通过紫外-可见光谱和荧光光谱滴定、稳态荧光猝灭和溴化乙啶竞争键合实验研究了Ru(Ⅱ)配合物[Ru(bpy)(H2iip)2](ClO4)2{bpy=2,2′-联吡啶, H2iip=2-(吲哚-3-基)-咪唑[4,5-f][1,10]-邻菲罗啉}的酵母RNA键合性质. 结果表明, 二者键合模式为嵌入键合, 其键合常数为7.09×106 L/mol, 比小牛胸腺DNA的键合常数大, 且比同类配合物[Ru(bpy)2(H2iip)](ClO4)2的酵母RNA键合常数大.  相似文献

合成了2,3-二甲基-1,4,8,9-四氮三联苯(dmtatp)和2,3-二苯基-1,4,8,9-四氮三联苯(dptatp)两种新配体及它们与2,2′-联吡啶和钌(Ⅱ)的混配物[Ru(bpy)2dmtatp]2+(1)和[Ru(bpy)2dptatp]2+(2),用电子吸收光谱、稳态荧光、粘度测定和圆二色谱研究了配合物与小牛胸腺DNA的相互作用。结合我们以前对配合物[Ru(bpy)2tatp]2+(3)(tatp为1,4,8,9-四氮三联苯)与小牛胸腺DNA的作用研究,得出配合物与DNA的键合强度顺序为:[Ru(bpy)2tatp]2+>[Ru(bpy)2dptatp]2+>[Ru(bpy)2dmtatp]2+,这与插入配体的位阻效应的减少趋势相一致。同时,还测定了配合物与DNA的键合常数。  相似文献

采用荧光猝灭法和毛细管区带电泳法研究了盐酸异丙肾上腺素(IH)与牛血清白蛋白(BSA)的相互作用。荧光猝灭法研究表明:IH对BSA有较强的荧光猝灭作用,根据Stern-Volmer方程得到猝灭速率常数(Kq)为2.53×1013 L·mol-1·s-1,该荧光猝灭属于静态猝灭。采用位点结合模型公式计算得结合常数为1.72×104 L·mol-1,结合位点数n为1;毛细管区带电泳法研究表明:IH与BSA的结合常数为4.07×104 L·mol-1,结合位点数n为1。  相似文献

合成并表征了一个新的Ru(II)配合物[Ru(bpy)2(hedppc)](ClO4)2 {bpy=2,2'-联吡啶, hedppc=二联吡啶[3,2-a: 2',3'-c]吩嗪-11-羧酸(2-羟乙基)酯}. 通过紫外-可见吸收光谱、与溴化乙锭竞争实验、粘度测量和DNA裂解实验研究了配合物与小牛胸腺DNA的相互作用性质. 结果表明配合物以插入模式与DNA键合,键合常数Kb=(6.99±1.34)×106 mol-1•L (s=2.03±0.04)与母体配合物[Ru(bpy)2 (dppz)]2+相近,但光致发光和溶剂变色等光学性质与[Ru(bpy)2 (dppz)]2+有明显的差别.  相似文献

合成了Co2+、Ni2+、Cu2+、Cd2+、Zn2+等金属的Schiff碱型二茂铁基多核配合物,并对其进行元素分析和光谱表征,发现该类配合物能有效地猝灭Ru(bpy)32+发光。Stern-Volmer作图呈线性关系并得猝灭速率常数kq为109L·mol-1·s-1数量级,且lgka与配合物的氧化还原电位有较好的线性关系;猝灭过程按电子转移机理进行。  相似文献

