In this study, silver nanoparticle (AgNP) synthesis was carried out using Onosma sericeum Willd. aqueous extract for the first time, with a simple, economical, and green method without the need for any other organic solvent or external reducing or stabilizing agent. A variety of AgNPs, all of different particle sizes, were synthesized by controlling the silver ion concentration, extract volume, temperature, and pH. It was determined that the optimum conditions for AgNP synthesis were 1 mM AgNO3, pH 8, 25 °C, 20 g/200 mL extract, silver nitrate, and extract ratio 5:1 (v/v). The AgNPs were defined using UV-Vis spectroscopy, field emission scanning electron microscopy (FESEM), energy dispersive X-ray analysis (EDAX), transmission electron microscopy (TEM), and X-ray diffraction (XRD). The particle size distribution and zeta potential measurements of the AgNPs were measured using the dynamic light scattering (DLS) technique. It was determined that the AgNPs with a particle size of less than 10 nm showed a higher catalytic effect in the reduction of 2-nitrobenzenamine. It was also found that these nanoparticles had a cytotoxic effect on the MCF-7 breast cancer cell line depending on dosage and time. The resulting IC50 values were between 76.63 µg/mL and 169.77 µg/mL. Furthermore, the biosynthesized AgNPs showed effective antibacterial activity against the Acinetobacter baumannii bacteria. The results of the study showed that synthesized AgNPs can have a promising role in biomedical and nanobiotechnology applications.  相似文献

Cannabis sativa L. (hemp) is a plant used in the textile industry and green building material industry, as well as for the phytoremediation of soil, medical treatments, and supplementary food products. The synergistic effect of terpenes, flavonoids, and cannabinoids in hemp extracts may mediate the biogenic synthesis of metal nanoparticles. In this study, the chemical composition of aqueous leaf extracts of three varieties of Romanian hemp (two monoecious, and one dioecious) have been determined by Fourier-Transformed Infrared spectroscopy (FT-IR), high-performance liquid chromatography, and mass spectrometry (UHPLC-DAD-MS). Then, their capability to mediate the green synthesis of silver nanoparticles (AgNPs) and their pottential antibacterial applications were evaluated. The average antioxidant capacity of the extracts had 18.4 ± 3.9% inhibition determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 78.2 ± 4.1% determined by 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS™) assays. The total polyphenolic content of the extracts was 1642 ± 32 mg gallic acid equivalent (GAE) L−1. After this, these extracts were reacted with an aqueous solution of AgNO3 resulting in AgNPs, which were characterized by UV−VIS spectroscopy, FT-IR, scanning electron microscopy (SEM-EDX), and dynamic light scattering (DLS). The results demonstrated obtaining spherical, stable AgNPs with a diameter of less than 69 nm and an absorbance peak at 435 nm. The mixture of extracts and AgNPs showed a superior antioxidant capacity of 2.3 ± 0.4% inhibition determined by the DPPH assay, 88.5 ± 0.9% inhibition as determined by the ABTS•+ assay, and a good antibacterial activity against several human pathogens: Escherichia coli, Klebsiella pneumoniae, Pseudomonas fluorescens, and Staphylococcus aureus.  相似文献

