根据多点氢键识别原理,设计合成了新的分子钳受体1~6。研究了其对巴比妥 、尿素、二苯甲酮、戊二酰亚胺等中性分子的识别性能。用差紫外光谱法测定了结 合常数和自由能变化(ΔG)。结果表明,所有分子钳受体与所考察的客体分子均 形成1:1型超分子配合物,识别作用的推动力主要为多重氢键的协同作用。讨论了 主客体间尺寸/形状、几何互补等因素对形成超分子配合物的影响。并利用~1H NMR、计算机模拟作辅助手段对实验结果和现象进行了解释。  相似文献

Anchoring of functionalized guest molecules to self-assembled monolayers (SAMs) is key to the development of molecular printboards for nanopatterning. One very promising system involves guest binding to immobilized beta-cyclodextrin (beta-CD) hosts, with guest:host recognition facilitated by a hydrophobic interaction between uncharged anchor groups on the guest molecule and beta-CD hosts self-assembled at gold surfaces. We use molecular dynamics free energy (MDFE) simulations to describe the specificity of guest:beta-CD association. We find good agreement with experimental thermodynamic measurements for binding enthalpy differences between three commonly used phenyl guests: benzene, toluene, and t-butylbenzene. van der Waals interaction with the inside of the host cavity accounts for almost all of the net stabilization of the larger phenyl guests in beta-CD. Partial and full methylation of the secondary rim of beta-CD decreases host rigidity and significantly impairs binding of both phenyl and larger adamantane guest molecules. The beta-CD cavity is also very intolerant of guest charging, penalizing the oxidized state of ferrocene by at least 7 kcal/mol. beta-CD hence expresses moderate specificity toward uncharged organic guest molecules by van der Waals recognition, with a much higher specificity calculated for electrostatic recognition of organometallic guests.  相似文献

Molecular receptors, consisting of either two parallel cofacially disposed terpyridyl-Pd-Cl+ or terpyridyl-Pt-Cl+ units, are described. Concerted rotation of these units about the molecular spacer can alter their separation between 6.4 and 7.2 A to accommodate the dimensions of molecular guests. Neutral and anionic planar complexes of platinum(II) were investigated as guests to determine if metal-metal interaction between the host and guest metals could stabilize host-guest association. With a neutral guest, it was found that host-guest formation is signaled by a color change from light yellow to deep red. For one of the anionic guests, a visible absorption band appears upon host-guest formation with the platinum receptor that is ascribed to transitions associated with a Pt-Pt interaction. The association constants found for the neutral guest with the palladium and platinum receptors are large, suggesting that metal-metal interaction contributes to the molecular recognition. The structures of the host-(neutral)guest complexes in solution have been determined by 1H NOESY spectra. A crystal structure of the platinum host-(neutral)guest complex is the same as that found in solution and confirms the presence of a Pt-Pt interaction. Temperature-dependent (195)Pt NMR spectra in solution provide a quantitative estimate of the conformational interconversions of the free platinum receptor.  相似文献

 A computational study of the mechanism of host–guest complexation between quaternary ammonium compounds and squaramido-based tripodal receptors has been carried out. Semiempirical molecular orbital calculations, which are in qualitative agreement with experimental results have been performed using the PM3 Hamiltonian. Molecular interaction potential (MIP) maps were used to analyze the suitability of both host and guest binding units for a high-affinity recognition process. MIP calculations were computed from PM3 wavefunctions of the corresponding ammonium cations and dimethyl squaramide as a model compound for the hydrogen-bond-acceptor unit of the receptors. MIP analyses are helpful for understanding the host–guest process from the point of view of the double-complementarity principle. Two new receptors incorporating a 4-n-butyl aniline moiety has been designed, synthesized and evaluated for their binding properties towards a series of ureido-glycine derivatives. The host design is based on an urea adamantyl host motif known from large generations of poly(propylene imine) dendrimers functionalized with urea adamantyl moieties on the periphery. The design of the host molecules was directed towards a study of the effects of basicity of an amine function versus the effect of molecular recognition on the binding strength as seen from comparing the results obtained in the present work with previously guest–host studies. The guest–host interaction features an electrostatic interaction and multiple hydrogen binding interactions, where the main difference between the hosts described here and previously described is a substitution from an amine to aniline. Anilines are weaker bases than aliphatic amines and they generally give lower binding constants when treated with acidic guest molecules. The association constants have been measured using NMR titrations and the nature of the guest–host system is discussed based on these results. A general decrease in binding affinities is observed upon changing from the trialkyl amine hosts to the dialkyl aniline based hosts. One exception was observed where the weaker base host had stronger affinity to one of the guests. Thus, when the basicity of the host is decreased other factors influence the binding such as a better geometric fit. A crystal structure of one of the receptors has been solved and it shows no intramolecular hydrogen bonding.  相似文献