比较研究了以C2O42-为共反应物时5个结构相关的Ru(II)配合物[Ru(bpy)2L1]2+, [Ru(bpy)2L2]2+, [Ru(bpy)2L3]2+, [Ru(phen)2L1]2+和[Ru(phen)2L2]2+(其中bpy=2,2′-联吡啶, phen=1,10-邻菲啰啉, L1=4-羧基苯基咪唑[4,5-f][1,10]邻菲啰啉, L2=3-羧基-4-羟基苯基咪唑[4,5-f][1,10]邻菲啰啉, L3=3,4-二羟基苯基咪唑[4,5-f][1,10]邻菲啰啉)的电致化学发光(ECL)性质. 结果表明, 酚羟基的存在能有效地淬灭Ru(II)配合物[Ru(bpy)2L2]2+, [Ru(bpy)2L3]2+和[Ru(phen)2L2]2+的ECL, 其它Ru(II)配合物的ECL量子效率与[Ru(bpy)3]2+相差不大.  相似文献

用荧光光谱法研究了生理酸度条件下,头孢噻肟对牛血清白蛋白,Cu(Ⅱ)对牛血清白蛋白以及Cu(Ⅱ)对头孢噻肟和牛血清白蛋白荧光光谱特性的影响。结果表明:Cu(Ⅱ)和头孢噻肟均可使牛血清白蛋白的荧光强度发生静态猝灭,并且在Cu(Ⅱ)存在下,头孢噻肟对牛血清白蛋白的荧光猝灭作用显著增强。根据荧光猝灭双倒数图计算头孢噻肟和牛血清白蛋白的结合常数为3.11×104L/mol,结合位点数为1.03;二元配合物Cu(Ⅱ)与牛血清白蛋白之间的结合常数为1.13×103L/mol,结合位点数为0.74。  相似文献

在pH=7.3的Tris-HCl缓冲溶液(模拟生理条件)中, 采用荧光光谱、 循环伏安曲线和紫外光谱研究了N-二(苯-二氨基甲酰基)甲基磷酸铕(Ⅲ)配合物[Eu(pic)3L]与牛血清白蛋白(BSA)的相互作用. 实验结果表明: 配合物与BSA可以形成1∶1结合型无荧光复合物Eu(pic)3L-BSA, Eu(pic)3L对 BSA 内源荧光的猝灭类型为静态猝灭. 根据双对数回归方程计算出二者在不同温度下的结合常数K及结合位点数n, 通过热力学参数得出配合物与 BSA 之间以氢键和范德华力为主. 根据Foster的偶极-偶极无辐射能量转移机理可知配合物与BSA之间可能以偶极-偶极无辐射能量转移方式进行能量传递. 分别考察了Fe3+和Cu2+对配合物与BSA结合作用的影响, 推测Fe3+和Cu2+可能在配合物与BSA间起“离子架桥”作用, 使Eu(pic)3L-BSA复合物的稳定性增强. 循环伏安法研究结果表明配合物与BSA相互作用形成无电活性的Eu(pic)3L-BSA复合物, 使得溶液中游离的配合物浓度降低.  相似文献

本文采用荧光法研究了水杨酸金属配合物与牛血清白蛋白的相互作用。观察到水杨酸金属配合物对牛血清白蛋白荧光产生猝灭现象,猝灭方式为静态猝灭。计算了结合常数和结合位点数。并且用圆二色谱法研究水杨酸金属配合物对牛血清白蛋白二级结构的影响,发现水杨酸金属配合物的存在明显改变牛血清白蛋白的构象。  相似文献

