In this communication, the surface activity of the ovalbumin (OVA) at the air/water interface was studied to establish the nature of the interaction with the stearic acid (SA) monolayer, based on Langmuir–Blodgett (LB) technique. The interaction was monitored by studying the time (t) variation of surface pressure (π) at constant area (A). The growth of π with time indicates a positive association between the SA and the OVA molecules. The surface compressibility analysis has been performed to specify the phase transition of OVA–SA mixed monolayer. Incorporation/association of OVA within the SA monolayer led to noteworthy changes in surface compressibility and was surface pressure as well as protein concentration dependent. Both the hydrophobic and the Vander wall type interactions are found to be responsible for the association. The quenching of tyrosine band in tryptophan excitation spectrum is observed in steady-state fluorescence spectroscopy. This suggests that the tyrosine is the probable binding site with SA. Due to incorporation of SA, the energy transfer from tyrosine to tryptophan is hindered. At higher pressure, OVA tend to squeeze out from the SA monolayer. The high-resolution field emission scanning electron microscope (FE-SEM) image confirms this observation. Aggregated protein structure observed at high pressure indicates unfolding of protein.  相似文献

Stearic acid (SA) and octadecylamine (ODA) monolayers at the air/liquid interface were used as template layers to adsorb glucose oxidase (GOx) from aqueous solution. The effect of the template monolayers on the adsorption behavior of GOx was studied in terms of the variation of surface pressure, the evolution of surface morphology observed by BAM and AFM, and the conformation of adsorbed GOx. The results show that the presence of a template monolayer can enhance the adsorption rate of GOx; furthermore, ODA has a higher ability, compared to SA, to adsorb GOx, which is attributed to the electrostatic attractive interaction between ODA and GOx. For adsorption performed on a bare surface or on an SA monolayer, the surface pressure approaches an equilibrium value (ca. 8 mN/m) after 2 to 3 h of adsorption and remains nearly constant in the following adsorption process. For the adsorption on an ODA monolayer, the surface pressure will increase further 1 to 2 h after approaching the first equilibrium pressure, which is termed the second adsorption stage. The measurement of circular dichroism (CD) spectroscopy indicates that the Langmuir-Blodgett films of adsorbed GOx transferred at the first equilibrium state (π = 8 mN/m) have mainly a β-sheet conformation, which is independent of the type of template monolayers. However, the ODA/GOx LB film transferred at the second adsorption stage has mainly an α-helix conformation. It is concluded that the specific interaction between ODA and GOx not only leads to a higher adsorption rate and adsorbed amount of GOx but also induces a conformation change in adsorbed GOx from β-sheet to α-helix. The present results indicate that is possible to control the conformation of adsorbed protein by selecting the appropriate template monolayer.  相似文献

Cholesterol is a main component of the cell membrane and could have significant effects on drug-cell membrane interactions and thus the therapeutic efficacy of the drug. It also plays an important role in liposomal formulation of drugs for controlled and targeted delivery. In this research, Langmuir film technique, atomic force microscopy (AFM) and Fourier transform infrared spectroscopy (FTIR) are employed for a systematic investigation on the effects of cholesterol component on the molecular interactions between a prototype antineoplastic drug (paclitaxel) and 1,2-dipalmitoyl-sn-glycerol-3-phosphocholine (DPPC) within the cell membrane by using the lipid monolayer at the air-water interface as a model of the lipid bilayer membrane and the biological cell membrane. Analysis of the measured surface pressure (pi) versus molecular area (a) isotherms of the mixed DPPC/paclitaxel/cholesterol monolayers at various molar ratios shows that DPPC, paclitaxel and cholesterol can form a non-ideal miscible system at the air-water interface. Cholesterol enhances the intermolecular forces between paclitaxel and DPPC, produces an area-condensing effect and thus makes the mixed monolayer more stable. Investigation of paclitaxel penetration into the mixed DPPC/cholesterol monolayer shows that the existence of cholesterol in the DPPC monolayer can considerably restrict the drug penetration into the monolayer, which may have clinical significance for diseases of high cholesterol. FTIR and AFM investigation on the mixed monolayer deposited on solid surface confirmed the obtained results.  相似文献

