利用辐照质粒DNA构象变化的分子模型,以DNA糖苷酶Fpg和AP核酸内切酶EndoIII识别并切割辐射所致DNA碱基损伤,将其转换为DNA断裂损伤,通过电泳分析DNA分子构象变化,研究比较γ射线、质子和7Li离子诱发DNA集簇性损伤。50Gy以上高剂量γ辐射对质粒DNA的损伤主要表现为单链断裂(SSB)和很少比例的双链断裂(DSB),并能产生一定水平的集簇性损伤。相比之下,高能质子束和高LET的7Li离子直接所致DNA的断裂损伤以及所产生的集簇性碱基损伤比γ射线的要严重,质子10Gy照射就可诱发明显的集簇损伤。  相似文献

We have investigated the damage of synthetic plasmid pBR322 DNA in dilute aqueous solutions induced by fast carbon ions. The relative contribution of indirect damage and direct damage to the DNA itself is expected to vary with linear energy transfer along the ion track, with the direct damage contribution increasing towards the Bragg peak. Therefore, 12C ions at the spread-out Bragg peak (dose averaged LET = 189 ± 15 keV/μm) and in the plateau region of the Bragg curve (LET = 40 keV/μm) were employed and the radical scavenger concentration in the plasmid solution was varied to quantify the indirect effect. In order to minimize the influence of 12C break-up fragments, a relatively low initial energy of 90 MeV/nucleon was employed for the carbon ions. DNA damage has been quantified by subsequent electrophoresis on agarose gels. We find that strand breaks due to both indirect and direct effects are systematically higher in the plateau region as compared to the Bragg peak region with the difference being smallest at high scavenging capacities. In view of the fact that the relative biological effectiveness for many biological endpoints is maximum at the Bragg peak our findings imply that DNA damage at the Bragg peak is qualitatively most severe.  相似文献

Interaction of ionizing radiation with plasmid DNA can lead to formation of single strand breaks, double strand breaks and clustered lesions. We have investigated the response of the synthetic plasmid pBR322 in aqueous solution upon irradiation with 12C ions under spread-out Bragg peak conditions (densely ionizing) and with 137Cs γ-photons (sparsely ionizing) as a function of dose. To evaluate the relevance of indirect effects, i.e. influences of diffusion limited radical induced DNA damage triggered by water radiolysis, the experiments were performed at various concentrations of the radical scavenger mannitol. Agarose gel electrophoresis was employed to quantify the DNA damage. At low scavenger concentration for a given dose DNA damage is higher for γ-photons than for 12C. For the latter, the microscopic dose distribution is inhomogeneous, with very high dose deposited along the few tracks through the solution. This is in agreement with the concept that scavengers efficiently reduce damage for γ-photons, implying that the underlying damage mechanism is single strand break induction by OH radicals. For 12C induced damage, the fraction of SSB and DSB that is unaffected by radical scavengers and thus due to direct effect is quantified.  相似文献

在地面模拟微重力的情况下, 应用碱性单细胞凝胶电泳(SCGE)技术对80 MeV/u Ne离子辐射诱发人血淋巴细胞DNA损伤修复效应进行了研究。 在不同时刻对相同剂量辐照后的淋巴细胞经单细胞电泳处理后显示, 在模拟微重力下孵育的彗星尾更长, 彗星头面积更小。 这表明, 相对地面环境而言, 模拟微重力环境对淋巴细胞的DNA损伤修复有一定的抑制作用。 Effect of the modeled microgravity (MMG) on heavy ion induced lymphocytes DNA repair by using single cell gel electrophoresis (SCGE) has been studied. The results showed that residual DNA damage induced by Ne ions irradiation increased more in cultures incubated in MMG than in 1 g, which indicated that MMG incubation after Ne ions irradiation reduce the DNA damage repair capacity.  相似文献

