Sesquiterpene esters from the fruits of Celastrus orbiculatus

Three new beta-dihydroagarofuran sesquiterpene esters, 6 alpha-acetoxy-9 beta-benzoyloxy-1 beta-cinnamoyloxy-8 beta-butanoyloxy-beta-dihydroagarofuran (1), 6 alpha-acetoxy-9 beta-benzoyloxy-1 beta-cinnamoyloxy-8 beta-(2-methylbutanoyloxy)-beta-dihydroagarofuran (2), and 6 alpha-acetoxy-1 beta,8 beta-dibenzoyloxy-9 beta-hydroxy-beta-dihydroagarofuran (6), together with three known compounds (3-5) were isolated from the fruits of Celastrus orbiculatus THUNB. Their structures were elucidated on the basis of spectroscopic methods. Compound 4 showed moderate activity in inhibiting LPS-induced nitric oxide production in murine macrophage RAW264.7 cells with an IC(50) value of 43.7 microM.     

A New and Efficient Synthetic Method and Antibacterial Activities of Oxazolidinone Analogues

A series of novel oxazolidinone analogues were prepared by a new and efficient synthetic method and their antibacterial activities were determined. These compounds wer echaracterized by LC-MS and ^1H NMR.     

Analysis of fatty oil in Semen Ziziphi Spinosae by capillary gas chromatography

A simple and fast capillary gas chromatographic (CGC) method with flame ionization detection is developed for the analysis of fatty oil in Semen Ziziphi Spinosae. After methyl-esterification, eight components are identified by gas chromatography-mass spectrometry. The derivatization condition is investigated in order to validate this method. Palmitic acid and stearic acid are quantitated simultaneously. The limits of detection are 5.024 microg/mL for palmitic acid and 6.957 microg/mL for stearic acid, respectively. The limits of quantitation are 16.76 microg/mL for palmitic acid and 23.19 microg/mL for stearic acid, respectively. The percent recoveries of palmitic and stearic acid are 97.4% and 96.6%. CGC is shown to be a quick and informative tool for the analysis of fatty oil in Semen Ziziphi Spinosae.     

用CR传输线模型研究涂层/金属体系阻抗谱

根据CR传输线模型和QR电路之间的关系,建立了拟合其初值的计算方法,借助Z-View软件,可求得各元件精确值.根据电容（Ci）和电阻（Ri）随特征频率（f*）的分布,推导了元件相对增量与恒相位角元件（Q）指数参数n的关系. 结果表明, 当n小于0.5时,Ci比Ri增加得更快,从新的角度说明了n的物理意义及其和界面脱层之间的关系.作为应用实例,拟合了不同特征的电化学阻抗谱,分析了有机涂层/金属腐蚀体系阻抗变化的具体过程,区分了点蚀和脱层因素对阻抗谱的影响,从高阻抗体系同时得到了与不同空隙率有关的涂层电容和电阻值,并根据涂层体系的不均匀特征探讨了模型结构的物理意义.     

PbO2阳极在硫酸溶液中的析氧失活行为

采用热分解鄄电镀法制备了以Sb 掺杂SnO₂(Sb-SnO₂)为底层的Ti 基PbO₂阳极(Ti/PbO₂). 采用加速电解寿命测试、电化学阻抗谱、XRD、SEM-EDX 等技术, 研究了Ti/PbO₂阳极在硫酸溶液中的电解失效行为和机制.结果表明,在新制备的PbO₂镀层中, 由于氧空位的存在, PbO₂镀层的内应力表现为拉应力, 随着电解的进行, 阳极表面生成的活性氧原子在向基底扩散的过程中, 将Pb³⁺态氧化为Pb⁴⁺态, 逐渐占据镀层内作为自由电子施主的氧空位, 这不仅导致镀层的导电性能下降, 同时使镀层的应力逐渐由拉应力转变为压应力, 镀层性质逐渐劣化. 这一过程基本结束时,活性氧原子才大量扩散至Ti基底导致基底的钝化, 在Ti 基底和镀层界面出现显著的界面应力, 在界面应力和镀层内压应力的共同作用下, 阳极出现鼓泡、脱落, 迅速进入失活阶段.     

