Summary High performance anion-exchange chromatography (HPAC) coupled with pulsed amperometric detection (PAD) under alkaline conditions (pH 13) separates neutral saccharides based upon their molecular size, saccharide composition, and glycosidic linkages. Carbohydrates were detected by oxidation with a gold-working electrode. HPAC-PAD was compared to high performance liquid chromatography (HPLC) with refractive index (RI) detection in terms of selectivity and sensitivity of saccharides. The results indicate that HPAC-PAD was more precise, two orders of magnitude more sensitive (pmol range) and gives better resolution of saccharides than HPLC-RI. HPAC-PAD required less sample preparation and was not subjected to matrix interferences. The use of HPAC-PAD was applied to the analysis of organic materials (plant residues, animal wastes and sewage sludge) and soil.  相似文献

High-performance anion-exchange chromatography (HPAEC) coupled with pulsed amperometric detection (PAD) under alkaline conditions (pH 9-13) separates aminosaccharides, neutral saccharides and glycuronic acids based upon their molecular size, saccharide composition and glycosidic linkages. Carbohydrates were extracted by utilizing 0.5 M H2SO4 (neutral monosaccharides), 0.25 M H2SO4 coupled with enzyme catalysis (glycuronic acids) and 3 M H2SO4 (aminosaccharides). Solid-phase extraction with strong cation and strong anion resins was used to partition the cationic aminosaccharides and anionic glycuronic acids and to deionize acid extracts for neutral saccharides. Separation was conducted on a medium-capacity anion-exchange column (36 mequiv.) utilizing sodium hydroxide (5-200 mM and sodium acetate (0-250 mM) as the mobile phase. The saccharides were detected by oxidation at a gold working electrode with triple-pulsed amperometry. HPAEC-PAD was found superior to high-performance liquid chromatography with refractive index (RI) detection for neutral monosaccharides and aminosaccharides and to low-wavelength UV detection for glycuronic acids in terms of resolution and sensitivity. HPAEC-PAD was not subject to interferences as was the case for low UV detection (210 nm) or RI analyses and was highly selective for mono- and aminosaccharides and glycuronic acids. The use of HPAEC-PAD was applied for the determination of the saccharide composition of organic materials (plant residues, animal wastes and sewage sludge), microbial polymers and soil.  相似文献

Summary Acid hydrolysis (0.25M H2SO4) coupled with enzyme catalysis (pectolyase and β-D-glucuronidase) were employed to extract galacturonic and glucuronic acids from microbial polysaccharides, plant residues, animal wastes, sewage sludge and soil. The glycuronic acids were separated by high-performance anion chromatography (HPAC) on a strong anion-exchange column using 0.1M sodium hydroxide with 0.25M sodium acetate as the mobile phase and determined by pulsed amperometric detection (PAD). HPAC-PAD was found to be superior to high-performance liquid chromatography with ultra-violet (UV) detection in terms of resolution and sensitivity of glycuronic acids. HPAC-PAD was not subject to interferences present with low UV detection (210 nm) and was highly selective for glycuronic acids. Enzymatic hydrolysis after treatment with mild acid (0.25M H2SO4) released galacturonic acids from orange peel and pectin, while glucuronic acid was released from Acacia powder. Large amounts of glycuronic acids were also extracted from plant materials. Low levels of uronic acids were detected in poultry manure, sewage sludge and organic-amended soils.  相似文献