To reveal the physical origin of the giant magneto-optical enhancement of Ni2+ ions in barium ferrite, quantitative calculations of the contributions of both the intra-ionic electric dipole transition between the 3d8 and 3d7 4p configurations of the Ni2+ ions and the intra-ionic electric dipole transition induced by odd-parity crystal field terms are presented. It is deduced that the transition is important in the origin of the considered magneto-optical enhancement. The most important factor is the Ni-Fe superexchange interaction; since it is strong enough, the Faraday rotation produced by the Ni2+ ions is large though the energy difference between the 3d8 and 3 d7 4 p configurations is large. It is demonstrated that though the intra-ionic electric dipole transition does produce Faraday rotation peaks in the visible range, their magnitude is too small to explain the observed Faraday rotation. The effect of the spin-orbit interaction on the Faraday rotation is analysed. The spin-orbit interaction of the ground configuration plays a very important role in the occurrence of Faraday effects, but the Faraday rotation does not increase linearly with the strength of the spin-orbit coupling. On the contrary, the spin-orbit interaction of the excited configuration has almost no effect on the Faraday rotation. It is shown that the mixing of the different multiplets of the ground term induced by the crystal field has a great influence on the magneto-optical properties. Received 7 January 1998  相似文献

Single nucleon transfer reactions in collisions of 156 MeV6Li ions with6Li nuclei, leading to unstable final nuclei (5Li,5He) have been experimentally studied. The measured6Li(6Li,7Li)5Ligr and6Li(6Li,7Be)5Hegr differential cross sections have been analysed on the basis of a FR-DWBA procedure, looking to which extent the entrance channel optical potentials may account for the unknown exit channel potentials.  相似文献

In this work, we have studied the effects of swift heavy ion irradiation on the creation of new functional groups in high and low density polyethylene (HDPE and LDPE). Polymers were irradiated with different ions (6.77 MeV He and 47 MeV Li) and fluences. The induced changes were analyzed by Fourier transform infrared (FTIR) spectroscopy. Creation and damage cross sections for some groups were compared for two different types of PE.  相似文献

Thermoluminescence (TL) of LiNaSO4:Eu phosphor, irradiated with 24 and 48 MeV 7Li ions at different fluences in the range 5×109-1×1012 ion/cm2, has been studied. The samples from the same batch were also exposed to γ-rays from a Cs137 source for comparative studies. The TL glow curves of the materials, irradiated with 7Li ions, have similar structures to that of γ-irradiated sample. They have a simple structure with a prominent peak at 412 K along with small one at around 481 K. The intensity ratios of 412-481 K peaks have been observed to increase with fluence increasing, while that of γ-irradiated sample shows a reverse trend. This could be attributed to the changes in the recombination center populations due to 7Li ions, that have been implanted inside the matrix of LiNaSO4:Eu, during irradiation and might also act as a source for new trapping and luminescent centers. The implantation has been confirmed by TRIM calculations. The penetration depths (where the ion comes to rest) are found to be 145 and 463 μm corresponding to 24 and 48 MeV ion beam energies, respectively, which are less than the thickness of the sample chips (∼800 μm). The efficiencies of LiNaSO4:Eu to 24 and 48 MeV 7Li ions measured relative to γ-rays of Cs137 are found to be 0.007 and 0.024, respectively. Theoretical analysis of the glow curves of the samples irradiated by 7Li ions and γ-rays were done by glow curve deconvolution method to determine trapping parameters of various peaks. The experimentally observed linearity/sublinearity has been discussed in the frame of track interaction model. Photoluminescence studies in the 7Li ions irradiated and un-irradiated samples show that europium ions have incorporated in the host in their divalent (emission at 440 nm) as well as trivalent (emissions at 594, 615 and 700 nm) forms. The intensities of the emission bands of these ions have been observed to increase with fluence increasing.  相似文献

Spectra and angular distributions have been measured for the light particles emitted from the reactions induced by 36 MeV 6Li and 7Li ions on targets of 12C, 13C and 27Al. Some measurements were also performed at beam energies of 28 and 32 MeV. The spectra are dominated by broad peaks centered at energies corresponding to the beam velocity, a characteristic of projectile breakup. A simple breakup model roughly reproduces the shapes of the measured spectra and angular distributions. The total cross sections observed for projectile breakup average about 600 mb and are almost as large as the fusion cross sections. The 7Li breakup yields are somewhat larger than those for 6Li, the same trend that was previously observed for the Li-induced fusion cross sections on C. As a result the measured total reaction cross sections induced by 7Li ions are about 200 mb larger than those of 6Li on the same targets. This difference is not predicted by conventional optical model calculations.  相似文献

