the synthesis of otherpeptides, the effects of reaction temperature, basicity of catalyst, and reaction time on the specific rota-tions and yields of these compounds are observed. The sequence of amino acids of all the protective poly-peptides is confirmed by FAB-MS.

多肽研究——Ⅰ.α-人心房肽(α-hANP)带保护基的片段的合成

本文报道了与α-人心房肽(α-hANP)的A_1—A_4四肽及A_5—A_(10),A_(11)—A_(16),A_(17)—A_(22),A_(23)—A_(28)四个六肽片段对应的带保护基片段的液相法合成。在有些肽段的合成中,比较了混合酸酐法和活泼酯法所得的结果。在另一些肽段的合成中,观察了反应温度、催化剂的碱性及反应时间对产物产率的影响。上述带保护基的片段均由快原子轰击质谱测定氨基酸顺序,确认结构准确无误。     

多肽研究(Ⅳ)——α-人心房肽片段的快原子轰击质谱(FAB-MS)研究

采用FAB-MS测定α-人心房肽片段的氨基酸序列。通过分析碎片离子的m/z值,可以明确表达各肽段的氨基酸序列。在分子离子区都是[M+H]~+峰,所有片段羰基的α断裂都有肽键断裂和碳碳键断裂两种方式。