A new technique for the assay of carbohydrates is described in which separation and quantification of neutral saccharides, aminosaccharides, glycuronic acids, and disaccharides may be accomplished in less than 50 min of total run time. This method involves optimized anion-exchange liquid chromatography coupled with integrated pulse amperometric detection. Complex carbohydrates from various sources, including dietary supplements, were hydrolyzed in a dilute solution of trifluoroacetic acid, freeze-dried, and reconstituted in water containing 2-deoxygalactose as the internal standard. The solution was filtered and separated on CarboPac PA20 column. The eluted saccharides were detected by oxidation on a gold electrode with quadruple-pulsed integrated amperometry. The calibration plots for the saccharides were linear with an average correlation coefficient of 0.999. Method precision regarding peak retention time and resolution used in the peak identifications was verified. With this method, previously difficult-to-separate saccharides, such as galactosamine, glucosamine, and N-acetylglucosamine, were successfully resolved from the neutral saccharides rhamnose, arabinose, and galactose. Mannose was also resolved from xylose, and de-acetylation of aminosaccharides prior to separation was not necessary. This technique provides an accurate and efficient means to assay carbohydrates in dietary supplements, which new federal regulations will soon mandate.  相似文献

Avian myosin light-chain kinase from smooth muscle of the gizzard and its catalytic domain, derived from the intact enzyme by trypsin digestion, was purified within 30-40 min by both analytical and preparative anion-exchange high-performance liquid chromatography. The proteins obtained were more than 95% pure and retained their biological activity. The high-performance anion-exchange chromatography protocols represent a significant decrease in purification time when compared with conventional ion-exchange chromatography.  相似文献

建立了高效、经济、大规模获得鸡卵黄免疫球蛋白(IgY)的生产方法。在对传统的水稀释法改良的基础上,结合聚乙二醇沉淀与离子交换色谱进行IgY的分离纯化。结果显示,用8倍无菌水稀释蛋黄液,用0.1 mol/L HCl调节pH为5.2,在4 ℃下静置8 h,于5000×g力离心可得上清粗IgY液,经测定回收率可达93.47%。然后用6%聚乙二醇沉淀后经DEAE-Toyopearl 650 M离子交换纯化,最佳的纯化条件: 0.05 mol/L磷酸盐缓冲液(PBS, pH 7)平衡上样,0.075 mol/L PBS(pH 7)洗脱。十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)分析结果显示所得的IgY的纯度为95.02%,活性保持率高达73.77%。本研究弥补了传统分离方法不能同时达到高纯度和高回收率的缺点,且可用于大规模生产。  相似文献

为了从猪血中分离纯化高纯度的猪血红蛋白,建立了通过超滤、DEAE-Sepharose Fast Flow离子交换色谱和Sephadex G-75凝胶 排阻色谱三步法制备高纯度猪血红蛋白的方法,并通过十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)、高效凝胶排阻色谱和反相高 效液相色谱方法,对纯化后的猪血红蛋白进行了鉴定。经三步分离纯化后,猪血红蛋白的纯度大于99%,含量为1.328 g/L。  相似文献

Summary Benzhydrylamine resin (BHAR) is a copolymer of (styrene-1% divinylbenzene) containing phenylmethylamine groups and commonly used as a solid support for peptide synthesis in organic solvents. We have demonstrated that a larger number of NH3 + groups distributed throughout the BHAR matrix improves bead solvation under more polar conditions. As this characteristic might allow this aminated-polymer to be used as a stationary phase for liquid chromatography, a hyghly substituted BHAR (2.4 mmol.g−1 of amine groups) was synthesized under forceful conditions. Neutral glycosphingolipids and negatively charged gangliosides and sulfatide obtained from rat brain extracts were successfully purified and fractionated in this BHAR batch by single-step, reverse-phase anion-exchange chromatography. Additionally, the anion-exchange potential of BHAR was also compared to that of commercial dimethylaminoethyl (DEAE)-Sephadex A25 resin.  相似文献

以偶氮二异丁腈( AIBN)为自由基引发剂,将甲基丙烯酸缩水甘油酯(GMA)单体和亚乙基二甲基丙烯酸酯(EDMA)交联剂通过原位聚合的方法,在不锈钢管柱(150 mm ×4.6 i.d.mm)中合成为具有一定机械性能和一定孔径结构的聚合物整体色谱柱;利用三甲胺动态修饰反应将整体柱改性为具有阴离子交换功能的整体型离子色谱分离柱.实验优化了制备条件和改性修饰条件,考察了相关离子交换容量、流体动力学参数和色谱性能等.采用直接紫外检测的方法,在205 nm检测波长下,常规阴离子乙酸根、溴酸根、亚硝酸根、溴离子、硝酸根均能得到较好的分离检测.结果表明,该阴离子交换整体色谱柱制备方法简便,成本较低,可以方便地与常规色谱系统进行联用,具有一定分析实用价值和较大的开发前景.  相似文献

