The presence of Fe‐bound cyanide ligands in the active site of the proton‐reducing enzymes [FeFe]‐hydrogenases has led to the hypothesis that such Brønsted–Lowry bases could be protonated during the catalytic cycle, thus implying that hydrogen isocyanide (HNC) might have a relevant role in such crucial microbial metabolic paths. We present a hybrid quantum mechanical/molecular mechanical (QM/MM) study of the energetics of CN? protonation in the enzyme, and of the effects that cyanide protonation can have on [FeFe]‐hydrogenase active sites. A detailed analysis of the electronic properties of the models and of the energy profile associated with H2 evolution clearly shows that such protonation is dysfunctional for the catalytic process. However, the inclusion of the protein matrix surrounding the active site in our QM/MM models allowed us to demonstrate that the amino acid environment was finely selected through evolution, specifically to lower the Brønsted–Lowry basicity of the cyanide ligands. In fact, the conserved hydrogen‐bonding network formed by these ligands and the neighboring amino acid residues is able to impede CN? protonation, as shown by the fact that the isocyanide forms of [FeFe]‐hydrogenases do not correspond to stationary points on the enzyme QM/MM potential‐energy surface.  相似文献

A detailed characterization of a close synthetic model of the [2 Fe]H subcluster in the [FeFe] hydrogenase active site is presented. It contains the full primary coordination sphere of the CO‐inhibited oxidized state of the enzyme including the CN? ligands and the azadithiolate (adt) bridge, [((μ‐S? CH2)2NR)Fe2(CO)4(CN)2]2?, R=CH2CH2SCH3. The electronic structure of the model complex in its FeIFeII state was investigated by means of density functional theory (DFT) calculations and Fourier transform infrared (FTIR) spectroscopy. By using a combination of continuous‐wave (CW) electron paramagnetic resonance (EPR) and hyperfine sublevel correlation (HYSCORE) experiments as well as DFT calculations, it is shown that, for this complex, the spin density is delocalized over both iron atoms. Interestingly, we found that the nitrogen hyperfine coupling, which represents the interaction between the unpaired electron and the nitrogen at the dithiolate bridge, is slightly larger than that in the analogous complex in which the CN? ligands are replaced with PMe3 ligands. This reveals, first, that the CN?/PMe3 ligands coordinated to the iron core are electronically coupled to the amine in the adt bridge. Second, the CN? ligands in this complex are somewhat stronger σ‐donor ligands than the PMe3 ligand, and thereby enable more spin density to be transferred from the Fe core to the adt unit, which might in turn affect the reactivity of the bridging amine.  相似文献

Catalytic H2 oxidation has been dissected by means of DFT into the key steps common to the Fe2 unit of both the [FeFe]-hydrogenase cofactor and selected biomimics. The aim was to elucidate the molecular details underlying the very different performances of the two systems. We found that the better enzyme performance is based on a single iron atom that is maintained electron-poor, favoring H2 binding, although embedded within a highly electron-rich cofactor, ensuring a facile oxidation of the Fe2–H2 adduct. This is due to 1) CN coordinating to both iron atoms, due to their amphipathic Lewis acid/bas e properties, and 2) the 4Fe4S subunit further withdrawing electrons from the Fe2 core. Preserving a moderate electron deficiency at a single iron also helps the cofactor preserve hydride affinity, which favors H2 cleavage. Such valuable characteristics allow the biocatalyst to turnover close to equilibrium conditions. All previous biomimicry has shown, in contrast, the impossibility to properly balance the two apparently contrasting aforementioned requisites, although evident progress has been made by the H2-ase community. Disclosure of the differences identified could inspire the design of novel biomimics, for instance, reconsidering the use of CN in the catalyst architecture. Indeed, in the presence of bases normally employed in oxidative catalysis, undesired stable protonation at coordinated CN, which affects the opposite process (proton reduction), could be overcome.  相似文献

