The present research work is designed to prepare and evaluate piperine liposomes and piperine–chitosan-coated liposomes for oral delivery. Piperine (PPN) is a water-insoluble bioactive compound used for different diseases. The prepared formulations were evaluated for physicochemical study, mucoadhesive study, permeation study and in vitro cytotoxic study using the MCF7 breast cancer cell line. Piperine-loaded liposomes (PLF) were prepared by the thin-film evaporation method. The selected liposomes were coated with chitosan (PLFC) by electrostatic deposition to enhance the mucoadhesive property and in vitro therapeutic efficacy. Based on the findings of the study, the prepared PPN liposomes (PLF3) and chitosan coated PPN liposomes (PLF3C1) showed a nanometric size range of 165.7 ± 7.4 to 243.4 ± 7.5, a narrow polydispersity index (>0.3) and zeta potential (−7.1 to 29.8 mV). The average encapsulation efficiency was found to be between 60 and 80% for all prepared formulations. The drug release and permeation study profile showed biphasic release behavior and enhanced PPN permeation. The in vitro antioxidant study results showed a comparable antioxidant activity with pure PPN. The anticancer study depicted that the cell viability assay of tested PLF3C2 has significantly (p < 0.001)) reduced the IC50 when compared with pure PPN. The study revealed that oral chitosan-coated liposomes are a promising delivery system for the PPN and can increase the therapeutic efficacy against the breast cancer cell line.  相似文献

研究了胡椒碱在碳纳米管修饰电极上的电化学行为,在pH为6.4的磷酸盐缓冲溶液中,胡椒碱在-1.12V(vs.SCE)处有一灵敏的还原峰.与裸电极相比,还原峰电位明显正移,峰电流显著增加,表明该修饰电极对胡椒碱的还原反应具有明显的催化作用.峰电流与胡椒碱的浓度在10-6~10-5mol/L范围内呈良好的线性关系(r=0.995),检出限为2.0×10-7mol/L.同时,计算了电荷转移数和扩散系数,考查了修饰电极的重现性,7次平行测量的RSD为4.96%.  相似文献

An attempt has been made to develop and validate a simultaneous HPLC method for novel approach of drug release via oil‐in‐water (o/w) nanoemulsion formulation and Habb‐e‐Khardal Unani tablet containing piperine and guggul sterones E and Z as main ingredients. Nanoemulsion was prepared by titration method using sefsol‐218 as an oily phase, cremophor‐EL as a surfactant, transcutol as a co‐surfactant and distilled water as an aqueous phase. The formulation was optimized on the basis of thermodynamic stability and dispersibilty test. The nanoformulation was evaluated for particle size, surface morphology, electrical conductivity and viscosity determination. The in vitro dissolution was carried out by dialysis bag method. Drugs were quantified using an HPLC method developed in‐house with a C18 column as stationary phase and acetonitrile and water as mobile phase at λmax of 240 nm. The optimized formulation showed higher drug release, lower droplet size and less viscosity as compared with the conventional Habb‐e‐Khardal Unani tablet. The present study illustrated the potential of nanoemulsion dosage form in improving biopharmaceutic performance of piperine and guggul sterone. The HPLC method was also found to be quite sufficient for the routine quality control of formulations containing piperine and guggul sterone E and Z as ingredients and also for in vitro drug release studies. Switchable‐hydrophilicity solvent liquid‐liquid microextraction and dispersive liquid‐liquid microextraction were compared for the extraction of piperine from Piper nigrum L. prior to its analysis by using high‐performance liquid chromatography with UV detection. Under optimum conditions, limits of detection and quantitation were found as 0.2–0.6 and 0.7–2.0 μg/mg with the two methods, respectively. Calibration graphs showed good linearity with coefficients of determination (R2) higher than 0.9962 and percentage relative standard deviations lower than 6.8%. Both methods were efficiently used for the extraction of piperine from black and white pepper samples from different origins and percentage relative recoveries ranged between 90.0 and 106.0%. The results showed that switchable‐hydrophilicity solvent liquid‐liquid microextraction is a better alternative to dispersive liquid‐liquid microextraction for the routine analysis of piperine in food samples. A novel scaled‐up dispersive liquid‐liquid microextraction method was also proposed for the isolation of piperine providing a yield of 102.9 ± 4.9% and purity higher than 98.0% as revealed by NMR spectroscopy.  相似文献