Interaction of native ovalbumin (OVA) with 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) Langmuir-Blodgett monolayer has been studied at the air-water interface. A compressibility study shows the positive association with DPPC. Adsorption kinetics shows that the protein adsorption is a one-step process and the amount of protein adsorbed depends on the concentration of protein at the water subphase. Incorporation of protein into the DPPC layer is surface-pressure dependent. The compressibility study indicates that the DPPC-OVA interaction is hydrophobic in nature and structural reorganization is eminent to adjust the hydrophobic mismatch between DPPC acyl chains and OVA hydrophobic moieties. At higher pressure, OVA tends to squeeze out from the DPPC monolayer. A nanometer scale FE-SEM image confirms this observation. Globular aggregates of protein of dimension 60-80 nm were observed in DPPC-OVA supported film. Steady-state fluorescence spectroscopy suggests that the tryptophan residues of OVA are main emitting species. The blue shift of tryptophan fluorescence in supported film may be due to the tryptophan molecule of protein exposed to the hydrophobic air phase.  相似文献

This study investigated the relaxation behaviors of octadecylamine (ODA), stearic acid (SA), and SA/ODA mixed monolayers at the air/water interface. Area relaxations of monolayers at constant surface pressure were studied by a nucleation and growth mechanism and by direct observation using a Brewster angle microscope (BAM). The results showed that ODA and SA monolayers exhibit different characteristics in the area loss and in the BAM morphology. In the initial relaxation stage, SA monolayer illustrates a more stable characteristic than ODA. But at the later stage, the area loss of SA monolayer increases more quickly than that for ODA due to significant nucleation and growth of 3D aggregates. The BAM results demonstrated that 3D aggregates of large scale domains are likely to form on a SA monolayer even when the area loss is insignificant. On the contrary, only dotlike aggregates of low density were found on the ODA monolayer when relaxation is carried out at higher surface pressure. The relaxation behavior of SA monolayer can be described by the Vollhardt model. However, the relaxation of ODA monolayer does not follow the nucleation model described by Vollhard but can reasonably be attributed to the effect of dissolution. For the SA/ODA mixed monolayers, the relaxation behaviors in the initial and final stages follow different mechanisms, which is attributed to the formation of distinct phases as observed from the BAM. This result also implied that SA and ODA are not completely miscible to be a homogeneous phase. Phases of various compositions were formed in the mixed monolayers, and thus, the relaxation mechanism was shifted during the relaxation process as dominated by different relaxation behaviors of various phases.  相似文献

Gold nanoparticles (Au NPs) were prepared and surface-modified by mercaptosuccinic acid (MSA) to render a surface with carboxylic acid groups (MSA-Au). Octadecylamine (ODA) was used as a template monolayer to adsorb the Au NPs dispersed in the subphase. The effect of MSA concentration on the incorporation of Au NPs on the ODA monolayer and the relevant behavior of the mixed monolayer were studied using the pressure-area (pi-A) isotherm and transmission electron microscopy (TEM) observations. The experimental results showed that the adsorbed density of Au NPs is low without the surface modification by MSA. When MSA was added into the Au NP-containing subphase, the incorporation amount of Au NPs increased with increasing MSA concentration up to approximately 1 x 10-5 M for the particle density of 1.3 x 1011 particles/mL. With a further increase in the MSA concentration, the adsorbed particle density decreases due to competitive adsorption between the free MSA molecules and the MSA-Au NPs. It is inferred that free MSA molecules adsorb more easily than the MSA-Au NPs on the ODA monolayer. Therefore, an excess amount of MSA present in the subphase is detrimental to the incorporation of gold particles. The study on the monolayer behavior also shows that the pi-A isotherm of the ODA monolayer shifts right when small amounts of Au NPs or free MSA molecules are incorporated. However, when larger amounts of particles are adsorbed at the air/liquid interface, a left shift of the pi-A isotherm appears, probably due to the adsorption of ODA molecules onto the particle surface and the transferring of the particles from beneath the ODA monolayer to the air/water interface. According to the present method, it is possible to prepare uniform particulate films of controlled densities by controlling the particle concentration in the subphase, the MSA concentration, and the surface pressure of a mixed monolayer.  相似文献

