双光子吸收是指在强光激发下,介质分子同时吸收两个光子,从基态跃迁到两倍光子能量的激发态的过程。荧光显微成像是研究活体生物的重要工具,而最通常的细胞成像方法则是使用单光子激发荧光团的单光子显微成像。近红外光源激发的双光子荧光探针克服了单光子荧光探针的光漂白与光致毒而更适于生物检测与成像,为生命科学研究提供了更为锐利的工具。双光子荧光探针的作用机理包括分子内电荷迁移(ICT)、荧光共振能量迁移(FRET)、光诱导电子迁移(PET)与基团转换(GC) 4种方式。该文综述了双光子阳离子探针(Mg2+, Ca2+, Pb2+, Hg2+, Ag+, Fe3+, Zn2+, Na+, Cr3+)、双光子阴离子探针(F-)、pH探针、双光子葡萄糖示踪器、双光子脂筏探针、双光子巯基探针、双光子半胱氨酸探针和双光子生物标记探针,以及双光子荧光探针在生物成像方面的应用,展望了双光子荧光探针的发展趋势与应用前景。  相似文献

Two-photon polymerization (2PP) represents a powerful technique for the fabrication of precise three-dimensional structures on a micro- and nanometer scale for various applications. While many review articles are focusing on the used polymeric materials and their application in 2PP, in this review the class of two-photon photo initiators (2PI) used for radical polymerization is discussed in detail. Because the demand for highly efficient 2PI has increased in the last decades, different approaches in designing new efficient 2PIs occurred. This review summarizes the 2PIs known in literature and discusses their absorption behavior under one- and two-photon absorption (2PA) conditions, their two-photon cross sections (σTPA) as well as their efficiency under 2PP conditions. Here, the photo initiators are grouped depending on their chromophore system (D-π-A-π-D, D-π-D, etc.). Their polymerization efficiencies are evaluated by fabrication windows (FW) depending on different laser intensities and writing speeds.  相似文献

通过多步反应制备了三个新型的双光子吸收化合物,测试了它们的光物理性质,发现该系列化合物具有较大的双光子吸收截面,并且随温度降低其双光子吸收截面增大.尤其是化合物TMVDB,其量子效率为0.83,常温时的双光子吸收截面达到1164GM,其双光子吸收截面除以分子量为2.08,表现出了优良的双光子吸收发光性能.进一步将TMVDB掺杂到固体材料中,得到强双光子诱导发光的固体材料.  相似文献

合成了一系列二噻吩衍生物类有机合物:5,5'-二(对-N,N-二甲胺基苯乙 烯基)-2,2'-二噻吩(BMSBT)与5,5'-二(对-N,N-二乙胺基苯乙烯基)- 2,2'-二噻吩(BESBT),5,5'-二(对N-环丁胺基苯乙烯基)-2,2'-二噻吩( BBSBT),5,5-二(对-N-咔唑基苯乙烯基)2-2'-二噻吩(BCSBT),并测定了 其吸收光谱、单光子荧光光谱和双光子荧光光谱。BMSBT,BESBT和BBSBT的单光子 荧光参数存在的很强的、规则的溶剂效应,表明其激发态的分子可能有较大的极性 。在800nm fs激光下测得BMSBT与BESBT在560nm附近(与单光子荧光发射峰波长接 近的位置)有较强的上转换荧光发射,上转换荧光强度与激发能量之间较好的平方 关系表明了双光子激发的机理,这种上转换荧光被称为双光子荧光。用以光子荧光 法测得BMSBT和BESBT的双光子吸收截面分别为54*10~(-50)(cm~4·s)/photon和 102*10~(-50)(cm~4·s)/photon。  相似文献

Lipophilic fluorophores are widely implemented in nonlinear microscopy; however, few existing membrane-specific probes combine the high brightness of two-photon excited fluorescence (2PEF) with pH sensitivity. Herein we describe four novel two-photon excited fluorophores, based on a coumarin 151 core structure, where lipophilicity is induced by a covalently attached phosphazene moiety. Changing the environmental acidity using trifluoromethanesulfonic (triflic) acid leads to profound changes in the linear fluorescence and 2PEF characteristics, due to chromophores’ switching between neutral- and protonated forms. We characterize this dependence by measuring the two-photon absorption (2PA) spectra over the region λ2PA=550–1000 nm, observing 2PA cross sections of σ2PA=10–20 GM, with an associated 2PEF brightness of 10–13 GM, in neutral solutions of both acetonitrile and n-octanol. Although quantum chemical modelling and NMR measurements show that, at high chromophore concentrations, protonation may be accompanied by a dimerization process, these dimers likely do not form at the lower concentrations used in optical spectroscopy.  相似文献

