Electrochemical oxidation of 3,4‐dihydroxybenzoic acid ( 1 ) and 4‐tert‐butylcatechol ( 5 ) in the presence of 4‐hydroxy‐6‐methyl‐2‐pyrone ( 2 ) as nucleophile in aqueous solution has been studied using cyclic voltammetry and controlled‐potential coulometry. The results indicate that 1 via Michael reaction under electro‐decarboxylation reaction converts to heterocyclic compound 4 , and the quinone derived from 4‐tert‐butylcatechol ( 5 ) participates in Michael reaction with 2 and through an ECE mechanism converts to the corresponding o‐quinone ( 6a ). The electrochemical synthesis of 4 and 6a has been successfully performed in an undivided cell.  相似文献

The surface of a gold (Au) disk electrode was modified with a self‐assembled monomolecular layer of dithiobis(4‐butylamino‐m‐phenylboronic acid) (DTBA‐PBA) to prepare L ‐lactate‐sensitive electrodes. The DTBA‐PBA‐modified electrodes exhibited an attenuated cyclic voltammogram (CV) for the Fe(CN)63? ion in the presence of L ‐lactate, as a result of the formation of phenylboronate ester of L ‐lactate accompanied with the addition of OH? ion to the boron atom. In other words, the negatively charged DTBA‐PBA monolayer blocked the electrode surface from the access of the Fe(CN)63?/4? ions. Thus, the DTBA‐PBA monolayer‐modified Au electrode can be used for determining L ‐lactate on the basis of the change in redox current of Fe(CN)63?/4? ions. The calibration graph useful for determining 1–30 mM L ‐lactate was obtained.  相似文献

A novel nanocomposite of quinone‐amine polymer and multiwalled carbon nanotubes was synthesized from iodate‐oxidation/Michael addition reaction of 1,2‐dihydroxybenzene with o‐phenylenediamine, which was characterized by TEM, FTIR and UV‐vis spectra. The nanocomposite modified Au electrode with well‐defined quinone redox peaks effectively mediated the oxidation of NADH in pH 7.0 phosphate buffer, with an overpotential decrease by ca. 470 mV (vs. bare Au), a limit of detection of 6.4 nmol L?1 and good antiinterferent ability.  相似文献

N,N′‐Pyromelliticdiimido‐di‐L ‐alanine ( 1 ), N,N′‐pyromelliticdiimido‐di‐L ‐phenylalanine ( 2 ), and N,N′‐pyromelliticdiimido‐di‐L ‐leucine ( 3 ) were prepared from the reaction of pyromellitic dianhydride with corresponding L ‐amino acids in a mixture of glacial acetic acid and pyridine solution (3/2 ratio) under refluxing conditions. The microwave‐assisted polycondensation of the corresponding diimide‐diacyl chloride monomers ( 5–7 ) with 4‐phenyl‐2,6‐bis(4‐aminophenyl) pyridine ( 10 ) or 4‐(p‐methylthiophenyl)‐2,6‐bis(4‐aminophenyl) pyridine ( 12 ) were carried out in a laboratory microwave oven. The resulting poly(amide‐imide)s were obtained in quantitative yields, and they showed admirable inherent viscosities (0.12–0.55 dlg?1), were soluble in polar aprotic solvents, showed good thermal stability and high optical purity. The synthetic compounds were characterized by IR, MS, 1H NMR, and 13C NMR spectroscopy, elemental analysis, and specific rotation. Copyright © 2008 John Wiley & Sons, Ltd.  相似文献

A novel chromogenic method to measure the peroxidase activity using para‐phenylenediamine dihydrochloride (=benzene‐1,4‐diamine hydrochloride; PPDD) and N‐(1‐naphthyl)ethylenediamine dihydrochloride (=N‐(2‐aminoethyl)naphthalen‐1‐amine; NEDA) is presented. The PPDD entraps the free radical and gets oxidized to electrophilic diimine, which couples with NEDA to give an intense red‐colored chromogenic species with maximum absorbance at 490 nm. This assay was adopted for the quantification of H2O2 between 20 and 160 μM . Catalytic efficiency and catalytic power of the commercial peroxidase were found to be 4.47×104 M ?1 min?1 and 3.38×10?4 min?1, respectively. The catalytic constant (kcat) and specificity constant (kcat/Km) at saturated concentration of the co‐substrates were 0.0245×103 min?1 and 0.0445 μM ?1 min?1, respectively. The chromogenic coupling reaction has a minimum interference from the reducing substances such as ascorbic acid, L ‐cystein, citric acid, and oxalic acid. The method being simple, rapid, precise, and sensitive, its applicability has been tested in the crude vegetable extracts that showed peroxidase activity.  相似文献

