This paper presents a low-capacity cation-exchange chromatography method for the analysis of UV-absorbing dipeptides and amino acids. A newly marketed low-capacity cation-exchange column packed with sulfo-functionalized highly cross-linked macroreticular poly(ethylstyrene-divinylbenzene) copolymer was used for the simultaneous determination of imidazole amino acids, aromatic amino acids, and creatinine in urine samples. A dual-mode binary gradient chromatography method was established using two solvents, A: 15 mM H3PO4/5 mM ethylenediamine and B: 15 mM H3PO4/5 mM ethylenediamine/40 (v/v) % CH3CN at 40 °C, with an optimized time program for changing the delivery ratio of A/B and the flow rate. Good chromatograms were obtained within an acceptable cycle time of 25 min. The quantification data were satisfactory for all analytes, showing the relative standard deviations (RSD) of retention times between 0.08 and 1.68 %; RSDs of area intensities between 0.23 and 2.60 %; and linear regression lines with r 2 more than 0.9994. The method could determine the creatinine ratios of the diagnostic markers on the single chromatographic run, which enabled to discriminate disease from health. For example, the creatinine ratios for phenylketonuria were significantly higher than those for controls. The method can provide highly cost-efficient information or useful knowledge for clinical and pharmaceutical studies.  相似文献

An MEKC method with LIF detection has been developed for the determination of seven neurotransmitter amino acids (NAAs) using 1,3,5,7‐tetramethyl‐8‐(N‐hydroxysuccinimidyl butyric ester)difluoroboradiaza‐S‐indacene as the labeling reagent. After derivatization at room temperature for 30 min, the seven target NAAs including glycine, alanine, γ‐aminobutyric acid, taurine, glutamine, glutamic acid, and aspartic acid were separated in running buffer, which consisted of 70 mM pH 4.00 H3PO4/Na3PO4 buffer, 5.5 mM cetyltrimethyl ammonium bromide and 20% v/v acetonitrile within 17 min. The LODs were 2 ~ 14 × 10?10 M without interference from other coexisting amino acids. The proposed method has been applied to the analysis of NAAs in the central nervous systems of healthy mice and those with Alzheimer's disease with recoveries of 92–104%.  相似文献

Abstract

A sensitive HPLC method with minimal sample preparation and good reproducibility for the determination of furosemide in plasma and urine is described. Acidified plasma samples were extracted using CH2Cl2 containing desmethylnaproxen as internal standard (IS). Fresh urine samples were incubated with β-gluc-uronidase for 15 minutes and then treated with CH3CN containing IS.

Chromatography was performed on a C18 column with 10 mcl sample injection, Mobile phases were: a) for plasma: 0.01 M NaH2PO4, pH 3.5 - CH3OH (65:35), and b) for urine: acetic acid, pH 3.5 - CHS3OH (60:40) at 3 ml/min and fluorescence detection at Ex 235/Em 389 nm. The plasma standard curve was linear from 0.01 to 15.0 mcg/ml and the urine from 0.5 to 200.0 mcg/ml. The within run CV's were 3,2% at 0.74 mcg/ml plasma and 2.0% at 10.7 mcg/ml urine. Recovery from plasma was 69.9% at 2.0 mcg/ml and 98.6% from urine at 5.0 mcg/ml. The stability of furosemide and its glucuronide were studied. Both methods have been applied to the analysis of plasma and urine samples obtained from human volunteers.  相似文献

The pretreatment of corn stover with H2SO4 and H3PO4 was investigated. Pretreatments were carried out from 30 to 120 min in a batch reactor at 121°C, with acid concentrations ranging from 0 to 2% (w/v) at a solid concentration of 5% (w/v). Pretreated corn stover was washed with distilled water until the filtrate was adjusted to pH 7.0, followed by surfactant swelling of the cellulosic fraction in a 0–10% (w/v) solution of Tween-80 at room temperature for 12 h. The dilute acid treatment proved to be a very effective method in terms of hemicellulose recovery and cellulose digetibility. Hemicellulose recovery was 62–90%, and enzymatic digestibility of the cellulose that remained in the solid was >80% with 2% (w/v) acid. In all cases studied, the performance of H2SO4 pretreatment (hemicellulose recovery and cellulose digestibility) was significantly better than obtained with H3PO4. Enzymatic hydrolysis was more effective using surfactant than without it, producing 10–20% more sugar. Furthermore, digestibility was investigated as a function of hemicellulose removal. It was found that digestibility was more directly related to hemicellulose removal than to delignification.  相似文献