Two methods have been used to isolate Hb F from red cells with low levels of Hb F (less than 2% FAD), namely an alkali denaturation procedure, as described by Tsuchiya et al. [Dokkyo J. Med. Sci., 10 (1983) 13], and anion-exchange chromatography. Analyses of these Hb F enriched hemoglobin solutions by high-performance liquid chromatography allowed quantitation of the different gamma chains in the Hb F. Although the data showed considerable variation, particularly for samples with low levels of Hb F, the final results obtained with the two approaches were comparable, suggesting that the much simpler and more economical alkali denaturation procedure can be used for this purpose.  相似文献

Chromatographic techniques are used in the purification step of human recombinant erythropoietin production process to obtain a reliable product with high purity. Anion-exchange chromatography supports have proved high efficient in removing contaminants such as DNA. For that reason, the DNA removal was determined by spike studies, on three anion-exchange chromatographic supports: gel, membrane, and monolithic column, which is used in intermediate purification stage. This study showed that membrane and monolith columns have very good results in the removal of contaminants at this step. Log removal values (LRV) greater than 3.5 were obtained from DNA spike clearance studies. Monolithic column was determined as the best technological proposal, with more than 4 LRV, 7.72?mg DNA per milliliter of adsorbent and 85% protein recovery in nonspike run. The results of this study may be used as a guide in the selection of commercially available chromatography supports for intermediate purification steps in recombinant protein production.  相似文献

Summary High-performance liquid chromatography using either reversed phase or anion-exchange techniques was used for the fractionation of hordein proteins extracted from European barley. A reversed phase method is presented which utilises an Ultrapore column packed with a wide pore (30 nm) C-3 alkylbonded silica support. Using this method up to 20 components may be separated in 54 min. Elution profiles were found to be reproducible. A further method using rapid anion-exchange chromatography indicated that up to 13 components may be separated, a number which is comparable to that found with electrophoresis. The separation of proteins extracted from different barley cultivars indicated that on the basis of elution profiles high-performance liquid chromatography using either reversed phase or anion-exchange offers considerable potential as a method for barley cultivar identification.  相似文献

Ion-exclusion/anion-exchange chromatography(IEC/AEC) on a combination of a strongly basic anion-exchange resin in the OH——form with basic eluent has been developed.The separation mechanism is based on the ion-exclusion/penetration effect for cations and the anion-exchange effect for anions to anion-exchange resin phase.This system is useful for simultaneous separation and determination of ammonium ion(NH+4),nitrite ion(NO-2),and nitrate ion(NO-3) in water samples.The resolution of analyte ions can be manipulated by changing the concentration of base in eluent on a polystyrene-divinylbenzene based strongly basic anion-exchange resin column.In this study,several separation columns,which consisted of different particle sizes,different functional groups and different anion-exchange capacities,were compared.As the results,the separation column with the smaller anion-exchange capacity(TSKgel Super IC-Anion) showed well-resolved separation of cations and anions.In the optimization of the basic eluent,lithium hydroxide(LiOH) was used as the eluent and the optimal concentration was concluded to be 2 mmol/L,considering the resolution of analyte ions and the whole retention times.In the optimal conditions,the relative standard deviations of the peak areas and the retention times of NH+4,NO-2,and NO-3 ranged 1.28%-3.57% and 0.54%-1.55%,respectively.The limits of detection at signal-to-noise of 3 were 4.10 μmol/L for NH+4,1.87 μmol/L for NO-2 and 2.83 μmol/L for NO-3.  相似文献

