TLC experiments have been performed to resolve the enantiomers of three β-blockers by complexation chiral TLC with Cu(II) cation and five l amino acids. Loading was carried out by using the Cu(II)–l amino acid complex as chiral mobile phase additive with untreated silica gel plates; by mixing the Cu complex with silica gel before preparing the TLC plates; by ascending development of untreated plates with solutions of the Cu complex; and by using a solution of Cu(II) acetate as mobile phase additive for plates prepared by mixing the l amino acid with silica gel. Spots were located by use of iodine vapour.  相似文献

Abstract

Procedures are described for the thin layer chromatographic (TLC) analysis of coal-derived liquids. After solvent selection using Selectosol (1) radial TLC, a series of compounds typically found in coal liquids was used to demonstrate their elution behavior on silica gel. It was determined that unactivated (Brockman 4) silica plates series developed with isooctane/tetrahydrofuran (THF) (80:20, v/v) provided the best separation. The order of elution was saturate >hydroaromatic >polynuclear aromatic (PNA)>phenols ≥ nitrogen bases. The use of two-dimensional TLC with reverse phase and silica contiguous on the same plate (Whatman Multi-K) is discussed for the rapid separation and identification of coal liquid components. Trimethylsilyl ether derivatives were prepared of phenols and alcohols present in coal conversion products. This technique proved most useful in that the TMS-derivatives behaved similarly to their parent PNA, in order of elution, on silica plates using a moderately active mobile phase (isooctane: THF,80:20, v/v). TLC and the reactions described provide a semi-quantitative measure of the degree of hydrogenation of coal-derived liquids.  相似文献

Fast resolutions of racemic compounds (sulfoxides, amino alcohols, and α-methylarylacetic acids derivatives) were achieved on a chiral microbore column using carbon dioxide and a polar methanol/dioxane modifier. The stationary phase used in this study contains the 3,5-dinitrobenzoyl derivative of R,R(?)-1,2-diaminocyclohexane (DACH-DNB) as the chiral moiety, anchored to a silica gel surface by covalent bonds. Both thermodynamic and kinetic separation performances were improved by using a super- or subcritical carbon dioxide mobile phase (SFC, SubFC).  相似文献

Abstract

Testosterone and its 6β-, 7α-, and 16α-hydroxylated metabolites were resolved by high pressure liquid chromatography and thin layer chromatography. Separation by HPLC was achieved in less than 45 min on a microparticulate silica gel column using isocratic elution with isopro-panol:tetrahydrofuran:hexane (5:15:80) as the mobile phase. TLC systems utilizing silica gel on glass and plastic plates, and polysilicic acid on glass fiber sheets are presented. The monohydroxylated metabolites of testosterone formed during incubation of (14C)-testosterone with liver postmitochondrial preparations from adult male rats pre-treated with phenobarbital or Aroclor 1254 were separated and quantitated by both HPLC and TLC. The results using both techniques are compared with those obtained by paper chromatography.  相似文献

A sensitive, selective, and robust high-performance TLC (HPTLC) method using chiral TLC plates for qualitative and quantitative analysis of phyllanthin (A), hypophyllanthin (B), niranthin (C), and nirtetralin (D), the active lignans of Phyllanthus species, was developed and validated. The effectiveness and role of various stationary phases viz TLC silica gel 60F(254), HPTLC silica gel 60F(254), and chiral TLC plates in the quantitation were evaluated. A precoated chiral TLC plate was found suitable for the simultaneous analysis of four pharmacologically active lignans. For achieving good separation, the optimized mobile phase of n-hexane/acetone/1,4-dioxane (9:1:0.5 by volume) was used (R(f) = 0.30, 0.36, 0.41, and 0.48 for compounds A, B, C, and D, respectively). A densitometric determination of the above compounds was carried out in reflection/absorption mode at 620 nm. Optimized chromatographic conditions provide well-separated compact bands for the tested lignans. The calibration curves were found linear in the concentration range of 100-500 ng/band. Recoveries of A-D were 99.98, 100.51, 99.22, and 98.74%, respectively. The method was validated according to ICH guidelines. The method reported here is reproducible and applied for the quantitative analysis of the above lignans in the leaves of four Phyllanthus species, i. e., P. amarus, P. maderaspatensis, P. urinaria, and P. virgatus.  相似文献

