A HPLC method with automated column switching and UV-diode array detection is described for the simultaneous determination of Vitamin D3 and 25-hydroxyvitamin D3 (25-OH-D3) in a sample of human plasma. The system uses a BioTrap precolumn for the on-line sample cleanup. A sample of 1 ml of human plasma was treated with 2 ml of a mixture of ethanol–acetonitrile (2:1 (v/v)). Following centrifugation, the supernatant was evaporated to dryness under a stream of dry and pure nitrogen. The residue was reconstituted in 250 μL of a solution of methanol 5 mmol l−1 phosphate buffer, pH 6.5 (4:1 (v/v)), and a 200 μl aliquot of this solution was injected onto the BioTrap precolumn. After washing during 5 min with a mobile phase constituted by a solution of 6% acetonitrile in 5 mmol l−1 phosphate buffer, pH 6.5 (extraction mobile phase), the retained analytes were then transferred to the analytical column in the backflush mode. The analytical separation was then performed by reverse-phase chromatography in the gradient elution mode with the solvents A and B (Solvent A: acetonitrile–phosphate buffer 5 mmol l−1, pH 6.5; 20:80 (v/v); solvent B: methanol–acetonitrile–tetrahydrofuran, 65:20:15 (v/v)). The compounds of interest were detected at 265 nm. The method was linear in the range 3.0–32.0 ng ml−1 with a limit of quantification of 3.0 ng ml−1. Quantitative recoveries from spiked plasma samples were between 91.0 and 98.0%. In all cases, the coefficient of variation (CV) of the intra-day and inter-day-assay precision was ≤2.80%. The proposed method permitted the simultaneous determination of Vitamin D3 and 25-OH-D3 in 16 min, with an adequate precision and sensitivity. However, the overlap of the sample cleanup step with the analysis increases the sampling frequency to five samples h−1. The method was successfully applied for the determination of Vitamin D3 and 25-OH-D3 in plasma from 46 female volunteers, ranging from 50 to 94 years old. Vitamin D3 and 25-OH-D3 concentrations in plasma were found from 4.30–40.70 ng ml−1 (19.74 ± 9.48 ng ml−1) and 3.1–36.52 ng ml−1 (7.13 ± 7.80 ng ml−1), respectively. A reversed flow injection colorimetric procedure for determining iron(III) at the μg level was proposed. It is based on the reaction between iron(III) with norfloxacin (NRF) in 0.07 mol l−1 ammonium sulfate solution, resulting in an intense yellow complex with a suitable absorption at 435 nm. Optimum conditions for determining iron(III) were investigated by univariate method. The method involved injection of a 150 μl of 0.04% w/v colorimetric reagent solution into a merged streams of sample and/or standard solution containing iron(III) and 0.07 mol l−1 ammonium sulfate in sulfuric acid (pH 3.5) solution which was then passed through a single bead string reactor. Subsequently the absorbance as peak height was monitored at 435 nm. Beer's law obeyed over the range of 0.2–1.4 μg ml−1 iron(III). The method has been applied to the determination of total iron in water samples digested with HNO3–H2O2 (1:9 v/v). Detection limit (3σ) was 0.01 μg ml−1 the sample through of 86 h−1 and the coefficient of variation of 1.77% (n=12) for 1 μg ml−1 Fe(III) were achieved with the recovery of the spiked Fe(III) of 92.6–99.8%.  相似文献