To confirm that the β-phenyl-α,β-unsaturated thiocarbonyl (PUSTC) scaffold, similar to the β-phenyl-α,β-unsaturated carbonyl (PUSC) scaffold, acts as a core inhibitory structure for tyrosinase, twelve (Z)-5-(substituted benzylidene)-4-thioxothiazolidin-2-one ((Z)-BTTZ) derivatives were designed and synthesized. Seven of the twelve derivatives showed stronger inhibitory activity than kojic acid against mushroom tyrosinase. Compound 2b (IC50 = 0.47 ± 0.97 µM) exerted a 141-fold higher inhibitory potency than kojic acid. Kinetic studies’ results confirmed that compounds 2b and 2f are competitive tyrosinase inhibitors, which was supported by high binding affinities with the active site of tyrosinase by docking simulation. Docking results using a human tyrosinase homology model indicated that 2b and 2f might potently inhibit human tyrosinase. In vitro assays of 2b and 2f were conducted using B16F10 melanoma cells. Compounds 2b and 2f significantly and concentration-dependently inhibited intracellular melanin contents, and the anti-melanogenic effects of 2b at 10 µM and 2f at 25 µM were considerably greater than the inhibitory effect of kojic acid at 25 µM. Compounds 2b and 2f similarly inhibited cellular tyrosinase activity and melanin contents, indicating that the anti-melanogenic effects of both were due to tyrosinase inhibition. A strong binding affinity with the active site of tyrosinase and potent inhibitions of mushroom tyrosinase, cellular tyrosinase activity, and melanin generation in B16F10 cells indicates the PUSTC scaffold offers an attractive platform for the development of novel tyrosinase inhibitors.  相似文献

A new cell‐printed scaffold consisting of poly(ϵ‐caprolactone) (PCL) and cell‐embedded alginate struts is designed. The PCL and alginate struts are stacked in an interdigitated pattern in successive layers to acquire a three‐dimensional (3D) shape. The hybrid scaffold exhibits a two‐phase structure consisting of cell (MC3T3‐E1)‐laden alginate struts able to support biological activity and PCL struts able to provide controllable mechanical support of the cell‐laden alginate struts. The hybrid scaffolds exhibit an impressive increase in tensile modulus and maximum strength compared to pure alginate scaffolds. Laden cells are homogeneously distributed throughout the alginate struts and the entire scaffold, resulting in cell viability of approximately 84%.  相似文献

The present study focuses on the biological synthesis, characterization, and antibacterial activities of silver nanoparticles (AgNPs) using extracellular extracts of Aspergillus japonicus PJ01.The optimal conditions of the synthesis process were: 10 mL of extracellular extracts, 1 mL of AgNO3 (0.8 mol/L), 4 mL of NaOH solution (1.5 mol/L), 30 °C, and a reaction time of 1 min. The characterizations of AgNPs were tested by UV-visible spectrophotometry, zeta potential, scanning electron microscope (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and thermogravimetric (TG) analyses. Fourier transform infrared spectroscopy (FTIR) analysis showed that Ag+ was reduced by the extracellular extracts, which consisted chiefly of soluble proteins and reducing sugars. In this work, AgNO3 concentration played an important role in the physicochemical properties and antibacterial properties of AgNPs. Under the AgNO3 concentration of 0.2 and 0.8 mol/L, the diameters of AgNPs were 3.8 ± 1.1 and 9.1 ± 2.9 nm, respectively. In addition, smaller-sized AgNPs showed higher antimicrobial properties, and the minimum inhibitory concentration (MIC) values against both E. coli and S. aureus were 0.32 mg/mL.  相似文献

通过原位沉淀法和冷冻相分离技术得到含有钙磷前驱体(CaP)的初始多孔支架, 利用多孔支架表面原位生成的壳聚糖(CS)膜减缓NaOH溶液中OH-离子的渗透速率, 以达到纳米羟基磷灰石(nHA)缓慢形成的目的, 从而制得nHA 分布均匀的CS/nHA多孔复合支架. 利用扫描电镜(SEM)和万能试验机研究复合支架的结构和性能, 发现nHA为针状结构, 长度为80200 nm, 宽度为2050 nm. 随着nHA含量的增加, 复合支架的孔隙率下降, 由(93.8±3.3)%降至(87.7±3.8)%, 压缩强度则逐渐提高, 由(0.5±0.09) MPa增加至(1.5±0.06) MPa. 当复合支架中nHA质量分数为25%时, 未发现nHA团聚现象, nHA均匀地分布于CS基体中. 通过红外光谱(FTIR)、 X射线衍射(XRD)及X射线光电子能谱(XPS)等分析推断, nHA与CS之间可能存在配位和氢键作用. 细胞实验结果表明, CS/nHA多孔复合支架具有良好的生物相容性, 细胞在支架内部贴壁黏附生长. CS/nHA多孔复合支架有望在骨组织工程领域具有良好的应用前景.  相似文献