The role of dipalmitoylphosphatic acid (DPPA) as a transfer promoter to enhance the Langmuir-Blodgett (LB) deposition of a dipalmitoylphosphatidylcholine (DPPC) monolayer at air/liquid interfaces was investigated, and the effects of Ca2+ ions in the subphase were discussed. The miscibility of the two components at air/liquid interfaces was evaluated by surface pressure-area per molecule isotherms, thermodynamic analysis, and by the direct observation of Brewster angle microscopy (BAM). Multilayer LB deposition behavior of the mixed DPPA/DPPC monolayers was then studied by transferring the monolayers onto hydrophilic glass plates at a surface pressure of 30 mN/m. The results showed that the two components, DPPA and DPPC, were miscible in a monolayer on both subphases of pure water and 0.2 mM CaCl2 solution. However, an exception occurs between X(DPPA)=0.2 and 0.5 at air/CaCl2-solution interface, where a partially miscible monolayer with phase separation may occur. Negative deviations in the excess area analysis were found for the mixed monolayer system, indicating the existence of attractive interactions between DPPA and DPPC molecules in the monolayers. The monolayers were stable at the surface pressure of 30 mN/m for the following LB deposition as evaluated from the area relaxation behavior. It was found that the presence of Ca2+ ions had a stabilization effect for DPPA-rich monolayers, probably due to the association of negatively charged DPPA molecules with Ca2+ ions. Moreover, the Ca2+ ions may enhance the adhesion of DPPA polar groups to a glass surface and the interactions between DPPA polar groups in the multilayer LB film structure. As a result, Y-type multilayer LB films containing DPPC could be fabricated from the mixed DPPA/DPPC monolayers with the presence of Ca2+ ions.  相似文献

To make ultrathin films for the fabrication of artificial olfactory systems, odorant biosensors, we have investigated mixed Langmuir and Langmuir-Blodgett films of odorant-binding protein/amphiphile. Under optimized experimental conditions (phosphate buffer solution, pH 7.5, OBP-1F concentration of 4 mg L(-1), target pressure 35 mN m(-1)), the mixed monolayer at the air/water interface is very stable and has been efficiently transferred onto gold supports, which were previously functionalized by self-assembled monolayers (SAMs) with 1-octadecanethiol (ODT). Atomic force microscopy and electrochemical impedance spectroscopy were used to characterize mixed Langmuir-Blodgett (LB) films before and after contact with a specific odorant molecule, isoamyl acetate. AFM phase images show a higher contrast after contact with the odorant molecule due to the new structure of the OBP-1F/ODA LB film. Non-Faradaic electrochemical spectroscopy (EIS) is used to quantify the effect of the odorant based on the electrical properties of the OBP-1F/ODA LB film, as its resistance strongly decreases from 1.18 MOmega (before contact) to 25 kOmega (after contact).  相似文献

This paper reports the pi-A isotherms and spectroscopic characteristics of mixed Langmuir and Langmuir-Blodgett (LB) films of nonamphiphilic carbazole (CA) molecules mixed with polymethyl methacrylate (PMMA) and stearic acid (SA). pi-A isotherm studies of mixed monolayer as well as the remarkable change in collapse pressure of the mixed monolayer isotherms definitely show that CA is incorporated into PMMA and SA matrices. However, CA is stacked in the PMMA/SA chains and forms microcrystalline aggregates, as is evidenced from the scanning electron micrograph picture. The nature of these aggregated species in the mixed LB films has been revealed by UV-vis absorption and fluorescence spectroscopic studies. The presence of two different kinds of band systems in the fluorescence spectra of the mixed LB films have been observed. This may be due to the formation of low-dimensional aggregates in the mixed LB films. Intensity distribution of different band systems is highly sensitive to the microenvironment of two different matrices as well as also on the film thickness.  相似文献