Cadmium concentration in lake sediments is determined by suspending the solid samples in a solution containing 5% (v/v) concentrated nitric acid and 0.1% (v/v) Triton X-100. Three modifiers were tested for the direct determination. The furnace temperature programmes and appropriate amount for each modifier were optimised to get the highest signal and the best separation between the atomic and background signals. The drying stage is performed by programming a 400 °C temperature, a ramp time of 25 s and hold time of 10 s on the power supply of the atomiser. No ashing step is used and platform atomisation is carried out at 2200 °C. W–Rh permanent modifier combined with conventional modifier by delivering 10 μl of 0.50% (w/v) NH4H2PO4 solution was the best chemical modifier for cadmium determination. This modifier also acts as a liquid medium for the slurry, thus simplifying the procedure. Calibration is performed using aqueous standards in the 1–5 μg l−1 range. The optimised method gave a limit of detection of 0.56 ng ml−1, characteristic mass of 10.1±0.8 pg for aqueous standard, 9.6±0.7 pg for slurry samples containing different Cd concentrations and good precision (7.6–5.2%). The method was validated by analysing four certified reference lake sediment materials: LKSD-1, LKSD-2, LKSD-3 and LKSD-4; satisfactory recoveries were obtained (90.0–96.3%) and no statistical differences were observed between the experimental and the certified cadmium concentration. The developed methodology was used to determine cadmium in three ‘real’ sediment samples from lakes in the area of Wielkopolski National Park, Poland.  相似文献

High-speed counter-current chromatography (HSCCC) coupled with evaporative light scattering detection (ELSD) was successfully applied to preparative separation and purification of verticine and verticinone from crude extracts of Bulbus Fritillariae Thunbergii by a one-step separation, using chloroform–ethanol–0.2 mol L−1 hydrochloric acid (3:2:2, v/v/v) as a solvent system. HPLC analysis of the fractions collected on the preparative HSCCC of 200 mg of crude extracts showed that the purity of verticine (25.6 mg) was 96.8% and that of verticinone (10.3 mg) was 95.4%. The chemical identities of these components were confirmed by 1H NMR and EI–MS.  相似文献

An on-line flow injection (FI) precipitation–dissolution system with microwave-assisted sample digestion has been developed for the electrothermal atomic absorption spectrometry (ETAAS) determination of trace or ultratrace amounts of molybdenum in human blood serum and whole blood samples. After the exposure of the sample to microwave radiation, the on-line precipitation of molybdenum was achieved by the merging-zone of a 0.5-ml plug of sample with a plug of potassium ferrocyanide, which were carried downstream with a solution of 0.5 mol l−1 of HNO3. The interfering effects of iron and copper were minimized by the introduction of a flow of a 5% (w/v) sodium potassium tartrate (for iron) and 2% (w/v) of thiourea (for copper and zinc) in a 5% (v/v) ammonia and 2% (v/v) ammonium chloride solution previous to the precipitation reaction. The reddish-brown precipitate of molybdenyl ferrocyanide was collected on the walls of a knotted reactor. The precipitate was dissolved with the introduction of 1 ml of a 3.0 mol l−1 NaOH solution and the best performance in terms of detection limit and precision was achieved when a sub-sample of 140 μl was collected in a capillary of a sampling arm assembly, to introduce 20 μl volumes into the atomizer by means of positive displacement with air through a time-based injector. A detection limit (3σ) of 0.1 μg Mo l−1 using an aqueous standard solution was obtained. The method is quantitative and is applied over the range 0.2–20.0 μg Mo l−1. The precision of the method evaluated by ten replicate analyses of aqueous standard solutions containing 0.5 and 1.0 μg Mo l−1 was 2.8 and 3.1% (relative standard deviation, RSD) (for n=5), respectively. Whereas, the precision evaluated by five replicate analysis of a serum and a whole blood sample were 3.3 and 3.8% RSD. An enrichment factor of ca. 3.5 was achieved with the introduction of 0.5 ml aqueous standard solutions at a sample flow rate of 1.0 ml min−1. Recoveries of spiked molybdenum in blood serum and whole blood were in the ranges 96–102 and 94–98%, respectively. The results obtained for two human whole blood certified reference materials were in good agreement with the indicative values.  相似文献

A reversed-phase liquid chromatography method for analysis of polymyxin B sulphate is described. The method uses a YMC-Pack Pro, C18, 5 μm, 250×4.6 mm I.D. column maintained at 30°C. The mobile phase comprises acetonitrile–sodium sulphate (0.7%, m/v)–phosphoric acid (6.8%, v/v dilution of 85%, m/m phosphoric acid)–water (22.25:50:5:22.75) at a flow-rate of 1.0 ml/min. Detection was by UV at 215 nm. The method is able to resolve polymyxin B1, the major component, from more than thirty other components present in the complex. Robustness was evaluated by performing a full-factorial design experiment. The method showed good selectivity, repeatability, linearity and sensitivity.  相似文献