A crown ether-based chiral stationary phase (CSP) without extra aminopropyl groups on the surface of silica gel was newly prepared by bonding (+)-(18-crown-6)-2,3,11,12-tetracarboxylic acid to silica gel. The new CSP was applied to the resolution of various racemic alpha-amino acids, amines, and amino alcohols. The chiral recognition efficiency of the new CSP was generally superior to that of the original CSP containing unreacted residual aminopropyl groups on the surface of silica gel in terms of the separation (alpha) and the resolution factors (Rs). The retention behaviors of analytes on the new CSP with the variation of the content of organic and acidic modifiers in aqueous mobile phase were consistent with those on the original CSP in the resolution of alpha-amino acids, but somewhat different in the resolution of racemic amines and amino alcohols from those on the original CSP and the difference was rationalized by the lipophilicity difference of the two CSPs. The effect of the column temperature on the chromatographic resolution behaviors on the new CSP was consistent with that on the original CSP.  相似文献

A simple and sensitive thin-layer chromatography (TLC) method coupled with an image analysis technique was developed for the simultaneous quantitative determination of L-proline and L-lysine in dietary supplement with good precision and accuracy. Separation was performed on silica gel plates using ethanol‒toluene (2:3, V/V) as the mobile phase. The visualization of chromatograms was based on iodine–azide reaction; therefore, pre-chromatographic derivatization reaction of amino acids with phenyl isothiocyanate was performed. Digital images of TLC plate chromatograms were converted into peak chromatograms, and quantitative analysis was conducted using TLSee software.

ABSTRACT

This review focuses on four unusual thin-layer chromatography (TLC) approaches for the determination of lipophilicity: (1) the use of medium-polar stationary phases: CN, NH2, and DIOL instead of RP plates, together with water-based mobile phase; (2) the use of silica gel in a typical normal-phase manner and treating extrapolated retention indices as the “reversed lipophilicity”; (3) the use of oil impregnated silica gel in the reversed-phase manner; and (4) the use of salting-out mobile phases. The chromatographic indices obtained in these systems are numerously reported as well correlated with lipophilicity and they are an interesting alternative to classical RP systems approaches.  相似文献

Atenolol and propranolol (the β-blocking agents) and salbutamol (broncho- and vasodilator) were resolved into their enantiomers by adopting different modes of loading/impregnating the Cu(II) complexes of l-proline (l-Pro), l-phenylalanine (l-Phe), l-histidine (l-His), N,N-dimethyl-l-phenylalanine (N,N-Me2-l-Phe), and l-tryptophan (l-Trp) on commercial precoated normal phase plates. The three different approaches were (A) using the Cu(II)-l-amino acid complex as chiral mobile phase additive, (B) ascending development of plain commercial plates in the solutions of Cu complex, and (C) using a solution of Cu(II) acetate as mobile phase additive for the commercial TLC plates impregnated with ascending development of plates in the solutions of amino acid. Spots were located using iodine vapour. The results obtained for the three methods have been compared for their efficiency and the issue of involvement of the Cu(II) cation for the best performance of the three methods has been discussed with respect to the same mobile phase. The detection limit is 0.18 μg for each enantiomer.  相似文献

Abstract

Impregnated thin-layer chromatography (TLC) layers based on silica gel are presented. Impregnating agents such as metal cations, inorganic ions, chelating agents, chiral selectors, surfactants, ion-pairing reagents, and ionic liquids are discussed. The role of impregnated TLC layers in medicinal chemistry is highlighted. The historical overview of TLC separations on physically coated layers is given and some future prospects ahead of this technique are discussed.  相似文献

