将5-取代胺基-2-巯基-1,3,4-噻二唑引入苯并[4,5]呋喃[3,2-d]嘧啶中,设计并合成了10个新型的4-(5-N-取代-1,3,4-噻二唑-2-巯基)-苯并[4,5]呋喃[3,2-d]嘧啶类衍生物(3a~3j),其结构经1H NMR,13C NMR,IR和MS确认。用MTT法测定了3a~3j对人胃腺癌细胞体(MGC)的体外增殖活性。结果表明,3a~3j均具有不同程度的抑制MGC的活性,其中4-(5-N-2’-甲氧基苯基-1,3,4-噻二唑-2-巯基)-苯并[4,5]呋喃[3,2-d]嘧啶(3j)在10μmol·L-1的浓度下对MGC的抑制率为86.4%。 相似文献

采用亚活性基团拼接原理,以取代苯胺为起始原料,合成了6个新型含1,3,4-噻二唑基的苯并呋喃嘧啶类化合物(5a~5f),其结构经1H NMR,13C NMR,IR和ESI-MS表征。采用MTT法对5进行了抑制人胃癌细胞MGC-803体外增殖活性测试。结果表明:在用药浓度为10μmol·L-1时,部分化合物对MGC-803细胞具有良好的抑制活性,其中4-[5-N-(2,6-二甲基苯基)-1,3,4-噻二唑-2-巯基]苯并[4,5]呋喃[3,2-d]嘧啶(5b),4-[5-N-(2,5-二甲氧基苯基)-1,3,4-噻二唑-2-巯基]苯并[4,5]呋喃[3,2-d]嘧啶(5c)和4-[5-N-(3-甲氧基苯基)-1,3,4-噻二唑-2-巯基]苯并[4,5]呋喃[3,2-d]嘧啶(5e)的抑制率分别为89.5%,88.9%和70.2%。 相似文献

以环己酮为原料,利用Gewald反应合成了6个3-烷基-5,6,7,8-四氢苯并[4,5]噻吩并[2,3-d]嘧啶-4-酮衍生物和17个3-亚苄基胺基四氢苯并[4,5]噻吩并[2,3-d]嘧啶-4-酮衍生物,通过1H NMR、13C NMR、IR和MS对所合成的化合物进行了结构表征,并对所合成化合物进行了抗肿瘤活性的筛选.结果表明4个化合物对人鼻咽癌细胞株(KB)和鼻咽鳞癌细胞株(CNE2)两种肿瘤细胞株表现出一定程度的抑制活性. 相似文献

为了寻找高效的抗肿瘤药物,设计并合成了一系列新型的1,2,3-三氮唑[4,5-d]嘧啶类衍生物,对这些化合物在人类五种癌细胞MGC-803(人胃癌细胞)、MCF-7(人乳腺癌细胞)、EC-109(人食管癌细胞)、PC-3(人前列腺癌细胞)、Hela(人宫颈癌细胞)进行抗肿瘤活性评价,结果显示大部分化合物具有较好的抗肿瘤活性,其中5-(((1H-苯并[d]咪唑-2-基)甲基)硫基)-3-苄基-N-(4-氯苯基)-3H-[1,2,3]-三氮唑[4,5-d]嘧啶-7-胺(11b)和2,2'-((5-(((1H-苯并[d]咪唑-2-基)甲基)硫基)-3-苄基-3H-[1,2,3]-三氮唑[4,5-d]嘧啶-7-基)氮烷二基)双(乙烷-1-醇)(11m)对MGC-803和Hela癌细胞的抗肿瘤活性优于对照品氟尿嘧啶. 相似文献

本文以环己酮为原料,通过氮杂Wittig反应合成了一系列结构新颖的取代四氢苯并噻吩并吡啶并嘧啶衍生物,并采用MTT法考察所合成目标化合物对CNE2、KB、MGC-803、MCF-7和PC3这5种肿瘤细胞的抑制活性。初步的生物活性结果表明,目标化合物对5种肿瘤细胞均有抑制活性,尤其是对胃癌MGC-803细胞展现出了更强的抑制活性。其中3-(4-氟苯基)-2-((4-氟苯基)氨基)-5-甲基-8,9,10,11-四氢苯并[4',5']噻吩并[3',2':5,6]吡啶并[4,3-d]嘧啶-4(3H)-酮[化合物8c,IC_(50)=(0. 9±0. 25)μmol·L~(-1)]对MGC-803的活性最强,是5-氟尿嘧啶[IC_(50)=(18. 4±1. 43)μmol·L~(-1)]的20倍;同时,目标化合物对正常的胃黏膜上皮细胞GES-1没有毒性。四氢苯并[4',5']噻吩并[3',2':5,6]吡啶并[4,3-d]嘧啶类化合物具有良好的抗肿瘤活性,值得进一步深入研究。 相似文献

为寻找以蛋白酪氨酸激酶为靶点的新型抗癌试剂,本文设计与合成了系列含哌嗪基的7-溴噻吩并[3,2-d]嘧啶衍生物。以巯基乙酸为原料,经六步反应得4-氯-7-溴噻吩并[3,2-d]嘧啶母核,再与不同的哌嗪衍生物在添加三乙胺作碱的条件下反应,制备了五个未见报道的含哌嗪基7-溴噻吩并[3,2-d]嘧啶化合物。结构经1H NMR和HRMS表征。 相似文献

Photocyclization of 3-chloro-N-phenylbenzo[b]thiophene-2-carboxamide 10 afforded [1]benzothieno[2,3-c]-quinolin-6(5H)-one 11 which was chlorinated to 6-chloro[1]benzothieno[2,3-c]quinoline 12 followed by dechlorination to give [1]benzothieno[2,3-c]quinoline 5 . A series of 6-substituted alkoxy and thioalkoxy[1]benzothieno[2,3-c]quinoline derivatives were prepared along with the N-methyl quaternary salt 13 of 5 . 6-Chloro[1]-benzothieno[2,3-c]quinoline 12 was converted into 6-hydrazino[1]benzothieno[2,3-c]quinoline 23 which upon treatment with formic acid yielded [1]benzothieno[2,3-c][1,2,4]triazolo[4,3-a]quinoline 6 . Treatment of 23 with nitrous acid resulted in [1]benzothieno[2,3-c]tetrazolo[1,5-a]quinoline 7 . Compounds 6 and 7 are novel heterocyclic ring systems. 相似文献
