The relevance of terms other than linear when deriving quantitative structure-activity relationship/quantitative structure-property relationship (QSAR/QSPR) models has been rarely considered so far. In this study, the impact of quadratic and interacting terms has been taken into account. The first effect of including such highly structured terms is a significant extension of the parametric domain that moves from the initial N to N(N + 3)/2 parameters. This substantial enlargement over the conventional linear boundaries involves a higher computational cost due to the increased combinatorial number of resulting theoretical QSAR/QSPR models. To face this issue, novel genetic-algorithm-based software, MGZ (multigenetic zooming), was developed and used for both variable selection and model building. To speed up the entire process of domain searching, MGZ was supported with multiple independent evolving populations and genetic storms to further QSAR/QSPR analyses. In addition, a novel fitness function was developed to score models on the basis of their inner predictive capability, assessed on the training set, structure complexity, and presence of nonlinear terms. The models were further validated by monitoring model redundancy and performing intensive randomization runs. The Selwood data set was used as a reference set to derive QSAR models. Furthermore, a QSPR study was conducted on the solubility data set of a large array of organic compounds. The results reported in the present paper demonstrate that our approach is successful in finding linear models, which are at least as good as the models previously derived using standard statistical approaches, and in deriving new nonlinear models with good statistical figures.  相似文献

提出拓扑化学行为概念,把对化合物性质最具影响力的因素归纳为拓扑生长力和拓扑阻滞力。根据该原理,利用价电子轨道能量对价电子距离矩阵进行修正。研究表明OET(轨道能量拓扑指数)对镧系元素和锕系元素的物理化学性质具有良好的应用,尤其是对尚未借助QSPR(定量结构性质关系)手段进行研究的光谱性质。LOO CV(留一法交叉检验)的结果验证了模型的良好稳定性和预测能力,采用的检验参数有:PRESS/SSY,SEPCVRCVSPRESS 和PSE,其中PRESS/SSY比值介于0.000 6和0.114 8之间。与文献进行比较,本文方法所得结果与之接近或更好。研究显示正是基于拓扑化学行为才有本文方法良好和较广的应用。  相似文献

取代喹啉类化合物抗菌活性的定量构效关系及分子设计   总被引:1,自引:0,他引:1  
采用密度泛函理论(DFT)和逐步回归分析法对15种新合成的取代喹啉类化合物进行了定量构效关系(QSAR)研究. 在B3LYP/6-31G(d,p)水平上计算了取代喹啉的量子化学参数, 通过逐步多元回归分析筛选出影响抗菌活性的主要因素, 建立了定量构效关系方程, 并用留一法交叉分析了模型的稳定性及预测能力. 结果表明, C5的亲核电子密度fNC5及C9-N1的键级BC9-N1是影响喹啉类化合物抗金黄色葡萄球菌活性的主要因素, 所得模型对该类化合物抗菌活性有较好的预测效果. 同时基于QSAR研究结果设计了4个活性较高的新喹啉衍生物.  相似文献

收集20种天然氨基酸的457种理化性质,按照疏水、电性特征、氢键贡献和立体特征分类后,对它们分别进行主成分分析(Principal component analysis,PCA),得到一个新的氨基酸残基结构描述符SVHEHS.用该描述符分别对血管紧张素转化酶(AngiotensinⅠconverting enzyme,ACE)抑制二肽、三肽、四肽进行序列表征,并用来与生物活性建立偏最小二乘(Partial least square regression,PLS)模型.ACE抑制二肽、三肽、四肽模型的相关系数、交叉验证相关系数、 均方根误差、外部验证相关系数分别为0.607,0.507,0.587,0.783;0.852,0.813,0.232,0.839;1,1,0,0.935.由此说明,采用SVHEHS描述符建立的PLS模型拟合、预测能力均较好,可用于血管紧张素转化酶抑制肽的定量构效关系研究.  相似文献

采用分子电性距离矢量(MEDV)表征有机磷酸酯类化合物的分子结构,运用多元线性回归建立定量结构-色谱保留关系(QSRR)模型,同时采用逐步回归结合统计检测对模型进行变量筛选,建立了35个有机磷酸酯类化合物在3种不同固定相(OV-101,DB-1701和DB-WX)上气相色谱保留指数(RI)与MEDV的定量相关模型.在3种固定相上的QSRR模型的建模计算值复相关系数(R)、留一法(leave-one-out)交互校验复相关系数(QCV)分别为0.998 0和0.995 1(OV-101);0.996 3和0.989 6(DB-1701);0.993 7和0.984 1(DB-WX),表明模型具有良好估计能力与稳定性.  相似文献

从20种天然氨基酸的41个randic molecular profiles非零描述符、44个eigenvalue based indices非零描述符和47个walk and path counts非零描述符分别进行主成分分析,得出一种新的氨基酸描述符-SVREW。将其应用于血管紧张素转化酶(ACE)抑制二肽和ACE抑制三肽、苦味二肽和苦味四肽、后叶催产素类似物、HLA-A*0201限制性CTL表位肽的结构表征,应用多元线性回归(MLR)建立定量构效关系模型,同时采用内部与外部双重验证的方法验证模型的稳定性。所建ACE抑制二肽、ACE抑制三肽、苦味二肽、苦味四肽、后叶催产素类似物、HLA-A*0201限制性CTL表位肽的模型复相关系数(R2cum)分别为0.994,0.797,0.948,0.878,0.686,0.720;留一法交互校验复相关系数(R2cv)分别为0.955,0.859,0.879,0.958,0.796,0.843;外部样本校验相关系数(Q2ext)分别为0.990,0.954,0.890,0.950,0.748,0.773。经研究表明SVREW描述符用于肽分子结构表征所建模型的稳定性与预测能力均较好,有望成为多肽定量构效关系研究中一种有效的结构表征方法,可对新药物的发现和研究提供指导。  相似文献