Determination of selenomethionine in selenized yeast by HPLC–ICP MS has been revisited with the focus on recovery of this amino acid during the proteolytic digestion and chromatography steps. Recovery of the extracted selenium from an anion-exchange column was 100% but selenomethionine quantified by the method of standard additions accounted only for 67% of the selenium injected. Analysis (by size-exclusion LC–ICP MS) of the eluate collected before and after the selenomethionine peak showed the presence of oxidized selenomethionine (ca. 3%) and selenomethionine likely to be unspecifically associated with the biological matrix continuum (ca. 11%). This finding was validated by two-dimensional LC–ICP MS using a different elution order, i.e. size-exclusion anion-exchange. The approach developed enabled demonstration that more than 80% of selenium in the selenized yeast is actually present in the form of selenomethionine and suggests that many results reported elsewhere for the concentration of this vital amino acid in selenized yeast may be negatively biased. The research also provided insight into speciation of selenium in the solid residue after proteolytic extraction but the additional amount of selenomethionine recovered was negligible (<1.5%).  相似文献

This article describes a new complementary peptide separation and purification concept that makes use of a novel mixed-mode reversed-phase/weak anion-exchange (RP/WAX) type stationary phase. The RP/WAX is based on N-(10-undecenoyl)-3-aminoquinuclidine selector, which is covalently immobilized on thiol-modified silica particles (5 microm, 100 A pore diameter) by radical addition reaction. Remaining thiol groups are capped by radical addition with 1-hexene. This newly developed separation material contains two distinct binding domains in a single chromatographic interactive ligand: a lipophilic alkyl chain for hydrophobic interactions with lipophilic moieties of the solute, such as in the reversed-phase chromatography, and a cationic site for anion-exchange chromatography with oppositely charged solutes, which also enables repulsive ionic interactions with positively charged functional groups, leading to ion-exclusion phenomena. The beneficial effect that may result from the combination of the two chromatographic modes is exemplified by the application of this new separation material for the chromatographic separation of the N- and C-terminally protected tetrapeptide N-acetyl-Ile-Glu-Gly-Arg-p-nitroanilide from its side products. Mobile phase variables have been thoroughly investigated to optimize the separation and to get a deeper insight into the retention and separation mechanism, which turned out to be more complex than any of the individual chromatography modes alone. A significant anion-exchange retention contribution at optimal pH of 4.5 was found only for acetate but not for formate as counter-ion. In loadability studies using acetate, peptide masses up to 200 mg could be injected onto an analytical 250 mm x 4 mm i.d. RP/WAX column (5 microm) still without touching bands of major impurity and target peptide peaks. The corresponding loadability tests with formate allowed the injection of only 25% of this amount. The analysis of the purified peptide by capillary high-performance liquid chromatography (HPLC)-UV and HPLC-ESI-MS employing RP-18 columns revealed that the known major impurities have all been removed by a single chromatographic step employing the RP/WAX stationary phase. The better selectivity and enhanced sample loading capacity in comparison to RP-HPLC resulted in an improved productivity of the new purification protocol. For example, the yield of pure peptide per chromatographic run on RP/WAX phase was by a factor of about 15 higher compared to the standard gradient elution RP-purification protocol.  相似文献

Two methods are described by which the enzymes 2-methyleneglutarate mutase and 3-methylitaconate delta-isomerase from Clostridium barkeri have been separated by high-performance liquid chromatography on a much larger scale than reported previously. First, the mutase eluted before the delta-isomerase after incubation with the mild detergent 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulphonate (CHAPS) followed by high-performance anion-exchange chromatography on Mono Q in the presence of the same detergent. Second, an even better separation, although with a lower yield of mutase, was obtained by hydrophobic interaction chromatography on phenyl-Sepharose HiLoad, whereby the enzymes were eluted in the reverse order. Final high-performance anion-exchange chromatography of the latter preparation on Mono Q at pH 8 gave highly purified 2-methyleneglutarate mutase (greater than 95% purity) which had a pink-orange colour (lambda max 280, 375, 470 and 532 nm). The enzyme was active in the absence of coenzyme B12 (adenosylcobalamin) and contained 2.1 mol of this coenzyme per homotetramer (molecular mass, m = 300 kilodalton).  相似文献

