Telomerase inhibition is an attractive strategy for cancer chemotherapy. In the current study, we have synthesized and characterized two chiral ruthenium(II) complexes, namely, Λ-[Ru(phen)(2)(p-MOPIP)](2+) and Δ-[Ru(phen)(2)(p-MOPIP)](2+), where phen is 1,10-phenanthroline and p-MOPIP is 2-(4-methoxyphenyl)-imidazo[4,5f][1,10]phenanthroline. The chiral selectivity of the compounds and their ability to discriminate quadruplex DNA were investigated by using UV/Vis, fluorescence spectroscopy, circular dichroism spectroscopy, fluorescence resonance energy transfer melting assay, polymerase chain reaction stop assay and telomerase repeat amplification protocol. The results indicate that the two chiral compounds could induce and stabilize the formation of antiparallel G-quadruplexes of telomeric DNA in the presence or absence of metal cations. We report the remarkable ability of the two complexes Λ-[Ru(phen)(2)(p-MOPIP)](2+) and Δ-[Ru(phen)(2)(p-MOPIP)](2+) to stabilize selectively G-quadruplex DNA; the former is a better G-quadruplex binder than the latter. The anticancer activities of these complexes were evaluated by using the MTT assay. Interestingly, the antiproliferative activity of Λ-[Ru(phen)(2)(p-MOPIP)](2+) was higher than that of Δ-[Ru(phen)(2)(p-MOPIP)](2+), and Λ-[Ru(phen)(2)(p-MOPIP)](2+) showed a significant antitumor activity in HepG2 cells. The status of the nuclei in Λ/Δ-[Ru(phen)(2) (p-MOPIP)](2+)-treated HepG2 cells was investigated by using real-time living cell microscopy to determine the effects of Λ/Δ-[Ru(phen)(2)(p-MOPIP)](2+) on intracellular accumulation. The results show that Λ/Δ-[Ru(phen)(2)(p-MOPIP)](2+) can be taken up by HepG2 cells and can enter into the cytoplasm as well as accumulate in the nuclei; this suggests that the nuclei were the cellular targets of Λ/Δ-[Ru(phen)(2)(p-MOPIP)](2+).  相似文献

We perform an extensive investigation on the α decay of heavy deformed even-even and odd-A nuclei within the multichannel cluster model. The dynamics effects of core nuclei are taken into account in dealing with the interaction matrix and the α-cluster formation. Enough decay channels are considered for convergence in solving the coupled equations. Systematic calculations have been performed for 36 even-even nuclei and 32 odd-A nuclei. The calculated results show good agreement with the available experimental data concerning total α-decay half-lives and branching ratios, including the branching ratios for high-spin states.  相似文献

An end-to-end differential measurement approach with capacitively coupled contactless conductivity detection (C(4)D) was applied to anion-exchange monolithic capillary column ion chromatography. The column was prepared by thermally initiated radical polymerization of poly(glycidyl methacrylate) in a fused-silica capillary of 320 μm i.d. and modified by quaternary ammonium latex surface coating. Two C(4)Ds were placed near both ends of the capillary column and the output difference between them was measured. With 15 mM potassium hydrogen phthalate used as the eluent, good separation of a mixture of inorganic anions (F(-), Cl(-), NO(2)(-), NO(3)(-)) was achieved. The detection limits of conventional C(4)D are 1.6, 0.28, 0.53, and 0.47 mg L(-1) for F(-), Cl(-), NO(2)(-), and NO(3)(-), respectively. To further enhance the sensitivity, the capacitive impedance from C(4)D was neutralized by an inductive impedance from a piezoelectric resonator. An increase in sensitivity by a factor of 7-8 was achieved in the resonating C(4)D in comparison with the conventional C(4)D. The detection limits of the resonating C(4)D are 0.23, 0.041, 0.065, and 0.059 mg L(-1) for F(-), Cl(-), NO(2)(-), and NO(3)(-), respectively. The response of the resonating C(4)D was analyzed based on an equivalent circuit model.  相似文献

We propose a half-cylindrical hyperlens with symmetrical metallodielectric structure which consists of five periods of the symmetrical GaP/Ag/GaP films. Such hyperlens can transfer subwavelength information to the output surface (about one wavelength) due to the flat dispersion. Compared with the half-cylindrical hyperlens composed of the resonant Ag/GaP structure, the symmetrical GaP/Ag/GaP metallodielectric structure shows higher transmission and resolution for the propagating waves and evanescent waves, which is also confirmed by the transfer matrix method. In addition, subwavelength imaging can be realized when the incident wavelength varies from 550 nm to 650 nm.  相似文献