Quantitative determination of vitamin B12 in B-complex tablets was performed by using multimode high-performance liquid chromatography. The multivitamin tablets (B1, B6 and B12) were sonicated for 30 min in methanol–water (50:50, v/v) and diluted to appropriated volume with the same solvent. The resulting solution was filtered and the filtrate was analysed on a phenylpropanolamine bonded silica column (15 cm×4.6 mm I.D., 5 μm). The optimized mobile phase was 30 mM phosphate buffer (pH 3.00) containing 6% (v/v) acetonitrile at a flow-rate of 1 ml min−1 and the detection was measured at 361 nm. The calibration graph prepared using standards was linear from 0.05 to 0.25 μg. The determination limit was 25 ng, the relative standard deviation was 0.47% and recovery from tablet solution was 100%. An analysis was completed in 5 min. The new method is simple, rapid and precise.  相似文献

An analytical method for separation and pre-concentration of lead in seawater for determination by inductively coupled plasma optical emission spectrometry has been investigated. Lead was retained in the solid phase (0.5 g) composed of co-precipitated naphthalene and alizarin red. The solid phase quantitatively sorbs Pb(II) at pH 8–9, and the metal was eluted using 5.0 ml of 2 mol l−1 nitric acid. The effect of NaCl, KCl, BaCl2, CaCl2, Na2SO4, MgCl2 and Na3PO4 on the sorption of Pb(II) in the solid phase was studied. A set of solutions containing varying amounts of electrolytes (0.5; 1.0; 3.0 and 5.0% m/v) with Pb (50 μg) was prepared and the recommended procedure applied. The Na3PO4 was found to interfere; the other electrolytes did not interfere up to 5% m/v. A pre-concentration factor of 40 was obtained in this analytical procedure. The limit of detection and limit of quantification for Pb(II) were 53 and 176 μg l−1, respectively. Lead was determined in seawater samples collected in Salvador city, Bahia, Brazil. The precision, expressed as R.S.D., was 1.8–4.6%, and the recovery of lead added to seawater samples was 95–97%.  相似文献

An on-line flow injection system has been developed for the selective determination of Se(IV) and Se(VI) in citric fruit juices and geothermal waters by hydride generation atomic absorption spectrometry with microwave-aided heating prereduction of Se(VI) to Se(IV). The samples and the prereductant solutions (4 mol l−1 HCl for Se(IV) and 12 mol l−1 HCl for Se(VI)) which circulated in a closed-flow circuit were injected by means of a time-based injector. This mixture was displaced by a carrier solution of 1% v/v of hydrochloric acid through a PTFE coil located inside the focused microwave oven and mixed downstream with a borohydride solution to generate the hydride. The linear ranges were 0–120 and 0–100 μg l−1 of Se(IV) and Se(VI), respectively. The detection limits were 1.0 μg l−1 for Se(IV) and 1.5 μg l−1 for Se(VI). The precision (about 2.0–2.5% RSD) and recoveries (96–98% for Se(IV) and 94–98% for Se(VI)) were good. Total selenium values were also obtained by electrothermal atomic absorption spectrometry which agreed with the content of both selenium species. The sample throughput was about 50 measurements per hour. The main advantage of the method is that the selective determination of Se(IV) and Se(VI) in citric fruit juices and geothermal waters is performed in a closed system with a minimum sample manipulation, exposure to the environment, minimum sample waste and operator attention.  相似文献