In this paper, a sensitive, simple and direct method for simultaneous determination of glucose, ribose, isomaltose and maltose in serum sample by high-performance anion-exchange chromatography coupled with integrated pulsed amperometric detection was developed. The four target analytes were easily and completely separated on an anion-exchange column at a flow-rate of 0.25 mL/min by binary step gradient elution in about 16 min and the two eluents were deionized water and 500 mM sodium hydroxide, respectively. The separated four analytes were detected directly by using a gold electrode and quadruple-potential waveform integrated pulsed amperometry without derivatization. Under the optimized conditions, when the injection volume was 25 microL, the detection limits (signal-to-noise ratio equal to 3) for glucose, ribose, isomaltose and maltose were 0.92, 7.50, 12.9 and 10.3 ng/mL, respectively. The calibration graphs of peak area for the four analytes were linear over two to three orders of magnitude with correlation coefficients greater than 0.998. R.S.D. of peak areas of the four analytes for five determinations were no more than 5.6%. The analytical method had been applied to the determination of glucose, ribose, isomaltose and maltose in real serum samples and good results with low relative standard deviation not more than 5.3% were obtained. The accuracy of the proposed method was tested by recovery measurements on spiked samples and good recovery results (98.1-107.9%) were obtained.  相似文献

The ion-pair, ligand-exchange and anion-exchange chromatography of the fungal metabolic tenuazonic acid (TA) and its related 3-acetyl 5-substituted pyrrolidine-2,4-diones were studied. Ion-pair chromatography was performed on a C18 column with a mobile phase composed of cetrimide, phosphate buffer in water-methanol and a metal complexant (ethylenediamine) to improve the peak sharpness. Addition of the same metal complexant to the mobile phase of the anion-exchange chromatographic system also improved its efficiency. TA and its 5-substituted analogues derived from valine and leucine were separated with the ion-pair and anion-exchange chromatographic systems. With ligand-exchange chromatography, TA could only be separated from its valine analogue. These chromatographic systems were used for the detection of TA in the culture filtrates of the fungus Pyricularia oryzae and in infected rice leaves. Deproteinated culture filtrates could be rapidly analysed for their TA content by anion-exchange chromatography. However, this system was not suitable for the detection of TA in the infected rice leaf as interfering compounds were coeluted with TA. Ion-pair and ligand-exchange chromatographic systems allowed the efficient quantification of TA in infected leaves.  相似文献

In anion-exchange chromatography using a high-concentration eluent and high-capacity ion-exchange resin, the effect of the countercation contained in the eluent was investigated. Cadmium(II) and zinc(II) ions were examined as additives in an aqueous potassium chloride eluent. The addition of these cations resulted in a reversed elution order of bromide and nitrate, as compared with conventional anion-exchange chromatography. The separation factor for these two anions increased as the cadmium concentration in the eluent was increased. Zinc(II) ion was also effective, but a relatively high concentration was necessary.  相似文献

By combining anion-exchange membrane purification with high-performance size-exclusion chromatography (HPSEC) analysis, a two-step chromatographic method was developed for the determination of vitellogenin (Vtg) in fish plasma. Most plasma protein interferences can be removed during anion-exchange membrane purification process. Vtg is eluted from the size-exclusion chromatography column with a retention time of about 9 min and is characterized based on the native molecular weight, with a limit of quantification of 20 g Vtg mL–1 plasma. The spiked recovery and interassay variability were better than 80% and 4.8%. This method was successfully applied to analyze the plasma Vtg levels of loach (Misgurnus angaillicaudatus) and sea catfish (Enchelyopus elongatus). In addition to all the female fish, Vtg is detected in 75% of male loaches and 100% of male sea catfish. The result indicates that some chemicals or unknown factors with estrogenic activity have induced male fish to produce Vtg.  相似文献