利用荧光光谱法及紫外吸收光谱法结合计算机模拟技术研究了在模拟生理条件下胡椒碱(piperine)与牛血清白蛋白(Bovine Serum Albumin, BSA)的相互作用. 根据荧光猝灭的有关方程分别求得不同温度下(298, 308和318 K)药物与蛋白相互作用的结合常数、结合位点数及键合距离. 实验所得到的热力学参数(ΔH&#1256;=-9.55 kJ/mol, ΔS&#1256;=46.75 J&#8226;mol-1&#8226;K-1)表明维持药物与蛋白质的相互作用力主要是疏水作用和静电作用. 分子模拟的结果显示了胡椒碱与BSA的键合机理和键合模式, 表明维持药物与蛋白质的相互作用力主要是疏水作用和氢键(位于氨基酸残基His 242, Arg 222和Arg 218位). 此外, 基于胡椒碱的荧光猝灭效应, 首次探讨了药物-蛋白质体系的几种物理化学参数, 包括电荷密度(δ)、离解常数(Kd)及量子产率(F)的变化效应.  相似文献

Abstract

GC-EIMS analysis, antifungal- and anti-aflatoxigenic activities of the ethanolic extract of Capsicum chinense and Piper nigrum fruits and their main bioactive compounds were evaluated upon Aspergillus parasiticus. The GC-EIMS analysis showed capsaicin (50.49%) and piperine (95.94%) as the major constituents in C. chinense and P. nigrum, respectively. MIC50 values revealed that capsaicin (39?μg/mL) and piperine (67?μg/mL) were lower than those from fruit extracts of C. chinense (381?μg/mL) and P. nigrum (68?μg/mL). Extracts and bioactive compounds showed anti-aflatoxigenic activity. Maximum aflatoxin inhibition occurred at 150?µg/mL of extracts and compounds. The present study showed satisfactory results concerning the effects of ethanolic extract of C. chinense and P. nigrum fruits upon A. parasiticus, showing the capabilities of inhibiting fungal growth development and altering aflatoxins production.  相似文献

运用荧光及紫外-可见吸收光谱法研究了胡椒酸丁二醇单酯(简称BPM)与牛血清白蛋白(BSA)的相互作用。实验结果表明,胡椒酸丁二醇单酯与BSA形成基态复合物导致BSA内源性荧光猝灭,猝灭机理主要为静态猝灭和非辐射能量转移,其猝灭速率常数为Kq为1.077×1013L/(mol.s)(25℃)、0.946×1013L/(mol.s)(37℃)。利用荧光猝灭反应测得结合常数KA为2.6×106(25℃)、3.4×106(37℃),结合位点数n为1.30(25℃)、1.33(37℃)。根据Frster能量转移理论得到结合距离r=2.92nm(25℃)、2.66nm(37℃)和能量转移效率E=0.45(25℃)、0.43(37℃)。通过热力学参数计算,确定胡椒酸丁二醇单酯与BSA的相互作用是熵增加和吉布斯自由能降低的自发过程,主要作用力是疏水作用力。  相似文献

Black pepper, though commonly employed as a spice, has many medicinal properties. It consists of volatile oils, alkaloids, pungent resins, etc., of which piperine is a major constituent. Though safe at low doses, piperine causes alteration in the activity of drug metabolising enzymes and transporters at high dose and is known to precipitate liver toxicity. It has a potential to form reactive metabolite(s) (RM) owing to the presence of structural alerts, such as methylenedioxyphenyl (MDP), α, β‐unsaturated carbonyl group (Michael acceptor), and piperidine. The present study was designed to detect and characterize stable and RM(s) of piperine formed on in vitro incubation with human liver microsomes. The investigation of RMs was done with the aid of trapping agents, viz, glutathione (GSH) and N‐acetylcysteine (NAC). The samples were analysed by ultra‐high performance liquid chromatography coupled with high resolution mass spectrometry (UHPLC‐HRMS) using Thermo Scientific Q Exactive Plus Orbitrap. Full scan MS followed by data‐dependent MS2 (Full MS‐ddMS2) mode was used to establish mass spectrometric fragmentation pathways of protonated piperine and its metabolites. In total, four stable metabolites and their isomers (M1a‐c, M2a‐b, M3a‐c, and M4a‐b) were detected. Their formation involved removal of carbon (3, M1a‐c), hydroxylation (2, M2a‐b), hydroxylation with hydrogenation (3, M3a‐c), and dehydrogenation (2, M4a‐b). Out of these metabolites, M1, M2, and M3 are reported earlier in the literature, but their isomers and two M4 variants are novel. In addition, six novel conjugates of RMs, including three GSH conjugates of m/z 579 and three NAC conjugates of m/z 435, were also observed.  相似文献

以胡椒碱为起始原料,与KOH反应生成胡椒酸钾盐,经酸化后制得胡椒酸(2); 2与乙醇经酯化反应制得胡椒酸乙酯(3); 3与己二胺在金属钠催化下经氨解反应合成了胡椒酸己二胺(4),其结构经1H NMR, 13C NMR和IR确证。合成4的最佳反应条件为:催化剂用量2 wt%, n(3):n(己二胺)=0.6, m(乙醇):m(3)=2.5,于65~75 ℃反应48 h,收率83.5%。该工艺在微型实验基础上放大十倍,收率大于79%。  相似文献