Summary A sensitive high performance liquid chromatographic method has been developed for the determination of cinnarizine in human plasma. Cinnarizine and clocinizine (internal standard) were extracted from acidified plasma (pH 4.7) into carbon tetrachloride and the organic layer was evaporated. The products were separated on a Microspher C18 (3 m) column, using a mixture of 0.04 % triethylamine in 0.01 M ammonium dihydrogen phosphate (NH4H2PO4), pH adjusted to 4.2 with orthophosphoric acid (H3PO4), and acetonitrile (2080, v/v) as mobile phase, at a flow rate of 1 ml/min at 40°C. Fluorescence detection (ex = 245 nm, em = 310 nm) was used; the detection limit was 0.5 ng/ml under the conditions used, and the calibration curve linear in the concentration range evaluated (1–60 ng/ml). The assay has been used to measure cinnarizine concentrations in plasma after oral administration to volunteers.  相似文献

The proposed simultaneous determination of thiols and disulfides requires 4- (aminosulfonyl)- 7-fluor-2,1,3,-benzoxadiazole (ABD-F) as the pre-column derivatization reagent for thiols and ammonium 7-fluoro-2,1,3,-benzoxadiazole-4-sulfonate (SBD-F) for disulfides followed by chromatography. The thiols and disulfides in solution are treated with ABD-F at 60°C for 5 min in pH 9.3 borate buffer containing 1 nM disodium-EDTA. After removal of excess of ABD-F with ethyl acetate, the remaining disulfides in the aqueous phase are treated with SBD-F at 60°C for 20 min in the presence of tri-n-butylphosphine, a reducing agent. The ABD-thiols and SBD-thiols thus produced are separated by reversed-phase chromatography and detected by fluorimetry (380- nm excitation, 510 - nm emission). SBD-cystein, SBD-homocystein, ABD-homocysteine, ABD-cysteine, SBD-glu- tathione, ABD-homocystein, SBD-N-acetylcystein, ABD-glutatione and ABD-N-acetylcysteine are well separated by linear gradient elution from 0.15 M H3PO4/CH3CN (96:4) to 0.15 M H3PO4/CH3CN (85:15) over 30 min followed by isocratic elution with 0.15 M H3PO4/CH3CN (85:15) fro 10 min. The detection limits for the derivatives are in the range 0.09–0.9 pmol. When the method was applied to the determination of thiols and disulfides in rat tissues, cystein (0.75 μmol g-1) and cystine (0.62 μmol g-1) were obtained in kidney and reduced glutathione (1.4–3.4 μmol g-1) was observed in liver, spleen, heart and testicle.  相似文献

A rapid and cost‐effective method based on microwave‐assisted extraction followed by capillary electrophoresis was developed for simultaneous quantification of seven alkaloids in Corydalis decumbens for the first time. The main parameters affecting microwave‐assisted extraction and capillary electrophoresis separation were investigated and optimized. The optimal microwave‐assisted extraction was performed at 40°C for 5 min using methanol/water (90:10, v/v) as the extracting solvent. Electrophoretic separation was achieved within 15 min using an uncoated fused‐silica capillary (50 μm internal diameter and 27.7 cm effective length) and a 500 mM Tris buffer containing 45% v/v methanol (titrated to pH* 2.86 with H3PO4). The developed method was successfully applied to the quantification of seven alkaloids in Corydalis decumbens obtained from different regions of China. The combination of microwave‐assisted extraction with capillary electrophoresis was an effective method for the rapid analysis of the alkaloids in Corydalis decumbens .  相似文献

Abstract

Adenine compounds can be measured in picomole amounts using liquid chromatography of the fluorescent 1, N6-etheno derivatives. The limit of detection for the etheno derivatives in tissue extracts, however, is tissue-dependent due to interference by nucleotides and fluorescent components which are normally present. Prior to derivatization nucleotides were partially removed from extracts of lymphocytes and erythrocytes by treatment with Dowex AG1-X2 anion exchange resin. Samples were analyzed using either a Partisil PXS 10/25 SCX column eluted with 100 mM NH4H2PO4, pH 4.5, at a flow rate of 2 ml/min; or using two μBondapak/C18 reversed-phase columns eluted with 5 mM KH2PO,4:25% methanol (V/V) pH 7.5, at a flow rate of 1 ml/min. Adenosine was found to be 0.07 nmole/ml in normal adult human plasma. The urine of a child with severe combined immunodeficiency disease associated with absence of adenosine deaminase contained a normal amount of adenosine (5–6 nmole/ml), but contained a high level (~60 nmole/ml) of deoxyadenosine. Deoxyadenosine was not detected (<0.01 nmole/ml) in normal adult urine. Because of its sensitivity and selectivity, this method of analysis should be applicable to studies of the physiological roles of adenine compounds.  相似文献

