Two new melampolide-type sesquiterpene lactones, 8beta-epoxyangeloyloxy-9alpha-ethoxy-14-oxo-acanthospermolide (1) and 8beta-angeloyloxy-9alpha-ethoxy-14-oxo-acanthospermolide (2), were isolated from the leaves of yacon [Smallanthus sonchifolia (POEPP. et ENDL.) H. Robinson] along with eleven known melampolides, allo-schkuhriolide (3), enhydrin (4), polymatin A (5), fluctuanin (6), 8beta-angeloyloxy-9alpha-acetoxy-14-oxo-acanthospermolide (7), 8beta-angeloyloxy-14-oxo-acanthospermolide (8), 8beta-methacryloyloxymelampolid-14-oic acid methyl ester (9), uvedalin (10), polymatin B (11), 8beta-tigloyloxymelampolid-14-oic acid methyl ester (12), and sonchifolin (13). Their structures were established on the basis of spectroscopic evidence including 1D- and 2D-NMR experiments. All isolates were evaluated for inhibition of LPS-induced nitric oxide production in murine macrophage RAW 264.7 cells.  相似文献

Three new abietane diterpenes, sugikurojins D (1), E (2), and F (3), and two new abietanes which incorporate cadinane, sugikurojins G (4) and H (5) were isolated from the bark of Cryptomeria japonica. These structures were elucidated primarily by extensive NMR experiments. The structure of sugikurojin D (1) was deduced to be 6alpha-acetoxy-7beta,11-dihydroxy-12-methoxy-8,11,13-abietatriene. Sugikurojin E (2) was deduced to be 6alpha-acetoxy-7beta,12-dihydroxy-8,11,13-abietatriene. Sugikurojin F (3) was 7alpha-methoxy-8,13-abietadien-11,12-dione. Sugikurojins G (4) and H (5) had a unique skeleton incorporating an alpha-cadinol or a 1alpha-hydroxy-T-cadinol in ferruginol, respectively. Also obtained in this investigation were the known diterpenes (6-14). An antibacterial activity of ten among these against Staphylococcus aureus and Escherichia coli was inactive at the (MIC: 125 microg/ml) level. Meanwhile, in the cytotoxic activity against HL-60, compounds 4, 8, and 11 showed moderate (IC50: 4, 35.4; 8, 28.0; 11, 52.4 microM) though weak (IC50: 4, 100; 8, 80.8; 11, 100 microM) activity against HCT-15.  相似文献

Six new ent-kaurane-type diterpenoids were isolated from the leaves of the endemic Vietnamese medicinal plant Croton tonkinensis GAGNEP. (Euphorbiaceae) together with three known ent-11alpha-acetoxy-7beta,14alpha-dihydroxykaur-16-en-15-one (1), ent-kaur-16-en-15-one 18-oic acid (5) and ent-18-hydroxykaur-16-ene (7). Their structures were determined by spectroscopic analyses to be ent-7beta-acetoxy-11alpha-hydroxykaur-16-en-15-one (2), ent-18-acetoxy-11alpha-hydroxykaur-16-en-15-one (3), ent-11alpha-acetoxykaur-16-en-18-oic acid (4), ent-15alpha,18-dihydroxykaur-16-ene (6), ent-11alpha,18-diacetoxy-7beta-hydroxykaur-16-en-15-one (8), and ent-(16S)-1alpha,14alpha-diacetoxy-7beta-hydroxy-17-methoxykauran-15-one (14). ent-Kaurane-type diterpenoids from Croton tonkinensis 2-4, 6, and 9-13, were tested for toxicity in the brine shrimp lethality assay. Compounds 9, 10, and 12 demonstrated significant activity, compounds 2, 3, 6, and 11 showed weak activity, and compounds 4 and 13 were inactive.  相似文献

A new taxoid, 5alpha,13alpha-diacetoxy-10beta-cinnamoyloxy-4(20),11-taxadien-9alpha-ol (1) along with its 9,10-isomer, taxinine NN-11 (2) were isolated from the callus cultures of Taxus cuspidata. The structures were identified by the analyses of the spectral data and chemical method. Their in vitro cytotoxicity against 3 cell lines (HepG2, WI-38 and VA-13) and multidrug resistance (MDR) reversal activity toward 2780AD tumor cells were preliminarily evaluated, the low cytotoxicities and potent MDR reversal activities suggested that they might be good lead compounds of tumor MDR reversal agent.  相似文献