The synthesis and reactivity of a CoI pincer complex [Co(ϰ3P,CH,P‐P(CH)PNMeiPr)(CO)2]+ featuring an η2‐ Caryl−H agostic bond is described. This complex was obtained by protonation of the CoI complex [Co(PCPNMeiPr)(CO)2]. The CoIII hydride complex [Co(PCPNMeiPr)(CNtBu)2(H)]+ was obtained upon protonation of [Co(PCPNMeiPr)(CNtBu)2]. Three ways to cleave the agostic C−H bond are presented. First, owing to the acidity of the agostic proton, treatment with pyridine results in facile deprotonation (C−H bond cleavage) and reformation of [Co(PCPNMeiPr)(CO)2]. Second, C−H bond cleavage is achieved upon exposure of [Co(ϰ3P,CH,P‐P(CH)PNMeiPr)(CO)2]+ to oxygen or TEMPO to yield the paramagnetic CoII PCP complex [Co(PCPNMeiPr)(CO)2]+. Finally, replacement of one CO ligand in [Co(ϰ3P,CH,P‐P(CH)PNMeiPr)(CO)2]+ by CNtBu promotes the rapid oxidative addition of the agostic η2‐Caryl−H bond to give two isomeric hydride complexes of the type [Co(PCPNMeiPr)(CNtBu)(CO)(H)]+.  相似文献

Nitrogenase cofactors can be extracted into an organic solvent to catalyze the reduction of cyanide (CN), carbon monoxide (CO), and carbon dioxide (CO2) without using adenosine triphosphate (ATP), when samarium(II) iodide (SmI2) and 2,6‐lutidinium triflate (Lut‐H) are employed as a reductant and a proton source, respectively. Driven by SmI2, the cofactors catalytically reduce CN or CO to C1–C4 hydrocarbons, and CO2 to CO and C1–C3 hydrocarbons. The C C coupling from CO2 indicates a unique Fischer–Tropsch‐like reaction with an atypical carbonaceous substrate, whereas the catalytic turnover of CN, CO, and CO2 by isolated cofactors suggests the possibility to develop nitrogenase‐based electrocatalysts for the production of hydrocarbons from these carbon‐containing compounds.  相似文献

[FeFe] hydrogenases carry out the redox interconversion of protons and molecular hydrogen (2H+ + 2e ⇌ H2) at a complex Fe–S active site known as the H-cluster. The H-cluster consists of a [4Fe–4S] subcluster, denoted here as [4Fe]H, linked via a cysteine sulfur to an interesting organometallic [2Fe]H subcluster thought to be the subsite where the catalysis occurs. This [2Fe]H subcluster consists of two Fe atoms, linked with a bridging CO and a bridging SCH2NHCH2S azadithiolate (adt), with additional terminal CO and CN ligands bound to each Fe. Synthesizing such a complex organometallic unit is a fascinating problem in biochemistry, complicated by the toxic nature of both the CO and CN species and the relative fragility of the azadithiolate bridge. It has been known for a number of years that this complex biosynthesis is carried out by a set of three essential Fe–S proteins, HydE, HydF, and HydG. HydF is a GTPase, while HydE and HydG are both members of the large family of radical S-adenosylmethionine (rSAM) enzymes. In this perspective we describe the history of research and discovery concerning these three Fe–S “maturase” proteins and describe recent evidence for a sequential biosynthetic pathway beginning with the synthesis of a mononuclear organometallic [Fe(ii)(CO)2CN(cysteine)] complex by the rSAM enzyme HydG and its subsequent activation by the second rSAM enzyme HydE to form a highly reactive Fe(i)(CO)2(CN)S species. In our model a pair of these Fe(i)(CO)2(CN)S units condense to form the [Fe(CO)2(CN)S]2 diamond core of the [2Fe]H cluster, requiring only the installation of the central CH2NHCH2 portion of the azadithiolate bridge, whose atoms are all sourced from the amino acid serine. This final step likely occurs with an interplay of HydE and HydF, the details of which yet remain to be elucidated.