Scaffold, an essential element of tissue engineering, should provide proper physical and chemical properties and evolve suitable cell behavior for tissue regeneration. Polycaprolactone/Gelatin (PCL/Gel)‐based nanocomposite scaffolds containing hydroxyapatite nanoparticles (nHA) and vitamin D3 (Vit D3) were fabricated using the electrospinning method. Structural and mechanical properties of the scaffold were determined by scanning electron microscopy (SEM) and tensile measurement. In this study, smooth and bead‐free morphology with a uniform fiber diameter and optimal porosity level with appropriate pore size was observed for PCL/Gel/nHA nanocomposite scaffold. The results indicated that adding nHA to PCL/Gel caused an increase of the mechanical properties of scaffold. In addition, chemical interactions between PCL, gelatin, and nHA molecules were shown with XRD and FT‐IR in the composite scaffolds. MG‐63 cell line has been cultured on the fabricated composite scaffolds; the results of viability and adhesion of cells on the scaffolds have been confirmed using MTT and SEM analysis methods. Here in this study, the culture of the osteoblast cells on the scaffolds showed that the addition of Vit D3 to PCL/Gel/nHA scaffold caused further attachment and proliferation of the cells. Moreover, DAPI staining results showed that the presence and viability of the cells were greater in PCL/Gel/nHA/Vit D3 scaffold than in PCL/Gel/nHA and PCL/Gel scaffolds. The results also approved increasing cell proliferation and alkaline phosphatase (ALP) activity for MG‐63 cells cultured on PCL/Gel/nHA/Vit D3 scaffold. The results indicated superior properties of hydroxyapatite nanoparticles and vitamin D3 incorporated in PCL/Gel scaffold for use in bone tissue engineering.  相似文献

Here, we demonstrated the fabrication of a composite scaffold (chitosan [CS], collagen [Col], and hydroxyapatite [HA]) with the incorporation of encapsulated Cissus quadrangularis (CQ) extract for tissue engineering applications. First, the crude extract of CQ loaded nanoparticles were synthesized via double emulsion technique using polycaprolactone (PCL) and polyvinyl alcohol (PVA) as oil and aqueous phases, respectively. Both PCL (20, 40, and 80 mg/mL) and PVA (0.5%, 1%, and 3% w/v) concentrations were varied to determine the optimum concentrations for CQ‐loaded nanoparticle preparation. The CQ‐loaded PCL nanoparticles (CQ‐PCL NPs), prepared with 20 mg/mL PCL and 0.5% (w/v) PVA, exhibited the smallest size of 334.22 ± 43.21 nm with 95.54 ± 1.49% encapsulation efficiency. Then, the CQ‐PCL NPs were incorporated into the CS/Col/HA scaffolds. These scaffolds were also studied for their ultrastructure, pore sizes, chemical composition, compressive modulus, water swelling, weight loss, and biocompatibility. The results showed that the addition of CQ‐PCL NPs into the scaffolds did not dramatically alter the ultrastructure and properties of the scaffolds, compared to CS/Col/HA scaffolds alone. However, incorporation of CQ‐PCL NPs in the scaffolds improved the release profile of CQ by preventing the initial burst release and prolonging the release rate of CQ. In addition, the CQ‐PCL NPs‐loaded CS/Col/HA scaffolds supported the attachment and proliferation of MC3T3‐E1 osteoblast cells.  相似文献

A hybrid technology that combines a three‐dimensional (3‐D) dispensing system with an electrospinning process was used to produce a hierarchical 3‐D scaffold consisting of micro‐sized polycaprolactone (PCL) strands and micro/nano‐sized fibres. The micro/nanofibre biocomposites electrospun with PCL/small intestine submucosa (SIS) and PCL/Silk fibroin were layered between melt‐plotted micro‐strands. The scaffold containing SIS exhibited a stronger hydrophilic property than other scaffolds due to the various hydrophilic components in SIS. The 3‐D hierarchical scaffold having biocomposites exhibited an incredibly enhanced initial cell attachment and proliferation of bone marrow‐derived mesenchymal stem cells relative to the normally designed 3‐D scaffold.

