氧亲和力是血红蛋白氧载体非常重要的参数之一,但其高低还没有统一的认识,有一种理论认为低氧亲和力是造成高血压的原因。为了进一步研究氧亲和力与血压之间的关系,本文制备了两种低氧亲和力的血红蛋白氧载体。用高碘酸钠氧化棉子糖(高碘酸钠:棉籽糖=6:1,摩尔比),得到氧化均一的开环棉子糖;以其作为交联剂聚合脱氧猪血红蛋白,在聚合1 h得到了低氧亲和力(P50 = 43.1 torr)主要为分子内交联的64 kDa的血红蛋白,在聚合2 4 h得到了低氧亲和力(P50 = 51.5 torr)平均分子量为600 kDa的聚合血红蛋白;分别进行大鼠50%等容量换血实验,前者显著引起血压升高而后者血压平稳,证实高血压主要和64 kDa血红蛋白有关,低氧亲和力不是造成高血压的主要原因。  相似文献

In this paper, the toxic influence of copper ions (II) on bovine hemoglobin was investigated by the combination of ultraviolet-visible absorption, fluorescence, time-resolved fluorescence, synchronous fluorescence, and circular dichroism spectra. Driven by hydrophobic and electrostatic forces, copper ions (II) could interact with bovine hemoglobin to form bovine hemoglobin-copper ions (II) complex with one binding site. The binding constant (K) was 1.57?×?104, 1.89?×?104 and 2.11?×?104?L/mol at 298, 304, and 310?K, respectively. The binding distance (r) was 4.24?nm. Fluorescence and time-resolved fluorescence spectra showed that bovine hemoglobin quenched by copper ions (II) was a static quenching process. Results of synchronous fluorescence spectra revealed that the microenvironment and the conformation of bovine hemoglobin were changed during the binding reaction. Data of circular dichroism spectra suggested that with the increasing concentration of copper ions (II), the secondary structure of bovine hemoglobin underwent a decrease in α-helix and alteration in backbone microenvironment. Copper ions (II) was thus evidenced to have a certain toxic effect on physical bodies.  相似文献

The synthesis of chiral polyaniline (PANI) induced by modified hemoglobin (Hb) was profoundly explored for the first time. Results revealed that after being separated, inactivated or immobilized, Hb can still induce the formation of chiral PANI successfully, suggesting that Hb can be used as the chiral inducers regardless of harsh reaction conditions. By examining the properties of PANI induced by modified Hb, it was found that Hb(inactivated)-PANI possessed excellent chirality, stability, and crystalline structure. The globin separated from Hb was demonstrated to have the ability of inducing the production of chiral PANI whereas the hematin from Hb had no capacity to direct enantio specificity for the PANI chains. Results indicated that Hb(immobilized)-PANI exhibited poor yield, doping state, and crystalline structure, indicating that the immobilization of Hb by entrapment was not bene cial to the polymerization reaction. Results also showed that the structure of Hb may have significant effects on the morphologies of chiral PANI.  相似文献

血细胞压积和平均血红蛋白浓度的检测对于心脑血管疾病以及贫血等疾病的预防和治疗起着非常重要的作用,为了能够无创、准确的对血细胞压积和平均血红蛋白浓度进行检测,本研究提出了一种基于光谱技术的检测方法。经解剖学研究表明,在微循环中,血流变的异常以及血液粘稠度的改变均能引起舌象的变化,即舌象和血液成分之间存在一定的相关性,所以本研究通过采集240例志愿者舌尖反射光谱,同时拍摄志愿者舌体图片和记录生化分析结果。在进行数学建模前,首先将240例志愿者随机分为校正集和验证集,运用偏最小二乘分别建立数学模型,校正集预测值和生化分析真实值之间相关系数分别为0.998和0.938,运用所建数学模型对验证集未知样本进行预测,验证集未知样本的预测结果与生化分析真实值之间相关系数分别为0.979和0.883,平均相对误差分别为1.65%和1.88%,均方根误差分别为4.066和4.139,由实验结果可见,所建立的模型可以较好的对HCT和MCHC进行预测,同时实验结果也表明近红外光谱分析结合偏最小二乘方法有可能能够为血细胞压积和平均血红蛋白浓度的检测提供一种无创的检测方法。  相似文献

采用微湿含浸法制备了有序中孔炭/四氧化三铁磁性材料.采用透射电镜和X射线衍射对复合材料进行了表征.将血红蛋白(Hb)固定于材料表面,对其直接电化学行为进行了研究,结果表明Hb在该材料内仍保持了其生物活性,在pH=7.0的PBS缓冲液中,血红蛋白表现出一对峰形良好的准可逆氧化还原峰,为Hb的Fe(Ⅲ)/Fe(Ⅱ)电对的特征峰,求出式电位E0'为-0.306 V,电子转移数为n=1.226,电荷传递系数为α=0.51,表观异相电子转移速率常数为KS=0.0144s-1.在3.00×10-6到1.50×10-4mol/L浓度范围内,血红蛋白的浓度与其响应电流呈良好的线性关系,线性相关系数为R=0.9924,最低检测限为0.270×10-6mol/L.  相似文献

FTIR-reflection absorption spectroscopy has been employed for investigation of absorption of hemoglobin, lysozyme and albumin on a copper surface. It was found that examined proteins absorb on copper surface from aqueous solutions to various extent depending on the way of preparation of the surface. Abstract

A novel hemoglobin (Hb) biosensor based on the remarkable synergistic effects of cerium dioxide (CeO2) and multiwalled carbon nanotubes (MWNTs) for detection of hydrogen peroxide (H2O2) is presented. The Hb/CeO2/MWNTs/CHIT nanocomposite was nanoengineered by selected matched material components and optimized composition ratio to produce a superior H2O2 sensor. The preparation method is quite simple and practical. This composite matrix combined the advantages of MWNTs, CeO2 nanoparticles, and chitosan (CHIT), with good electron-transfer ability, attractive biocompatibility, and fine film-forming ability, which could increase Hb attachment quantity and H2O2 detection sensitivity. In the optimum pH 7.0 phosphate buffer, the electrocatalytic response exhibited a linear dependence on H2O2 concentration in a wide range from 5.0 × 10?6 to 4.6 × 10?4 mol L?1 with a detection limit of 6.5 × 10?7 mol/L (3σ).  相似文献

Abstract

A new hemoglobin (Hb) modified carbon paste (CP) electrode was fabricated by simply mixing the hemoglobin with carbon powder and paraffin homogeneously. To prevent the leakage of Hb from the electrode surface, a Nafion film was further applied on the surface of Hb-carbon composite paste electrode. Direct electrochemistry of hemoglobin in the paste electrode was easily achieved, and a pair of well-defined quasi-reversible redox peak of heme Fe(III)/Fe(II) couple appeared with the formal potential (E0′) as ?0.335 V (vs. Saturated calomel electrode; CE) in pH 7.0 phosphate buffer solution (PBS). The fabricated Hb modified electrode showed good electrocatalytic ability to the reduction of trichloroacetic acid (TCA) and H2O2.  相似文献

基于血红蛋白(Hb)的过氧化物酶活性,催化过氧化氢氧化对甲基酚的反应体系,发现表面活性剂SDS对该体系反应速率有明显的增强效应;从而建立了新的高灵敏测定Hb的催化荧光分析法。在1.5×10-3mol/L SDS存在下,测定Hb的线性范围1×10-9~8×10-8mol/L,检出限为3.4×10-10mol/L,用于尿中Hb的测定,结果满意。  相似文献