Copper(I) halides with triphenyl phosphine and imidaozlidine‐2‐thiones (L ‐NMe, L ‐NEt, and L ‐NPh) in acetonitrile/methanol (or dichloromethane) yielded copper(I) mixed‐ligand complexes: mononuclear, namely, [CuCl(κ1‐S‐L ‐NMe)(PPh3)2] ( 1 ), [CuBr(κ1‐S‐L ‐NMe)(PPh3)2] ( 2 ), [CuBr(κ1‐S‐L ‐NEt)(PPh3)2] ( 5 ), [CuI(κ1‐S‐L ‐NEt)(PPh3)2] ( 6 ), [CuCl(κ1‐S‐L ‐NPh)(PPh3)2] ( 7 ), and [CuBr(κ1‐S‐L ‐NPh)(PPh3)2] ( 8 ), and dinuclear, [Cu21‐I)2(μ‐S‐L ‐NMe)2(PPh3)2] ( 3 ) and [Cu2(μ‐Cl)21‐S‐L ‐NEt)2(PPh3)2] ( 4 ). All complexes were characterized with analytical data, IR and NMR spectroscopy, and X‐ray crystallography. Complexes 2 – 4 , 7 , and 8 each formed crystals in the triclinic system with P$\bar{1}$ space group, whereas complexes 1 , 5 , and 6 crystallized in the monoclinic crystal system with space groups P21/c, C2/c, and P21/n, respectively. Complex 2 has shown two independent molecules, [(CuBr(κ1‐S‐L ‐NMe)(PPh3)2] and [CuBr(PPh3)2] in the unit cell. For X = Cl, the thio‐ligand bonded to metal as terminal in complex 4 , whereas for X = I it is sulfur‐bridged in complex 3 .  相似文献

The N‐carboxyanhydrides (NCAs) of sarcosine (Sar), D ,L ‐leucine (D ,L ‐Leu), D ,L ‐phenylalanine (D ,L ‐Phe), and L ‐alanine (L ‐Ala) were polymerized in dioxane. Imidazole served as initiator and the NCA/initiator ratio was varied from 1/1 to 40/1. The isolated polypeptides were characterized by 1H NMR spectroscopy, by MALDI‐TOF mass spectrometry, by viscosity measurements, and by SEC measurements in the case of poly(sarcosine). Cyclic oligopeptides were found in all reaction products and in the case of polySar, poly(D ,L ‐Leu), and poly(D ,L ‐Phe) the cycles were the main products. In the case of poly(L ‐Ala), rapid precipitation of β‐sheet lamellaes prevented efficient cyclizations and stabilized imidazolide endgroups. © 2005 Wiley Periodicals, Inc. Starting from methyl 2,3‐O‐isopropylidene‐α‐D ‐mannofuranoside ( 5 ), methyl 6‐O‐benzyl‐2,3‐O‐isopropylidene‐α‐D ‐lyxo‐hexofuranosid‐5‐ulose ( 12 ) was prepared in three steps. The addition reaction of dimethyl phosphonate to 12 , followed by deoxygenation of 5‐OH group, provided the 5‐deoxy‐5‐dimethoxyphosphinyl‐α‐D ‐mannofuranoside derivative 15a and the β‐L ‐gulofuranoside isomer 15b . Reduction of 15a and 15b with sodium dihydrobis(2‐methoxyethoxy)aluminate, followed by the action of HCl and then H2O2, afforded the D ‐mannopyranose ( 17 ) and L ‐gulopyranose analog 21 , each having a phosphinyl group in the hemiacetal ring. These were converted to the corresponding 1,2,3,4,6‐penta‐O‐acetyl‐5‐methoxyphosphinyl derivatives 19 and 23 , respectively, structures and conformations (4C1 or 1C4, resp.) of which were established by 1H‐NMR spectroscopy.  相似文献