Porous nano-hydroxyapatite/polycaprolactone (nHA/PCL) scaffolds with different composition ratios of nHA/PCL were fabricated via a melt-molding/porogen leaching technique. All scaffolds were characterized before and after degradation in vitro for six months. The original scaffolds had high porosity at around 70% and showed decreasing compressive modulus (from 24.48 to 2.69 MPa for hydrated scaffolds) with the introduction of nHA. It was noted that the scaffolds could retain relatively stable architecture and mechanical properties for at least six months, although some slight changes happened with the nHA/PCL scaffolds in the mass, the nHA content, the PCL molecular weight and the crystallinity. Moreover, during the 7 days culture of bone marrow stromal cells (BMSCs) on scaffolds, the cell adhesion and proliferation of BMSCs were presented well on both the surface and the cross-section of the scaffolds. All of these results suggested the nHA/PCL scaffolds to be promising in bone tissue engineering.  相似文献

Silver nanoparticles (AgNPs) are favoured antibacterial agents in nano-enabled products and can be released into water resources where they potentially elicit adverse effects. Herein, interactions of 10 and 40 nm AgNPs (10-AgNPs and 40-AgNPs) with aquatic higher plant Salvinia minima at 600 µg/L in moderately hard water (MHW), MHW of raised calcium (Ca2+), and MHW containing natural organic matter (NOM) were examined. The exposure media variants altered the AgNPs’ surface properties, causing size-dependent agglomeration. The bio-accessibility in the ascending order was: NOM < MHW < Ca2+, was higher in plants exposed to 10-AgNPs, and across all exposures, accumulation was higher in roots compared to fronds. The AgNPs reduced plant growth and the production of chlorophyll pigments a and b; the toxic effects were influenced by exposure media chemistry, and the smaller 10-AgNPs were commonly the most toxic relative to 40-AgNPs. The toxicity pattern was linked to the averagely higher dissolution of 10-AgNPs compared to the larger counterparts. The scanning electron microscopy and X-ray fluorescence analytical techniques were found limited in examining the interaction of the plants with AgNPs at the low exposure concentration used in this study, thus challenging their applicability considering the even lower predicted environmental concentrations AgNPs.  相似文献

Fibrous scaffolds, which can mimic the elastic and anisotropic mechanical properties of native tissues, hold great promise in recapitulating the native tissue microenvironment. We previously fabricated electrospun fibrous scaffolds made of hybrid synthetic elastomers (poly(1,3‐diamino‐2‐hydroxypropane‐co‐glycerol sebacate)‐co‐poly (ethylene glycol) (APS‐co‐PEG) and polycaprolactone (PCL)) to obtain uniaxial mechanical properties similar to those of human aortic valve leaflets. However, conventional electrospinning process often yields scaffolds with random alignment, which fails to recreate the anisotropic nature of most of the soft tissues such as native heart valves. Inspired by the structure of native valve leaflet, we designed a novel valve leaflet‐inspired ring‐shaped collector to modulate the electrospun fiber alignment and studied the effect of polymer formulation (PEG amount [mole %] in APS‐co‐PEG; ratio between APS‐co‐PEG and PCL; and total polymer concentration) in tuning the biaxial mechanical properties of the fibrous scaffolds. The fibrous scaffolds collected on the ring‐shaped collector displayed anisotropic biaxial mechanical properties, suggesting that their biaxial mechanical properties are closely associated with the fiber alignment in the scaffold. Additionally, the scaffold stiffness was easily tuned by changing the composition and concentration of the polymer blend. Human valvular interstitial cells (hVICs) cultured on these anisotropic scaffolds displayed aligned morphology as instructed by the fiber alignment. Overall, we generated a library of biologically relevant fibrous scaffolds with tunable mechanical properties, which will guide the cellular alignment.  相似文献