合成了一个新的双光子吸收聚合引发剂,9-乙基-3-{2-[4-(2-吡啶基-4-乙烯基)-苯基]-乙烯基}-9氢咔唑 (EPVPVC),用1H NMR谱、13C NMR谱和元素分析进行了表征.测试了紫外吸收光谱、单光子荧光光谱、单光子荧光寿命、单光子荧光量子产率和双光子荧光光谱.量化计算结果表明,EPVPVC的双光子吸收截面为56.6×10-50 cm4·s·photon-1.在780 nm的飞秒脉冲激光激发下,EPVPVC发出较强的上转换荧光,荧光峰位于536 nm.用EPVPVC做引发剂,加入丙烯酸酯型齐聚物(cN120C80),在脉冲宽度为200 fs,重复频率为76 MHz,中心波长为780 nm 飞秒脉冲激光下,制作了一个三维周期性微结构.在理论计算的基础上,定性地探讨了其可能的聚合机理.  相似文献

用密度泛涵方法和ZINDO方法,从理论上研究了一系列四羧酸二萘嵌苯的衍生物(PTCDS)。对其平衡几何、电子结构和单双光子吸收性质进行了详细的计算研究。结果表明,此系列衍生物的双光子吸收截面主要由其主体结构(二萘嵌苯)决定。△EH-Lgap (HOMO与LUMO间的能隙)的减小,HOMO轨道能量的增大,分子主轴两端取代基给电子强度的增强,共轭长度的增大,分子刚性的增强都有利于此系列化合物双光子吸收截面的增大。并设计了比文献[22]报道的分子C有更大的双光子吸收截面的两个分子D1,D2。  相似文献

The structure of the dipolar compound has been determined by X-ray diffraction. 以三苯胺或硝基苯为端基, 合成了三个卟啉多枝分子: 5-(4-硝基苯甲酰氧基)苯基-10,15,20-三-(4-溴苯基)卟啉(TPP-NO2)、5-(4-硝基苯甲酰氧基)苯基-10,15,20-三-(4-二苯胺基-1-苯乙烯基)苯基卟啉(TPP-X3)和5,10,15,20-四-(4-二苯胺基-1-苯乙烯基)苯基卟啉(TPP-X4), 进行了红外光谱、核磁共振光谱和质谱表征. 比较研究了分子“枝”、“核”不同键合方式与不同对称结构对分子的线性光谱、非线性光谱以及分子内能量转移行为的影响. 在钛宝石激光器(800 nm)和Nd∶YAG倍频光(532 nm)泵浦下, 样品溶液均发出卟啉环特有的红色荧光——前者系双光子吸收机制“上转换”荧光, 后者则为双光子吸收与分子内能量转移机制“下转换”荧光. 飞秒Z-scan技术测得样品双光子吸收截面最大可达130 GM, 与四苯基卟啉(TPP)同等测试条件下的双光子吸收截面相比增大了两个数量级.  相似文献

Summary: This article describes the development and evolution of two-photon lithography of 3-dimensional (3D) polymeric structures through two-photon photopolymerization (TPP) and the materials used to facilitate the same. The first part elaborates on the advantages of TPP over conventional lithographic techniques used in the fabrication of 3D microelectromechanical systems (MEMS). The second part introduces the preparation of highly efficient organic two-photon absorbing materials based on the centro-symmetric π-conjugated system and their performance as sensitizers in two-photon polymerization.  相似文献

The photophysical properties and the photoinitiating reactivity of a ditopic alkoxynitrostilbene were compared to those of its single branch chromophore used as a reference. Whereas a trivial additive effect is observed when considering the one- and two-photon absorption properties, a clear and very significant amplification has been highlighted for the photoreactivity of this free radical photoinitiator which was used as a hydrogen abstractor in presence of an aliphatic amine co-reactant. We indeed demonstrate that the proximity of two nitroaromatics moieties within the same molecular architecture gives rise to an original cycling mechanism based on a stepwise photo triggering of each photoredox center followed by a subsequent regenerative process. The combination of a high two-photon absorption cross-section (δ780nm≈330 GM) with a strong enhancement in photoreactivity makes this nitrostilbene bichromophore a very suitable candidate for two-photon polymerization applications.  相似文献

自2000年以来,双光子技术开始应用于光生酸剂体系中,并取得了一定的研究进展。双光子技术在光生酸剂中的应用主要有两种情况:一是单分子体系,即光生酸剂分子本身具有较高的双光子吸收截面,可以在双光子激发下产生光酸;二是双分子体系,由具有较高吸收截面的敏化剂分子和光生酸剂分子组成,通过分子间电子转移的方式产生光酸。本文就这一类具有特殊光学性质的有机分子体系的构成及其应用进行了综述,介绍了不同类型的可以利用双光子技术的光生酸剂体系,总结了双光子技术在光生酸剂发展中面临的一些关键问题,展望了双光子技术在光生酸剂中的发展方向。  相似文献