A simple and rapid thin layer chromatographic (TLC)-image analysis method was developed for simultaneous quantification of three curcuminoids; curcumin (CUR), desmethoxycurcumin (DES) and bisdesmethoxycurcumin (BIS), in Curcuma longa (turmeric). Chromatographic separation of the curcuminoids was achieved on silica gel 60 F254 TLC plates, using chloroform–hexane–methanol (1:1:0.1, v/v/v) as the mobile phase. Image analysis of the scanned TLC plate was performed by Photoshop 7.0 to quantify the amount of each curcuminoid. The method was validated and found to be accurate, reliable and convenient for the analysis of CUR, DES and BIS in turmeric.

Abstract

A chiral separation of N(+)-and S(-)-benzyl-3-tetrahydrofuroate (I) and p-nitrobenzyl-3-tetrahydrofuroate (II) using a Chiralcel OB© (cellulose tribenzoate) column with a hexane/2-propanol (60:40 v/v) mobile phase is described. Enantiomeric purity of R(+)-I was evaluated using the same chromatographic conditions. I was also separated using a Chiralspher© (polyamides bonded to silica gel) column with an ethanol/distilled water (50:50 v/v) mobile phase.  相似文献

Abstract

The enantiomeric separation by high-performance liquid chromatography of underivatized non-protein amino acids was investigated by using a column packed with octadecylsilanized silica coated with N,S-dioctyl-D-penicillamine as a chiral ligand-exchange phase (Sumichiral OA-5000). Excellent to good separations of enantiomers were achieved with a variety of nonprotein amino acids carrying aliphatic or aromatic side-chains by optimizing the amount (0-20%, v/v) of the organic component (2-propanol) and the concentration (1-5 mM) of the complexing metal ion (Cu2+) in the hydro-organic eluent.  相似文献

Cellulose tris(4-methylphenylcarbamate),amylose tris(3,5-dimethylphenylcarbamate) and amylose tris (phenylcarbamate) were prepared by the method reported by Okamoto and were coated onto an aminopropylated mesoporous spherical silica gel.These final products were used as chiral stationary phases of high performance liquid chromatography for the eighteen structurally related biphenyl compounds.The resolution was made using normal-phase methodology with a mobile phase consisting of n-hexane-alcohol(ethanol,1-propanol,2-propanol or 1-butanol).The effects of various aliphatic alcohols in the mobile phase were studied.The structural features of the solutes that influence their k′ were discussed.A dominant effects of trifluoroacetic acid on chiral separation of acidic solutes was noted.  相似文献

Abstract

The TLC data for different solutes chromatographed in methanol-acetone mobile phase on silica gel at 293 K are analysed by means of the theoretical equations discussed in the previous paper (1). This analysis shows that for the above systems the solute-solvent and solvent-solvent association and the composition of the surface phase play an important role. The composition of the surface phase has been determined by utilizing the excess adsorption data of methanol from acetone on silica gel.  相似文献

TLC of a fifteen component mixture of amino acids has been carried out in two ways; firstly, the amino acids were treated with halides below their isoelectric points and chromatographed on plain silica plates, and secondly the amino acids in their cationic forms were chromatographed on silica plates impregnated with halides, keeping the same solvent system. The resolution is considered to be affected by hydrophobic interactions between silica gel and amino acid molecule and by the polarity and the flow of the mobile phase. The method provides resolution of 10–11 amino acids from the fifteen component mixture.  相似文献

Resolution of three commonly used beta-blockers, (+/-)-atenolol, (+/-)-metoprolol and (+/-)-propranolol, into their enantiomers has been achieved using normal-phase TLC on silica gel plates impregnated with L-aspartic acid as the chiral selector. Different combinations of acetonitrile-methanol-water as mobile phase were found to be successful in resolving the enantiomers. The spots were detected with iodine and the detection limits were found to be 0.26 microg for atenolol and 0.23 microg for each of metoprolol and propranolol as racemate.  相似文献