Three‐dimensional (3D) printing becomes an attractive technique to fabricate tissue engineering scaffolds through its high control on fabrication and repeatability using the printing parameters. This technique can be combined by the finite element method (FEM), and tissue‐specific scaffolds with desirable morphological and mechanical properties can be designed and manufactured. In this study, the influential 3D printing parameters on the morphological and mechanical properties of polycaprolactone (PCL) filament and scaffold were studied experimentally and numerically. First, the effects of printing parameters and process on the properties of extruded PCL filament were investigated. Then, using FEM, the effects of filament specifications on the overall characteristics of the scaffold were evaluated. Results showed that both the printing process in terms of resting time and remaining time and the printing parameters like pressure, printing speed, and printing path length have influenced the filament properties. In addition, both the filament diameter and elastic modulus had significant effects on the properties of scaffold especially, a 20% increase in the filament diameter caused the scaffold compressive elastic modulus to rise by around 72%. It is concluded that the printing parameters and process must be tuned very well in fabricating scaffolds with the desired morphology and mechanical property.  相似文献

The ability to control the architecture and strength of a bone tissue engineering scaffold is critical to achieve a harmony between the scaffold and the host tissue. Rapid prototyping (RP) technique is applied to tissue engineering to satisfy this need and to create a scaffold directly from the scanned and digitized image of the defect site. Design and construction of complex structures with different shapes and sizes, at micro and macro scale, with fully interconnected pore structure and appropriate mechanical properties are possible by using RP techniques. In this study, RP was used for the production of poly(ε-caprolactone) (PCL) scaffolds. Scaffolds with four different architectures were produced by using different configurations of the fibers (basic, basic-offset, crossed and crossed-offset) within the architecture of the scaffold. The structure of the prepared scaffolds were examined by scanning electron microscopy (SEM), porosity and its distribution were analyzed by micro-computed tomography (µ-CT), stiffness and modulus values were determined by dynamic mechanical analysis (DMA). It was observed that the scaffolds had very ordered structures with mean porosities about 60%, and having storage modulus values about 1 × 107 Pa. These structures were then seeded with rat bone marrow origin mesenchymal stem cells (MSCs) in order to investigate the effect of scaffold structure on the cell behavior; the proliferation and differentiation of the cells on the scaffolds were studied. It was observed that cell proliferation was higher on offset scaffolds (262000 vs 235000 for basic, 287000 vs 222000 for crossed structure) and stainings for actin filaments of the cells reveal successful attachment and spreading at the surfaces of the fibers. Alkaline phosphatase (ALP) activity results were higher for the samples with lower cell proliferation, as expected. Highest MSC differentiation was observed for crossed scaffolds indicating the influence of scaffold structure on cellular activities.  相似文献

A group of polyethers containing electroactive pendent 4,7-diarylfluorene chromophores have been prepared by the multi-step synthetic route. Full characterization of their structures has been presented. The polymeric materials represent derivatives of high thermal stability with initial thermal degradation temperatures in a range of 392–397 °C. Glass transition temperatures of the amorphous polymers range from 28 °C to 63 °C and depend on structures of the 4,7-diarylfluorene chromophores. Electron photoemission spectra of thin layers of the electroactive derivatives showed ionization potentials in the range of 5.8–6.0 eV. Hole injecting/transporting properties of the prepared polymeric materials were confirmed during formation of organic light-emitting diodes with tris(quinolin-8-olato)aluminium (Alq3) as a green emitter, which also serves as an electron transporting layer. The device using hole-transporting polymer with electronically isolated 2,7-di(4-biphenyl)fluorene chromophores demonstrated the best overall performance with low turn on voltage of 3 V, high current efficiency exceeding 1.7 cd/A, and with maximum brightness over 200 cd/m2. The organic light-emitting diode (OLED) characteristics were measured in non-optimized test devices. The efficiencies could be further improved by an optimization of device structure, formation conditions, and encapsulation of the devices.  相似文献

