Liposomes play an important role in medical and pharmaceutical science as, nanoscale drug carriers. One of the most important features is their size and size distribution, influencing both their bio-distribution and their targeting efficiency to tumors and also therapeutic activity of liposomal antitumor drugs. In this study, the effect of preparation method and molecular interaction on size and shape of liposome was studied. The size and shape characterization of liposomes was performed by asymmetrical flow field-flow fractionation coupled online with multi-angle light scattering (AF4-MALS). The size distributions obtained by AF4-MALS were compared to mean particle sizes and size distribution measured with other standard method such as Photon Correlation Spectroscopy (PCS). Furthermore, the effect of molecular interaction (hydrophilic and hydrophobic model drugs) on liposomal structure was assessed.  相似文献

One of the most important name reactions in organic chemistry, is the Diels‐Alder cycloaddition reaction. It is a chemical reaction between a conjugated diene and a substituted alkene, commonly termed the dienophile to construct a substituted cyclohexene derivative. It is the stereotypical example of a pericyclic reaction with a concerted mechanism. In synthesis, the use of electricity instead of stoichiometric amounts of oxidant or reducing agents is definitely appealing for economic, ecological and selective, reasons. In this review, we try to underscore the combination of the electrosynthesis with Diels‐Alder cycloaddition reaction to establish of a powerful synthetic tool which may encourage synthetic organic chemists to use it in the future.  相似文献

This study presents the application of the mixture design technique to develop an optimal liposome formulation by using the different lipids in type and percentage (DOPC, POPC and DPPC) in liposome composition. Ten lipid mixtures were generated by the simplex-centroid design technique and liposomes were prepared by the extrusion method. Liposomes were characterized with respect to size, phase transition temperature, ζ-potential, lamellarity, fluidity and efficiency in loading calcein. The results were then applied to estimate the coefficients of mixture design model and to find the optimal lipid composition with improved entrapment efficiency, size, transition temperature, fluidity and ζ-potential of liposomes. The response optimization of experiments was the liposome formulation with DOPC: 46%, POPC: 12% and DPPC: 42%. The optimal liposome formulation had an average diameter of 127.5 nm, a phase-transition temperature of 11.43 °C, a ζ-potential of -7.24 mV, fluidity (1/P)(TMA-DPH)((?)) value of 2.87 and an encapsulation efficiency of 20.24%. The experimental results of characterization of optimal liposome formulation were in good agreement with those predicted by the mixture design technique.  相似文献

New hydroxyl‐terminated amphiphilic block copolymers (HO‐ABPs) having pendant t‐butyl groups for pH‐responsiveness and terminal OH groups for bioconjugation are reported. These HO‐ABPs consist of hydrophilic poly(oligo(ethylene oxide) monomethyl ether methacrylate) and hydrophobic poly(t‐butyl methacrylate) blocks and were synthesized by a consecutive atom transfer radical polymerization in the presence of an OH‐terminated bromine initiator. Aqueous self‐assembly of HO‐ABPs resulted in colloidally stable micellar aggregates being capable of encapsulating hydrophobic guest molecules. They were nontoxic to cells and destabilized in response to low pH. A facile bioconjugation of HO‐ABP micelles for active targeting is demonstrated by conjugation with biotin (vitamin H) and competitive assay exhibiting >93% ABP chains conjugated with biotin in each micelle. © 2013 Wiley Periodicals, Inc.