[FeFe]-hydrogenases are known for their high rates of hydrogen turnover, and are intensively studied in the context of biotechnological applications. Evolution has generated a plethora of different subclasses with widely different characteristics. The M2e subclass is phylogenetically distinct from previously characterized members of this enzyme family and its biological role is unknown. It features significant differences in domain- and active site architecture, and is most closely related to the putative sensory [FeFe]-hydrogenases. Here we report the first comprehensive biochemical and spectroscopical characterization of an M2e enzyme, derived from Thermoanaerobacter mathranii. As compared to other [FeFe]-hydrogenases characterized to-date, this enzyme displays an increased H2 affinity, higher activation enthalpies for H+/H2 interconversion, and unusual reactivity towards known hydrogenase inhibitors. These properties are related to differences in active site architecture between the M2e [FeFe]-hydrogenase and “prototypical” [FeFe]-hydrogenases. Thus, this study provides new insight into the role of this subclass in hydrogen metabolism and the influence of the active site pocket on the chemistry of the H-cluster.

Reversible proton‐ and electron‐transfer steps are crucial for various chemical transformations. The electron‐reservoir behavior of redox non‐innocent ligands and the proton‐reservoir behavior of chemically non‐innocent ligands can be cooperatively utilized for substrate bond activation. Although site‐decoupled proton‐ and electron‐transfer steps are often found in enzymatic systems, generating model metal complexes with these properties remains challenging. To tackle this issue, we present herein complexes [(cod?H)M(μ‐L2?) M (cod?H)] (M=PtII, [ 1 ] or PdII, [ 2 ], cod=1,5‐cyclooctadiene, H2L=2,5‐di‐[2,6‐(diisopropyl)anilino]‐1,4‐benzoquinone), in which cod acts as a proton reservoir, and L2? as an electron reservoir. Protonation of [ 2 ] leads to an unusual tetranuclear complex. However, [ 1 ] can be stepwise reversibly protonated with up to two protons on the cod?H ligands, and the protonated forms can be stepwise reversibly reduced with up to two electrons on the L2? ligand. The doubly protonated form of [ 1 ] is also shown to react with OMe? leading to an activation of the cod ligands. The site‐decoupled proton and electron reservoir sources work in tandem in a three‐way cooperative process that results in the transfer of two electrons and two protons to a substrate leading to its double reduction and protonation. These results will possibly provide new insights into developing catalysts for multiple proton‐ and electron‐transfer reactions by using metal complexes of non‐innocent ligands.  相似文献

Combined molecular dynamics (MD) and quantum mechanical/molecular mechanical (QM/MM) calculations were performed on the crystal structure of the reduced membrane‐bound [NiFe] hydrogenase (MBH) from Ralstonia eutropha to determine the absolute configuration of the CO and the two CN? ligands bound to the active‐site iron of the enzyme. For three models that include the CO ligand at different positions, often indistinguishable on the basis of the crystallographic data, we optimized the structures and calculated the ligand stretching frequencies. Comparison with the experimental IR data reveals that the CO ligand is in trans position to the substrate‐binding site of the bimetallic [NiFe] cluster.  相似文献