This research aimed to enhance the antibacterial activity of silver nanoparticles (AgNPs) synthesized from silver nitrate (AgNO3) using aloe vera extract. It was performed by means of incorporating AgNPs on an activated carbon nanoparticle (ACNPs) under ultrasonic agitation (40 kHz, 2 × 50 watt) for 30 min in an aqueous colloidal medium. The successful AgNPs synthesis was clarified with both Ultraviolet-Visible (UV-Vis) and Fourier Transform Infrared (FTIR) spectrophotometers. The successful AgNPs–ACNPs incorporation and its particle size analysis was performed using Transmission Electron Microscope (TEM). The brown color suspension generation and UV-Vis’s spectra maximum wavelength at around 480 nm confirmed the existence of AgNPs. The particle sizes of the produced AgNPs were about 5 to 10 nm in the majority number, which collectively surrounded the aloe vera extract secondary metabolites formed core-shell like nanostructure of 8.20 ± 2.05 nm in average size, while ACNPs themselves were about 20.10 ± 1.52 nm in average size formed particles cluster, and 48.00 ± 8.37 nm in average size as stacking of other particles. The antibacterial activity of the synthesized AgNPs and AgNPs-immobilized ACNPs was 57.58% and 63.64%, respectively (for E. coli); 61.25%, and 93.49%, respectively (for S. aureus). In addition, when the AgNPs-immobilized ACNPs material was coated on the cotton and polyester fabrics, the antibacterial activity of the materials changed, becoming 19.23% (cotton; E. coli), 31.73% (polyester; E. coli), 13.36% (cotton; S. aureus), 21.15% (polyester; S. aureus).  相似文献

A simple, eco-friendly, cost-effective and rapid microwave-assisted method has been developed to synthetize dendritic silver nanostructures, composed of silver nanoparticles (AgNPs), using white grape pomace aqueous extract (WGPE) as both reducing and capping agent. With this aim, WGPE and AgNO3 (1 mM) were mixed at different ratio, and microwave irradiated at 700 W, for 40 s. To understand the role of bioactive compounds involved in the green synthesis of AgNPs, preliminary chemical characterization, FT-IR analysis and 1H NMR metabolite profiling of WGPE were carried out. The effects of bioactive extract concentration and stability over time on AgNPs formation were also evaluated. WGPE-mediated silver nanostructures were then characterized by UV–vis, FTIR analyses, and scanning electron microscopy. Interestingly, the formation of dendritic nanostructures, originated from the self-assembly of Ag rounded nanoparticles (average diameter of 33 ± 6 nm), was observed and ascribed to the use of microwave power and the presence of organic components within the used WGPE, inducing an anisotropic crystal growth and promoting a diffusion-limited aggregation mechanism. The bio-dendritic synthetized nanostructures were also evaluated for potential applications in bio-sensing and agricultural fields. Cyclic voltammetry measurements in 0.5 M phosphate + 0.1 M KCl buffer, pH 7.4 showed that green AgNPs possess the electroactive properties typical of AgNPs produced using chemical protocol. The biological activity of synthetized AgNPs was evaluated by in-vitro antifungal activity against F. graminearum. Additionally, a phytotoxicity evaluation of synthetized green nanostructures was carried out on wheat seed germination. Results highlighted the potential of WGPE as green agent for bio-inspired nanomaterial synthesis, and of green Ag nanostructures, which can be used as antifungal agent and in biosensing applications.  相似文献