In this work, we report a series of cyclophane fluorescent sensors based on acridine combining with imidazolium through ether linkages. X-ray crystal structures demonstrated the self-assembly behavior of these cyclophanes in the solid state driven by hydrogen bond and π–π interactions. Sensors showed excellent selectivity towards Fe3+ in aqueous solution (H2O/CH3CN?=?49:1, v/v) and H2PO4? in acetonitrile solution with notable color change under UV light, evident changes were also noticed in fluorescence spectra. In fluorescence emission, the obvious turn-off was induced by Fe3+ in aqueous solution and the obvious turn-on as well as bathochromic-shift was induced by H2PO4? in acetonitrile solution.  相似文献

The development of a method for the separation of 2′-2′-difluorodeoxycytidine (gemcitabine, dFdC), 2′-2′-difluorodeoxyuridine (dFdU) and their mono-, di- and triphosphates using a porous graphitic carbon column (Hypercarb), without ion-pairing agent, is described. The retention of dFdC and dFdU could be controlled with an organic modifier (acetonitrile, CH3CN) and the retention of the anionic nucleotides with an eluting ion (bicarbonate). Separation of all analytes was achieved using a 0–25 mM ammonium bicarbonate gradient in CH3CN–H2O (15:85, v/v). Under these conditions, however, very long re-equilibration times were required. Injection of an acidic solution (100 μL 10% formic acid in H2O, v/v; 2.65 M) after running a gradient directly restored the separation capabilities of the column. Still, separation between the analytes slowly deteriorated over a period of months. These problems were solved by preconditioning the column with a pH buffered hydrogen peroxide (H2O2) solution (0.05% H2O2 in CH3CN–H2O (15:85, v/v), pH 4) before starting an analytical run. The oxidation of the stationary phase with H2O2 prevented its slow reduction, which most likely caused the decreasing retention times. The analytes were detected using tandem mass spectrometry.  相似文献

In this paper chromatographic systems for the accurate and reliable determination of the percentage of different species (free pertechnetate, particulate99mTc and labelled99mTc) in99mTc-labelled radiopharmaceuticals are described. The procedures are based on the simultaneous run of two different chromatograms, one in 85% CH3OH and the other in 15% H3PO4. Also, 0.9% NaCl can be used instead of 15% H3PO4 for the second chromatogram. Alternative procedures applying two dimensional chromatography on Whatman No. 1 paper or silica G plates with 85% CH3OH for the first run and 15% H3PO4 for the second are given. The applicability of these procedures is limited to water-soluble radiopharmaceuticals.  相似文献

In the study, epoxidation of dodecanol monomaleate, hexadecanol monomaleate, and octadecanol monomaleate were carried out using [π-C5H5N(CH2)15CH3]3PO4(WO3)4 as phase-transfer catalyst with 30% hydrogen peroxide as oxidant. The experimental results showed that [π-C5H5N(CH2)15CH3]3PO4(WO3)4 had excellent catalytic effect on translating C=C double-bond into an epoxy group and had a good yield rate. The synthesized epoxides of decanol monomaleate, hexadecanol monomaleate, and octadecanol monomaleate have enormous potential application in organic synthesis, and they could be readily transformed into various synthetically useful intermediates or final products in many fields of chemistry.   相似文献

Tetraalkylammonium salts [R3RN]+X (R=R, XCH3, Cl; C2H5, Br; C3H7, Br; C4H9, H2PO4; C5H11 Br; R, R, XCH3, C16H33, Br) were investigated as ion-pairing reagents for the reversed-phase ion-pair chromatographic separation of selenite and selenate. Other chromatographic parameters such as composition and pH of the mobile phase and concentration of the ion-pairing reagent were also investigated. The compatibility of the proposed chromatographic procedures with various selenium-specific detectors is discussed. The absolute detection limits were found to be 31 ng Se for selenite and 51 ng Se for selenate (100 l injection) with a solution of 5 mM tetrabutylammonium dihydrogen phosphate in 5050 (v/v) methanol/water as mobile phase at a flow rate of 1 ml/min when a flame atomic absorption spectrometer was used as detector. The HPLC/FAAS system was employed for the determination of selenite in solutions serving as selenium supplement for animals.  相似文献

The chromatofocusing technique has been applied for the formation of ascending linear and quasilinear pH gradients within columns with reversed-phase sorbents Diasorb D-130-C18 and Diasorb-100-CN-mono for the first time. It was found that the optimum composition of mobile phases was 25 mM KH2PO4 + 25% CH3CN (pH 3.0) for the starting solution and 3.15 mM KH2PO4 + 25% CH3CN (pH 6.8–7.0) for the eluent. Separation of components of Maxicold and Efferalgan-C pharmaceuticals has been carried out by the chromatofocusing technique on the Diasorb-100-CN-mono sorbent.  相似文献