Syntheses of the copper and gold complexes [Cu{Fe(CO)5}2][SbF6] and [Au{Fe(CO)5}2][HOB{3,5-(CF3)2C6H3}3] containing the homoleptic carbonyl cations [M{Fe(CO)5}2]+ (M=Cu, Au) are reported. Structural data of the rare, trimetallic Cu2Fe, Ag2Fe and Au2Fe complexes [Cu{Fe(CO)5}2][SbF6], [Ag{Fe(CO)5}2][SbF6] and [Au{Fe(CO)5}2][HOB{3,5-(CF3)2C6H3}3] are also given. The silver and gold cations [M{Fe(CO)5}2]+ (M=Ag, Au) possess a nearly linear Fe-M-Fe’ moiety but the Fe-Cu-Fe’ in [Cu{Fe(CO)5}2][SbF6] exhibits a significant bending angle of 147° due to the strong interaction with the [SbF6] anion. The Fe(CO)5 ligands adopt a distorted square-pyramidal geometry in the cations [M{Fe(CO)5}2]+, with the basal CO groups inclined towards M. The geometry optimization with DFT methods of the cations [M{Fe(CO)5}2]+ (M=Cu, Ag, Au) gives equilibrium structures with linear Fe-M-Fe’ fragments and D2 symmetry for the copper and silver cations and D4d symmetry for the gold cation. There is nearly free rotation of the Fe(CO)5 ligands around the Fe-M-Fe’ axis. The calculated bond dissociation energies for the loss of both Fe(CO)5 ligands from the cations [M{Fe(CO)5}2]+ show the order M=Au (De=137.2 kcal mol−1)>Cu (De=109.0 kcal mol−1)>Ag (De=92.4 kcal mol−1). The QTAIM analysis shows bond paths and bond critical points for the M−Fe linkage but not between M and the CO ligands. The EDA-NOCV calculations suggest that the [Fe(CO)5]→M+←[Fe(CO)5] donation is significantly stronger than the [Fe(CO)5]←M+→[Fe(CO)5] backdonation. Inspection of the pairwise orbital interactions identifies four contributions for the charge donation of the Fe(CO)5 ligands into the vacant (n)s and (n)p AOs of M+ and five components for the backdonation from the occupied (n-1)d AOs of M+ into vacant ligand orbitals.  相似文献

The kinetics and the mechanism of the cyanide‐induced demetallation of a series of Ni2+ complexes with macrocyclic ligands of different ring size (12‐ to 14‐membered; see 1 – 4 ) and steric constraints was studied. Although the rates differ by almost five orders of magnitude when compared to each other under fixed experimental conditions (pH 10.5, [CN?]=10?2 M ), all reactions proceed through the relatively rapid formation of cyano adducts [Ni(CN)nL] (n=1, 2), which then react with additional CN? or HCN to give the final products. Of paramount importance for the reaction rate is the geometry and configuration of the cyano adducts [Ni(CN)nL] (n=1,2). cis‐Dicyano derivatives with a folded macrocycle react faster than trans‐compounds. In the case of (1,4,8,11‐tetraazacyclotetradecane)nickel(2+) ([Ni ( 4 )]2+), which gives a trans‐ dicyano adduct, the base‐catalyzed N‐inversion necessary to obtain the cis‐dicyano derivative becomes rate determining at high CN? concentrations.  相似文献

The novel bulky diphosphite (P∩P) ligands ( 3 and 4 ) based on the 2,7,9,9‐tetramethyl‐9H‐xanthene‐4,5‐diol ( 2 ) backbone were investigated in the Rh‐catalyzed hydroformylation of oct‐1‐ene, styrene, and (E)‐oct‐2‐ene. These diphosphites gave rise to very active and selective catalysts for the hydroformylation of oct‐1‐ene to nonanal with average rates>10000 (mol aldehyde)(mol Rh)−1h−1 (P(CO/H2)=20 bar, T=80°, [Rh]=1 mM ) and maximum selectivities of 79% for the linear product. Relatively high selectivities towards the linear aldehyde (up to 70%, linear/branched up to 2.3) but very high activities (up to 39000 (mol aldehyde)(mol Rh)−1h−1) were observed for the hydroformylation of styrene in the presence of these bidentate ligands (P(CO/H2)=2 – 10 bar, T=120°, [Rh]=0.2 mM ). Remarkable activities (up to 980 (mol aldehyde)(mol Rh)−1h−1) were achieved with these diphosphites for the hydroformylation of (E)‐oct‐2‐ene with selectivities for the linear product of 74% (l/b up to 2.8, P(CO/H2)=2 bar, T=120°, [Rh]=1 mM ). A detailed study of the solution structure of the catalyst under catalytic conditions was performed by NMR and high‐pressure FT‐IR. The spectroscopic data revealed that under hydroformylation conditions, the bidentate ligands rapidly formed stable, well‐defined catalysts with the structure [RhH(CO)2(P∩P)]. All the ligands showed a preference for an equatorial‐apical ( ea ) coordination mode in the trigonal bipyramidal Rh‐complexes, indicating that a bis‐equatorial ( ee ) coordination is not a prerequisite for highly selective catalysts.  相似文献

