Paeoniflorin is a well‐known monoterpene glucoside in the herbal drug that exhibits a number of biological activities. The pharmacokinetic characteristics of paeoniflorin from total glucosides of paeony in spontaneously hypertensive rats (SHR) are still unclear. It is essential to investigate the in vivo and in vitro pharmacokinetic differences of paeoniflorin from total glucosides of paeony in Sprague–Dawley (SD) and SHR. The in vivo pharmacokinetic data were analyzed using DAS 2.0 software and the in vitro metabolic characteristics were measured using rat hepatic microsomes. The concentration of paeoniflorin in biological samples was determined using high‐performance liquid chromatography–electrospray ionization tandem mass spectrometry method, which showed good precision and stability. The plasma concentration–time profiles of paeoniflorin following oral administration of total glucosides of paeony showed a single peak and there were significant differences in the mean values of AUC(0–t), AUC(0–∞), CLz/F and Tmax between SD and SHR (p < 0.05). The metabolic rate of paeoniflorin from total glucosides of paeony was slower in SHR than in SD rats (p < 0.05). Abstract

We study the inverse problem of parameter identification in noncoercive variational problems that commonly appear in applied models. We examine the differentiability of the set-valued parameter-to-solution map using the first-order and the second-order contingent derivatives. We explore the inverse problem using the output least-squares and the modified output least-squares objectives. By regularizing the noncoercive variational problem, we obtain a single-valued regularized parameter-to-solution map and investigate its smoothness and boundedness. We also consider optimization problems using the output least-squares and the modified output least-squares objectives for the regularized variational problem. We give a complete convergence analysis showing that for the output least-squares and the modified output least-squares, the regularized minimization problems approximate the original optimization problems suitably. We also provide the first-order and the second-order adjoint method for the computation of the first-order and the second-order derivatives of the output least-squares objective. We provide discrete formulas for the gradient and the Hessian calculation and present numerical results.  相似文献

建立火焰原子吸收分光光度法测定维血康颗粒中总铁的含量,火焰类型选择空气–乙炔,测定波长为248.3 nm,测定时间为4.0 s,燃气流量为0.9 L/min,以氘灯作为背景校正。铁的质量浓度在2.4~28.8μg/mL范围内与吸光度呈良好的线性,线性相关系数r~2=0.999 8。6次测定结果的相对标准偏差小于1.0%,样品加样回收率为93.89%,方法检出限为0.011 3μg/mL。该方法具有准确度高,稳定性、重复性好等优点,可用于维血康颗粒中总铁含量的测定。  相似文献

The total synthesis of 7,10‐epimer of the proposed structure of amphidinolide N was accomplished. The requisite chiral C17–C29 subunit was assembled stereoselectively via Keck allylation, Shi epoxidation, diastereoselective 1,3‐reduction, and a later oxidative synthesis of the THF framework. The C1–C13 and C17–C29 subunits were successfully coupled using a Enders RAMP “linchpin” as the C14–C16 three carbon unit, thereby controlling the chirality at C14 and C16. The labile allyl epoxy moiety was successfully constructed by Grieco–Nishizawa olefination at a final stage of the synthesis.  相似文献

Polytheonamide B ( 1 ) is a natural peptide that displays potent cytotoxicity against P388 mouse leukemia cells (IC50=0.098 nm ). Linear 48‐mer 1 is known to form monovalent cation channels on binding to lipid bilayers. We previously developed a fully synthetic route to 1 , and then achieved the design and synthesis of a structurally simplified analogue of 1 , namely, dansylated polytheonamide mimic 2 . Although the synthetically more accessible 2 was found to emulate the channel function of 1 , its cytotoxicity was decreased 120‐fold. Herein, the chemical preparation and biological evaluation of seven analogues 3 – 9 of 2 are reported. Compounds 3 – 9 were modified at their N terminus and/or the side chain of residue 44 of 2 to alter their physicochemical properties. The total synthesis of 3 – 9 was accomplished in a unified fashion by a combination of solid‐phase and solution‐phase chemistry. Systematic evaluation of the hydrophobicities, single‐channel currents, ion‐exchange activities, and cytotoxicities of 3 – 9 revealed that their hydrophobicities are correlated with the total magnitude of ion exchange and determine their cytotoxic potency. Consequently, the most hydrophobic analogue 9 exhibited the lowest IC50 value, which is comparable to that of 1 . Therefore, these results clarified that the bioactivity of the polytheonamide‐based peptides can be rationally controlled by changing their hydrophobicity at the N and C termini of the 48‐amino‐acid sequence.  相似文献

Abstract

Four kinds of mercury species (inorganic mercury (Hginorg), methylmercury (MeHg), total organic mercury (ΣHgorg), and insoluble mercury, deemed to be mercuric selenide (HgSe), were determined in the livers of dolphins from the Brazilian coast. The MeHg was identified and quantified in the toluene layer on a Gas Chromatograph with an Electron Capture Detector (GC-ECD). The ΣHgorg was isolated by acid leaching (H2SO4-KBr-CuSO4) and then extracted into CH2Cl2. The ΣHgorg and Hginorg were determined by Cold-Vapor Atomic Absorption Spectroscopy (CV-AAS). The MeHg was the smallest fraction of Hgtot, with a median of 9%, whereas the highest fraction of the Hgtot was as HgSe, corresponding to 53%. The fractions of Hginorg and ΣHgorg corresponded to 30% and 39%, respectively. The lowest fraction of MeHg and the highest fraction of HgSe in the liver of all animals are related to different capacities or strategies of detoxification of methylmercury in this organ.  相似文献