Measurements of transient magnetic fields (TF) were performed on swift heavy ions of56Fe and80Se, with Coulomb excitation of their first 2+ state as probe, traversing thin Fe layers with segmented and unsegmented structures. The 50 µm×50 µm squares of the segments were accomplished applying the techniques of photolithography and ion etching. The magnitude of the TF deduced clearly shows that by segmentation of the targets the ion beam induced attentuations can be eliminated. This finding has direct applications tog-factor measurements.  相似文献

Radiotherapy employs ionizing radiation to induce lethal DNA lesions in cancer cells while minimizing damage to healthy tissues. Due to their pattern of energy deposition, better therapeutic outcomes can, in theory, be achieved with ions compared to photons. Antiprotons have been proposed to offer a further enhancement due to their annihilation at the end of the path. The work presented here aimed to establish and validate an experimental procedure for the quantification of plasmid and genomic DNA damage resulting from antiproton exposure. Immunocytochemistry was used to assess DNA damage in directly and indirectly exposed human fibroblasts irradiated in both plateau and Bragg peak regions of a 126 MeV antiproton beam at CERN. Cells were stained post irradiation with an anti-γ-H2AX antibody. Quantification of the γ-H2AX foci-dose relationship is consistent with a linear increase in the Bragg peak region. A qualitative analysis of the foci detected in the Bragg peak and plateau region indicates significant differences highlighting the different severity of DNA lesions produced along the particle path. Irradiation of desalted plasmid DNA with 5 Gy antiprotons at the Bragg peak resulted in a significant portion of linear plasmid in the resultant solution.  相似文献

Pulsed discharge is used for sterilization and disinfection, but the details of the molecular mechanisms remain largely unknown. Since pulsed discharge generates reactive oxygen species (ROS), we analyzed the oxidative DNA damages after pulsed discharge treatment to consider the involvement of ROS in the damaging process. We applied pulsed discharge with cavitation to plasmid DNA molecules and estimated the yields of the damages by agarose gel electrophoresis. The treated DNA contained various oxidative DNA damages, including single and double strand breaks and base lesions. The yields of the damages increased in response to the energy used for pulsed discharge. We also measured the yield of 8-hydroxyguanine (8-OH-G), one of the major oxidative base lesions, in the treated plasmid DNA by mass spectrometry quantitatively and found that the yield of the oxidative base lesion corresponded to the increment of the applied energy. In addition, we observed the involvement of mutM gene, which is responsible for repair of 8-OH-G, in the increased sensitivity of Escherichia coli to pulsed discharge. Therefore, ROS seem to mediate the sterilization ability of pulsed discharge.  相似文献

Abstract

Boron was implanted into several solids in the dose regime 1014 to 1016 ions/cm2 and at energies between some 10 keV and some MeV. Measurements of the corresponding depth distributions were performed by means of the 10B(n, α0)7Li(gnd) and 10B(n, α1)7Li?(1st) nuclear reaction techniques with thermal neutrons, and by SIMS. Dynamic nuclear polarization parameters, obtained at 75 G, are reported for 7Li ions in collision with several radical anions and with one radical cation. All systems show large negative 7Li N.M.R. enhancements indicative of weak scalar relaxation. However, radical induced relaxation rates derived from 7Li T 1 measurements suggest stronger complexing of lithium ions with radical anions than with the radical cation as would be expected from simple coulombic considerations. Translational modulation of the dipolar interaction best accounts for proton and radical cation dipolar relaxation rates while rotational modulation best accounts for the corresponding radical anion rates; this supports the interpretation above. A model for the lithium radical collision is proposed which implies that, for radical anions, scalar coupling is only apparently weak and that the low 7Li scalar relaxation rates observed result from scalar correlation times (τc = 10-8-10-9 s) longer than any yet observed by this technique. The model predicts that, for certain ranges of τc, increasing strength of complex formation should lead to smaller scalar relaxation rates and more negative enhancements, in contrast with the behaviour of fluorocarbons where the reverse was true. The predicted dependence of enhancement upon τc also suggests that 7Li enhancements should be extremely sensitive to variations in the chemical properties of the system.  相似文献