Two new lupane-triterpene carboxylic acids, called acantrifoic acid A (1) and acantrifoside C (2) have been isolated from the leaves of Acanthopanax trifoliatus. Based on extensive 1D and 2D NMR spectroscopic data, their chemical structures were determined as 3 alpha-acetoxy-30-hydroxylup-20(29)-ene-23,28-dioic acid and 3 alpha-acetoxy-30-hydroxylup-20(29)-ene-23,28-dioic acid 28-O-alpha-L-rhamnopyranosyl-(1-->4)-beta-D-glucopyranosyl-(1-->6)-beta-D-glucopyranosyl ester.  相似文献

A new lathyrane-type diterpene 8alpha,15beta-diacetoxy-7beta-benzoyloxy-3beta-(2-methylpropanoyloxy)-4alphaH,9alphaH, 11alphaH-lathyra-5E,12E-dien-14-one (latazienone) has been isolated from the latex of Euphorbia latazi Kunth. Further study of the methanol extract of the aerial parts of Physalis angulata (Solanaceae) resulted in the isolation of new withanolides, designated physagulins L, M and N, together with known withanolide, physagulin D and flavonol glycoside, quercetin 3-O-rhamnosyl-(1-->6)-galactoside. The chemical structures of these new withanolides were elucidated by detailed spectroscopic analyses to be (20R,22R)-15alpha-acetoxy-5alpha,6beta,14beta,17beta,27-pentahydroxy-1-oxo-witha-2, 24-dienolide, (20S,22S)-15alpha-acetoxy-5alpha,6beta,14alpha,23beta-tetrahydroxy-1-oxo-witha-2,16,24-trienolide and (20S,22R)-15alpha-acetoxy-5beta,6beta-epoxy-14alpha-hydoxy-3beta-methoxy-1-oxo-witha-16,24-dienolide, respectively. All these compounds showed weak trypanocidal activity against trypomastigotes, an infectious form of Trypanosoma cruzi, the etiologic agent for Chagas' disease. Withanolides obtained in the previous paper showed considerable trypanocidal activity, suggesting the structure-activity relationships.  相似文献

Five new lanostane-type triterpenes, named fomefficinic acid A-E (1-5), were isolated from the dried sclerotium of Fomes officinalis, respectively. Their structures were established as 24-methylene-3-oxo-lanost-8-en-21-oic acid (1), 3alpha,15alpha-dihydroxy-24-methylene-lanosta-7,9(11)-dien-21-oic acid (2), 3alpha,15alpha-dihydroxy-24-methylene-lanost-8-en-21-oic acid (3), 15alpha-hydroxy-3-oxo-24-methylenelanost-8-en-21-oic acid (4), 15alpha-acetoxy-3-oxo-24-methylenelanosta-7,9(11)-dien-21-oic acid (5), by spectral analysis and chemical methods as well as comparison with known compounds.  相似文献

A new germacranolide, inulacappolide (1), was isolated from the EtOH extract of the whole plant of Inula cappa along with 16 known compounds. The structure of inulacappolide was a rare 1(10)-saturated type of germacran-6,12-olide, identified as 2alpha-acetoxy-3beta-hydroxy-9beta-angeloyloxygermacra-4-en-6alpha,12-olide by spectral analysis (IR, HR-ESI/MS, (1)H-NMR, (13)C-NMR, HMQC, HMBC, NOESY). In vitro, it showed antiproliferative effects against human cervical cancer HeLa, human leukemia K562 and human nasopharyngeal carcinoma KB cell lines with IC(50) values of 1.2 microM, 3.8 microM and 5.3 microM, respectively.  相似文献