The spectroscopic and photophysical properties of [Ru(bpy)(3)](2)[[Mo(18)O(54)(SO(3))(2)], where bpy is 2,2'-bipyridyl and [Mo(18)O(54)(SO(3))(2)](4-) is either the α or β-sulfite containing polyoxomolybdate isomer, have been measured and compared with those for the well known but structurally distinct sulfate analogue, α-[Mo(18)O(54)(SO(4))(2)](4-). Electronic difference spectroscopy revealed the presence of new spectral features around 480 nm, although they are weak in comparison with the [Ru(bpy)(3)](2)[Mo(18)O(54)(SO(4))(2)] analogue. Surprisingly, Stern-Volmer plots of [Ru(bpy)(3)](2+) luminescence quenching by the polyoxometallate revealed the presence of both static and dynamic quenching for both α and β-[Mo(18)O(54)(SO(3))(2)](4-). The association constant inferred for the ion cluster [Ru(bpy)(3)](2)α-[Mo(18)O(54)(SO(4))(2)] is K = 5.9 ± 0.56 × 10(6) and that for [Ru(bpy)(3)](2)β-[Mo(18)O(54)(SO(4))(2)] is K = 1.0 ± 0.09 × 10(7). Unlike the sulfate polyoxometalates, both sulfite polyoxometalate-ruthenium adducts are non-luminescent. Despite the strong electrostatic association in the adducts resonance Raman and photoelectrochemical studies suggests that unlike the sulfato polyoxometalate analogue there is no sensitization of the polyoxometalate photochemistry by the ruthenium centre for the sulfite anions. In addition, the adducts exhibit photochemical lability in acetonitrile, attributable to decomposition of the ruthenium complex, which has not been observed for other [Ru(bpy)(3)](2+) -polyoxometalate adducts. These observations suggest that less electronic communication exists between the [Ru(bpy)(3)](2+) and the sulfite polyoxoanions relative to their sulfate polyoxoanion counterparts, despite their structural and electronic analogy. The main distinction between sulfate and sulfite polyoxometalates lies in their reversible reduction potentials, which are more positive by approximately 100 mV for the sulfite anions. This suggests that the capacity for [Ru(bpy)(3)](2+) or analogues to sensitize photoreduction in the adducts of polyoxometalates requires very sensitive redox tuning.  相似文献

Luminescence quenching of the metallopolymers [Ru(bpy)(2)(PVP)(10)](2+) and [Ru(bpy)(2)(PVP)(10)Os(bpy)(2)](4+), both in solution and as thin films, is reported, where bpy is 2,2'-bipyridyl and PVP is poly(4-vinylpyridine). When the metallopolymer is dissolved in ethanol, quenching of the ruthenium excited state, Ru(2+*), within [Ru(bpy)(2)(PVP)(10)](2+) by [Os(bpy)(3)](2+) proceeds by a dynamic quenching mechanism and the rate constant is (1.1 +/- 0.1) x 10(11) M(-1) s(-1). This quenching rate is nearly two orders of magnitude larger than that found for quenching of monomeric [Ru(bpy)(3)](2+) under the same conditions. This observation is interpreted in terms of an energy transfer quenching mechanism in which the high local concentration of ruthenium luminophores leads to a single [Os(bpy)(3)](2+) centre quenching the emission of several ruthenium luminophores. Amplifications of this kind will lead to the development of more sensitive sensors based on emission quenching. Quenching by both [Os(bpy)(3)](2+) and molecular oxygen is significantly reduced within a thin film of the metallopolymer. Significantly, in both optically driven emission and electrogenerated chemiluminescence, emission is observed from both ruthenium and osmium centres within [Ru(bpy)(2)(PVP)(10)Os(bpy)(2)](4+) films, i.e. the ruthenium emission is not quenched by the coordinated [Os(bpy)(2)](2+) units. This observation opens up new possibilities in multi-analyte sensing since each luminophore can be used to detect separate analytes, e.g. guanine and oxoguanine.  相似文献

Treatment of a thiolato-bridged Ru(II)Ag(I)Ru(II) trinuclear complex, [Ag{Ru(aet)(bpy)(2)}(2)](3+) (aet = 2-aminoethanthiolate; bpy = 2,2'-bipyridine), with NaI in aqueous ethanol under an aerobic condition afforded a mononuclear ruthenium(II) complex having an S-bonded sulfinato group, [1](+) ([Ru(aesi-N, S)(bpy)(2)](+) (aesi = 2-aminoethanesulfinate)). Similar treatment of optically active isomers of an analogous Ru(II)Ag(I)Ru(II) trinuclear complex, Δ(D)Δ(D)- and Λ(D)Λ(D)-[Ag{Ru(d-Hpen-O,S)(bpy)(2)}(2)](3+) (d-pen = d-penicillaminate), with NaI also produced mononuclear ruthenium(II) isomers with an S-bonded sulfinato group, Δ(D)- and Λ(D)-[2](+) ([Ru(d-Hpsi-O,S)(bpy)(2)](+) (d-psi = d-penicillaminesulfinate)), respectively, retaining the bidentate-O,S coordination mode of a d-Hpen ligand and the absolute configuration (Δ or Λ) about a Ru(II) center. On refluxing in water, the Δ(D) isomer of [2](+) underwent a linkage isomerization to form Δ(D)-[3] (+) ([Ru(d-Hpsi-N,S)(bpy)(2)](+)), in which a d-Hpsi ligand coordinates to a Ru(II) center in a bidentate-N,S mode. Complexes [1](+), Δ(D)- and Λ(D)-[2](+), and Δ(D)-[3](+) were fully characterized by electronic absorption, CD, NMR, and IR spectroscopies, together with single-crystal X-ray crystallography. The electrochemical properties of these complexes, which are highly dependent on the coordination mode of sulfinate ligands, are also described.  相似文献