We investigate anionic [Co,CO2,nH2O] clusters as model systems for the electrochemical activation of CO2 by infrared multiple photon dissociation (IRMPD) spectroscopy in the range of 1250–2234 cm−1 using an FT-ICR mass spectrometer. We show that both CO2 and H2O are activated in a significant fraction of the [Co,CO2,H2O] clusters since it dissociates by CO loss, and the IR spectrum exhibits the characteristic C−O stretching frequency. About 25 % of the ion population can be dissociated by pumping the C−O stretching mode. With the help of quantum chemical calculations, we assign the structure of this ion as Co(CO)(OH)2. However, calculations find Co(HCOO)(OH) as the global minimum, which is stable against IRMPD under the conditions of our experiment. Weak features around 1590–1730 cm−1 are most likely due to higher lying isomers of the composition Co(HOCO)(OH). Upon additional hydration, all species [Co,CO2,nH2O], n≥2, undergo IRMPD through loss of H2O molecules as a relatively weakly bound messenger. The main spectral features are the C−O stretching mode of the CO ligand around 1900 cm−1, the water bending mode mixed with the antisymmetric C−O stretching mode of the HCOO ligand around 1580–1730 cm−1, and the symmetric C−O stretching mode of the HCOO ligand around 1300 cm−1. A weak feature above 2000 cm−1 is assigned to water combination bands. The spectral assignment clearly indicates the presence of at least two distinct isomers for n ≥2.  相似文献

It is common and chemically intuitive to assign cations electrophilic and anions nucleophilic reactivity, respectively. Herein, we demonstrate a striking violation of this concept: The anion [B12Cl11] spontaneously binds to the noble gases (Ngs) xenon and krypton at room temperature in a reaction that is typical of “superelectrophilic” dications. [B12Cl11Ng] adducts, with Ng binding energies of 80 to 100 kJ mol−1, contain B−Ng bonds with a substantial degree of covalent interaction. The electrophilic nature of the [B12Cl11] anion is confirmed spectroscopically by the observation of a blue shift of the CO stretching mode in the IR spectrum of [B12Cl11CO] and theoretically by investigation of its electronic structure. The orientation of the electric field at the reactive site of [B12Cl11] results in an energy barrier for the approach of polar molecules and facilitates the formation of Ng adducts that are not detected with reactive cations such as [C6H5]+. This introduces the new chemical concept of “dipole-discriminating electrophilic anions.”  相似文献

Developments in the chemistry of weakly coordinating anions enabled isolation of numerous unique metal complexes with unusual ligands. An important example is the family of metal-Fe(CO)5 complexes. In the current paper we present synthesis and thorough characterization of the first truly homoleptic {Cu[Fe(CO)5]2}+ cation obtained as a salt of weakly coordinating [Al(ORF)4] (RF=C(CF3)3) anion. TGA/DSC/MS study show that its decomposition becomes noticeable only above 110 °C, thus it can be stored as powder in air-free conditions for months. The crystal structure of {Cu[Fe(CO)5]2}+ shows strong asymmetry of the cation and very short Cu-CO bonds in comparison to analogous {M[Fe(CO)5]2}+ where M=Ag or Au. Characterization is complemented with analysis of vibrational spectra and extensive DFT calculations which give insight into the energetics of Cu+-Fe(CO)5 systems. Our results show that {Cu[Fe(CO)5]2}+ is homoleptic only as salt of [Al(ORF)4]. Furthermore, in the presence of additional, even weakly basic ligands, the Cu+-Fe(CO)5 bond strength decreases what may contribute to the complex's instability in liquid SO2 or in the presence of [SbF6]. These conclusions point at the key role of selection of proper anion and solvent in stabilization of these types of complexes.  相似文献