Two armed chiral calix[4]arenes 8-16 functionalized at the lower rim with chiral aminonaphthol units have been prepared and the structures of these receptors characterized by FTIR, (1)H, and (13)C, DEPT and COSY NMR spectroscopy and elemental analysis. The enantioselective recognition of these receptors with various carboxylic acids has been studied by (1)H NMR and UV/Vis spectroscopy. The receptors exhibited different chiral recognition abilities towards the enantiomers of racemic materials and formed 2 : 1 or 1 : 1 complexes between host and guest. It was also demonstrated that chiral calix[4]arenes 9 and 16 could be used as chiral NMR solvating agents to determine the enantiomeric purity of mandelic acid.  相似文献

Three beta-cyclodextrin (beta-CyD) derivatives with crown ether units, that is N-(4'-benzo-15-crown-5)-6-imino-6-deoxy-beta-CyD (2), 6,6'-[N-(4,4'-dibenzo-18-crown-6)-imino]-bridged bis(beta-CyD)(3), and 2,2'-[O-(4',5'-benzo-15-crown-5)-ethyl]-bridged bis (beta-CyD)(5), were synthesized as cooperative recognition receptor models. Their molecular binding behavior with four representative fluorescent dyes, i.e., ammonium 8-anilino-1-naphthalenesulfonate (ANS), sodium-6-toluidino-2-naphthalene-sulfonate (TNS), Acridine Red (AR) and Rhodamine B (RhB), was investigated in buffer solutions (pH = 7.20) at 25 degreesC by means of circular dichroism, NMR and fluorescence spectroscopy. 2D-ROESY experiments showed that dyad host 2 and triad host 3 adopted a CyD-guest-crown ether binding mode, while triad host 5 adopted a CyD-guest-CyD binding mode, upon inclusion complexation with guest molecules. Therefore, hosts 2 and 3 showed high molecular recognition ability towards charged guests, giving an enhanced binding ability up to 115 times for ANS by 3 and fairly high molecular selectivity up to 1450 times for the ANS/AR pair by 2 as compared with native beta-CyD in an aqueous phosphate buffer solution. On the other hand, host 5 was found to be able to effectively recognize the shape of a guest molecule, showing significantly higher binding ability towards linear guests. The binding affinities and molecular recognition abilities of these CyD-crown ether conjugates towards guest molecules are discussed from the viewpoint of electrostatic and/or hydrophobic interactions, size/shape-fit concept, and multiple recognition mechanism between host and guest.  相似文献

A series of modified beta-cyclodextrins with nucleobase substituents, that is, mono(6-ade-6-deoxy)-beta-cyclodextrin (2) and mono(6-ura-6-deoxy)-beta-cyclodextrin (3) as well as mono(6-thy-6-deoxy)-beta-cyclodextrin (4), were selected as molecular receptors to investigate their conformation and inclusion complexation behaviors with some chiral molecules, that is, (+)-camphor, (-)-camphor, (+)-borneol, and (-)-borneol, by spectrophotometric and microcalorimetric titrations in aqueous phosphate buffer solution (pH 7.2) at 298.15 K. Circular dichroism and NMR studies demonstrated that these nucleobase-modified beta-cyclodextrins adopted a co-inclusion mode upon complexation with guest molecules; that is, the originally self-included nucleobase substituents of the host did not move out from the beta-cyclodextrin cavity, but coexisted with guest molecule in the beta-cyclodextrin cavity upon inclusion complexation. Significantly, these nucleobase-modified beta-cyclodextrins efficiently enhanced the molecular binding ability and the chiral recognition ability of native beta-cyclodextrin, displaying enantioselectivity up to 3.7 for (+)-camphor/(-)-camphor pair by 2 and 3.5 for (-)-borneol/(+)-borneol pair by 3. The enhanced molecular/chiral recognition abilities of 2-4 toward (+/-)-camphor were mainly attributed to the increased entropic gains due to the extensive desolvation effects, while the favorable enthalpic gains originating from the good size-fit relationship as well as the hydrogen bond interactions between host and guest result in the enhanced molecular/chiral recognition abilities of 2-4 toward (+/-)-borneol.  相似文献