A new withanolide, withangulatin I (2), was isolated from the whole plant of Physalis angulata. Its structure was established as (20S,22R)-15alpha-acetoxy-5beta,6beta-epoxy-14alpha-hydroxy-1,4-dioxo-witha-2,16,24-trienolide on the basis of chemical and spectroscopic methods including 2D-NMR and circular dichroism (CD) experiments. Withangulatin A (1) and withangulatin I (2) were tested for their cytotoxic activities against two human cancer cell lines, colorectal carcinoma COLO 205 and gastric carcinoma AGS, in vitro. Compounds 1 and 2 exhibited inhibitory activities against these two human cancer cells with IC(50) values of 16.6 and 1.8 and 53.6 and 65.4 muM, respectively.  相似文献

n-Hexane and dichloromethane extracts obtained from Laurencia dentroidea (Rhodophyta) and Canistrocarpus cervicornis (Phaeophyta) were investigated for their acaricidal and repellent properties against Tetranychus urticae under laboratory conditions. The two extracts displayed moderate toxicity and good repellent proprieties, and were significantly more toxic (36-fold) than the positive control (eugenol), whereas eugenol was tenfold more repellent than either seaweed extract. The sesquiterpenoid elatol (1) was isolated from L. dentroidea and the diterpenoid seco-dolastane (4R,9S,14S)-4alpha-acetoxy-9beta, 14alpha-dihydroxydolast-1(15),7-diene (2) from C. cervicornis, the chemical structures of which were characterized by NMR spectroscopic data (1H and 13C) and by comparison with literature data. These compounds exhibited moderate toxicity, but a high degree of repellent activity against T. urticae. The findings suggest that marine natural products, specifically terpenes, can be employed for the development of new pesticides and become prototype agrochemical agents.  相似文献

Several new acyclonucleoside purine and 8-azapurine analogs have been prepared from 2-amino-4,6-dichloropyrimidine ( 1 ) and 3-amino-1,2-propanediol ( 2a ) and 4-amino-1-butanol ( 2b ), respectively, as the starting materials. The new target compounds are: 2-amino-6-chloro-9-(2,3-dihydroxypropyl)purine ( 6a ), 2-amino-6-chloro-9-(4-hydroxybutyl)purine ( 6b ), 2-amino-6-chloro-9-(2,3-dihydroxypropyl)-8-azapurine ( 7a ), 2-amino-6-chloro-9-(4-hydroxybutyl)-8-azapurine ( 7b ), 9-(2,3-dihydroxypropyl)-8-azaguanine ( 8a ), 9-(4-hydroxybutyl)-8-azaguanine ( 8b ), 9-(2,3-dihydroxypropyl)-8-azathioguanine ( 9a ), and 9-(4-hydroxybutyl)-8-azathioguanine ( 9b ). Also, the requisite intermediate pyrimidine derivatives, 2,5-diamino-4-(2,3-dihydroxypropylamino)-6-chloropyrimidine ( 5a ) and 2,5-diamino-4-(4-hydroxybutylamino)-6-chloropyrimidine ( 5b ) are novel.  相似文献

Various 6-substituted 9-benzyl-8-hydroxypurines were synthesized in order to investigate the structure-activity relationship at the 6-position of 9-benzyl-8-hydroxyadenine (1), which is a lead compound for the screening of interferon (IFN)-inducing activity. 6-Unsubstituted, mercapto-, methylthio- and hydroxy-9-benzyl-8-hydroxypurines (2-5) were prepared from 5-amino-1-benzyl-4-cyano-2-hydroxyimidazole (9). Synthesis of a 6-methoxy analog (6) was conducted from 5-amino-4-benzylamino-6-chloropyrimidine (13). 6-Alkylamino and acylaminopurines (7 and 8) were also prepared by alkylation and acylation of 1, respectively. Since these compounds (2-8) indicated no activity, it was found that a free amino group of 1 is required for the expression of IFN-inducing activity.  相似文献

Three new disialo- and trisialo-gangliosides, CEG-6 (6), CEG-8 (8), and CEG-9 (9), were obtained, together with one known ganglioside, HLG-3 (7), from the lipid fraction of the chloroform/methanol extract of the sea cucumber Cucumaria echinata. The structures of the new gangliosides were determined on the basis of chemical and spectroscopic evidence to be 1-O-[alpha-L-fucopyranosyl-(1-->11)-(N-glycolyl-alpha-D-neuraminosyl)-(2-->4)-(N-acetyl-alpha-D-neuraminosyl)-(2-->6)-beta-D-glucopyranosyl]-ceramide (6) and 1-O-[(N-glycolyl-D-neuraminosyl)-(2-->11)-(N-glycolyl-D-neuraminosyl)-(2-->4)-(N-acetyl-D-neuraminosyl)-(2-->6)-D-glucopyranosyl]-ceramide (8, 9). The ceramide moieties of each compound were composed of an homogeneous sphingosine or phytosphingosine base and heterogeneous 2-hydroxy or nonhydroxylated fatty acid units. These gangliosides showed neuritogenic activity toward the rat pheochromocytoma cell line PC-12 in the presence of nerve growth factor.  相似文献