Angular distributions of the differential cross section and the second-rank tensor analysing power T20 have been measured for the one-proton transfer reaction induced by polarized 7Li on 208Pb at 33 MeV leading to several states in 209Bi. The experimental data were analysed simultaneously with data for elastic and inelastic scattering of 7Li by means of CCBA and CRC calculations. Hydroxyapatite crystalline nanoparticles were created by a precipitation hydrothermal technique and the majority of crystal particles were in the size range of 40–60nm and exhibited a colloidal feature when suspended in water. The gastric cancer SGC-7901 cell line cells were cultivated in the presence of10–100 μg ml−1 hydroxyapatite nanoparticle suspension and verified by MTT evaluation for their biocompatibility in vitro. The agarose gel electrophoresis analysis demonstrated that the HA nanoparticles potentially adsorb the green fluorescence protein EGFP-N1 plasmid DNA at pH 2 and 7, but not at pH 12. The DNA–nanoparticle complexes transfected EGFP-N1 pDNA into SGC-7901 cells in vitro with the efficiency about 80% as referenced with Lipofectmine TM 2000. In vivo animal experiment revealed no acute toxic adverse effect 2weeks after tail vein injection into mice, and TEM examination demonstrated their biodistribution and expression within the cytoplasm and also a little in the nuclei of the liver, kidney and brain tissue cells. These results suggest that the HA nanoparticle is a promising material that can be used as gene carrier, vectors.  相似文献

Local environments and dynamics of lithium ions in the binary lithium silicide Li13Si4 have been studied by 6Li MAS-NMR, 7Li spin-lattice relaxation time and site-resolved 7Li 2D exchange NMR measurements as a function of mixing time. Variable temperature experiments result in distinct differences in activation energies characterizing the transfer rates between the different lithium sites. Based on this information, a comprehensive picture of the preferred ionic transfer pathways in this silicide has been developed. With respect to local mobility, the results of the present study suggests the ordering Li6/Li7>Li5>Li1>Li4 >Li2/Li3. Mobility within the z=0.5 plane is distinctly higher than within the z=0 plane, and the ionic transfer between the planes is most facile via Li1/Li5 exchange. The lithium ionic mobility can be rationalized on the basis of the type of the coordinating silicide anions and the lithium-lithium distances within the structure. Lithium ions strongly interacting with the isolated Si4− anions have distinctly lower mobility than those the coordination of which is dominated by Si26− dumbbells.  相似文献

As-quenched and stress field annealed FINEMET ribbons were irradiated with 246?MeV energy Kr, 470?MeV energy Xe and 720?MeV energy Bi ions and investigated by 57Fe M?ssbauer spectroscopy and XRD methods. The change in relative areas of the 2nd and 5th lines in the M?ssbauer spectra indicated significant changes in the magnetic anisotropy of both as-quenched and stress annealed FINEMET due to irradiation with swift heavy ions. Differences were observed between the effect of irradiations with various ions having different energy and fluence. The effect of irradiation on the magnetic orientation in FINEMET was explained in terms of radiation induced defects. The swift heavy ion irradiation can be applied to produce FINEMET ribbons with more favorable soft magnetic properties for technological applications.  相似文献

Electrophoretic ratchets have been developed for both analytical and preparative electrophoresis. These ratchets use a new type of pulsed field. The quality of the fractionations meets the usual standards for biochemistry-based electrophoresis. The supporting medium is either an agarose gel or a capillary-contained polymer solution. The electrophoretic ratchets are effective with a particle that has an electrophoretic mobility (μ=velocity/electrical field) that varies as the electrical field varies. A ratchet developed for DNA molecules is effective because μ increases in magnitude as the electrical field increases in magnitude. Ratchets developed for both DNA–protein complexes and spheres are effective because of the opposite dependence of μ on the electrical field. Ratchet-based gel electrophoresis can be performed in a continuous, preparative mode. Ratchet-based capillary electrophoresis provides a necessary component for cyclic capillary electrophoresis. Cyclic capillary electrophoresis of DNA is a procedure for analyzing a DNA profile in several segments. These segments are separated by electrophoretic enhancements of the DNA profile. Cyclic capillary electrophoresis is being developed for increasing both the length and the accuracy of the analysis of a DNA-sequencing ladder.