A low solvent consumption method for Fourier transform infrared spectroscopy (FT-IR) determination of doxylamine succinate in pharmaceuticals has been developed. The analyte was continuous and selectively extracted with a 13% (v/v) ethanol:chloroform solvent mixture, recirculating the solvent through the sample and monitoring the process by FT-IR. Doxylamine succinate was determined by on-line standard addition measuring the peak area in the regions 1730–1710 and 1485–1462 cm−1 corrected with a two-point baseline established between 2000 and 1800 cm−1. This new method implies low volumes of chloroformic solvent mixture, only 2.6 mL per sample, in front of classical batch FT-IR methods, improving analytical efficiency and reducing waste generation. The on-line extraction and standard addition determination of doxylamine succinate allowed a throughput of 10 h−1.  相似文献

An inductively coupled plasma mass spectrometer (ICP-MS) was used as an ion chromatographic (IC) detector for the speciation analysis of arsenic and selenium. The arsenic and selenium species studied included arsenite [As(III)], arsenate [As(V)], monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), arsenobetaine (AsB), selenite [Se(IV)] and selenate [Se(VI)]. Gradient elution using (NH4)2CO3 and methanol at pH 9 allowed the chromatographic separation of all species in less than 12 min. Effluents from the IC column were delivered to the nebulization system of ICP-DRC-MS for the determination of arsenic and selenium. The potentially interfering 38Ar40Ar+ and 40Ar40Ar+ at the selenium masses m/z 78 and 80 were reduced in intensity by approximately 3 orders of magnitude by using 0.6 mL min−1 CH4 as reactive cell gas in the DRC while an Rpq value of 0.3 was used. Meanwhile, arsenic was determined as the adduct ion 75As12CHH+ at m/z 89, which is more sensitive than 75As. The limits of detection for arsenic and selenium were in the range of 0.002–0.01 ng mL−1 and 0.01–0.02 ng mL−1, respectively, based on peak height. The relative standard deviation of the peak areas for five injections of 5 ng mL−1 As and Se mixture was in the range of 2–4%. The concentrations of arsenic and selenium species have been determined in urine samples collected locally. The major As and Se species in urines were AsB, DMA and probably selenosugar at concentration of 20–40, 15–19 and 17–31 ng mL−1, respectively. The recoveries were in the range of 94–105% for all the determinations. This method has also been applied to determine various arsenic compounds in two fish samples. In this study, a simple and rapid microwave-assisted extraction method was used for the extraction of arsenic compounds from fish. The arsenic species were quantitatively leached with an 80% v/v methanol solution in a focused microwave field during a period of 5 min.  相似文献

A mild, efficient and convenient extraction method of using 2-mercaptoethanol contained extractant solution combined with an incubator shaker for determination of mercury species in biological samples by HPLC–ICP-MS has been developed. The effects of the concentration of 2-mercaptoethanol, the composition of the extractant solution and the shaking time on the efficiency of mercury extraction were evaluated. The optimization experiments indicated that the quantitative extraction of mercury species from biological samples could be achieved by using 0.1% (v/v) HCl, 0.1% (v/v) 2-mercapoethanol and 0.15% (m/v) KCl extractant solution in an incubator shaker for shaking overnight (about 12 h) at room temperature. The established method was validated by analysis of various biological certified reference materials, including NRCC DOLT-3 (dogfish liver), IAEA 436 (tuna fish), IAEA MA-B-3/TM (garfish filet), IAEA MA-M-2/TM (mussel tissue), GBW 08193 (bovine liver) and GBW 08572 (prawn). The analytical results of the reference materials were in good agreement with the certified or reference values of both methyl and total mercury, indicating that no distinguishable transformation between mercury species had occurred during the extraction and determination procedures. The limit of detection (LOD) for methyl (CH3Hg+) and inorganic mercury (Hg2+) by the method are both as 0.2 μg L−1. The relative standard deviation (R.S.D.s) for CH3Hg+ and Hg2+ are 3.0% and 5.8%, respectively. The advantages of the developed extraction method are that (1) it is easy to operate in HPLC–ICP-MS for mercury species determination since the extracted solution can be directly injected into the HPLC column without pH adjustment and (2) the memory effect of mercury in the ICP-MS measurement system can be reduced.  相似文献