A novel agarofuran sesquiterpene polyol ester, 1beta,2beta,6alpha,15beta-tetracetoxy-8 beta,9alpha-dibenzoyloxy-beta- dihydroagarofuran (celahin D) (1), two known analogues of 1,1beta-acetoxy-8beta,9alpha-dibenzoyloxy-4al pha6alpha-dihydroxy-2beta(alphamethylbutanoyloxy)-beta-++ +dihydroagarofuran (2) and beta-acetoxy-8beta,9alpha-dibenzoyloxy-6alpha-hy droxy-2beta(alpha -methylbutanoyloxy)-beta-dihydroagarofuran (3), and a known cytotoxic sesquiterpene pyridine alkaloid, emarginatine E (4) were isolated from the stems of Celastrus hindsii Benth. Three known triterpenes, loranthol (5), lupenone (6) and friedelinol (7) were also obtained from the titled plant. Structural elucidation of compound 1 was established by 2D NMR spectra.  相似文献

From leaves of Turpenia ternata (Staphylaceae), one megastigmane and seven of its glucosides (1-8) were isolated. Megastigmane and two of the glucosides were found to be known compounds, namely, 3S,5R,6R,9S-tetrahydroxymegastigmane (1), corchoionoside C (2), and icariside B4 (3). The structures of compounds 4-8 (turpinionosides A-E, respectively) were elucidated by means of spectroscopic analyses, and then their absolute structures were determined by the modified Mosher's method to be (3S,5R,6S,9S)-3,6,9-trihydroxymegastigman-7-ene 3-O- and 9-O-beta-D-glucopyranosides (4, 5, respectively), (1S,3S,5R,6S,9R)-3,9,12-trihydroxymegastigmane 3-O-beta-D-glucopyranoside (6), (3S,4R,9R)-3,4,6-trihydroxymegastigman-5-ene 3-O-beta-D-glucopyranoside (7), and (2S,9R)-2,9-dihydroxymegastigman-5-en-4-one 2-O-beta-D-glucopyranoside (8).  相似文献

From the whole plant of Dracocephalum kotschyi BOISS., two new monoterpene glycosides (9, 10), together with seven known terpenoids and a phytosterol (1-8), were isolated. Their structures were determined to be limonen-10-al (1), geranial (2), neral (3), beta-sitosterol (4), oleanolic acid (5), ursolic acid (6), p-mentha-8-en-1,2-diol (7), colosolic acid (8), limonen-10-ol 10-O-beta-D-glucopyranoside (9), and limonen-10-ol 10-O-beta-D-glucopyranosyl-(1-->2)-beta-D-glucopyranoside (10). Compounds 1 (3.1 microM), 2 (3.1 microM), 3 (3.1 microM), 5 (6.2 microM), 6 (6.2 microM), and 8 (6.2 microM) were effective against epimastigotes of Trypanosoma cruzi.  相似文献