Here we report the synthesis of luminescent ruthenium complexes that bind DNA base pair mismatches. [Ru(bpy)2(tpqp)]Cl2 (tpqp = 7,8,13,14-tetrahydro-6-phenylquino[8,7-k][1,8]phenanthroline), [Ru(bpy)2(pqp)]Cl2 (pqp = 6-phenylquino[8,7-k][1,8]phenanthroline), and [Ru(bpy)2(tactp)]Cl2 [tactp = 4,5,9,18-tetraazachryseno[9,10-b]triphenylene] have been synthesized, and their spectroscopic properties in the absence and presence of DNA have been examined. While [Ru(bpy)2(pqp)]2+ shows no detectable luminescence, [Ru(bpy)2(tpqp)]2+ is luminescent in the absence and presence of DNA with an excited-state lifetime of 10 ns and a quantum yield of 0.002. Although no increase in emission intensity is associated with binding to mismatch-containing DNA, luminescence quenching experiments and measurements of steady-state fluorescence polarization provide evidence for preferential binding to oligonucleotides containing a CC mismatch. Furthermore, by marking the site of binding through singlet oxygen sensitized damage, the complex has been shown to target a CC mismatch site directly with a specific binding affinity, Kb = 4 x 10(6) M(-1). [Ru(bpy)2(tactp)]2+, an analogue of [Ru(bpy)2(dppz)]2+ containing a bulky intercalating ligand, is luminescent in aqueous solution at micromolar concentrations and exhibits a 12-fold enhancement in luminescence in the presence of DNA. The complex, however, tends to aggregate in aqueous solution; we find a dimerization constant of 9.8 x 10(5) M(-1). Again, by singlet oxygen sensitization it is apparent that [Ru(bpy)2(tactp)]2+ binds preferentially to a CC mismatch; using a DNase I footprinting assay, a binding constant to a CC mismatch of 8 x 10(5) M(-1) is found. Hence results with these novel luminescent complexes support the concept of using a structurally demanding ligand to obtain selectivity in targeting single base mismatches in DNA. The challenge is coupling the differential binding we can obtain to differential luminescence.  相似文献

A new Ru(II) complex of [Ru(bpy)2(Hmspip)]Cl2 {in which bpy=2,2'-bipyridine, Hmspip=2-(4-(methylsulfonyl)phenyl)-1H-imidazo[4,5-f][1,10]phenanthroline} have been synthesized and characterized. The ground- and excited-state acid-base properties of [Ru(bpy)2(Hmspip)]Cl2 and its parent complex of [Ru(bpy)2(Hpip)]Cl2 {Hpip=2-phenyl-1H-imidazo[4,5-f][1,10]phenanthroline} have been studied by UV-visible (UV-vis) and emission spectrophotometric pH titrations. [Ru(bpy)2(Hmspip)]Cl2 acts as a calf thymus DNA intercalators with a binding constant of 4.0×10(5) M(-1) in buffered 50 mM NaCl, as evidenced by UV-vis and luminescence titrations, steady-state emission quenching by [Fe(CN)6]4-, DNA competitive binding with ethidium bromide, reverse salt titrations and viscosity measurements.  相似文献