Herein we describe an extensive study of the response of a set of closely related dynamic combinatorial libraries (DCLs) of macrocyclic receptors to the introduction of a focused range of guest molecules. We have determined the amplification of two sets of diastereomeric receptors induced by a series of neutral and cationic guests, including biologically relevant compounds such as acetylcholine and morphine. The host–guest binding affinities were investigated using isothermal titration calorimetry. The resulting dataset enabled a detailed analysis of the relationship between the amplification of selected receptors and host–guest Gibbs binding energies, giving insight into the factors affecting the design, simulation and interpretation of DCL experiments. In particular, two questions were addressed: Is amplification by a given guest selective for the best receptor? And does the best guest induce the largest amplification of a given receptor? Our experimental results and computer simulations showed that the relative levels of amplification of hosts by a guest are well‐correlated with their relative affinities, and simulations have confirmed previous observations that amplification can be selective for the best receptor when only modest amounts of guest are used. In contrast, the correlation between guest binding and the extent of amplification of a given receptor across a wide range of guests tends to be poorer, because every guest has its own unique set of affinities for competing receptors in the DCL. This implies that the results of screening a DCL for selective receptors by comparing the response of the mixture to two different guests should be interpreted with caution. DCLs are complex mixtures in which all compounds are connected through a set of equilibria. Obtaining quantitative information about all host–guest binding constants from such systems will require the explicit and simultaneous consideration of all of the main equilibria within a DCL.  相似文献

It is still a challenge to achieve both excellent mechanical strength and biocompatibility in hydrogels. In this study, we exploited two interactions to form a novel biocompatible, slicing‐resistant, and self‐healing hydrogel. The first was molecular host–guest recognition between a host (isocyanatoethyl acrylate modified β‐cyclodextrin) and a guest (2‐(2‐(2‐(2‐(adamantyl‐1‐oxy)ethoxy)ethoxy)ethoxy)ethanol acrylate) to form “three‐arm” host–guest supramolecules (HGSMs), and the second was covalent bonding between HGSMs (achieved by UV‐initiated polymerization) to form strong cross‐links in the hydrogel. The host–guest interaction enabled the hydrogel to rapidly self‐heal. When it was cut, fresh surfaces were formed with dangling host and guest molecules (due to the breaking of host–guest recognition), which rapidly recognized each other again to heal the hydrogel by recombination of the cut surfaces. The smart hydrogels hold promise for use as biomaterials for soft‐tissue repair.  相似文献

A novel type of host–guest recognition systems have been developed on the basis of a Au(III) molecular tweezer receptor and chiral Pt(II) guests. The complementary host–guest motifs display high non-covalent binding affinity (Ka: ~104 L/mol) due to the participation of two-fold intermolecular π–π stacking interactions. Both phosphorescence and chirality signals of the Pt(II) guests strengthen in the resulting host–guest complexes, because of the cooperative rigidifying and shielding effects rendered by the tweezer receptor. Their intensities can be reversibly switched toward pH changes, by taking advantage of the electronic repulsion effect between the protonated form of tweezer receptor and the positive-charged guests in acidic environments. Overall, the current study demonstrates the feasibility to enhance and modulate phosphorescence and chirality signals simultaneously via molecular tweezer-based host–guest recognition.  相似文献