Deprotonation of the phosphamonocarbaborane, exo-6-R-arachno-6,7-PCB(8)H(12) (R = Ph 1a or Me 1b), yields exo-6-R-arachno-6,7-PCB(8)H(11)(-), which when reacted with appropriate transition-metal reagents affords new metallaphosphamonocarbaborane complexes in which the metals adopt endo-eta(1), exo-eta(1), eta(4), eta(5), or eta(6) coordination geometries bonded to the formal R-arachno-PCB(8)H(11)(-), R-arachno-PCB(8)H(10)(2-), R-arachno-PCB(8)H(9)(3-), or R-nido-PCB(8)H(9)(-) ligands. The reaction of exo-6-(C(6)H(5))-arachno-6,7-PCB(8)H(11)(-) (1a-) with Mn(CO)(5)Br generated the eta(1)-sigma product exo-6-[Mn(CO)(5)]-endo-6-(C(6)H(5))-arachno-6,7-PCB(8)H(11) (2) having the [Mn(CO)(5)] fragment in the thermodynamically favored exo position at the P6 cage atom. On the other hand, reaction of 1a- with (eta(5)-C(5)H(5))Fe(CO)(2)I resulted in the formation of two products, an eta(1)-sigma complex endo-6-[(eta(5)-C(5)H(5))Fe(CO)(2)]-exo-6-(C(6)H(5))-arachno-6,7-PCB(8)H(11) (3) having the (eta(5)-C(5)H(5))Fe(CO)(2) fragment attached at the endo-P6 position and an eta(6)-closo complex, 1-(eta(5)-C(5)H(5))-2-(C(6)H(5))-closo-1,2,3-FePCB(8)H(9) (4a). Rearrangement of the endo-compound 3 to its exo-isomer 5 was observed upon photolysis of 3. Synthesis of the methyl analogue of 4a, 1-(eta(5)-C(5)H(5))-2-CH(3)-closo-1,2,3-FePCB(8)H(9) (4b), along with a double-insertion product, 1-CH(3)-2,3-(eta(5)-C(5)H(5))(2)-2,3,1,7-Fe(2)PCB(8)H(9) (6), containing two iron atoms eta(5)-coordinated to a formal R-arachno-PCB(8)H(9)(3-), was achieved by reaction of exo-6-CH(3)-arachno-6,7-PCB(8)H(11)(-) (1b-) with FeCl(2) and Na(+)C(5)H(5)(-). Complexes 4a and 4b can be considered ferrocene analogues, in which an Fe(II) is sandwiched between C(5)H(5)(-) and 6-R-nido-6,9-PCB(8)H(9)(-) anions. Reaction of exo-6-(C(6)H(5))-arachno-6,7-PCB(8)H(11)(-) (1a-) with cis-dichlorobis(triphenylphosphine)platinum (II) afforded two compounds, an eta(1)-sigma complex with the metal fragment again in the endo-P6 position, endo-6-[cis-(Ph(3)P)(2)PtCl]-exo-6-(C(6)H(5))-arachno-6,7-PCB(8)H(11) (7) and an eta(4)-complex, 7-(C(6)H(5))-11-(Ph(3)P)(2)-nido-11,7,8-PtPCB(8)H(10) (8) containing the formal R-arachno-PCB(8)H(10)(2)(-) anion. The structures of compounds 2, 3, 4a, 4b, 6, 7, and 8 were crystallographically confirmed.  相似文献

以腺苷为母体,对其N6-位进行结构改造,首先经邻位双羟基保护,N6-位氯代,再在N6-位引入哌嗪环制得中间体2',3'-异丙叉-6-哌嗪嘌呤核苷(4);4与N-氯乙酰苯胺类似物(6a~6h)偶联后脱除邻位双羟基保护合成了8个新型的N6-哌嗪取代腺苷衍生物(8a~8h),其结构经1H NMR,13C NMR和HR-ESI-MS表征。采用MTT法研究了8a~8h对Hela肿瘤细胞的抑制活性。结果表明:大部分目标化合物对Hela肿瘤细胞具有较好的抑制活性,其中2-{4-[9-(3,4-二羟基-5-羟甲基-四氢呋喃-2-基)-9H-嘌呤-6-基]-哌嗪-1-基}-N-(3-氟苯基)-乙酰胺(8e)的活性最好,IC50为21.74μmol·L-1。  相似文献

The first phytochemical study of Simira eliezeriana Peixoto (Rubiaceae) allowed the isolation and structural determination of two new diterpenes named simirane A [(5R,6R,8R,9R,10S,11S,13S)-6β,11β-dihydroxy-2,4(18),15-erythroxylatrien-1-one] (1) and simirane B [(5S,8R,9R,10S,11S,13S)-11β-hydroxy-2,4(18),15-erythroxylatrien-1-one] (2), together with seven known compounds: sitosterol (3), stigmasterol (4), campesterol (5), coniferaldehyde (6), vanillin (7), pinoresinol (8) and harman (9) from the bark of the plant. The structures of the compounds were established on the basis of spectroscopic methods, including 1-D and 2-D NMR, HRESI-MS and CD analysis and comparisons with available literature data of known compounds.  相似文献