The electrochemical behavior of cytochrome c (cyt‐c) that was electrostatically immobilized onto a self‐assembled monolayer (SAM) of captopril (capt) on a gold electrode has been investigated. Cyclic voltammetry, scanning electrochemical microscopy (SECM) and electrochemical impedance spectroscopy were employed to evaluate the blocking property of the capt SAM. SECM was used to measure the bimolecular electron transfer (ET) kinetics (kBI) between a solution‐based redox probe and the immobilized protein. In addition, the tunneling ET between the immobilized protein and the underlying gold electrode was calculated. A kBI value of (5.0±0.6)×108 mol?1 cm3 s?1 for the bimolecular ET and a standard tunneling rate constant (k0) of 46.4±0.2 s?1 for the tunneling ET have been obtained.  相似文献

The present work describes oxidation of ascorbic acid (AA) at octacyanomolybdate‐doped‐glutaraldehyde‐cross‐linked poly‐L ‐lysine (PLL‐GA‐Mo(CN) film modified glassy carbon electrode in 0.1 M H2SO4. The modified electrode has been successfully prepared by means of electrostatically trapping Mo(CN) mediator in the cationic film of glutaraldehyde‐cross‐linked poly‐L ‐lysine. The dependence of peak current of modified electrode in pure supporting indicates that the charge transfer in the film was a mixed process at low scan rates (5 to 200 mV s?1), and kinetically restrained at higher scan rates (200 to 1000 mV s?1). Cyclic voltammetry and rotating disk electrode (RDE) techniques are used to investigate the electrocatalytic oxidation of ascorbic acid and compared with its oxidation at bare and undoped PLL‐GA film coated electrodes. The rate constant of catalytic reaction k obtained from RDE analysis was found to be 9.5×105 cm3 mol?1 s?1. The analytical determination of ascorbic acid has been carried out using RDE technique over the physiological interest of ascorbic acid concentrations with a sensitivity of 75 μA mM?1. Amperometric estimation of AA in stirred solution shows a sensitivity of 15 μA mM?1 over the linear concentration range between 50 and 1200 μM. Interestingly, PLL‐GA‐Mo(CN) modified electrode facilitated the oxidation of ascorbic acid but not responded to other electroactive biomolecules such as dopamine, uric acid, NADH, glucose. This unique feature of PLL‐GA‐Mo(CN) modified electrode allowed for the development of a highly selective method for the determination of ascorbic acid in the presence of interferents.  相似文献

To develop a novel polycondensation method for the preparation of poly (amino acid)s, we screened a transition metal or a rare‐earth triflate as a Lewis acid for the polycondensation of activated amino acid esters in N,N‐dimethylformamide solutions at room temperature. The polymerizations of 4‐nitrophenyl L ‐leucinate ( 1a ) and 4‐nitrophenyl L ‐valinate ( 1b ) scarcely proceeded without any Lewis acid at room temperature. In the presence of 5 mol % metal triflates, especially scandium(III) trifluoromethanesulfonate, the polymerizations of both monomers were promoted effectively. The products, which were collected by the reaction mixture being poured into water, were recognized as poly(L ‐valine)s by Fourier transform infrared spectroscopy, gel permeation chromatography analysis, and 1H NMR spectroscopy. These results showed that a metal triflate as a Lewis acid could coordinate to a carbonyl oxygen of activated L ‐valinate and L ‐leucinate even in a highly polar solvent, such as N,N‐dimethylformamide; therefore, the polymerizations of activated L ‐valinate and L ‐leucinate were promoted. Because steric hindrance derived from the isobutyl group in 1b was less than that of the isopropyl unit in 1a , the effect of the metals was not as sensitive for the polymerization of 1b . © 2006 Wiley Periodicals, Inc. This paper reports the fabrication of Au nanoparticles (Au NPs)‐Ni‐Al layerd double hydroxide (LDH) composite film by one step electrochemical deposition on the surface of a glass carbon electrode from the mixture solution containing HAuCl4 and nitrate salts of Ni2+ and Al3+. Improved conductivity was obtained by Au NPs codeposited on LDH film. The synergic effect of LDHs and Au NPs dramatically improves the performance of L ‐cysteine electro‐oxidation, displaying low oxidation peak potential (0.16 V) and high current response. Thus the electrode was used to sense L ‐cysteine, showing good sensitivity and selectivity.  相似文献