Development and validation of a liquid chromatography/tandem mass spectrometry procedure for the quantification of adefovir in human plasma

To investigate the pharmacokinetics of adefovir as an anti-hepatitis B virus drug, a sensitive and specific liquid chromatography/tandem mass spectrometry method was developed and validated. After a simple protein precipitation using methanol, the post-treatment samples were analyzed on a C(18) column interfaced with a triple-quadrupole tandem mass spectrometer using positive electrospray ionization. A structural analogue, PMPA, was used as the internal standard. The method was linear in the concentration range 0.25-100 ng/mL. The lower limit of quantification was 0.25 ng/mL. The intra- and inter-day relative standard deviation over the entire concentration range was 5.7% or less. The accuracy determined at three concentrations (0.75, 10 and 80 ng/mL for adefovir) was within +/-2.5% relative error. The method described here was successfully applied for the evaluation of pharmacokinetic profiles of adefovir after single oral administration doses of 5, 10 and 20 mg adefovir dipivoxil to ten healthy volunteers.     

Simultaneous determination of chlorpheniramine and pseudoephedrine in human plasma by liquid chromatography-tandem mass spectrometry

A sensitive and specific procedure for simultaneous quantitation of chlorpheniramine and pseudoephedrine in human plasma has been developed and validated. Analytes were extracted from plasma samples by liquid-liquid extraction, separated on a Diamonsil C18 column (250 x 4.6 mm i.d.) and detected by tandem mass spectrometry with an atmospheric pressure chemical ionization interface. Diphenhydramine was used as the internal standard. The method has a lower limit of quantitation of 0.2 and 2.0 ng/mL for chlorpheniramine and pseudoephedrine, respectively. The intra- and inter-day relative standard deviation, calculated from quality control (QC) samples were below 4.3% for chlorpheniramine and below 9.5% for pseudoephedrine. The inter-day relative error as determined from QC samples was within 4.7% for each analyte. The overall extraction recoveries of chlorpheniramine and pseudoephedrine were 77 and 61% on average, respectively. The method was successfully applied to pharmaockinetic study of chlorpheniramine and pseudoephedrine in volunteers receiving formulations containing 4 mg of chlorpheniramine maleate and 60 mg of pseudoephedrine hydrochloride.     

New selective O-debenzylation of phenol with Mg/MeOH

Carboxylate-benzyl and nitro-benzyl groups used in phenols protection were selectively debenzylated with 3-25 equiv of Mg in methanol at room temperature. Good yields of the desired phenols were obtained within 3-10 h from a wide variety of O-(carboxylate-benzyl)- or O-(nitro-benzyl)-phenols. Selective O-debenzylation was possible in the presence of O-(carboxylate-benzyl)- or O-(nitro-benzyl)-phenols with Mg/MeOH.     

A new lignan from the seeds of Sphaerophysa salsula

A novel lignan: sphaerophyside SC was isolated from ethanolic extract of the seeds of Sphaerophysa salsula (Pall.) DC. The structure of the compound was elucidated mainly on the basis of the 1D NMR and 2D NMR data.     

HPLC determination of ferulic acid in rat plasma after oral administration of Rhizoma Chuanxiong and its compound preparation

An HPLC method is described for determination of ferulic acid in rat plasma. The concentration of ferulic acid in rat plasma was determined after deproteinization with acetonitrile using sulfamethoxazole as internal standard. Chromatographic separations were performed on a C(18) stationary phase with a mobile phase composed of acetonitrile-water (16:84, v/v) with 1% glacial acetic acid. The UV detection wavelength was set at 320 nm. The method was successfully applied to the determination of pharmacokinetic parameters in rat plasma after oral administration of Rhizoma Chuanxiong and and its compound preparation Suanzaoren decoctions. The calibration curve was linear over the range 0.0510-4.08 micro g/mL in rat plasma. Within-day and between-day precisions were less than 4.5% RSD. Mean recovery was determined as 96.9%. The limit of quantitation was 0.0510 micro g/mL. The pharmacokinetic parameters of the two preparations were different significantly (p < 0.05), which may attribute to the effects of other ingredients present in Suanzaoren decoction.