Bone‐derived extracellular matrix (ECM) is widely used in studies on bone regeneration because of its ability to provide a microenvironment of native bone tissue. However, a hydrogel, which is a main type of ECM application, is limited to use for bone graft substitutes due to relative lack of mechanical properties. The present study aims to fabricate a scaffold for guiding effective bone regeneration. A polycaprolactone (PCL)/beta‐tricalcium phosphate (β‐TCP)/bone decellularized extracellular matrix (dECM) scaffold capable of providing physical and physiological environment are fabricated using 3D printing technology and decoration method. PCL/β‐TCP/bone dECM scaffolds exhibit excellent cell seeding efficiency, proliferation, and early and late osteogenic differentiation capacity in vitro. In addition, outstanding results of bone regeneration are observed in PCL/β‐TCP/bone dECM scaffold group in the rabbit calvarial defect model in vivo. These results indicate that PCL/β‐TCP/bone dECM scaffolds have an outstanding potential as bone graft substitutes for effective bone regeneration.  相似文献

A new all‐aqueous and green process is described to form three‐dimensional porous silk fibroin matrices with control of structural and morphological features. Silk‐based scaffolds are prepared using lyophilization. Gelatin is added to the aqueous silk fibroin solution to change the silk fibroin conformation and silk fibroin–water interactions through adjusting the hydrophilic interactions in silk fibroin–gelatin–water systems to restrain the formation of separate sheet like structures in the material, resulting in a more homogenous structure. Water annealing is used to generate insolubility in the silk fibroin–gelatin scaffold system, thereby avoiding the use of organic solvents such as methanol to lock in the β‐sheet structure. The adjusting of the concentration of gelatin, as well as the concentration of silk fibroin, leads to control of morphological and functional properties of the scaffolds. The scaffolds were homogeneous in terms of interconnected pores, with pore sizes ranging from 100 to 600 µm, depending on the concentration of silk fibroin used in the process. At the same time, the morphology of the scaffolds changed from lamellar sheets to porous structures based on the increase in gelatin content. Compared with salt‐leaching aqueous‐derived scaffolds and hexafluoroisopropanol (HFIP)‐derived scaffolds, these freeze‐dried scaffolds had a lower content of β‐sheet, resulting in more hydrophilic features. Most of gelatin was entrapped in the silk fibroin–gelatin scaffolds, without the burst release in PBS solution. During in vitro cell culture, these silk fibroin–gelatin scaffolds had improved cell‐compatibility than salt‐leaching silk fibroin scaffolds. This new process provides useful silk fibroin‐based scaffold systems for use in tissue engineering. Furthermore, the whole process is green, including all‐aqueous, room temperature and pressure, and without the use of toxic chemicals or solvents, offering new ways to load bioactive drugs or growth factors into the process.

Tissue engineering approach aims to overcome the transplant drawbacks and facilitate tissue repair and regeneration. Here, a new conductive, highly porous, and flexible polycaprolactone/gelatin/polypyrrole/graphene 3D scaffolds for nerve tissue repair is presented. A simple and efficient porogen leaching fabrication method is applied to create a 3D network with a pore radius of 3.8 ± 0.7 to 4.2 ± 0.8 μm with an exceptional uniform circular porous structure. The conductivity of the polymeric scaffold without graphene, in wet conditions, was found to be 0.78 ± 0.1 S.m−1 and it increased to 3.3 ± 0.2 S.m−1 for the optimized sample containing 3wt% graphene (G3). Tensile strength was measured at 163 KPa for the base sample (without graphene) and improved to 526 KPa for G3 sample. Following 42 days of incubation in PBS, 32.5% degradation for the base sample (without graphene) was observed. The cell study demonstrated a non-cytotoxic nature of all scaffolds tested and the cells had mostly stretched and covered the surface. Overall, the sum of results presented in this study demonstrate a simple fabrication platform with extraordinary aspects that can be utilized to mimic the native conductive tissue properties, and also because of its flexibility it can easily be rolled into a nerve conduit to fill gaps in nerve tissue regeneration.  相似文献