Cyclodextrins (CDs) are widely utilized in studies of chiral and molecular recognition. By changing the functionality of the guest molecule, the effect of such changes on recognition by the host CD molecule can be examined. We report crystal structure determinations for two nearly isomorphous complexes of phenylalanine derivatives: beta-CD/N-acetyl-L-phenylalanine methyl ester and beta-CD/N-acetyl-L-phenylalanine amide. The complexes crystallize as hydrated head-to-head host dimers with two included guest molecules in space group P1. The crystal packing is such that it presents a nonconstraining hydrophobic pocket adjacent to a hydrophilic region, where potential hydrogen-bonding interactions with hydroxyl groups of neighboring cyclodextrin molecules and waters of hydration can occur. The two host molecules display very similar conformations; only a few of the primary hydroxyl groups are conformationally disordered. There are a number of changes in the location of water of hydration molecules, some of which are the result of different hydrogen-bonding interactions. For the different guest molecules, similar modes of penetration are observed in the CD torus; however, there is a 0.985-A shift in the position of the guest molecules in the host torus, which takes place without changing the hydrophobic interactions displayed by the phenyl side chains. This observation and the thermal motion of the guest molecules in the ester complex are taken as evidence that complex binding forces are weak. The pseudopeptides experience a significant degree of flexibility in the crystalline environment provided by CD dimers. Conformational differences of the pseudopeptide backbones and the presence of disordered water molecules in the host-guest interface provide examples of different hydrogen-bonding schemes of similar potential energy. The crystal system presents an opportunity to establish a database of molecular interactions for small peptides and peptide analogues with waters of hydration and functional groups in nonconstraining binding environments.  相似文献

Turning molecular recognition into an effective mechanical response is critical for many applications ranging from molecular motors and responsive materials to sensors. Herein, we demonstrate how the energy of the molecular recognition between a supramolecular host and small alkylammonium salts can be harnessed to perform a nanomechanical task in a univocal way. Nanomechanical Si microcantilevers (MCs) functionalized by a film of tetra-phosphonate cavitands were employed to screen as guests the compounds of the butylammonium chloride series 1-4, which comprises a range of low molecular weight (LMW) molecules (molecular mass < 150 Da) that differ from each other by one or a few N-methyl groups (molecular mass 15 Da). The cavitand surface recognition of each individual guest drove a specific MC bending (from a few to several tens of nanometer), disclosing a direct, label-free, and real-time mean to sort them. The complexation preferences of tetraphosphonate cavitands toward ammonium chloride guests 1-4 were independently assessed by isothermal titration calorimetry. Both direct and displacement binding experiments concurred to define the following binding order in the alkylammonium series: 2 > 3 ≈ 1 ? 4. This trend is consistent with the number of interactions established by each guest with the host. The complementary ITC experiments showed that the host-guest complexation affinity in solution is transferred to the MC bending. These findings were benchmarked by implementing cavitand-functionalized MCs to discriminate sarcosine from glycine in water.  相似文献

Supramolecular surfactants self-aggregate via noncovalent interactions. The process is rapid and reversible which makes them potentially useful for the design of stimuli-responsive systems. Here we focus on host:guest interactions using calixarenes as molecular receptors. Calixarenes are an important family of molecular receptors because of their ease of preparation and post-modification. The calixarenes discussed here cannot self-aggregate, but when combined with an appropriate guest molecule readily self-associate. Different types of aggregates are obtained, e.g., micelles or vesicles, depending upon the structures of the calixarene and guest.  相似文献

设计合成了3种新型间苯二甲酰腙类化合物,利用UV-Vis及1H NMR考察了其与F-、Cl-、Br-、I-、CH3COO-、HSO4-、H2PO4-、ClO4-阴离子的相互作用。结果表明,主体分子4a(双对硝基苯并呋喃甲醛间苯二甲酰腙)在DMSO溶液中对F-和CH3COO-有显著识别效果,溶液颜色由黄色变为深黄色和棕红色。通过1H NMR滴定及质子溶剂效应进一步证明,主体分子与阴离子之间是以氢键作用方式相结合。Job曲线表明,主客体间形成1:1型氢键络合物。基于实验结果,探讨了主客体间形状和大小匹配对识别能力的影响以及主客体之间的识别模式。  相似文献

The objective of this work is to study both the dynamics and mechanisms of guest incorporation into the pores of 2D supramolecular host networks at the liquid-solid interface. This was accomplished by adding molecular guests to prefabricated self-assembled porous monolayers and the simultaneous acquisition of scanning tunneling microscopy (STM) topographs. The incorporation of the same guest molecule (coronene) into two different host networks was compared, where the pores of the networks either featured a perfect geometric match with the guest (for trimesic acid host networks) or were substantially larger than the guest species (for benzenetribenzoic acid host networks). Even the moderate temporal resolution of standard STM experiments in combination with a novel injection system was sufficient to reveal clear differences in the incorporation dynamics in the two different host networks. Further experiments were aimed at identifying a possible solvent influence. The interpretation of the results is aided by molecular mechanics (MM) and molecular dynamics (MD) simulations.  相似文献