Mixed monolayers of an acylated derivative of hepatitis A synthetic peptide VP3(110-121) with neutral, cationic or anionic lipids were spread at the air/water interface. Deviations from ideality as well as thermodynamic values were calculated at different surface pressures using the free-excess energy, the interaction parameter and the enthalpy. The miscibility at the collapse point was also checked. Maximum deviations from ideality were found for mixtures containing the anionic lipid phosphatidylglycerol (PG), and it seems that the monolayer composition is not stable through compression, as the peptide is ejected from the film. Films containing neutral [dipalmitoylphosphatidylcholine (DPPC)] or cationic [stearylamine (SA)] lipids showed more regular behaviour. As the peptide has a net negative charge it is probable that electrostatic interactions are in part responsible of the good miscibility of palmitoyl VP3(110-121) with SA. In order to prepare liposomes containing palmitoyl VP3(110–121), lipids such as SA or DPPC/SA will be a more suitable choice than anionic lipids such as PG. Mixed monolayers of stearic acid (SA) and octadecylamine (ODA) at the air/water interface were investigated in this article. The miscibility of the two compounds was evaluated by the measurement of surface pressure-area per molecule (pi-A) isothems and the direct observation of Brewster angle microscopy (BAM) on the water surface. The two compounds were spread individually on the subphase (method 1) or premixed first in the spreading solvent and then cospread (method 2). The effect of spreading method on the miscibility of the two compounds was also studied. The results show that the mixed monolayers prepared by method 1 cannot get a well-mixed state. The isotherms of mixed monolayers preserve both characteristics of SA and ODA and exhibit two collapse points. The calculated excess surface area is very small. Besides, distinguished domains corresponding to those of pure SA and ODA can be inspected from the BAM images. Such results indicate that SA and ODA cannot get a well-mixed phase via 2-dimensional mixing. On the contrary, in the mixed monolayer prepared by cospreading, the two compounds exhibit high miscibility. In the pi-A isotherms, the individual characteristics of SA and ODA disappear. The calculated excess area exhibits a highly positive deviation which indicates the existence of special interaction between the two compounds. The low compressibility of isotherm implies the highly rigid characteristic of the mixed monolayer. which was also sustained by the striplike collapse morphology observed from the BAM. The rigid characteristic of SA/ODA mixed monolayer was attributed to the formation of "catanionic surfactant" by electrostatic adsorption of headgroups of SA and ODA or to the formation of salt by acid-base reaction.  相似文献

The interfacial tensions of mixed α-dipalmitoylphosphatidylcholine (DPPC)/β-lactoglobulin layers at the chloroform/water interface have been measured by the pendent drop and drop volume techniques. In certain intervals, the adsorption kinetics of these mixed layers was strongly influenced by the concentrations of both protein and DPPC. However, at low protein concentration, Cβ-lactoglobulin=0.1 mg l−1, the adsorption rate of mixed interfacial layers was mainly controlled by the variation of the DPPC concentration. As Cβ-lactoglobulin was increased to 0.8 mg l−1, the interfacial activity was abruptly increased, and within the concentration range of CDPPC=10−4–10−5 mol l−1, the DPPC has very little effect on the whole adsorption process. In this case, the adsorption rate of mixed layers was mainly dominated by the protein adsorption. This phenomenon also happened as the protein concentration was further increased to 3.6 mg l−1. When CDPPC>3 · 10–5 mol l−1, the adsorption behaviour was very similar to that of the pure DPPC although the protein concentration was changed. The equilibrium interfacial tensions of the mixed layers are dramatically effected by the lipid as compared to the pure protein adsorption at the same concentration. It reveals the estimation of which composition of lipid and protein decreases the interfacial tension. The combination of Brewster angle microscopy (BAM) with a conventional LB trough was applied to investigate the morphology of the mixed DPPC/β-lactoglobulin layers at the air/water interface. The mixed insoluble monolayers were produced by spreading the lipid at the water surface and the protein adsorbed from the aqueous buffer subphase. The BAM images allow to visualise the protein penetration and distribution into the DPPC monolayer on compression of the complex film. It is shown that a homogeneous distribution of β-lactoglobulin in lipid layers preferentially happens in the liquid fluid state of the monolayer while the protein can be squeezed out at higher surface pressures.  相似文献

