为建立研究基因表达差异的抑制性消减杂交(SSH)方法,以未加诱导剂处理的结肠癌LoVo细胞提取的mRNA为模板合成的cDNA作为驱赶子(driver),联合使用10.0μmol/LATRA和0.1μmol/L1,25-(OH)2D3诱导2d的LoVo细胞提取的mRNA为模板合成的cDNA作为测试子(tester)进行SSH实验,结果为:消减产物与未消减样品(对照)在琼脂糖凝胶电泳图谱上明显不同,前者可见一些扩增条带,其大小范围在100-1000bp之间,本实验说明SSH方法对于识别差异表达的基因具有高效性。 相似文献

Based on the linear viscoelastic differential constitutive law and cable structural model, the coupled longitudinal-transverse
waves that propagate along a viscoelastic cable with small curvature is investigated. A mathematical model is presented that
describes the three-dimensional nonlinear response of a viscoelastic cable. An asymptotic form of this model is obtained for
the linear response of cables having small equilibrium curvature. The spectral relation governing the propagating waves is
derived using transform methods. The spectral relation is employed in deriving a Green's function that is then used to construct
solutions for in-plane response under distributed harmonic excitation. Analysis of forced response reveals the existence of
two types of periodic waves that propagate through the cable, one characterizing extension-comprehensive deformation and the
other characteristic transverse deformation.

鉴于“方形”势阱描述量子阱中的电子运动行为过于简单、过于理想,引入了正切平方势来代替,使结果得到了改善。在量子力学框架内,利用正切平方势把电子的Schrdinger方程化为超几何方程,利用系统参数和超几何函数严格地求解了电子的本征值和本征函数,并以Ga1-xAlxAs-GaAs-Ga1-xAlxAs量子阱为例计算了电子的能级和能级之间的跃迁。结果表明,电子在量子阱中的能量是量子化的,而相邻能级之间的跃迁给出与实验进一步符合的结果。 相似文献

It has become very important to study and find optimal conditions for imaging electron-beam (e-beam) sensitive materials in scanning transmission electron microscopy under low electron-dose with high signal-to-noise ratio (SNR). Convergence and collection angles and electron-probe current are essential parameters. However, these parameters have rarely been discussed in a systematic way. In this paper, the illumination and collection conditions are optimized according to the resolution requirement of different materials by adjusting the condenser and intermediate lenses in a commercial transmission electron microscope. To demonstrate the significance of optimizing these parameters, two examples, zeolite MFI and metal–organic framework (MOF) MIL-101, are taken among the sensitive materials, with the most important electron incidences along the [010] and <110> directions, respectively. High SNR atomic resolution images of MFI are obtained with e-beam current as low as 0.50 pA, reaching information transfer for reflection up to 18 0 2 corresponding to d-spacing of 0.11 nm, close to the resolution limit of 0.098 nm from resolvable diffraction limit. MOF MIL-101 is characterized under an even lower e-beam 0.2 pA to avoid severe beam damage. High-quality annular dark and bright field images are obtained, which proves the wide applicability of this method on more e-beam sensitive materials. 相似文献

Because quantum critical systems are very sensitive to the variation of parameters around the quantum phase transition (QPT), quantum criticality has been presented as an efficient resource for metrology. In this paper, we address the issue whether the divergent feature of the inverted variance is realizable in the presence of noise when approaching the QPT. Taking the quantum Rabi model (QRM) as an example, we obtain the analytical result for the inverted variance with single-photon relaxation. We show that the inverted variance may be convergent in time due to the noise. Since the precision of the metrology is very sensitive to the noise, as a remedy, we propose squeezing the initial state to improve the precision under decoherence. In addition, we also investigate the criticality-based metrology under the influence of the two-photon relaxation. Strikingly, although the maximum inverted variance still manifests a power-law dependence on the energy gap, the exponent is positive and depends on the dimensionless coupling strength. This observation implies that the criticality may not enhance but weaken the precision in the presence of two-photon relaxation, due to the non-linearity introduced by the two-photon relaxation. 相似文献