在常规大气环境条件下,基于单极性纳秒脉冲电源对表面介质阻挡放电特性进行了实验研究.结果表明:纳秒脉冲表而介质阻挡放电的本质是丝状放电,放电集中在电压脉冲的上升沿;激励电压和脉冲重复频率越大,放电越强烈,越接近均匀放电,但电压的作用更侧重于均匀性,而频率的作用则侧重于放电的强度;电极间隙的优化可以使表面介质阻挡放电特性最好;玻璃作为阻挡介质时容易发生沿面闪络.  相似文献

设计制作了全固态高重复频率磁脉冲压缩发生器,最高重复频率5 kHz,脉宽70 ns,通过调节初始储能电容上的电压可在500 阻性负载上获得4~40 kV连续可调的输出电压。通过分析简化的磁压缩末级回路,分析了预脉冲产生的过程,得出了预脉冲的电压表达式,选取适当的磁芯相对磁导率,经过求解,得出在磁开关未饱和电感一定时预脉冲随负载阻值变化的曲线簇,从曲线中可以看出:随着负载的阻值的增大,预脉冲的峰值绝对值也增大;在负载恒定的情况下,增大磁开关未饱和电感的大小可以显著地减小负载两端预脉冲的峰值绝对值,这要求磁开关磁芯有更高的相对磁导率。  相似文献

从理论分析、软件模拟、实验研究等方面研究了氮气介质高气压开关的工作特性。根据强度理论计算所设计并加工的开关的理论压强可达10 MPa。对氮气介质高气压开关进行实验,得到在气压范围2,4,6 MPa、电极间隙0.1,0.5,1.0 mm时,气隙的击穿延时小于14 ns,抖动小于1 ns;在气压2 MPa、电极间隙0.1 mm时50%击穿电压约为34 kV。与常压空气开关相比较,该开关具有很好的稳定性和绝缘特性。  相似文献

Efficient white light-emitting diodes (WOLEDs) were fabricated with a solution-processed single emission layer composed of a molecular and polymeric material mixed-host (MH). The main host used was a blue-emitting molecular material of 4,4′-bis(2,2′-diphenylvinyl)-1,1′-biphenyl (DPVBi) and the assisting host used was a hole-transport-type polymer of poly(9-vinylcarbazole) (PVK). By co-doping 4,4′-bis[2-(4-(N,N-diphenylamino)phenyl)vinyl]biphenyl and 5,6,11,12-tetraphenylnaphacene into the MH, the performances of the fabricated devices made with different mixing ratio of host materials were investigated, and were to depend on the mixing ratios. Under the optimal PVK:DPVBi ratio (3:7), we achieved a maximum luminance of 14 110 cd/m2 and a maximum current efficiency of 9.5 cd/A. These improvements were attributed to the MH structure, which effectively improved the thermal stability of spin-coated film and enhanced the hole-injection/transporting properties of WOLEDs.  相似文献

应用基于气相色谱-质谱联用(GC-MS)的代谢组学方法结合细胞周期实验,研究羽扇豆醇体外抑制人乳腺癌细胞MCF-7增殖的作用机理。代谢组学的研究结果表明:通过正交偏最小方差判别分析(OPLS-DA)可以很好地区分羽扇豆醇作用的MCF-7细胞代谢谱与对照组细胞代谢谱,模型参数为:R2Ycum=0.988,Q2Ycum=0.964。VIP(variable importance in the projection)值大于1的差异代谢物进一步用t检验进行单位分析,选择t<0.05(VIP>1)的代谢物作为羽扇豆醇作用组的生物标志物,得到琥珀酸、磷酸、亮氨酸、异亮氨酸等11种代谢差异物。结合羽扇豆醇将细胞周期抑制在G1期这一现象,推测羽扇豆醇可能是主要抑制了三羧酸循环中的琥珀酰辅酶A的生成和底物磷酸化生成ATP的反应来抑制MCF-7细胞的增殖。本实验从代谢组学角度为乳腺癌抗肿瘤机制提供新的线索。  相似文献