Docetaxel (DCT) is an antineoplastic drug for the treatment of a wide spectrum of cancers. DCT surface properties as well as miscibility studies with l-alpha-dipalmitoyl phosphatidylcholine (DPPC), which constitutes the main component of biological membranes, are comprehensively described in this contribution. Penetration studies have revealed that when DCT is injected under DPPC monolayers compressed to different surface pressures, it penetrates into the lipid monolayer promoting an increase in the surface pressure. DCT is a surface active molecule able to decrease the surface tension of water and to form insoluble films when spread on aqueous subphases. The maximum surface pressure reached after compression of a DCT Langmuir film was 13 mN/m. Miscibility of DPPC and DCT in Langmuir films has been studied by means of thermodynamic properties as well as by Brewster angle microscopy (BAM) analysis of the mixed films at the air-water interface, concluding that DPPC and DCT are miscible and they form non-ideally mixed monolayers at the air-water interface. Helmholtz energies of mixing revealed that no phase separation occurs. In addition, Helmholtz energies of mixing become more negative with decreasing areas per molecule, which suggests that the stability of the mixed monolayers increases as the monolayers become more condensed. Compressibility values together with BAM images indicate that DCT has a fluidizing effect on DPPC monolayers.  相似文献

The antimalarial agent halofantrine penetrates dipalmitolylphosphatidylcholine (DPPC) monolayers resulting in an increase in surface pressure and an expansion in area occupied by the lipid components of the monolayer. This phenomenon is observed at concentrations (0.05-0.2 microm) of halofantrine that have no surface activity. Penetration increases with drug concentration and is greatest at low initial surface pressures of the monolayer. A critical surface pressure of the DPPC monolayer has been determined from constant area and constant pressure conditions. The magnitude of these values support the hypothesis that halofantrine readily penetrates the DPPC monolayers. The presence of cholesterol in the DPPC monolayer hampers penetration and a lower critical surface pressure is obtained under such conditions. Even then, a slower rate of penetration is observed only in monolayers maintained at high initial surface pressures (10, 15 mN/m), corresponding to the liquid condensed phase of the monolayer, and not at low surface pressures (2.5, 5.0 mN/m). These results help to give a better understanding of the dynamics of the halofantrine-phospholipid interaction as well as the pharmacodynamic character of the drug.  相似文献

本文制备了卟啉-紫精与花生酸镉混合LB膜, 用紫外可见光谱研究了膜中卟啉基团的聚集及取向, X射线衍射说明混合LB膜具有层状有序的周期结构。扫描电镜结果表明: 通过调节膜的表面压可使聚集成"微畴"的卟啉-紫精均匀分布在花生酸镉单分子层中, 随膜表面压的增大, 小的"微畴"相互连接形成更大的"微畴"。电子衍射说明混合膜中两组份分相存在, 且都为六角对称的有序结构。  相似文献

Reported here is the study on the structure of Langmuir-Blodgett (LB) films of double-armed dibenzo-18-crown-6 contain biphenyl (DACE) which are newly synthesized and mixed with stearic acid (SA). In addition, the miscibility of the two compounds was also tested by the measurement of the surface pressure-area (pi-A) isotherms of DACE and DACE/SA with various proportions. It is noted that there is no phase segregation in the mixed monolayer film of DACE/SA. Upon calculation of the excess surface area, it is concluded that the SA molecule can enter into the crown ether ring, while an 18C6 ring can host a maximum of one SA molecule. The difference of spectroscopic properties of DACE in LB films and bulk solution has been investigated by ultraviolet-visible (UV-vis) and Fourier-transfer infrared (FTIR) measurements. Molecules of DACE exist in the mixed LB films as monomers in contrast to those in the concentrated solution as aggregates. The hydrocarbon chains in DACE and DACE/SA LB films are tilted to the normal of the substrate surface, but perpendicular to the dipole moment of CO. Both CO bonds in the phenyl ethers and carboxylic ester of DACE, and the long axes of phenyl rings are aligned nearly perpendicular to the substrate surface. Infrared spectra of mixed LB films of DACE/SA present further evidence that the SA molecules enter into the crown ether rings of DACE.  相似文献