The two enantiomers of [Ru(bpy)2(bbtb)]2+{bpy = 2,2'-bipyridine; bbtb = 4,4'-bis(benzothiazol-2-yl)-2,2'-bipyridine} have been isolated and fully characterised. Both enantiomers have been shown to have a strong association with calf thymus DNA by UV/visible absorption, emission and CD spectroscopy, with the Lambda enantiomer having the greater affinity. The binding of both enantiomeric forms of [Ru(bpy)2(Me2bpy)]2+ and [Ru(bpy)2(bbtb)]2+{Me(2)bpy = 4,4'-dimethyl-2,2'-bipyridine} to a range of oligonucleotides, including an octadecanucleotide and an icosanucleotide which contain hairpin-sequences, have been studied using a fluorescent intercalator displacement (FID) assay. The complex [Ru(bpy)2(bbtb)]2+ exhibited an interesting association with hairpin oligonucleotides, again with the Lambda enantiomer binding more strongly. A (1)H NMR spectroscopic study of the binding of both enantiomers of [Ru(bpy)2(bbtb)]2+ to the icosanucleotide d(CACTGGTCTCTCTACCAGTG) was conducted. This sequence contains a seven-base-pair duplex stem and a six-base hairpin-loop. The investigation gave an indication of the relative binding of the complexes between the two different regions (duplex and secondary structure) of the oligonucleotide. The results suggest that both enantiomers bind at the hairpin, with the ruthenium centre located at the stem-loop interface. NOE studies indicate that one of the two benzothiazole substituents of the bbtb ligand projects into the loop-region. A simple model of the metal complex/oligonucleotide adduct was obtained by means of molecular modelling simulations. The results from this study suggest that benzothiazole complexes derived from inert polypyridine ruthenium(II) complexes could lead to the development of new fluorescent DNA hairpin binding agents.  相似文献

Two novel chiral ruthenium(II) complexes, Δ‐[Ru(bpy)2(dmppd)]2+ and Λ‐[Ru(bpy)2(dmppd)]2+ (dmppd = 10,12‐dimethylpteridino[6,7‐f] [1,10]phenanthroline‐11,13(10H,12H)‐dione, bpy = 2,2′‐bipyridine), were synthesized and characterized by elemental analysis, 1H‐NMR and ES‐MS. The DNA‐binding behaviors of both complexes were studied by UV/VIS absorption titration, competitive binding experiments, viscosity measurements, thermal DNA denaturation, and circular‐dichroism spectra. The results indicate that both chiral complexes bind to calf‐thymus DNA in an intercalative mode, and the Δ enantiomer shows larger DNA affinity than the Λ enantiomer does. Theoretical‐calculation studies for the DNA‐binding behaviors of these complexes were carried out by the density‐functional‐theory method. The mechanism involved in the regulating and controlling of the DNA‐binding abilities of the complexes was further explored by the comparative studies of [Ru(bpy)2(dmppd)]2+ and of its parent complex [Ru(bpy)2(ppd)]2+ (ppd = pteridino[6,7‐f] [1,10]phenanthroline‐11,13 (10H,12H)‐dione).  相似文献

A new technique for investigating drug-protein binding was developed employing capillary electrophoresis (CE) coupled with tris(2,2'-bipyridyl) ruthenium(II) [Ru(bpy)(3) (2+)] electrochemiluminescence (ECL) (CE-ECL) detection after equilibrium dialysis. Three basic drugs, namely pridinol, procyclidine and its analogue trihexyphenidyl, were successfully separated by capillary zone electrophoresis with end-column Ru(bpy)(3) (2+) ECL detection. The relative drug binding to human serum albumin (HSA) for each single drug as well as for the three drugs binding simultaneously was calculated. It was found that the three antiparkinsonian drugs compete for the same binding site on HSA. This work demonstrated that Ru(bpy)(3) (2+) CE-ECL can be a suitable technique for studying drug-protein binding.  相似文献