The catalytic activity of l ‐arginine‐coated nano‐Fe3O4 particles (Fe3O4@l ‐arginine) proves they are a novel magnetic catalyst without the use of heat and reflux for the synthesis of 1,3‐diaryl‐2‐N‐azaphenalene derivatives and n‐acyl‐1,3‐diaryl‐2‐N‐azaphenylene derivatives in a one‐pot pseudo‐five‐component condensation reaction of compounds of 2,7‐naphthalene diol, aldehydes, and ammonia derivatives (ammonium acetate or ammonium hydrogen phosphate) and solvent (water and alcohol) with high yield and short reaction times, economical, and simple workup. The structure and magnetic properties of the obtained nanoparticles were characterized via Fourier transform infrared spectroscopy (IR) and field emission scanning electron microscopy (FE‐SEM). The results demonstrated that the average size of the synthesized magnetite nanoparticles is about 21 nm. In addition, the heterogeneous catalyst can be easily recovered magnetically and can be reused for further runs without significant loss of its catalytic activity.  相似文献

We have developed and validated a high‐performance liquid chromatography method that uses monolithic silica disk‐packed spin columns and a monolithic silica column for the simultaneous determination of NG‐monomethyl‐l ‐arginine, NG,NG‐dimethyl‐l ‐arginine, and NG,NG′‐dimethyl‐l ‐arginine in human plasma. For solid‐phase extraction, our method employs a centrifugal spin column packed with monolithic silica bonded to propyl benzenesulfonic acid as a cation exchanger. After pretreatment, the methylated arginines are converted to fluorescent derivatives with 4‐fluoro‐7‐nitro‐2,1,3‐benzoxadiazole, and then the derivatives are separated on a monolithic silica column. l ‐Arginine concentration was also determined in diluted samples. Standard calibration curves revealed that the assay was linear in the concentration range 0.2–1.0 μM for methylated arginines and 40–200 μM for l ‐arginine. Linear regression of the calibration curve yielded equations with correlation coefficients of 0.999 (r2). The sensitivity was satisfactory, with a limit of detection ranging from 3.75 to 9.0 fmol for all four compounds. The RSDs were 4.3–4.8% (intraday) and 3.0–6.8% (interday). When this method was applied to samples from six healthy donors, the detected concentrations of NG‐monomethyl‐l ‐arginine, NG,NG‐dimethyl‐l ‐arginine, NG,NG′‐dimethyl‐l ‐arginine and l ‐arginine were 0.05 ± 0.01, 0.41 ± 0.07, 0.59 ± 0.11, and 83.8 ± 30.43 μM (n = 6), respectively.  相似文献

D ,L ‐3‐Methylglycolide (MG) was synthesized via two step reactions with a good yield (42%). It was successfully polymerized in bulk with stannous octoate as a catalyst at 110 °C. The effects of the polymerization time and catalyst concentration on the molecular weight and monomer conversion were studied. Poly(D ,L ‐lactic acid‐co‐glycolic acid) (D ,L ‐PLGA50; 50/50 mol/mol) copolymers were successfully synthesized from the homopolymerization of MG with high polymerization rates and high monomer conversions under moderate polymerization conditions. 1H NMR spectroscopy indicated that the bulk ring‐opening polymerization of MG conformed to the coordination–insertion mechanism. 13C NMR spectra of D ,L ‐PLGA50 copolymers obtained under different experimental conditions revealed that the copolymers had alternating structures of lactyl and glycolyl. © 2000 John Wiley & Sons, Inc. Two types of three‐arm and four‐arm, star‐shaped poly(D,L ‐lactic acid‐alt‐glycolic acid)‐b‐poly(L ‐lactic acid) (D,L ‐PLGA50‐b‐PLLA) were successfully synthesized via the sequential ring‐opening polymerization of D,L ‐3‐methylglycolide (MG) and L ‐lactide (L ‐LA) with a multifunctional initiator, such as trimethylolpropane and pentaerythritol, and stannous octoate (SnOct2) as a catalyst. Star‐shaped, hydroxy‐terminated poly(D,L ‐lactic acid‐alt‐glycolic acid) (D,L ‐PLGA50) obtained from the polymerization of MG was used as a macroinitiator to initiate the block polymerization of L ‐LA with the SnOct2 catalyst in bulk at 130 °C. For the polymerization of L ‐LA with the three‐arm, star‐shaped D,L ‐PLGA50 macroinitiator (number‐average molecular weight = 6800) and the SnOct2 catalyst, the molecular weight of the resulting D,L ‐PLGA50‐b‐PLLA polymer linearly increased from 12,600 to 27,400 with the increasing molar ratio (1:1 to 3:1) of L ‐LA to MG, and the molecular weight distribution was rather narrow (weight‐average molecular weight/number‐average molecular weight = 1.09–1.15). The 1H NMR spectrum of the D,L ‐PLGA50‐b‐PLLA block copolymer showed that the molecular weight and unit composition of the block copolymer were controlled by the molar ratio of L ‐LA to the macroinitiator. The 13C NMR spectrum of the block copolymer clearly showed its diblock structures, that is, D,L ‐PLGA50 as the first block and poly(L ‐lactic acid) as the second block. © 2001 John Wiley & Sons, Inc. Iodination of N2‐isobutyryl‐5‐aza‐7‐deazaguanine ( 7 ) with N‐iodosuccinimide (NIS) gave 7‐iodo‐N2‐isobutyryl‐5‐aza‐7‐deazaguanine ( 8 ) in a regioselective reaction (Scheme 1). Nucleobase‐anion glycosylation of 8 with 2‐deoxy‐3,5‐di‐O‐toluoyl‐α‐D ‐ or α‐L ‐erythro‐pentofuranosyl chloride furnished anomeric mixtures of D ‐ and L ‐nucleosides. The anomeric D ‐nucleosides were separated by crystallization to give the α‐D ‐anomer and β‐D ‐anomer with excellent optical purity. Deprotection gave the 7‐iodo‐5‐aza‐7‐deazaguanine 2′‐deoxyribonucleosides 3 (β‐D ; ≥99% de) and 4 (α‐D ; ≥99% de). The reaction sequence performed with the D ‐series was also applied to L ‐nucleosides to furnish compounds 5 (β‐L ; ≥99% de) and 6 (α‐L ; ≥95% de).  相似文献