用作“表面离子”的钌螯合物Ru(dpphen)32+与脂肪酸类成膜分子以1:2混合时能够得到稳定的混合单分子膜.以十八烷基三氯硅烷(octadecayl trichloro silane, OTS)分子部分取代Ru(dpphen)32+,得到功能分子组份可控的混合单分子膜.研究表明,OTS分子在纯水表面上可以形成交联网状单分子膜结构,混入硬脂酸(SA)分子后,网状结构逐渐被破坏.SA含量增加,破坏的程度就增大,直至SA/OTS为3:1时,完全没有网状交联结构,形成可以用来沉积LB膜的均匀致密的单分子膜.表面离子Ru(dpphen)32+与OTS和SA一起构成三组份混合单分子膜,OTS和Ru(dpphen)32+为表面离子.单分子膜中混有Ru(dpphen)32+分子,可以有效地阻止OTS的交联发生,同时Ru(dpphen)32+/SA基团与OTS/SA基团是均匀共混的.改变Ru(dpphen)32+/SA基团与OTS/SA基团的混合比,即可以做到Ru(dpphen)32+的组份精确可控,得到可用来沉积LB膜的均一、稳定的单分子膜.  相似文献

A Langmuir-Blodgett (LB) inducing method was firstly used to prepare single layer and multilayer conducting composite PEDOT-PSS film. The film-forming ability of ionization ODA and ODA-SA monolayer spread on PEDOT-PSS nanoparticle sub-phase and the behavior of ODA/PEDOT-PSS assembly particles on pure water were firstly investigated. The results indicated that nanoparticles in the suphase are packed in the ionization monolayer and stable complex Langmuir film is formed at the air/water interface. It has been found that the best film-forming conditions for composite film are as follows:distinct interface was formed between ODA and PEDOT-PSS layer and single layer thickness of PEDOT-PSS was about 23 nm,well accordant to the size of PEDOT-PSS nanoparticles. Different structures were designed to test the conductive ability of these composite films and a variable range hopping (VRH) model was used to explain the film conductive mechanism. The results indicated that a 3D-VRH model explained well the transferring of charge carrier in the multilayer film.  相似文献

The structure and properties of mixed monolayer or LB film of 5,10,15,20-tetra-4-oxy(2-stearic acid)phenyl porphyrin/C60 were studied. A isotherms and small angle X-ray diffraction (SAXD) results show that C60 molecules were incorporated into the cavities of por-phyrin molecules in mixed monolayer and LB film . UV-vis spectra of mixed LB films show that the absorption intensity of porphyrin varied compared with pure porphyrin film, probably arising from the interactions between C60 and porphyrin ring. C60 molecules in mixed systems are well-dispersed. The TPP(CO2H)/C60 mixed LB film is a kind of two-dimensional host-guest system.  相似文献

In this work, surface film balance and Brewster angle microscopy techniques have been used to analyze the structural characteristics (structure, topography, reflectivity, thickness, miscibility, and interactions) of hydrolysates from sunflower protein isolate (SPI) and dipalmitoylphosphatidylcholine (DPPC) mixed monolayers spread on the air-water interface. The degree of hydrolysis (DH) of SPI, low (5.62%), medium (23.5%), and high (46.3%), and the protein/DPPC mass fraction were analyzed as variables. The structural characteristics of the mixed monolayers deduced from the surface pressure (pi)-area (A) isotherms depend on the interfacial composition and degree of hydrolysis. At surface pressures lower than the equilibrium surface pressure of SPI hydrolysate (pi(e)(SPI hydrolysate)), both DPPC and protein are present in the mixed monolayer. At higher surface pressures (at pi > pi(e)(SPI hydrolysate)), collapsed protein residues may be displaced from the interface by DPPC molecules. The differences observed between pure SPI hydrolysates and DPPC in reflectivity (I) and monolayer thickness during monolayer compression have been used to analyze the topographical characteristics of SPI hydrolysates and DPPC mixed monolayers at the air-water interface. The topography, reflectivity, and thickness of mixed monolayers confirm at microscopic and nanoscopic levels the structural characteristics deduced from the pi-A isotherms.  相似文献