The 17e monoradical [Mn(CO)5] is widely recognized as an unstable organometallic transient and is known to dimerize rapidly with the formation of a Mn Mn single bond. As a result of this instability, isolable analogues of [Mn(CO)5] have remained elusive. Herein, we show that two sterically encumbering isocyanide ligands can destabilize the Mn Mn bond leading to the formation of the isolable, manganese(0) monoradical [Mn(CO)3(CNArDipp2)2] (ArDipp2=2,6‐(2,6‐(iPr)2C6H3)2C6H3). The persistence of [Mn(CO)3(CNArDipp2)2] has allowed for new insights into nitrosoarene spin‐trapping studies of [Mn(CO)5].  相似文献

The coordination of 10-electron diatomic ligands (BF, CO N2) to iron complexes Fe(CO)2(CNArTripp2)2 [ArTripp2=2,6-(2,4,6-(iso-propyl)3C6H2)2C6H3] have been realized in experiments very recently (Science, 2019 , 363, 1203–1205). Herein, the stability, electronic structures, and bonding properties of (E1E2)Fe-(CO)2(CNArTripp2)2 (E1E2=BF, CO, N2, CN, NO+) were studied using density functional (DFT) calculations. The ground state of all those molecules is singlet and the calculated geometries are in excellent agreement with the experimental values. The natural bond orbital analysis revealed that Fe is negatively charged while E1 possesses positive charges. By employing the energy decomposition analysis, the bonding nature of the E2E1–Fe(CO)2(CNArTripp2)2 bond was disclosed to be the classic dative bond E2E1→Fe(CO)2(CNArTripp2)2 rather than the electron-sharing double bond. More interestingly, the bonding strength between BF and Fe(CO)2(CNArTripp2)2 is much stronger than that between CO (or N2) and Fe(CO)2(CNArTripp2)2, which is ascribed to the better σ-donation and π back-donations. However, the orbital interactions in CN→Fe(CO)2(CNArTripp2)2 and NO+→Fe(CO)2(CNArTripp2)2 mainly come from σ-donation and π back-donation, respectively. The different contributions from σ donation and π donation for different ligands can be well explained by using the energy levels of E1E2 and Fe(CO)2(CNArTripp2)2 fragments.  相似文献

Green Fluorescent Protein (GFP) is known to undergo excited-state proton transfer (ESPT). Formation of a short H-bond favors ultrafast ESPT in GFP-like proteins, such as the GFP S65T/H148D mutant, but the detailed mechanism and its quantum nature remain to be resolved. Here we study in vacuo, light-induced proton transfer from the GFP chromophore in hydrogen-bonded complexes with two anionic proton acceptors, I and deprotonated trichloroacetic acid (TCA). We address the role of the strong H-bond and the quantum mechanical proton-density distribution in the excited state, which determines the proton-transfer probability. Our study shows that chemical modifications to the molecular network drastically change the proton-transfer probability and it can become strongly wavelength dependent. The proton-transfer branching ratio is found to be 60 % for the TCA complex and 10 % for the iodide complex, being highly dependent on the photon energy in the latter case. Using high-level ab initio calculations, we show that light-induced proton transfer takes place in S1, revealing intrinsic photoacid properties of the isolated GFP chromophore in strongly bound H-bonded complexes. ESPT is found to be very sensitive to the topography of the highly anharmonic potential in S1, depending on the quantum-density distribution upon vibrational excitation. We also show that the S1 potential-energy surface, and hence excited-state proton transfer, can be controlled by altering the chromophore microenvironment.  相似文献