The new copper(II) tetraphosphonate, [Cu3(HO3PCH2)2N–CH2C6H4CH2–N(CH2PO3H)2)(H2O)4] n (1) was hydrothermally synthesized from the reaction of Cu(NO3)2 and (H2O3PCH2)2N–CH2C6H4CH2–N(CH2PO3H2)2, (H8L). Compound 1 was structurally characterized by means of X-ray single crystal diffraction. The structure of 1 showed a 3D structure constructed from two types of Cu(II) and chelating and bridging modes of (HO3PCH2)2N–CH2C6H4CH2–N(CH2PO3H)2, and features a (44.610.8) topological network. The 3D compound is further stabilized by hydrogen bonds. UV–Vis diffuse reflectance and infrared spectroscopy as well as elemental analysis of compound 1 are also presented.  相似文献

Kinetic study on the cleavage of N‐(4′‐methoxyphenyl)phthalamic acid (NMPPAH) in mixed H2O‐CH3CN and H2O‐1,4‐dioxan solvents containing 0.05 M HCl reveals the formation of phthalic anhydride (PAn)/phthalic acid (PA) as the sole or major product. Pseudo first‐order rate constants (k1) for the conversion of NMPPAH to PAn decrease nonlinearly from 60.4 × 10?5 to 2.64 × 10?5 s?1 with the increase in the contents of 1,4‐dioxan from 10 to 80% v/v in mixed aqueous solvents. The rate of cleavage of NMPPAH in mixed H2O‐CH3CN solvents at ≥50% v/v CH3CN follows an irreversible consecutive reaction path: NMPPAH PA. Abstract

A high performance liquid chromatographic method for quantitating pentamidine in plasma has been developed. Sample clean-up involved precipitating plasma with acetonitrile containing the internal standard, hexamidine. The supernatant was passed through a C8 Bond Elut column and eluted with a methanolic solution of sodium 1-heptanesulfonate. The eluate was then analyzed on an Altex C8 column with a mobile phase consisting of 45% CH8CN, 0.02% detramethylammonium chloride and 0.1% H3PO4. Using fluorescence detection (EX: 275 nm and EM: 340 nm), the detection limit was 1.25 ng/ml for 0.5 ml of plasma. The coefficients of variation for interday and intraday were around 10%.  相似文献

Wheat straw was pretreated by phosphoric acid plus hydrogen peroxide (PHP), in which temperature, time, and H3PO4 proportion for pretreatment were investigated by using response surface method. Results indicated that hemicellulose and lignin removal positively responded to the increase of pretreatment temperature, H3PO4 proportion, and time. H3PO4 proportion was the most important variable to control cellulose recovery, followed by pretreatment temperature and time. Moreover, these three variables all negatively related to cellulose recovery. Increasing H3PO4 proportion can improve enzymatic hydrolysis; however, reduction on cellulose recovery results in decrease of glucose yield. Extra high temperature or long time for pretreatment was not beneficial to enzymatic hydrolysis and glucose yield. Based on the criterion for minimizing H3PO4 usage and maximizing glucose yield, the optimized pretreatment conditions was 40 °C, 2.0 h, and H3PO4 proportion of 70.2 % (H2O2 proportion of 5.2 %), by which glucose yielded 299 mg/g wheat straw (946.2 mg/g cellulose) after 72-h enzymatic hydrolysis.  相似文献

Inductively coupled plasma mass spectrometry (ICP-MS) used for the detection of most common selenium isotopes 78Se (23.8%, abundance) and 80Se (49.6%, abundance) is interfered in the presence of 38Ar40Ar+ and 40Ar40Ar+ in argon (Ar) gas. To address this issue, ICP-MS with an octopole reaction system (ORS) was explored for the detection of selenite and selenate, which was separated by anion-exchange chromatography. Results indicate that it was possible to detect 78Se using no collusion gas, while to detect 80Se a H2 as the collusion/reaction gas was recommended since the background and noise were significantly reduced using H2 as the gas. The selenium speciation interested was separated on a new column (G3154/101A, Agilent technologies) within 5 min using a mobile phase containing 10 mM NH4H2PO4 and 20 mM NH4NO3 at pH 6.5. Linear plots were obtained in a concentration range of 1–200 μg/L with detection limits less than of 0.4 μg/L for 80Se (IV) and 0.6 μg/L for 80Se (VI) using H2 as the reaction gas. Finally, the proposed method was used in the determination of selenium in water and soil.  相似文献