A highly selective and sensitive rhodamine-based colorimetric chemosensor (1) for quantification of divalent copper in aqueous solution has been investigated in this work. It was designed using salicylaldehyde hydrazone and rhodamine 6G as copper-chelating and signal-reporting groups, respectively. In environmentally friendly media (50% (v/v) water/ethanol and 10 mM NaAc–HAc neutral buffer (pH 7.0)), the sensor exhibited selective absorbance enhancement to Cu2+ over other metal ions at 529 nm, with a dynamic working range of 0.05–5.00 μM and a detection limit of 10 nM Cu2+, respectively. To achieve fluorometric determination of Cu2+, the Cu2+-induced absorbance enhancement of 1 was efficiently converted to fluorescence quenching by fluorescence inner filter effects using rhodamine B (RB) as a fluorophore. The selectivity and sensitivity of fluorescence analysis were similar to those of absorptiometric measurement. Both absorptiometric and fluorometric methods were successfully applied to the detection of Cu2+ in three water samples.  相似文献

A new aqueous two-phase system is developed consisting of sodium perfluorooctanoate (SPFO) and dodecyltriethylammonium bromide (C12NE) cationic–anionic surfactant mixture. The two phases with a clear interfacial boundary formed when SPFO to C12NE molar ratio is 1.2:1 in the presence of 5% (v/v) nitric acid. The top phase is transparent and the bottom phase is opalescent. Extractions of dyes, porphyrin compounds with the two-phase system were performed. The results show that hydrophobic molecules were extracted into the surfactant-rich bottom phase with high extraction efficiencies. Positively charged porphyrins were extracted into the bottom phase with higher extraction efficiencies than negatively charged porphyrins. Such a new anionic surfactant two-phase system would be complementary to the C12NE–SDS (sodium dodecyl sulfate) cationic two-phase which has been proven to be effective for extractions of porphyrins with substituted groups like carboxyl or sulfonic acid groups.  相似文献

The absorption and emission properties of rhodamine 101 in polar, aprotic propionitrile-butyronitrile (4:5 v/v) solvent have been investigated. The equilibrium between the neutral zwitterionic and lactonic (colourless) forms is strongly affected by the temperature in the range 293–144 K. In dilute solution (1.12×10−5M), at room temperature the lactonic form is 98%, while at 144 K the zwitterionic form predominates, being 79%. These findings are discussed in terms of differential stabilization of the lactonic and zwitterionic forms through non-specific solute-solvent interactions involving the COO−1 group.  相似文献

Simple and efficient analytical HPLC methods using Chiralpak AS-H as chiral stationary phase were developed for direct enantioseparation of 11 novel phenylethanolamine derivatives. The chromatographic experiments were performed in normal phase mode with n-hexane–ethanol–triethylamine (TEA) as mobile phase. Excellent baseline enantioseparation was obtained for most of compounds. The effects of the concentration of organic modifiers and column temperature were studied for the enantiomeric separation. The mechanism of chiral recognition was discussed based on the relationship between the thermodynamic parameters and structures of compounds. It was found that the enantioseparations were all enthalpy driven, and the tert-butyl groups of compounds had significant influence on the chiral recognition. Trantinterol enantiomers were resolved (Rs = 2.73) within 14 min using n-hexane–ethanol–TEA (98:2:0.1, v/v/v) as mobile phase with a flow rate of 0.8 mL min−1 at 30 °C. The optimized method was validated for linearity, precision, accuracy and stability in solution and proved to be robust. The limits of detection (LOD) and quantification (LOQ) for (+)-trantinterol were 0.15 and 0.46 μg mL−1. The method was applied for enantiomeric impurity determination of (−)-trantinterol bulk samples.  相似文献