Fluorinases offer an environmentally friendly alternative for selective fluorination under mild conditions. However, their diversity is limited in nature and they have yet to be engineered through directed evolution. Herein, we report the directed evolution of the fluorinase FlA1 for improved conversion of the non‐native substrate 5′‐chloro‐5′‐deoxyadenosine (5′‐ClDA) into 5′‐fluoro‐5′‐deoxyadenosine (5′‐FDA). The evolved variants, fah2081 (A279Y) and fah2114 (F213Y, A279L), were successfully applied in the radiosynthesis of 5′‐[18F]FDA, with overall radiochemical conversion (RCC) more than 3‐fold higher than wild‐type FlA1. Kinetic studies of the two‐step reaction revealed that the variants show a significantly improved kcat value in the conversion of 5′‐ClDA into S‐adenosyl‐l ‐methionine (SAM) but a reduced kcat value in the conversion of SAM into 5′‐FDA.  相似文献

N‐Methyl‐L ‐phenylalanine ( 5 ), N‐methyl‐4‐nitro‐L ‐phenylalanine ( 6 ), and N,N‐dimethyl‐4‐nitro‐L ‐phenylalanine ( 7 ?H+) were investigated as substrates or inhibitors of phenylalanine ammonia lyase from Petroselinum crispum. Whereas the former was a reluctant substrate (Km =6.6 mM , kcat =0.22 s?1), no reverse reaction could be detected by using methylamine and (E)‐cinnamate ( 2 ). The Km value for ammonia in the reverse reaction by using (E)‐cinnamate ( 2 ) was determined to be 4.4 and 2.6M at pH 8.8 and 10, respectively. The N‐methylated 4‐nitro‐L ‐phenylalanines 6 and 7 showed only strong inhibitory effects (Ki =130 nM and 8 nM , resp.). These and former results are discussed in terms of the mechanism of action of phenyalalanine and histidine ammonia lyases.  相似文献

The formation of an inclusion complex between 4‐aminobiphenyl (4‐AB) and β‐cyclodextrin molecules (β‐CD), allows the use of thiolated β‐CDs as chemi‐adsorbed material on a Au electrode as a self‐assembled submonolayer for the selective square wave voltammetric determination of 4‐AB. The submonolayer was characterized by reductive desorption and an association constant of 1.2×104 L/mol was obtained. The optimization of variables yielded a linear dependence of ip/4‐AB concentration in the range of 10?5 to 10?4 mol/L. The selectivity of the method was evaluated in the presence of other aromatic amines obtaining better results with the modified electrode. This methodology was applied to the voltammetric determination of 4‐AB in wastewater samples.  相似文献