Two-photon excited fluorescent (TPEF) materials are highly desirable for bioimaging applications owing to their unique characteristics of deep-tissue penetration and high spatiotemporal resolution. Herein, by connecting one, two, or three electron-deficient zinc porphyrin units to an electron-rich triazatruxene core via ethynyl π-bridges, conjugated multipolar molecules TAT-(ZnP) n (n=1–3) were developed as TPEF materials for cell imaging. The three new dyes present high fluorescence quantum yields (0.40–0.47) and rationally improved two-photon absorption (TPA) properties. In particular, the peak TPA cross section of TAT-ZnP (436 GM) is significantly larger than that of the ZnP reference (59 GM). The δTPA values of TAT-(ZnP)2 and TAT-(ZnP)3 further increase to 1031 and up to 1496 GM, respectively, indicating the effect of incorporated ZnP units on the TPA properties. The substantial improvement of the TPEF properties is attributed to the formation of π-conjugated quadrapole/octupole molecules and the extension of D -π-A-D systems, which has been rationalized by density function theory (DFT) calculations. Moreover, all of the three new dyes display good biocompatibility and preferential targeting ability toward cytomembrane, thus can be superior candidates for TPEF imaging of living cells. Overall, this work demonstrated a promising strategy for the development of porphyrin-based TPEF materials by the construction and extension of D -π-A-D multipolar array.  相似文献

Exploration of simple and economical synthetic routes to construct active two-photon initiators (2PIs) with long conjugation length is highly desirable to promote the development of two-photon-induced photopolymerization technology. In this article, several conjugated carbazole-based derivatives containing N=C bonds as π bridges and electron donor/acceptor groups were prepared through one-step Schiff base reaction. The structure–activity relationship was systematically investigated through calculations and experimental tests. The results showed that compared to the D-π-D-π-D molecules, the A-π-D-π-A molecules exhibited stronger charge transfer, which induce redshifted linear absorption and enhanced two-photon absorption at 800 nm. The 2PIs displayed aggregation-induced emission property and show potential for the fabrication of luminescent microdevices. In two-photon-induced microfabrication tests, the formulations containing novel 2PIs exhibited lower threshold energy than the widely used commercial photoinitiator Irgacure 369 and the two-photon resists IP-L is the brand name of the two-photon resist from Nanoscribe, specially designed for nanoscribe's laser lithography systems. © 2018 Wiley Periodicals, Inc. E,E-1,4-Bis(4'-N,N-diphenylaminostyryl)-2,5-dimethoxybenzene (DPAMOB) has been synthesized by a simple and effective solid phase Wittig reaction and characterized by 1^H NMR spectra and elemental analysis, Linear absorption, single-photon induced fluorescence and two-photon induced fluorescence spectra were experimentally studied. The new dye has a large two-photon absorption (TPA) cross-section of σr= 1007,2 GM [1 GM= 1 × 10^-50 cm^4·s/(photon molecule)] at 800 nm measured by the two-photon induced fluorescence method. The experimental results confirm that DPAMOB is a good TPA chromophore and can successfully initiate two-photon photopolymerization of ethoxylated trimethylolpropane triacrylate esters (SR454). Finally, a microstructure has been fabricated by use of DPAMOB as initiator.  相似文献

双光子荧光显微成像兼具诸如近红外激发、暗场成像、避免荧光漂白和光致毒、定靶激发、高横向分辨率与纵向分辨率、降低生物组织吸光系数及降低组织自发荧光干扰等特点而显著地优于单光子荧光显微成像,为生命科学研究提供了更为锐利的工具。而用于像离子的含量及其对生理的影响、离子参与的生理活动机制、离子与分子的作用、特定分子的分布及其相互作用等方面研究的双光子荧光探针,是实现成像的关键。双光子荧光探针的研究旨在促进双光子荧光显微镜应用的发展,促进生命科学、医学科学的快速发展,同时也带动双光子荧光探针所隶属的化学这一学科的发展。因此对双光子荧光探针的研究具有重要的理论和实践意义。该文综述了双光子荧光显微成像的优点、双光子荧光探针设计的原理及双光子荧光探针在离子分析方面的应用,并展望了这类荧光探针的发展趋势与应用前景。  相似文献

Two carbazole-based polymerization initiators possessing blue fluorescence emission have been synthesized via Wittig reaction in the solid phase at room temperature. Two-photon excited fluorescence (TPEF) spectra for them were investigated under 800 nm fs laser pulse and two-photon absorption cross sections were determined by the Z-scan technique. Then two-photon initiating polymerization (TPIP) microfabrication experiments were successfully carried out. Three-dimensional lattice and artificial defects were gained, indicating that they were viable candidates for the two-photon polymerization initiator in practical application of microfabrication.