We have evaluated the hydroxyl group–solvent and carbonyl group–solvent specific interactions by using mostly an Alltima C18 stationary phase and subsidiarily squalane-adsorbed C18 phase, and by measuring the retention data of carefully selected solutes in 60:40, 70:30, 80:20 and 90:10 (%, v/v) methanol–water eluents at 25, 30, 35, 40, 45 and 50°C. The selected solutes are four positional isomers of phenylbutanol, 5-phenyl-1-pentanol, three positional isomers of alkylarylketone derived from butylbenzene, and 1-phenyl-2-hexanone. The magnitudes of carbonyl group–solvent specific interaction enthalpies are larger than those of hydroxyl group–solvent specific interaction enthalpies in general. We observed clear discrepancies in functional group–solvent specific interactions among positional isomers. The spatial accessibility of the functional group by the solvent molecules seems to govern the strength of interaction. The relationships between molecular structures and functional group accessibilities have been discussed. The specific functional group–mobile phase interactions obtained by the Alltima C18 stationary phase were systematically different from those obtained by the squalane-impregnated C18 stationary phase, which may be due to structural differences between the two phases.  相似文献

In the present work, a reversed-phase high-performance liquid chromatographic procedure has been developed for the determination of water-soluble vitamins (thiamine hydrochloride, pyridoxine hydrochloride, nicotinamide, riboflavin phosphoric ester and cyanocobalamine) and fat-soluble vitamins (retinol palmitate, cholecalciferol, -tocopherol acetate) in multi-vitamin pharmaceutical formulations. The sample treatment proposed consists of a solid-phase extraction with C18 AR cartridges that allow the separation of fat-soluble vitamins, which were retained on the sorbent, from water-soluble vitamins. Afterwards, the water-soluble vitamins were analysed by HPLC on a Nova-Pack C18 (150×3.9 mm, 4 μm) analytical column, using CH3OH–0.05 M CH3COONH4 as mobile phase The chromatographic analysis of the fat-soluble vitamins was carried out after their sequential elution with methanol and chloroform from C18 sorbent, on the above column. The mobile phase employed was MeOH–CH3CN (95:5, v/v) working at a flow-rate of 2 ml min−1 in isocratic mode. The solid-phase extraction for these vitamins had been previously optimised. The experimental variables studied were: application volume, elution solvents and cleaning solutions. The UV–Vis detection of vitamins was made at 270 nm for all the water-soluble vitamins (362 nm for B12) and 285 nm for the water-soluble and fat-soluble vitamins present in real samples at different concentration levels. The accuracy of the method was tested obtaining an average recovery ranging between 78 and 116%.  相似文献

A solid-phase extraction (SPE) method followed by a reversed-phase high-performance liquid chromatography (HPLC) procedure is reported for the assay of a wide polarity range acaricide residues in honey. After selection of suitable chromatographic and detection conditions, most steps of the SPE procedure that may affect to the recovery were investigated. Honey sample was buffered at pH 6 and then applied to the preconditioned C18 sorbent. A washing step was performed with 1 ml of a mixture of tetrahydrofuran (THF)–phosphate buffer (10:90, v/v) and finally, the analytes were eluted with 1 ml of THF. The extract was evaporated to dryness, reconstituted in mobile phase and chromatographed on a reversed-phase C18 column with diode array detection. The recoveries of the more polar acaricides were higher than 80% and 60–70% for the more apolar ones. Limits of detection obtained ranged from 1 to 200 ng/g.  相似文献

The ab initio molecular structures and vibrational frequencies for several transition metal heptoxides X2O7n (n=0, 2, 4) were calculated using effective core potentials at the HF and DFT (B3LYP) levels. The relative merits of different valence basis set arrangements were tested by comparison with experimental results available, in particular with gas-phase Re2O7 molecular structure and vibrational frequencies. The calculations were then extended to other heptoxides of the VB, VIB and VIIB transition metal groups. The results indicate that a staggered geometry (either D3d or C2) is the energy minimum for most of the heptoxides studied. The only exceptions are Mn2O7, which clearly prefers an eclipsed C2v(syn) configuration, and Tc2O7, for which C2 and C2v(syn) geometries have nearly the same energy.

Particular attention was given to the magnitude of the X–O–X bond angle, as this structural parameter has been a matter of some controversy. The calculated values range from 125° (Mn2O7) to 180° (Group VB heptoxides) and depend on the position of the transition metal in the Periodic Table. A tendency for linearity of the X–O–X moiety on going both downwards the group and backwards the period was observed and discussed.  相似文献
